【発明の詳細な説明】N−(4−シアノフェニルカルバモイル若しくは2−シアノピリジ−5−イルカ ルバモイル)−L−アスパラギン酸又はL−グルタミン酸の3,4−二置換α− ベンゼンアミド由来の新規甘味料
本発明の目的は、N−(4−シアノフェニルカルバモイル若しくは2−シアノ
ピリジ−5−イルカルバモイル)−L−アスパラギン酸α−ベンゼンアミド又は
L−グルタミン酸α−ベンゼンアミド由来の新規甘味料そのものである。非常に
甘味の強いこれらの新規甘味料は、様々な製品、特に炭酸飲料、食品、砂糖菓子
、練り粉菓子、チューインガム、衛生用品、美容用品、化粧用品、薬用及び獣医
用製品、並びに、これらに相当するものに甘味を与えるのに好適である。
周知のとおり、甘味料の工業的規模の利用が可能となるには、その使用コスト
が制限されるようにとりわけ強い甘味能を有する必要がある。
本発明の新規化合物は、重量基準でショ糖(砂糖)の最大50000倍強力な
、極めて高い甘味活性を持つ。現在市販されている人工甘味料の中で最も良く利
用されているものは、アスパルテームの名でよりよく知られているジペプチド誘
導体N−L−α−アスパルチル−L−フェニルアラニン1−メチルエステルであ
る。この化合物の主な利点は、優れた感覚受容性、並びに、L−アスパラギン酸
及びL−フェニルアラニンという2つの天然アミノ酸を基礎に置いた化学構造で
ある。しかしながら、その甘味能は比較的弱く、重量基準でショ糖の120〜1
80倍にすぎない。このことは、120〜180gの砂糖に代わるには、アスパ
ルテームが1g必要である事を示す。この比較的弱い甘味能、及び、ジペプチド
構造が元で比較的高価になる製造コストが、アスパルテームが高価な甘味料であ
る原因であり、工業的に利用する際の大きな欠点である。
本発明の化合物は、甘味能が非常に強く、更に製造が簡便であるため、低コス
トの甘味料を製造できるという利点がある。実際、本発明の好ましい甘味料は、
重量基準で砂糖の最大50000倍の甘味を持つ。このことは、本発明の好まし
い化合物1グラムで、砂糖50キログラムに充分代替できることを意味する。本
発明の好ましい甘味料は、アスパルテーム自身よりも約250〜400倍強力で
あるので、その使用コストはアスパルテームよりはるかに低いものとなり、その
工業的利用を有利なものとしている。
JP86−260052で、ツチヤらは、以下の一般式を持つ甘味料を開示し

上記公報では、甘味能を表す数字は、特許請求された2つの化合物でしか明ら
かにされていない。すなわち、以下の式のN−(4−ニトロフェニルカルバモイ
ル)−L−アスパラギン酸−α−4−クロロベンゼンアミド:
は、ショ糖の5400倍の甘味能を持つと記述されており、以下の式のN−(4
−ニトロフェニルカルバモイル)−L−アスパラギン酸−α−4−シアノベンゼ
ンアミド:
は、ショ糖の4800倍の甘味能を持つと記述されている。
N−(4−シアノフェニルカルバモイル若しくは2−シアノピリジ−5−イル
カルバモイル)−L−アスパラギン酸−α−ベンゼンアミド又はL−グルタミン
酸−α−ベンゼンアミドのいくつかの誘導体で、ベンゼンアミド基の3(メタ)
位と4(パラ)位を適切な置換基で置換すると、全く予期しないことにその甘味
能が著しく増加することが見いだされ、これが本発明の基礎を構成する。ところ
で、甘味料の構造をたとえわずかでも修飾すると、その活性が全く失われてしま
う可能性があるのは周知のことである。それゆえ、ベンゼンアミド基の2箇所を
置換することによって、甘味活性が抑制されるのではなく、逆に従来の一置換化
合物の活性を強力に高めることになるとは全く予想されなかった。
従って、本発明の目的は、次式で表されることを特徴とする新規甘味料:

本発明の特に好ましい第二の態様は、次式のN−(2−シアノピリジ−5−イ
ルカルバモイル)−L−アスパラギン酸−α−3,4−ジメチルベンゼンアミド

その甘味能は、2%ショ糖溶液との比較によると、重量基準でショ糖の4000
0倍である。
本発明の他の好ましい化合物は、N−(4−シアノフェニルカルバモイル)−
L−アスパラギン酸−α−3,4−ジメチレンジオキシベンゼンアミド、N−(
2−シアノピリジ−5−イルカルバモイル)−L−アスパラギン酸−α−3,4
