抗生物質
本発明は、一般式1(a)(式中、Mはポリエンマクロライド抗生物質残基を
表わし、Rは糖残基の可変部分を表わし、そして同一かまたは異なることができ
るR1およびR2はそれぞれC1-4アルキル基を表わす)のポリエンマクロライド
群抗真菌性抗生物質のアルキルエステルのN−アルキル−N−グリコシル誘導体
、一般式2(a)(式中、M,R,R1およびR2は前記同様で、Aは無機または
有機酸のアニオンを表わす)によって表わされる化合物1(a)の塩類、一般式
1(a)および2(a)の化合物の調製法ならびに薬剤におけるその使用に関す
る。
とくに興味ある化合物は、一般式1(b)(式中、Mはポリエンマクロライド
抗生物質残基を表わし、Rは糖残基の可変部分を表わす)のポリエンマクロライ
ド群抗真菌性抗生物質のメチルエステルのN−メチル−N−グリコシル誘導体お
よび一般式2(b)(式中、Mはポリエンマクロライド抗生物質残基を表わし、
Rは糖残基の可変部分を表わし、Aは無機または有機酸のアニオンを表わす)の
、その塩類を含み、その調製法および薬剤としての使用にも興味がある。
出発原料の抗生物質のアミノ基をアルキル基で置換するポリエンマクロライド
群抗生物質のN−アルキル誘導体は公知である。
出発原料の抗生物質のアミノ基を糖残基で置換するポリエンマクロライドのN
−グリコシル誘導体も公知である(L.Falkowski、J.Golik、P.Kolodziejczyk、J.P
awlak、J.Zielinski、T.Ziminski、E.Borowski の J.Antibiotics 28、244(197
5);L.Falkowski、J.Pawlak、J.Golik、P.Kolodziejczyk、B.Stefanska、E.Byl
ec、E.Borowski の Acta Polon.Pharm.37、517(1980))。これら誘導体の調製に
用いられる糖類の例にはD−グルコース、D−マンノース、L−ラムノース、D
−リボース、およびマルトースがある。ポリエンマクロライドを適当な糖と反応
させると同時にアマドリ転位が起って、対応するN−グリコシル誘導体を生成す
る。該化合物は出発原料の抗生物質と同様の生物学的活性を示し、水溶性塩を生
じる。しかし、それに付随する高毒性は顕著な欠点を示す。

のActa Polon.Pharm.37、517(1980)の論文から出発原料の抗生物質のアミノ基
をD−グルコース、D−マンノース、L−ラムノース、D−リボース、マルトー
スのような糖残基で置換するポリエンマクロライドのN−グリコシル誘導体が公
知である。該化合物は、ポリエンマクロライドと前記の糖類との反応および同時
のアマドリ転位によって調製される。該化合物は出発原料の抗生物質と同様の生
物学的活性を示し、水溶性塩を生じる。しかし、該化合物は著しい毒性を示す。
出発原料の抗生物質のアミノ基を完全にメチル化して第四級アンモニウム塩と
するポリエンマクロライドメチルエステルのトリメチルアンモニウム誘導体も公
知である(L.Falkowski、 B.Stefanska、 J.Zielinski、 E.Bylec、 J.Gol
ik、P.Kolodziejczyk、 E.Borowski の J.Antibiotics 32、1080(1979))。
該化合物は、出発原料の抗生物質のジメチル硫酸による完全メチル化によって調
製される。該誘導体の有利な性質には水への溶解度、出発原料の抗生物質と同様
の抗真菌性活性がある。
別の種類の誘導体は、L.Falkowski、B.Stefanska、J.Zielinski、E.Bylec
、J.Golik、P.Kolodziejczyk、E.Borowskiの論文のJ.Antibiotics 32 1080(1
979)により公知のポリエンマクロライドメチルエステルのトリメチルアンモニウ
ム誘導体であって、該化合物では、抗生物質のメチルエステルのアミノ基は完全
