Claims (1)
- 【特許請求の範囲】 1.GalNAc-トランスフェラーゼ活性検出用のシンチレーション近接アッセ イ(SPA)。 2.取り込み放射能定量用のシンチレーション近接アッセイ(SPA)ビーズ を含む請求項1記載のアッセイ。 3.酵素、GalNAc-トランスフェラーゼとの反応の結果産生された適当なリ ガンドと相互作用する適当なレセプターを含む請求項2記載のアッセイ。 4.レクチン被覆SPAビーズである適当なレセプターを含み、リガンドがコ ンジュゲートしていない受容体ペプチドである請求項3記載のアッセイ。 5.サルビア・スクラレア(Salvia Sclarea)、ヘリックス・ポマティア(He lix Pomatia)およびビシア・ビローサ(Vicia Villosa)よりなる群から採取さ れるレクチンを含む請求項4記載のアッセイ。 6.抗体被覆SPAビーズである適当なレセプターを含み、リガンドがコンジ ュゲーテッドしていない受容体ペプチドである請求項3記載のアッセイ。 7.Tn抗原を認識する抗体を含む請求項6記載のアッセイ。 8.抗体被覆SPAビーズである適当なレセプターを含み、リガンドが抗原性 ペプチド配列に融合した修飾受容体ペプチドである請求項3記載のアッセイ。 9.MYC(例えば、AEEQKLISEEDLLRKRREQLKHKLE QLRNSC)またはFLAG(DYKDDDK)の一部分のいずれかである抗原 性ペプチド配列を含む請求項8記載のアッセイ。 10.アビジンまたはストレプトアビジン被覆SPAビーズである適当なレセ プターを含み、リガンドがビオチンコンジュゲーテッド受容体ペプチドである請 求項3記載のアッセイ。 11.3アミノ酸のうちの1が受容体ペプチド、すなわち、SerまたはThrで ある少なくとも3アミノ酸のペプチド配列を有するビオチンコンジュゲーテッド 受容体ペプチドを含む請求項10記載のアッセイ。 12.ビオチンコンジュゲーテッド受容体ペプチドを含み、ペプチド配列が、 以下:R-1-R0-R1-R2-R3-R4-R5(またはPO)-R6-R7-R8-R9-R10-R11[ 式中、R5(またはPO)は受容体アミノ酸で、SerまたはThr; R4およびR6は、独立して、Asp、Asn、Glu、Gln、Ser、Gly、His、A rg、Thr、Ala、Pro、Tyr、Val、Met、Ile、Leu、Phe、Lys、Cysまた はTrpで; R-1-R3およびR7-R11は、独立して、アミノ酸を含まないか、または以下の アミノ酸:Asp、Asn、Glu、Gln、Ser、Gly、His、Arg、Thr、Ala、P ro、Tyr、Val、Met、Ile、Leu、Phe、Lys、CysまたはTrpのいずれか] で示されるアミノ酸R-1-R11を有すると記載し得る請求項11記載のアッセイ 。 13.ビオチンコンジュゲーテッド受容体ペプチドを含み、ペプチド配列が、 以下:R1-R2-R3-R4-R5(またはPO)-R6-R7-R8-R9 [式中、R5(またはPO)は受容体アミノ酸で、SerまたはThr; R4およびR6は、独立して、Asp、Asn、Glu、Gln、Ser、Gly、His、A rg、Thr、Ala、Pro、Tyr、Val、Met、Ile、Leu、Phe、Lys、Cysまた はTrp; R1-R3およびR7-R9は、独立して、アミノ酸を含まないか、または以下のア ミノ酸:Asp、Asn、Glu、Gln、Ser、Gly、His、Arg、Thr、Ala、Pro 、Tyr、Val、Met、Ile、Leu、Phe、Lys、CysまたはTrpのいずれか] で示されるアミノ酸R1-R9を有すると記載し得る請求項12記載のアッセイ。 14.ペプチド配列がチャートIに示すいずれかの配列である請求項13載の アッセイ。 15.受容体ペプチド中のアミノ酸R1ないしR9が、以下のアミノ酸:Asp、 Asn、Glu、Gln、Ser、Gly、His、Arg、Thr、Ala、Pro、Tyr、Val、 Met、Ile、Leu、Phe、Lys、CysまたはTrpのいずれかである請求項14記 載のアッセイ。 16.アミノ酸R1ないしR9が、以下のアミノ酸:Ser、Thr、Pro、Alaま たはGlyのいずれかである請求項14記載のアッセイ。 17.ペプチド配列が、以下:RTPPP、RSPPP、PPASTSAPG またはPPASSSAPGのいずれかである請求項12記載のアッセイ。 18.ペプチド配列がPPASTSAPGである請求項16記載のアッセイ。 19.ペプチド配列中に9のアミノ酸が存在する請求項13記載のアッセイ。 20.アミノ酸がSer、Thr、Pro、AlaまたはGlyよりなる請求項19記載 のアッセイ。 21.ペプチド配列がPPASTSAPGまたはPPASSSAPGである請 求項20記載のアッセイ。