−メチレンジオキシベンゼンアミド、N−(4−シアノフェニルカルバモイル)
−L−アスパラギン酸−α−3−ヒドロキシ−4−メトキシベンゼンアミド、N
−(2−シアノピリジ−5−イルカルバモイル)−L−アスパラギン酸−α−3
−ヒドロキシ−4−メトキシベンゼンアミド、N−(4−シアノフェニルカルバ

以上のように、本発明の化合物は、非常に特殊な性質、及び、ベンゼンアミド
基上の置換基の位置という二点で、JP86−260052で開示された化合物
とは異なるものである。実際、この従来技術を扱った公報の中では、開示・特許
請求された化合物はベンゼンアミド基のベンゼン環が一置換であり、その置換基
も公報中の例では4位(パラ)にしかない。
本発明によると、フェニル基Rは、3位(メタ)及び4位(パラ)が両者とも
に特定の基でもって置換されている。本発明の化合物では、この新規の基Rの選
択次第で、甘味能が劇的に増加し従来の化合物の10倍に達することも可能であ
り、その結果、この種の化合物の使用コストが著しく減少する。
本発明の甘味料は、所望の強さの甘味が得られるのに充分な量を添加するので
あれば、甘味を与えたいあらゆる食用製品に添加することができる。上記甘味料
の最適使用濃度は、例えば、上記甘味料の甘味能、製品の保存及び使用条件、製
品の主要構成要素、並びに、所望の甘味の程度等の種々の要素で決定される。食
用製品の製造にあたって必要な甘味料の最適量は、当業者ならば、通常の知覚分
析を実行することにより容易に決定することが出来る。本発明の甘味料は、一般
に、使用化合物の甘味能に応じて、食用製品1キログラム又は1リットル当たり
、0.5mg〜50mgの量を食用製品に添加する。当然のことながら、濃縮製
品は甘味料をより大量に含有しており、これを最終使用濃度にまで希釈する。
本発明の甘味料は、純品として目的製品に添加することが出来るが、甘味が強
力であるため、一般には適切な添加剤又は充填剤と共に混合する。
好ましくは、適切な添加剤又は充填剤は、ポリデキストロース、デンプン、マ
ルトデキストリン、セルロース、メチルセルロース、カルボキシメチルセルロー
ス及びその他のセルロース誘導体、アルギン酸ナトリウム、ペクチン、ゴム、ラ
クトース、マルトース、グルコース、ロイシン、グリセロール、マンニトール、
ソルビトール、炭酸水素ナトリウム、リン酸、クエン酸、酒石酸、フマル酸、安
息香酸、ソルビン酸及びプロピオン酸並びにそれらのナトリウム、カリウム及び
カルシウム塩、並びに、これらに相当するものからなる群より選択される。
本発明の甘味料は、食料製品に用いるに当たり、甘味料単独で用いてもよいし
、ショ糖、コーンシロップ、果糖、甘味性ジペプチド類縁体又は誘導体(アスパ
ルテーム、アリテーム)、ネオヘスペリジンジヒドロカルコン、水素化イソマル
トース、ステビオシド、L体の糖、グリシルリチン、キシリトール、ソルビトー
ル、マンニトール、アセスルフェーム(acesulfame)−K、サッカリ
ン並びにそのナトリウム、カリウム、アンモニウム及びカルシウム塩、シクラメ
ン酸(cyclamic acid)並びにそのナトリウム、カリウム及びカル
シウム塩、スクラロース(sucralose)、モネリン、タウマチン、並び
に、これらに相当するもの等の他の甘味料と共に用いてもよい。
本発明の化合物は、酸として用いられてもよいし、生理学的に許容される無機
又は有機塩基との反応により形成される塩として用いられてもよく、これにより
その溶解度が向上する。これらの化合物は、ナトリウム、カリウム、アンモニウ
ム、カルシウム又はマグネシウム塩とされるのが好ましい。
本発明の化合物の製法は、とりわけ、一般的にペプチド合成において用いられ
る方法(例えば、M.ボダンスキー(Bodansky)とA.ボダンスキー、
ザ・プラクティス・オブ・ペプチド・シンセシス(The Practice
of Peptide Synthesis)、シュプリンガー・フェルラーク
(Springer Verlag)、ベルリン、1984を参照)を利用する
。
従って、本発明の化合物の製法は、市販されているもの、例えば、N−ベンジ
ルオキシカルボニル−L−アスパラギン酸β−ベンジルエステル(n=1)、N
