HIV−TATの検出方法
本発明は、体試料中のHIV−TATの検出方法及び本目的に使用されるキッ
トに関する。
AIDSの場合、T細胞は、しばしばプログラムされた細胞死をこうむること
がよく知られている。該細胞死はT細胞アポトーシスとして述べられる。最近の
よってT細胞アポトーシスが増すという事実に言及している。
この知見は、現在、HIV感染及びAIDSを、それぞれ、より良く理解すし
、新しい治療方法を考える可能性を提供する。しかしながら、本目的のために、
体試料中のHIV−TATを検出することができることが必要である。しかし、
これは、これまでは可能ではなかった。
したがって、本発明の目的は、体試料中にHIV−TATを検出できる方法を
提供することにある。
本発明によって、これは下記工程を有する方法により達成される:
(a)体試料を採取し、該体試料をデターゼント(Detergens)及び高
濃度の塩と共にインキュベートする工程、
(b)工程(a)由来の体試料をサイズ分画する工程、及び
(c)工程(b)由来の体試料を抗HIV−TAT抗体と共にインキュベートす
る工程。
「体試料」という用語は、人間及び動物由来のいかなる種類でもよい体試料を
有する。特に、これらは、血液、痰、尿、大便、体液、胆汁、胃腸の分泌物、臓
器の吸引物又は斑(Organpunktate)、生検試料及びリンパ液であ
る。
体試料を採取するために、慣用の方法を使うことができる。ついで、該体試料
を、慣用のデターゼント、例えば、NP−40又はtriton X−100と
共にインキュベートする。また複数のデターゼントを同時に又は連続的に使用す
ることが可能である。全体で1%のデターゼントを用いることが好ましい。さら
に、該体試料に高濃度の塩を添加する。それは慣用の塩、例えばNaClであっ
てもよい。また、複数の塩を同時に又は連続的に用いることが可能である。全体
で0.1〜1Mの塩を使用することが好ましい。HIV−TATを、デターゼン
ト及び高濃度の塩の使用によって、結合したタンパク質及び核酸から遊離する。
さらに、デターゼントによって、該体試料中でHIV粒子を不活性化する。
さらに、該体試料をサイズ分画に供する。本目的のために、慣用の方法、例え
ば、Filtron社、ノースボロー(Northborough)、マサチュ
ッツ州、U.S.Aの遠心分離(分画)カラムによる分画を使用することが可能
である。5〜30kDの範囲以内で分画を行うことが好ましい。
また、該体試料を、抗HIV−TAT抗体と共にインキュベートする。「HI
V−TAT」という用語は、HIV感染を引き起こす及び/又はAIDSを生じ
るウイルスから生じるいかなるTAT及びその断片をも有する。該用語は、合成
的に作成した、いかなるTAT及びその断片にも関する。該抗体はポリクローナ
ル又はモノクローナルであってよく、モノクローナル抗体が好ましい。さらに、
該抗体は、合成であってもよく、任意にHIV−TATを認識するために必要で
ない部分を一部又は全部欠損してもよく、これらの部分をそれぞれ、さらに好ま
しい性質を有する抗体を提供する他の部分によって置き換えてもよい。また、多
様な抗HIV−TAT抗体を同時に又は連続的に使用することが有利であろう。
特に、該1又は複数の抗HIV−TAT抗体をインキュベートすることと並行し
て、該体試料中のHIV−TATの定量的検出をしやすいように、一定のTAT
標品と共に前記インキュベートすることが好ましい。
1又は複数の抗HIV−TAT抗体との該体試料のインキュベーション並びに
TAT標品との後者のインキュベーションは、ウエスタンブロット、ELISA
、例えばサンドウィッチ若しくは競合ELISA、免疫蛍光法又は免疫沈降法等
の慣用の方法の一部となってもよい。本目的のために、該抗体はラベルしてもよ
く、もし適切ならば、又は該抗体に対するラベルされた抗体を組み合わせて用い
てもよい。
本発明による方法は、HIV−TATを体試料中で検出できるという点が特徴
である。該方法は、特異的に及び迅速に行うことができる。したがって、HIV
感染を診断すること及び/又はAIDS症状を追跡することのために完全に適す
る。また、後者は、特にHIV−TATに対しておこなわれる治療方法の効果を
直接的に追跡する大きな利点を有する。
また、本発明によって、体試料中のHIV−TATの検出のためのキットが提
供される。かかるキットは、好ましくは、下記を有する。
(a)デターゼント、
(b)高濃度の塩の溶液、
(c)サイズ分画カラム、
(d)1又は複数の抗HIV−TAT抗体、
(e)HIV−TAT標品、並びに
(f)慣用の補助剤及び担体材料。
本発明による方法と関連して作られた前記説明は、ここで相応して考慮されな

本発明を以下の実施例により説明する。実施例:HIV−1感染した人間の体試料中のTATの検出
33人のHIV−1感染した人間から血清試料を採取した。20人のHIVの
人間由来の血清及びHIV−1感染したH9細胞の上清と共に、それらをTAT
の存在を試験した。さらに、一定の標品を得るために、対照血清に合成TATを
添加した。
前記血清を1%triton X−100及び1MNaClと共に、それぞれ
