5−HT4アゴニストとしての置換
ジヒドロベンゾフラン誘導体
本発明は、胃腸障害およびCNS障害の治療に5−HT4リセプタアゴニスト
として作用する置換ジヒドロベンゾフラン誘導体の使用、5−HT4リセプタア
ゴニスト活性を有する或る種の新規な化合物、その製造方法、およびそれらを含
有する医薬組成物に関するものである。
非古典的な5−ヒドロキシトリプタミン リセプタが5−HT4リセプタとし
て考えられている[トレンズ・ファーマコロジカル・サイエンス(1992)、
第13巻、第141〜5頁]。
胃腸運動の刺激剤として永年にわたり臨床的に使用されていた置換ベンズアミ
ド メトクロプラミドのプロキネチック(prokinetic)作用は、5−
HT4リセプタに対するそのアゴニスト効果に基づくと思われる[ドラグ・デザ
イン・アンド・デリベリー(1988)、第3巻、第273〜295頁]。
幾種かの5−HT4リセプタアゴニストは記憶機能障害に関
する適切な動物挙動試験にて活性であった[ゲラルジニ等、19thC.I.N.
P.コングレス、ワシントン、1994年6月;ゲーラルジニ等、第10回ヨー
ロピアン・ソサエティ・フォー・ニューロケミストリー、エルサレム、1994
年8月]。
国際特許出願WO 93/16072号はベンゾピラン、ベンゾチオピランも
しくはベンゾフラン核から誘導された5−HT4リセプタアンタゴニストを記載
している。
国際特許出願WO 94/08995号は、5−HT4アンタゴニスト活性を
有するベンゾフランもしくはジベンゾフランの新規なカルボキシレートもしくは
カルボキサミドに関するものである。 本発明者等は、たとえば5−[HT4ア
ンタゴニスト活性を有するWO 93/16072号およびWO 94/089
95号に開示された化合物のような最も近縁の従来化合物と類似した構造を有す
るにも拘らず、5−HT4リセプタアゴニスト特性を有する種類の置換ジヒドロ
ベンゾフランカルボン酸誘導体を同定した。
本発明の化合物は、5−HT4アゴニスト活性により5−HT4リセプタの刺激
が必要とされる全ゆる病変に有用であり、
したがって本発明の化合物はたとえば消化不良、胃食道逆流病(GORD)およ
び軽症胃アトニーのような胃腸障害の治療および/または学習や記憶機能障害を
特徴とするCNS障害の治療にも治療プロキネチック剤として有用である。
したがって本発明は、式(I)
[式中、R1、R2およびR3はそれぞれ独立して水素、C1〜C6アルキル、ハロ
ゲン、ヒドロキシ、C1〜C4アルコキシ、アミノ、C1〜C4アルキルアミノもし
くはC1〜C4ジ−アルキルアミノであり;
XはO、NHもしくはCH2であり;
Zは基(a)、(b)、(c)もしくは(d)
であり、ここで
nは1、2、3もしくは4であり;
mは0もしくは1であり;
qは0、1もしくは2であり;
R4は水素、C1〜C6アルキル、ベンジル、シクロヘキシルメチルもしくは−C
H2−CH2−SO2NH−R6であり、ここでR6はC1〜C6アルキルもしくはベ
ンジルであり;
R5はC1〜C6アルキルであり;
Tはハロゲンであり;
ただしZが基(c)のとき、XはOもしくはCH2である]
の、5−HT4アゴニストとして使用するためのジヒドロベンゾフラン誘導体ま
たはその医薬上許容しうる塩に関するものである。したがって式(I)の化合物
は5−HT4リセプタの刺激が必要とされる全ゆる病変の治療に有用である。例
として、式(I)の化合物はたとえば消化不良、胃食道逆流病(GORD)もし
くは軽症胃アトニーのような胃腸障害の治療に治療プロキネチック剤として有用
である。さらに式(I)の化合物は、その5−HT4アゴニスト特性のため、学
習および/または記憶機能障害を特徴とするCNS障害の治療にも認識活性化剤
として有用である。
アルキル基、アルコキシ基およびアルキルアミノ基は側鎖もしくは直鎖とする
