Abstract
PURPOSE: To prepare adenosine monophosphate deaminase by culturing fungi belonging to Saccharomyces genus, etc.
CONSTITUTION: Fungi belonging to Saccharomyces, Hansenula, Kloechera, Candida, Torulopsis, or Endomycopsis genus, and capable of producing adenosine monophosphate deaminase, are inoculated in a nutrient medium, and cultured at 25W 35°C for 8W48hrs, and the accumulated adenosine monophosphate deaminase is separated from the medium. The adenosine monophosphate deaminase has a molecular weight of 280,000W320,000 by the gel-filtration and about 80,000 by SDS- electrophoresis, the optimum pH range, the stable pH range, the optimum temperature range, and the stable temperature range in a phosphate buffer solution of 7.5W7.8, 7W8, about 35°C, and lower than 40°C, respectively, and a Km value to adenosine-monophosphate of 2×10-6 M, and is capable of hydrolyzing adenosine monophosphate to inosine-monophosphate and ammonia.
