Abstract

PURPOSE: To produce a highly pure heparinase free from other acidic mucopolysaccharase, in large quantities, by treating a roughly purified heparinase solution with column chromatography using a cationic exchanging material as adsorbent.
CONSTITUTION: Bacterial cells obtained by the culturing of heparinase-producing bacteria, e.g. Flavobacterium heparinum, in a medium containing heparin, are disintegrated by ultrasonic treatment, etc. The disintegrated cells are extracted, and preliminary purified by protamin sulfate treatment, hydroxyapatite fractionation, etc. The purified heparinase is obtained by the column chromatography of the preliminary purified heparin using a cation exchange resin, as an adsorbent. The cation exchanger is, e.g. a cation exchange cellulose, cation exchange crosslinked dextran, etc., and pref. phosphocellulose, phosphonomethylcellulose, etc.
