Abstract

PURPOSE: To prepare immune gamma-globulin derivative which can be administered by intravenous injection, and having low anticomplimentary titer, by reacting human immune globulin with trithionic aid salt and sulfurous acid (salt) in water, thereby breaking the interchain S-S bond, and at the same time, sulfonating the S atom.
CONSTITUTION: Human immune gamma-globulin is made to react with a trithionate- ion-producing substance (e.g. sodium trithionate) and a sulfite-ion-producing substance (e.g. sulfurous acid (salt)). The interchain S-S bond of the immune globulin is broken, and at the same time, the S atom is sulfonated, thereby converting the S-S bond to S-sulfonate group (-S-SO3-). After the reaction, it is purified by dialysis salting out, column chromatography, etc. to obtain the purified objective compound. The amounts of the sulfite and the trithionate is ≥ 2 moles, pref. ≥ 10 moles, and ≥ 1 mole, pref. ≥ 2 moles, per 1 mole of S-S bond, respectively. The reaction is carried out at pH6W10, and 50°C, pref. 10W45°C, for 0.5W24 hours.
