Abstract
PURPOSE: To obtain enzyme compsn. useful as immobilized enzyme for analysis of a pyruvate-containing system or a pyruvate-free system, by a method wherein a thiol reagent is blended with immobilized enzyme of pyruvate oxidase included in phtot-setting resin.
CONSTITUTION: Pyruvate oxidase is uniformly mixed with nonionic prepolymer having at least two photo-polymerizable ethylenically unsaturated gp. per molecule in an aq. medium such as tris-hydrochloric acid buffer solution or phosphate buffer solution. The mixt. is irradiated with light of a wavelength of 250W600mμ for 10secW30min to obtain immobilized enzyme of pyruvate oxidaze included in photo- setting resin. This immobilized enzyme is blended with a thiol reagent such as mercaptoethanol, dithiothreitol or dithioerythritol as a stabilizer to obtain immobilized enzyme composition of pyruvate oxidase.