にメチル化されて第四級アンモニウム塩となっている。該化合物は、出発原料の
抗生物質のジメチル硫酸による完全メチル化によって調製される。該誘導体は水
に可溶で、出発原料の抗生物質の活性と同様の抗真菌性活性を特徴とするが、極
めて毒性があって、不安定である。
他のタイプのN−アルキル誘導体には、抗生物質とN−エチルマレイミド、N
,N'−ヘキサメチレンジマレイミド、N−(3−ジメチルアミノプロピル)マレ
イミドのようなN置換マレイミドとのマイケル付加反応によって生成するN−ス
クシンイミジル誘導体があり、該化合物はA.Czerwinski、W.A.Konig、T.Zie
niawa、P.Sowinski、V.Sinnwell、S.Milewski、E.Borowskiの論文J.Antibio
tics
44 979(1991)によって公知である。該化合物は出発原料の抗生物質よりも毒性が
少ないが、抗真菌性活性が低下する。
最後に、ポリエンマクロライドのN−アルキル誘導体の最後の公知の群は、抗
生物質と、アセチルアセトン、エチルアセチルアセテート、ジメチルアセタール
またはジメチルホルムアミドとの反応によって生成するN−エナミンおよびアミ
ジン誘導体であり、該化合物は、B.Stefanska、J.Zielinski、E.Borowski、L
.Falkowskiの論文Acta Polonica Phann.45、71(1988)に示されている。該誘
導体は、出発原料の抗生物質と同様の抗真菌性活性および有機溶剤へのすぐれた
溶解度を示すが、該化合物は依然として著しい毒性があり、また極めて不安定で
ある。
本発明者らは今やポリエンマクロライド抗生物質のアルキルエステルの混合N
−アルキル−N−グリコシル誘導体を調製した。これら混合化合物の調製法も確
立した。これら新規化合物は、出発原料の抗生物質と同様のすぐれた抗真菌性活
性を有し、酸と水に可溶な塩類を生成し、かつ著しく低毒性であることが判明し
た。従来技術のポリエンマクロライドのN−アルキル誘導体はすべて、かなりの
欠点である高毒性を示すので、これらの性質は予期しないことである。本発明に
より含まれる化合物はこの欠点がない。
現在まで、ポリエンマクロライド抗生物質のメチルエステルのN−メチル−N
−グリコシル誘導体およびその調製法は知られていなかった。驚くべきことに、
該化合物は、出発原料の抗生物質と同様の高抗真菌性活性を保持し、酸と水に可
溶な塩を生成し、かつ驚異的に低毒性である。これら化合物は、従来技術のN−
アルキル誘導体と同様の毒性を示さない。高毒性は以前に公知であったすべての
ポリエンマクロライドN−アルキル誘導体の基本的な欠点をなすが、本発明によ
り含まれる化合物はこの欠点がない。
本発明の第1の態様は、一般式1(a)(式中、Mはポリエンマクロライド群
抗生物質の残基を表わし、Rは抗生物質と単糖類またはオリゴ糖との反応によっ
て生成する糖残基の一部分を表わし、そして同一かまたは異なることができるR1
およびR2はそれぞれC1-4アルキル基を表わす)のポリエンマクロライド群抗
生物質のアルキルエステルのN−アルキル−N−グリコシル誘導体を提供する。
好ましくは、ポリエンマクロライド群抗生物質の残基MはアンホテリシンB、
カンジジン、カンジドイン、カンジジニン、マイコヘプチン、ナイスタチン、ポ
リフンギン、オーレオファシン、バシジン、トリコマイシンまたはカンジシジン
から選ばれる。
糖残基Rが誘導される単糖類またはオリゴ糖はD−グルコース、またはL−グ
ルコース、またはD−マンノース、またはD−ガラクトース、またはラクトース
、またはマルトースから選ぶのが好ましい。ポリエンマクロライドを適当な糖と
反応させると、同時にアマドリ転位が起って本発明の化合物に対するN−グリコ
シル前駆物質を生成する。
本発明の好ましい態様は、一般式1(b)(式中、Mはポリエンマクロライド
群抗生物質の残基を表わし、Rは抗生物質と、単糖類またはオリゴ糖、好ましく
