Claims (1)
- 【特許請求の範囲】 1.一種または複数種の細胞から細胞内物質を放出させる方法であって、前記一 種または複数種の細胞を含有する懸濁液に対して50ボルト以下の電圧を印加す る方法。 2.前記電圧が0.5−50ボルトである、請求項1記載の方法。 3.前記電圧が0.5−15ボルトである、請求項1記載の方法。 4.前記電圧が1−10ボルトである、請求項1記載の方法。 5.前記電圧を、前記懸濁液中において、5mm以下離れている複数の電極の間 で印加する、前記のいずれか一つの請求項に記載の方法。 6.前記電圧を、前記懸濁液中において、5mm以下離れている複数の電極の間 で印加する、前記のいずれか一つの請求項に記載の方法。 7.前記電極の間隔が1.5mm以下である、請求項6記載の方法。 8.前記電極の間隔が0.5mm以下である、請求項6記載の方法。 9.前記細胞が、細菌細胞、酵母菌細胞、植物細胞、動物細胞、昆虫細胞または ヒト細胞である、前記のいずれか一つの請求項に記載の方法。 10.前記電圧を少なくとも30秒の時間印加する、前記のいずれか一つの請求 項に記載の方法。 11.前記電圧を少なくとも2分の時間印加する、請求項10記載の方法。 12.前記電圧を、前記の時間の間、連続的に印加する、請求項11記載の方法 。 13.一本鎖核酸を生産する方法であって、電極によって前記細胞の懸濁液に対 して50ボルト以下の電圧を印加して、前記細胞から核酸を放出させることによ って、細胞から二本鎖核酸を放出させ、前記電極によって前記懸濁液に対して前 記と同じかまたは相異なる電圧を印加して、前記二本鎖核酸を一本鎖核酸へと変 換することによって、二本鎖核酸を変性させる方法。 14.前記変性を生じさせる際に、0.5−3ボルトの電圧を印加する、請求項 13記載の方法。 15.前記変性を生じさせる際に、1.5−2.5ボルトの電圧を印加する、請 求項13記載の方法。 16.変性を補助するプロモーターの存在下に、変性を実施する、請求項13− 15のいずれか一つの請求項に記載の方法。 17.前記プロモーター化合物が、メチル ビオロゲンまたはその 塩、または多価無機陽イオンである、請求項16記載の方法。 18.核酸の標的配列を増殖する方法であって、核酸の変性、ハイブリッド形成 および複製を含んでおり、ここで前記核酸を、請求項1−12のいずれか一つの 請求項に記載の方法によって細胞から放出させ、前記核酸を含有する溶液を、最 大2分間、前記核酸の少なくとも一部を前記溶液中で全体的なまたは部分的な一 本鎖形に変換するような条件下で、複数の電極の間の印加電圧に対して供するこ とによって、前記変性を実施する方法。 19.前記増殖法がPCR法またはLCR法である、請求項18記載の増殖方法 。 20.核酸を複製する方法であって:請求項1−12のいずれか一つの請求項に 記載の方法によって細胞から二本鎖核酸を放出させ;溶液中の試料二本鎖核酸の 鎖を、電極から溶液に印加される電圧の影響下に分離し;前記核酸の分離された 鎖を、前記の変性された核酸の鎖の少なくとも一種とハイブリッド形成する少な くとも一種のオリゴヌクレオチドプライマーとハイブリッド形成し;これとハイ ブリッド形成する核酸の各鎖に対して十分に相補的な前記のまたは各プライマー の伸長産物を合成し;および核酸鎖とハイブリッド形成して伸長産物を得た核酸 鎖から、前記のまたは各伸長産物を分離する方法。 21.細胞中の所定の核酸配列の存在または不存在を検出する方法であって、請 求項1−12のいずれか一つの請求項に記載の方法によって前記細胞から核酸を 放出させ;放出された二本鎖核酸を、前 記核酸に対して電圧を印加することによって変性させ;この変性された核酸を、 前記配列に対するオリゴヌクレオチドプローブとハイブリッド形成させ;および 前記ハイブリッド形成が生じたか否かを測定する方法。