ことができる。
C1〜C6アルキル基の代表例はメチル、エチル、n−およびイソ−プロピル、
n−、イソ−、sec−およびt−ブチルを包含する。
C1〜C4アルコキシ基の代表例はメトキシおよびエトキシを包含する。
C1〜C4アルキルアミノ基は特にメチルアミノもしくはエチルアミノである。
C1〜C4ジ−アルキルアミノ基は特にジメチルアミノもしくはジエチルアミノ
である。
ハロゲンは弗素、臭素、塩素もしくは沃素、特に塩素もしくは臭素を包含する
。
式(I)の化合物の医薬上許容しうる塩は無機酸(たとえば塩酸、臭化水素酸
、硫酸、燐酸)または有機酸(たとえば酢酸、プロピオン酸、乳酸、シュウ酸、
リンゴ酸、マレイン酸、酒石酸、クエン酸、安息香酸、マンデリン酸、サリチル
酸およびフマル酸)との酸付加塩を包含する。
Zが基(a)もしくは(b)である式(I)の化合物の医薬上許容しうる塩の
例は第四級誘導体、たとえば式Rx−W[式中、RxはC1〜C6アルキルもしくは
フェニル−C1〜C6アルキルであり、Wは酸の陰イオンに対応する基である]の
化合物により四級化された化合物を包含する。
好ましくはRxはC1〜C4アルキルもしくはフェニル−C1〜C4アルキルであ
り、特にメチル、エチル、n−プロピル、n−ブチル、ベンジルもしくはフェニ
ルエチルである。
好ましくはWはたとえば塩素、臭素もしくは沃素のようなハロゲンである。
Zが基(a)、(b)もしくは(c)である式(I)の化合物の医薬上許容し
うる塩の例はたとえばN−オキサイドのような内部塩をも包含する。式(I)の
化合物、その医薬上許容しうる塩(第四級誘導体およびN−オキサイドを包含す
る)はたとえば水和物のような医薬上許容しうる溶媒和物をも形成することがで
き、これらも本発明に包含される。
Zが基(c)である式(I)の化合物は不整炭素原子を有し、従って光学異性
体の混合物(ラセミ混合物)として或いは単一の光学異性体(エナンチオマー)
として存在することができる。これらエナンチエマーは光学上純粋な出発物質か
ら別々に合成することができ、或いはラセミ混合物から常法で分離することもで
きる。
さらに本発明はその範囲内に式(I)の化合物の代謝物および医薬上許容しう
るバイオプレカーサー(プロドラッグとしても知られる)の両者を包含する。
さらに本発明は好適種類の式(I)の化合物を新規な化合物と称する。
本発明の他の主題を構成するこれら化合物は、式(I)
[式中、R1は水素であり;

本発明による好適化合物の例は次の通りである:
N−[1−ブチル−1−アザビシクロ[2,2,2]オクト−3−イル]−4−
アミノ−5−クロロ−2,3−ジヒドロベンゾ[b]フラン−7−カルボキサミ
ド ブロマイド;

の化合物を式R5T[式中、R5はC1〜C6アルキルであり;Tはハロゲンである
]のハロゲン化アルキルと反応させて、式(I)[式中、R1、R2およびR3は
上記の意味を有し、XはO、NHもしくはCH2であり、Zは基(d)である]
の化合物を得;
さらに所望ならば式(I)の化合物が不整炭素原子を有する場合は
(D)式(I)の化合物のラセミ混合物を単一異性体に分割し;および/または
所望ならば
(E)式(I)の化合物をその医薬上許容しうる塩に変換することからなる方法
により得ることができる。
工程(A)における、式(II)の化合物と式(III)、(IV)もしくは(V)
の化合物との反応は類似方法であって当業界で周知の方法により行うことができ
る。たとえば式(II)のハロゲン化アシルをたとえばジクロロメタン、テトラヒ
ドロフランもしくはアセトニトリルのような適する有機溶剤中で約0℃から混合
物のほぼ還流温度に至る範囲の温度にてたとえばト
リエチルアミン、炭酸水素ナトリウムもしくは炭酸カリウムのようなプロトン掃