はD−グルコース、もしくはL−グルコース、もしくはD−マンノース、もしく
はD−ガラクトース、もしくはラクトース、もしくはマルトースとの反応、およ
び同時のアマドリ転位によって生成する糖残基の一部分を表わす)によって示さ
れるポリエンマクロライド群抗生物質のメチルエステルのN−メチル−N−グリ
コシル誘導体を含む。
好ましい形態の本発明は、ポリエンマクロライド群抗生物質がアンホテリシン
B、またはカンジジン、またはカンジドイン、またはカンジジニン、またはマイ
コヘプチン、またはナイスタチン、またはポリフンギン、またはオーレオファシ
ン、またはバシジン、またはトリコマイシン、またはカンジシジンである誘導体
に関する。
本発明の第2の態様は、一般式2(a)(式中、M,R,R1およびR2は前記
本発明の第1の態様に示した通りであり、Aは有機または無機酸のアニオンを表
わす)のポリエンマクロライド群抗生物質のN−アルキル−N−グリコシル誘導
体の塩類を提供する。該塩は生理学的に許容できる塩であることが好ましく、ま
たAがL−アスパラギン酸のアニオンである化合物がとくに好ましい。R1およ
びR2がメチル基である塩がとりわけ好ましい。
本発明の第2の態様の好ましい実施態様は、一般式2(b)(式中、Mはポリ
エンマクロライド群抗生物質の残基を表わし、Rは抗生物質と、単糖類またはオ
リゴ糖、好ましくD−グルコース、もしくはL−グルコース、もしくはD−マン
ノース、もしくはD−ガラクトース、もしくはラクトース、もしくはマルトース
との反応および同時のアマドリ転位によって生成する糖残基の一部分を表わし、
かつAは有機または無機酸のアニオンを表わす)によって示されるポリエンマク
ロライド群抗生物質のN−メチル−N−グリコシル誘導体の塩類を含む。
好ましい形態の本発明は、ポリエンマクロライド群抗生物質がアンホテリシン
B、またはカンジジン、またはカンジドイン、またはカンジジニン、またはマイ
コヘプチン、またはナイスタチン、またはポリフンギン、またはオーレオファシ
ン、またはバシジン、またはトリコマイシン、またはカンジシジンである塩に関
し、また好ましくAがL−アスパラギン酸のアニオンに関する。
本発明の第3の態様は、式1(a)(式中、M,R,R1およびR2は本発明の
第1および第2の態様でさきに述べた通りである)の化合物の調製法において、
ポリエンマクロライド抗生物質を単糖類またはオリゴ糖と反応させ、該反応は同
時にアマドリ転位を生起して、ポリエンマクロライド抗生物質のN−グリシジル
誘導体を生成することを特徴とし、該アマドリ転位の生成物を単離し、該生成物
をアルキル化剤で処理し、さらに該組成物を精製する工程を含む方法を含む。
本発明の第3の態様の1つの実施態様では、アマドリ転位の生成物を、該転位
が生起する溶液から沈澱させて、懸濁物として単離させる。アマドリ転位を維持
させるにはN,N−ジメチルホルムアミドのような有機溶剤が好ましい。アマド
リ転位の生成物を沈澱させて懸濁液を生成させるにはジエチルエーテルのような
有機溶剤が適切である。
本発明の第3の態様の第2の実施態様では、アマドリ転位の生成物のアルキル
化を低温で行う。−5℃ないし+5℃の温度が好ましい。ジアゾアルカンのよう
なアルキル化剤を使用することができる。ジアゾメタンのエーテル溶液が好まし
いアルキル化剤である。
溶剤を除去し、ジエチルエーテルで沈澱させるとN−アルキル−N−グリコシ
ル粗製物を単離することができる。精製産物は、公知の精製工程を用いて単離す
ることができる。
本発明の第3の態様の好ましい実施態様は、一般式1(b)(式中、Mはポリ
エンマクロライド群抗生物質の残基を表わし、Rは抗生物質と、単糖類またはオ
リゴ糖、好ましくはD−グルコース、もしくはL−グルコース、もしくはD−マ
ンノース、もしくはD−ガラクトース、もしくはラクトース、もしくはマルトー
スとの反応、および同時のアマドリ転位によって生成する糖残基の一部分を表わ
す)によって示されるポリエンマクロライド群抗生物質のメチルエステルのN−