−ベンジルオキシカルボニル−L−グルタミン酸γ−ベンジルエステル(n=2
)、及び、Rが上述したような二置換のフェニル基であるアミンR−NH2等を
前駆体として、以下の反応スキームに従って行われる:
本発明の化合物は、再結晶やクロマトグラフィー等の標準的な技術を用いて精
製する。その構造及び純度は、通常用いる技術(薄膜クロマトグラフィー、高速
液体クロマトグラフィー、赤外分光光度法、核磁気共鳴、元素分析)により確認
した。
実施例中に示した化合物の甘味能は、八人の経験者が評価した。この評価は、
化合物を様々な濃度の水溶液にし、コントロールとして濃度が2%のショ糖溶液
と味覚を比較することにより行う。ショ糖との比較による試験化合物の甘味能は
、同程度の甘味を持つ時の(すなわち、多数の人が、試験化合物溶液とコントロ
ールのショ糖溶液の甘味を同程度であると考えた時)試験化合物とショ糖の重量
比に当たる。
以下に実施例を掲げて本発明の実施方法及び利点を更に詳しく説明するが、本
発明はこれら実施例にのみ限定されるものではない。実施例
N−(4−シアノフェニルカルバモイル)−L−アスパラギン酸−α−3,4
−ジメチルベンゼンアミドの調製:
1. N−ベンジルオキシカルボニル−L−アスパラギン酸−α−3,4−ジメ
チルベンゼンアミドβ−ベンジルエステルの調製:
−15℃に冷却した、3g(8.4mmol)のN−ベンジルオキシカルボニ
ル−L−アスパラギン酸β−ベンジルエステル(バーヘム(Bachem)社製
No.C−1350)の50cm3テトラヒドロフラン溶液に、0.84g(8
.4mmol)のN−メチルモルホリンと、1.14g(8.4mmol)のク
ロロギ酸イソブチルを連続して加えた。この温度で2分間攪拌した後、3,4−
ジメチルアニリン(アルドリッチ(Aldrich)社製No.12637−3
)を1g(8.4mmol)加えた。反応混合物を緩やかに温め、室温で2時間
攪拌した。沈殿したN−メチルモルホリン塩酸塩をろ過により除去し、20cm3
のテトラヒドロフランで洗浄した。この濾液を減圧下で濃縮・乾燥させ、得ら
れた残滓を50cm3のジエチルエーテル中で粉砕した。得られた白色の固体を
ろ過により分離し、さらに20cm3のジエチルエーテルで洗浄し、N−ベンジ
ルオキシカルボニル−L−アスパラギン酸−α−3,4−ジメチルベンゼンアミ
ドβ−ベンジルエステルを3.3g(85%収率)得た。その融点は146℃で
あった。
2. L−アスパラギン酸−α−3,4−ジメチルベンゼンアミドの調製:
上で得られたN−ベンジルオキシカルボニル−L−アスパラギン酸−α−3,
4−ジメチルベンゼンアミドβ−ベンジルエステル3g(5.6mmol)の1
00cm3メタノール溶液を、活性炭素に担持させた10%パラジウムの存在下
、大気圧下で18時間、水素化した。ろ過により触媒を除去し、次いで、溶液を
減圧下で濃縮した。得られた白色の固体を、アセトン中で粉砕しながら洗浄し、
1.3gのL−アスパラギン酸−α−3,4−ジメチルベンゼンアミド(85%
収率)を融点が204℃の白色固体として得た。この純度は、薄膜クロマトグラ
フィー(G60シリカゲル(メルク(Merck)社No.1.05554シリ
カ)、溶離液はブタノール−酢酸−水(8−2−2)、ニンヒドリンにより呈色
、Rf=0.49)を用いて調べた。
3. N−(4−シアノフェニルカルバモイル)−L−アスパラギン酸−α−3
,4−ジメチルベンゼンアミドの調製:
0.7g(3mmol)のL−アスパラギン酸−α−3,4−ジメチルベンゼ
ンアミドと、0.63gの(6mmol)の炭酸ナトリウムの30cm3水溶液
に、0.47g(3.2mmol)の4−シアノフェニルイソシアネートの30
cm3ベンゼン溶液を、激しく攪拌しながら加えた。この溶液を室温で30分間
攪拌したのち、30cm3のジエチルエーテルで3度洗浄した。6N塩酸溶液で
pH2〜3まで酸性化すると、白色の沈殿物が生じた。これをろ過し5〜6cm3
の水で洗浄した。ろ過・乾燥させて、N−(4−シアノフェニルカルバモイル
)−L−アスパラギン酸−α−3,4−ジメチルベンゼンアミドを0.98g(
86%収率)得た。95%エタノールから再結晶したものでは、融点は172℃
であった。
