Claims (1)
- 【特許請求の範囲】 1.a.還元性SDS−PAGEで測定したときの分子量が約30kDであり、 b.本質的に配列番号2のアミノ酸配列を有する、 ことを特徴とする実質的に純粋な繊維芽細胞増殖因子相同因子−1(FHF− 1)。 2.請求項1に記載のFHF−1ポリペプチドをコードする、単離されたポリヌ クレオチド配列。 3.FHF−1ヌクレオチド配列が、 a.配列番号1(ここでTはUでもよい); b.配列番号1に相補的な核酸配列; c.鎖長が少なくとも15塩基であり、かつストリンジェント条件下で配列 番号2のFHF−1タンパク質をコードするDNAと選択的にハイブリダイズす る、aまたはbの断片; よりなる群から選択される、請求項2に記載のポリヌクレオチド。 4.前記ポリヌクレオチドが哺乳動物細胞から単離されたものである、請求項2 に記載のポリヌクレオチド配列。 5.哺乳動物細胞がヒト細胞である、請求項4に記載のポリヌクレオチド。 6.請求項2に記載のポリヌクレオチドを含有する発現ベクター。 7.前記ベクターがプラスミドである、請求項6に記載のベクター。 8.前記ベクターがウイルスである、請求項6に記載のベクター。 9.請求項6に記載のベクターで安定に形質転換された宿主細胞。 10.前記細胞が原核細胞である、請求項9に記載の宿主細胞。 11.前記細胞が真核細胞である、請求項9に記載の宿主細胞。 12.FHF−1ポリペプチドと結合する抗体またはその免疫反応性フラグメント 。 13.前記抗体がポリクローナルである、請求項12に記載の抗体。 14.前記抗体がモノクローナルである、請求項12に記載の抗体。 15.FHF−1関連細胞増殖性疾患が疑われる患者の検体を、FHF−1と結合 する薬剤と接触させ、この薬剤のFHF−1への結合を検出することを含んで なる、FHF−1関連細胞増殖性疾患の検出方法。 16.前記細胞が脳、精巣または眼の細胞よりなる群から選択される、請求項15に 記載の方法。 17.前記薬剤がFHF−1と結合する抗体である、請求項15に記載の方法。 18.前記薬剤がFHF−1ポリペプチドをコードするポリヌクレオチドまたはそ の断片である、請求項15に記載の方法。 19.前記検出をin vivo で行う、請求項15に記載の方法。 20.前記検出をin vitroで行う、請求項15に記載の方法。 21.前記薬剤が検出可能に標識されている、請求項17または18に記載の方法。 22.検出可能な標識が放射性同位元素、蛍光化合物、生物発光化合物および化学 発光化合物よりなる群から選択される、請求項21に記載の方法。 23.FHF−1の発現と関連した細胞増殖性疾患を有するかまたは該疾患が疑わ れる細胞を、FHF−1活性を抑制する薬剤と接触させることを含んでなる、F HF−1関連細胞増殖性疾患の治療方法。 24.前記薬剤が抗FHF−1抗体である、請求項23に記載の方法。 25.前記薬剤がFHF−1アンチセンス配列である、請求項23に記載の方法。 26.前記細胞が精巣、脳または眼の細胞である、請求項23に記載の方法。 27.FHF−1活性を抑制する薬剤がベクターを使って細胞に導入される、請求 項23に記載の方法。 28.前記ベクターがコロイド分散系である、請求項27に記載の方法。 29.コロイド分散系がリポソームである、請求項28に記載の方法。 30.リポソームが本質的に標的特異性である、請求項29に記載の方法。 31.リポソームが解剖的にターゲッティングされる、請求項30に記載の方法。 32.リポソームが機械的にターゲッティングされる、請求項30に記載の方法。 33.機械的ターゲッティングが受動的である、請求項32に記載の方法。 34.機械的ターゲッティングが能動的である、請求項32に記載の方法。 35.リポソームが糖、糖脂質およびタンパク質よりなる群から選択される成分と カップリングすることにより能動的にターゲッティングされる、請求項34に記載 の方法。 36.タンパク質成分が抗体である、請求項35に記載の方法。 37.前記ベクターがウイルスである、請求項27に記載の方法。 38.前記ウイルスがRNAウイルスである、請求項37に記載の方法。 39.RNAウイルスがレトロウイルスである、請求項38に記載の方法。 40.レトロウイルスが本質的に標的特異性である、請求項39に記載の方法。