メチル−N−グリコシル誘導体の調製法において、アマドリ転位によって得られ
るポリエンマクロライド群抗生物質のN−グリコシル誘導体を、有機溶剤、好ま
しくはN,N−ジメチルホルムアミド中の該誘導体の溶液から溶剤、好ましくは
ジエチルエーテルで沈澱させて懸濁液に変え、次いで低温、好ましくは−5℃な
いし+5℃の範囲でジアゾメタンのエーテル溶液で処理し、撹拌し、溶剤の蒸発
および濃溶液、好ましくは過剰のジエチルエーテルによる沈澱によって単離し、
さらに粗製物を公知の方法で精製する方法を含む。
本発明の第4の態様は、一般式2(a)(式中、M,R,R1およびR2は本発
明の第1、第2、および第3の態様についてさきに述べた通りである)のポリエ
ンマクロライド抗生物質のアルキルエステルのN−アルキル−N−グリコシル誘
導体の塩類調製法において、本発明の第3の態様によって調製したN−アルキル
−N−グリコシル誘導体を均質懸濁液を生成させるのに十分な水に懸濁させ、得
られた懸濁液を酸性化して、生成物を単離させる工程を含む方法を提供する。ポ
リエンマクロライド抗生物質のメチルエステルのN−メチル−N−グリコシル誘
導体の塩類の調製がとくに好ましい。
懸濁液を酸性化するのに有機または無機酸を使用することができる。L−アス
パラギン酸が好ましい。
精製産物の単離は、有機溶液による粗製物の沈澱に続く補足的な適当な溶剤に
よる洗浄および乾燥によって行うことができる。粗製物を沈澱させるのに用いる
溶剤は水と混和できるものが好ましく、アセトンが好ましい。生成物を洗うのに
用いる典型的な溶剤にはアセトンおよびジエチルエーテルがある。生成物は減圧
で乾燥するのが好ましい。
本発明の第4の態様の好ましい実施態様は、一般式2(b)(式中、Mはポリ
エンマクロライド群抗生物質の残基を表わし、Rは抗生物質と、単糖類またはオ
リゴ糖、好ましくはD−グルコース、もしくはL−グルコース、もしくはD−マ
ンノース、もしくはD−ガラクトース、もしくはラクトース、もしくはマルトー
スとの反応および同時のアマドリ転位によって生成する糖残基の一部分を表わす
)によって示されるポリエンマクロライド群抗生物質のメチルエステルのN−メ
チル−N−グリコシル誘導体の塩類の調製法において、アマドリ転位によって得
られるポリエンマクロライド群抗生物質のN−グリコシル誘導体を、有機溶剤、
好ましくはN,N−ジメチルホルムアミド中の該誘導体の溶液から、溶剤、好ま
しくはジエチルエーテルで沈澱させて懸濁液に変え、次いで低温、好ましくは−
5℃ないし+5℃の範囲で、ジアゾメタンのエーテル溶液で処理し、撹拌し、さ
らに溶剤の蒸発および濃溶液、好ましくは過剰のジエチルエーテルによる沈澱に
より単離させ、さらに公知の方法により粗製物を精製し、次に固形物として得た
誘導体を少量の水に懸濁させ、理論量の有機または無機酸を加え、次いで生成し
た溶液から水と混和する過剰の有機溶剤、好ましくはアセトンで生成物を沈澱さ
せ、さらに固形物を、好ましくはアセトンで洗った後、好ましくはジエチルエー
テルで洗い、好ましくは減圧で乾燥する方法を含む。
本発明の第5の態様は、治療に使用する式1(a)のN−アルキル−N−グリ
コシル誘導体またはその塩類を含む。
本発明の第6の態様は、ヒトおよび動物の真菌感染症の処置法において、式1
(a)のN−アルキル−N−グリコシル誘導体またはその塩類を本明細書でさき
に述べたようにヒトおよび動物への投与を含む方法を提供する。式1(b)のN
−メチル−N−グリコシル誘導体またはその塩類がとくに興味がある。
本発明の第6の態様の第1の好ましい実施態様は、ヒトおよび動物の外因性お
よび内因性真菌感染症の処置法において、一般式1(b)(式中、Mはポリエン
マクロライド群抗生物質の残基を表わし、Rは抗生物質と単糖類またはオリゴ糖
、好ましくはD−グルコース、もしくはL−グルコース、もしくはD−マンノー
ス、もしくはD−ガラクトース、もしくはラクトース、もしくはマルトースとの
