Abstract

PURPOSE: To prepare high purity cholinesterase in high recovery, from human blood-plasma or a cholinesterase-containing fraction originated therefrom.
CONSTITUTION: (1) Human blood-plasma or a fraction of blood-plasma protein containing cholinesterase is fractionated in an aqueous medium containing 0.5W5% of proteins at pH3W7 with polyethylene glycol, and the precipitate is separated from the solution; (2) the precipitate is subjected to ion exchange chromatograpy in an aqueous medium at pH4W6; and (3) the eluted liquid is fractionated at a protein content of 0.01W5% and pH4W10 using ammonium sulfate and sodium sulfate, and the precipitate is separated from the solution. The steps (1) to (3) can be carried out in arbitrary order. The aqueous solution of cholinesterase thus obtained may be converted to a stable solid esterase preparation by adding a neutral amino acid, sugar, etc. as a stabilizing agent, followed by freeze-drying.
