Abstract
PURPOSE: To determine with high sensitivity oxobarbituric acid derivative, which is a substance to be tested, from a trace amount of a sample by enzyme immunoassay method employing alkaliphosphatase as labelled enzyme.
CONSTITUTION: A sample contg. oxobarbituric acid derivative and an enzyme- labelled phenobarbital of the formula (wherein ALP is alkaliphosphatase residue; n is an integer of 1W20) are competitively reacted with anti-oxobarbiturate antibody. After the addition of buffer solution to the reaction mixt., ALP-labelled phenobarbital combined with insoluble antibody and uncombined ALP-labelled phenobarbital are separated by means of centrifugal separation or the like. After the separation, enzyme activity of one of them is measured by a conventional method such as Bessey-Lowry method to determine the amount of oxobarbituric acid derivative in the sample.