反応、および同時のアマドリ転位によって生成する糖残基の一部分を表わす)に
よって示されるポリエンマクロライド群抗生物質のメチルエステルのN−メチル
−N−
グリコシル誘導体を用いて該感染症を処置する方法を含む。
本発明の第6の態様の第2の好ましい実施態様は、ヒトおよび動物の外因性お
よび内因性真菌感染症の処置法において、一般式2(b)(式中、Mはポリエン
マクロライド群抗生物質の残基を表わし、Rは抗生物質と、単糖類またはオリゴ
糖、好ましくはD−グルコース、もしくはL−グルコース、もしくはD−マンノ
ース、もしくはD−ガラクトース、もしくはラクトース、もしくはマルトースと
の反応、および同時のアマドリ転位によって生成する糖残基の一部分を表わし、
Aは有機または無機酸のアニオンを表わす)によって示されるポリエンマクロラ
イド抗生物質のメチルエステルのN−メチル−N−グリコシル誘導体の塩類を使
用して該感染症を処置する方法を含む。
本発明の第7の態様は、さきに本明細書でさきに述べたように真菌感染症を処
置するのに使用するための式1(a)のN−アルキル−N−グリコシル誘導体ま
たはその塩類を提供する。式1(b)のN−メチル−N−グリコシル誘導体また
はその塩類がとくに興味がある。
処置が講じられる感染症は内因性であっても外因性であってもよい。投与の方
式は感染症の性質による。したがって、本発明の化合物は静脈内、腹腔内、経口
、局所、皮下、直腸または膣投与用に処方することができる。
本発明の第8の態様は、真菌感染症の処置に使用するための式1(a)のN−
アルキル−N−グリコシル誘導体またはその塩、および生理学的に許容できるキ
ャリヤーを含む組成物を提供する。N−アルキル−N−グリコシル誘導体および
キャリヤーの性質は投与方式による。該組成物は、場合により、必要に応じて他
の公知の抗真菌剤とともに、本発明による1種以上の化合物から処方することが
できる。式1(b)のN−メチル−N−グリコシル誘導体またはその塩類を含有
する組成物がとくに興味がある。
本発明の第9の態様は、医薬用または獣医学用に処方される1種以上の式1(
a)のN−アルキル−N−グリコシル誘導体またはその塩類および生理学的に許
容できるキャリヤーを含む単位用量形態を提供する。単位用量形態は、場合によ
り他の公知の抗真菌剤とともに本発明による1種以上の化合物から処方すること
ができる。本発明の前記態様の類推から式1(b)のN−メチル−N−グリコ
シル誘導体を含有する単位用量形態がとくに興味がある。
前述のように、単位用量形態の性質は投与方式による。典型的には、錠剤やカ
プセルは経口投与に適し、クリームやパッチは局所投与に適し、またペッサリー
は直腸や膣投与に適する。
本発明の第10の態様は、真菌感染症の処置に用いるための薬剤調製への式1
(a)のN−アルキル−N−グリコシル誘導体またはその塩類の使用を提供する
。N−アルキルおよびアルキルエステル置換基が共にメチル基である化合物がと
くに興味がある。
本発明の化合物について分光法を用いて行った構造決定は出発原料の抗生物質
の安全性が反応中保持されることを示す。
ポリエンマクロライド群抗生物質のメチルエステルのN−メチル−N−グリコ
シル誘導体の本発明による調製法は、出発原料の抗生物質の構造を変えることな
く所望の生成物をもたらす。得られた化合物の構造は分光法を用いて確認された
。式3のN−メチル−N−D−フルクトシルアンホテリシンBメチルエステルの
構造決定によって実証される。
N−メチル−N−D−フルクトシルアンホテリシンBメチルエステルの電子ス
ペクトルは、アンホテリシンBを意味する出発原料の抗生物質の電子スペクトル
と同一であって、本発明による方法が、ポリエン発色団の分解をもたらさず、大
きい吸光値(E1% 1cm=1300(382nmにおいて))は得られた生成物の
高純度を確証することを示す。N−メチル−N−D−フルクトシルアンホテリシ
ンBメチルエステルの赤外吸収スペクトルは、1730cm-1におけるエステル
カルボニル基の伸縮振動に関連するバンドおよび遊離カルボキシル基のバンドの
欠除を示し、これはカルボキシル基がメチルエステル基に完全に変ったことを意
味す
る。N−メチル−N−D−フルクトシルアンホテリシンBメチルエステルの構造
に関する完全な情報は1H(DQF−COSY、ROESY)、13C(DEPT
)の核磁気共鳴スペクトル(NMR)および化合物の式3への帰属を可能にした
ヘテロ相関スペクトル(Varian 300MHZスペクトロメーター)によっても
たらされた。
もっとも重要な1Hおよび13Cの情報をそれぞれが表1および2に示す。アン
ホテリシンBのアグリコンのNMRデータは文献値(P.Sowinski、J.Pawlak、
E.Borowski、P.Gariboldiの Magn.Reson.Chem.30,275(1992))に完全に
一致する。N−CH3(δ=2.35ppm)およびH−1”(2.30および
3.15ppm)の1H化学シフト(DMSO/MeOD溶剤系中)はアミノ置換
基の効果の特徴である。酸性化後、アミノ基のプロトン化によって(H−3'のδ
が3.19ppmに変る)、δはN−CH3の2.92ppmおよびH−1”の3.
64に変る。これらのデータはプロトンNCH3/H3'、NCH3/H2'、NC
H3/H1”bおよび1″a/H3'間のROE効果によっても支持される。結合
定数およびROEは、以前に遊離アンホテリシンBに認められたように、マイコ
サミン部分の4C1立体配座を示す。
表1はN−メチル−N−D−フルクトシルアンホテリシンBメチルエステルの
二糖類フラグメントの化学シフト1HおよびROE効果を示す。
表2は、N−メチル−N−D−フルクトシルアンホテリシンBメチルエステル
の二糖類フラグメントの13C−NMR化学シフトとD−フルクトースのデータと
の比較を示す。
表2からのフルクトシルフラグメントの13C−NMRデータをD−フルクトー
スの文献値と比較すると、糖置換基がピラノシド形であることがわかる。表3に
示すカップリング定数の観察値は式4によって表わされるフルクトピラノシド環
の舟形立体配座を立証する。該環のこのコンホメーションだけが測定したカップ
リング定数と完全に一致する。
表3はN−メチル−N−D−フルクトシルアンホテリシンBメチルエステルの
二糖類フラグメントのプロトンのカップリング定数JH,Hを示し(ピリジン−d5
:メタノール−d49:1)、密に結合したH3”−H6”スピン系のカップリ
ング定数および化学シフトはコンピュータのシミュレーションによって反復改善
した。
示された結果は式4によって表わされるN−メチル−N−D−フルクトシルアン
ホテリシンBメチルエステルのフルクトシルフラグメントの立体配座を立証する
。他の、ポリエンマクロライド群抗生物質のメチルエステルのN−メチル−N−
グリコシル誘導体は前記と同様の方法によって特徴づけた。
これから、下記の非限定例を参照しながら本発明を述べる。本発明の範囲に入
るさらに他の態様は当業者には明らかとなろう。抗真菌活性
調製したすべての化合物について、生体外で抗真菌活性さらにまた毒性も測定
した。N−メチル−N−D−フルクトシルアンホテリシンBメチルエステルアス
パラテートを指していう最良の性質を有する化合物の場合には、生体内の活性お
よび毒性をも測定した。生体外の抗真菌活性
ポリエンマクロライド処置の標準法に従って化合物の抗真菌活性を測定した。
液状Sabouraud培地に104細胞/mlの試験生物 Candida albicans ATCC 262778
を接種して供試抗生物質(無菌希釈液)とともに30℃で24時間培養した。対
照としてアンホテリシンBを使用した。化合物はDMFに溶解し、溶液の適当な
量を培地に加えた。成育阻止の程度を測定するのに濁度測定法(660nm)を
適用した。真菌の成育を50%阻止した抗生物質の濃度を投与量反応曲線から求
めた。得られたIC50値は化合物の抗真菌活性を特徴づけた。生体外(in vitro)毒性
動物細胞に対する生体外の化合物の毒性を、ヒトの赤血球の溶血度の測定によ
り、ポリエンマクロライド処置の標準法を用いて測定した。新鮮なクエン酸添加

生体内毒性は、N−メチル−N−D−フルクトシルアンホテリシンBメチルエ
ステルL−アスパラテートについて、最大耐用量(MTD)および急性毒性(LD50
)として求めた。MTD用量を求めるために、化合物をグルコースの5%溶液
に溶解し、Balb/cマウスに単一および多重用量で静脈内および腹腔内に投
与した。単一最大耐用量は静脈内投与の場合には100mg/kg、腹腔内投与
の場合には200mg/kg以上であった。5日間で100mg/kgという腹
腔内投与の多重最大耐用量は極めて多かった。このような用量の場合には、観察

N−メチル−N−D−フルクトシルアンホテリシンBメチルエステルL−アス
パラテートの急性毒性LD50は平均重量が20gのSwiss Websterのメスのマウ
スについて試験した。種々の用量の供試化合物および比較のため5%グルコース
に溶解したファンギゾンの形のアンホテリシンBをマウスの静脈内に投与した。
溶液の投与量は0.5mlであった。対照として5%グルコース溶液の0.5mlを
マウスに投与した。両方の配合物の各用量を5匹のマウスに投与した。マウスは
7日間観察した。次に、マウスを殺して、若干の血清指数を求めた。対照と比較
して、アスパラテートアミノトランスフェラーゼまたはクレアチニンのレベルが
増大しないことが判明した。N−メチル−N−D−フルクトシルアンホテリシン
BメチルエステルL−アスパラテートの場合にはLD50が400mg/kgであ

マウスの全身性カンジタ症モデルを用いてN−メチル−N−D−フルクトシル
アンホテリシンBメチルエステルL−アスパラテートの化学療法効果を調べた。
Candida albicans をSabouraudデキストロースブロス中で室温下で一夜間生育さ
せた。真菌細胞を遠心分離し、0.9%塩化ナトリウム溶液で2回洗って、生理
的食塩水中に懸濁させた。体重25gのメスのSwiss Websterマウスに、0.9
%塩化ナトリウム溶液0.2ml中の105個のCandida の細胞を静脈内に注射し
た。当初、感染症は全身的であったが、2ないし3日までには腎臓に局限された
。無処置のマウスは通常感染後7日から14日の間に死亡した。感染後3日間は
2回、続けて5日間は5ないし6時間間隔でマウスの静脈内に処置した。配合物
は5%グルコース溶液として投与した。マウスは感染の日から始めて5週間観察
した。この時が過ぎると生存したマウスを殺して、腎臓を取り出し、滅菌水で均
質にしてホモジネートをSabouraudデキトスロース寒天の上に置いて、Candidaの

ポリエンマクロライド群抗生物質のメチルエステルのN−メチル−N−グリコ
シル誘導体、その塩類、および調製法を下記実施例によって説明する。実施例I
1gのアンホテリシンB(E1% 1cm=1350(382nm、MeOHにおい
て))を15mlのN,N−ジメチルホルムアミドに溶解し、0.3gのD−グ
ルコースを加え、混合物を暗所内で、37℃で40時間撹拌した。次に、反応物
を冷却し、過剰のジエチルエーテルで固形物を沈澱させた。固形物を遠心分離し
、ジエチルエーテルで2回洗い、減圧乾燥した。過剰のグルコースを除くために
固形物を20mlの水に懸濁させ、遠心分離し、少量の水で2回、アセトンで2

ール−水10:6:1(容量/容量)溶剤系でシリカゲルを用いる薄層クロマト
グラフィーを行った。約2時間を要した反応完了後、過剰のジアゾメタンおよび
ジエチルエーテルを40℃以下の温度で減圧下で蒸発させた。過剰のジエチルエ
ーテルを用いて残留物から粗製物を沈澱させ、遠心分離し、ジエチルエーテルで
2回、さらにn−ヘキサンで洗い、減圧乾燥して、0.95gの粗製物を得た。
クロロホルム−メタノール−水20:8:1(容量/容量)溶剤系で70−23

0.5−0.54の画分を集めた。集めた画分を減圧下で蒸発させた。乾燥残留

験したCandida albicansに対するこの化合物の抗真菌活性はIC50=0.12μ
g/mlを示し、前記のように測定したヒトの赤血球に対する毒性は350μg
/mlよりも大きいEH50を示した。比較すると、出発原料のアンホテリシンB
のEH50値は1.5μg/mlであった。350μg/mlを上回るとこの化合
物はこの実験条件では不溶であるので、N−メチル−N−D−フルクトシルアン
ホテリシンBメチルエステルのEH50の正確な値は求めることができなかった。
実施例II
0.5gのカンジジン(E1% 1cm=1175(382nm、MeOHにおいて))
および0.15gのD−グルコースを10mlのN,N−ジメチルホルムアミド
に溶解して、37℃で36時間撹拌した。以降の手順は実施例Iと同様であって
、0.43gのN−D−フルクトシルカンジジン(E1% 1cm=1100(382
nm)MeOH中において))が得られた。実施例Iと同様にこの生成物をジア
ゾメタンのジエチルエーテル溶液でメチル化して、0.4gの粗製物を得た。実

実施例III
2gのナイスタチン(E1% 1cm=870(304nm、MeOH中において))お
よび0.6gのD−グルコースを35mlのN,N−ジメチルアセトアミドに溶
解して、37℃で40時間撹拌した。反応完了後、粗製物をジエチルエーテルで
沈澱させ、遠心分離して、真空乾燥した。次に過剰のグルコースを少量の水−ア
セトン1:1混合物、ついでアセトン、ジエチルエーテルで洗い去り、生成物を
遠心分離し、真空デシケータで乾燥して、1.2gのN−D−フルクトシルナイ
スタチン(E1% 1cm=720(304nm、MeOH中において))を得た。実
施例Iと同様にジアゾメタンを用いてこの生成物をメチル化して、1.25gの
粗製物を得た。この生成物を実施例Iと同様にシリカゲルカラムで精製した。純
粋なN−メチル−N−D−フルクトシルナイスタチンメチルエステルを含有する
画分を実施例Iのように薄層クロマトグラフィーにかけてRf=0.49−0.5
2と確認した。該画分を集め、減圧下30℃で蒸発乾固し、少量のN,N−ジメ
チルホルムアミドに溶解し、生成物をジエチルエーテルで沈澱させた。生成物を

のN−メチル−N−D−フルクトシルナイスタチンメチルエステル(E1% 1cm=

実施例IV
15mlのN,N−ジメチルホルムアミド中の0.79gのバシジン(抗生物

反応完了後、混合物を冷却し、過剰のジエチルエーテルで固形物を沈澱させた。

N−D−フルクトシルバシジン(E1% 1cm=720(378nm、MeOH中に
おいて))を得た。実施例Iと同様にジアゾメタンを用いて得られた誘導体をメ
チル化して、0.5gの粗製物を得た。実施例Iに述べたと同様に、シリカゲル
カラムのクロマトグラフィーによって純粋なN−メチル−N−D−フルクトシル
バシジンメチルエステルを単離した。ただしカラムはクロロホルム−メタノール
−水30:8:1溶剤系で展開した。純粋なバシジン誘導体を含有し、かつ薄層
クロマトグラフィーにかけてクロロホルム−メタノール−水13:8:1溶剤系
でRf=0.53−0.55値を有する画分を集めて一緒にした。以後の手順は
実施例IIIと同様であった。0.07gのN−メチル−N−D−フルクトシル
バシジンメチルエステル(E1% 1cm=900(378nm、MeOH中において
))が得られた。この化合物はIC50=0.01μg/mlおよびEH50=17
0μg/mlを示した。この化合物の構造はN−メチル−N−D−フルクトシル
アンホテリシンBメチルエステルについて述べた方法によって決定された。
実施例V
0.79gのカンジシジンD(抗生物質複合カンジシジンの主成分)を実施例
IVに述べたと同様に処理した。0.1gのN−メチル−N−D−フルクトシル