去剤の存在下に式(III)、(IV)もしくは(V)のアルコールもしくはアミン
と反応させることができる。
工程(B)における式、(VI)の化合物と式(VII)、(VIII)もしくは(IX
)の化合物との反応も当業界で周知の方法により行うことができる。たとえば式
(VI)のハロゲン化アシルをたとえばテトラヒドロフランもしくはジエチルエー
テルのような適する有機溶剤中でたとえばFe(アセチルアセトネート)3もし
くはCuIの存在下に約−78℃〜約30℃の範囲の温度にて式(VII)、(VII
I)もしくは(IX)のグリニヤール試薬と反応させることができる。
工程(C)における、式(X)の化合物とハロゲン化アルキルR5Tとの反応
は標準的方法により行うことができる。たとえば式(X)の化合物をたとえばメ
タノールもしくはエタノールのような適する有機溶剤の存在下に約30℃から混
合物のほぼ還流温度に至る範囲の温度にて上記化合物R5Tと反応させることが
できる。EおよびYがOHである式(II)および(VI)のカルボン酸は市販され
ているか或いは公知化合物である。
酸誘導体(II)および(VI)は公知化合物(EP0 234
872 A1号、アドリア・ラボラトリース社)であるか、或いは対応の酸か
ら当業界で周知の方法により作成することもできる。
式(III)、(IV)および(V)のアルコールおよびアミンは市販されている
か、或いは公知化合物である。
式(VII)、(VIII)および(IX)の有機金属誘導体は、市販されているか或
いは対応する式(III)、(IV)および(V)のアルコールから容易に作成しう
る対応のハロゲン化アルキルから標準方法によって作成することができる。Xが
NHである式(X)の化合物は公知化合物である(EP 0 234872 A
1号、アドリア・ラボラトリース社)。式R5Tのハロゲン化アルキルは市販さ
れている製品である。
本発明による化合物の異性体混合物から単一異性体への分離および式(I)の
化合物からその医薬上許容しうる塩への変換は当業界で周知された方法により行
うことができる。
上記したように、本発明の化合物は5−HT4リセプタに対する5−HT(セ
ロトニン)の強力なアゴニストであり、したがって5−HT4リセプタの刺激が
必要とされる病変の治療に使用することができる。特に5−HT4アゴニストは
胃腸運
動の刺激剤であることが知られているので、本発明の化合物はたとえば消化不良
、胃食道逆流病(GORD)もしくは軽症胃アトニーのような胃腸病の治療に治
療プロキネチック剤として使用することができる。さらに5−HT4リセプタは
シナプス可塑性現象および記憶過程に関与すると考えられ[CNS Drugs
(1994)、第1巻、第6〜15頁]、さらに5−HT4リセプタ刺激はイン
ビボでラット前頭皮質におけるアセチルコリン放出を促進することが示されてい
る[Neuro Report(1994)、第5巻、第1230〜2頁]という事実に鑑み
、本発明による化合物の他の用途は学習および記憶機能障害を特徴とするCNS
障害の治療における認識活性化剤とすることもできる。
本発明による化合物の5−HT4リセプタ親和性は、グロスマン等の方法[ブ
リティッシュ・ジャーナル・ファーマコロジー(1993)、第109巻、第6
18〜624頁]にしたがいラット線条体にて5−HT4リセプタ放射性リガン
ド[3H]−GR−113808の結合の抑制により判定された。
5−HT4アゴニストとしての本発明による化合物の活性は、バクスター等の
方法[ノインヒン・シュミーデベルグ・アーカ
イブ・ファーマコロジー(1991)、第343巻、第439〜446頁]にし
たがいラットのカルバコール予備収縮食道粘膜筋のリセプタ媒介弛緩応答により
インビトロにて評価された。
例として本発明による化合物の代表群、すなわち(S)−(+)−N−[1−
ブチル−1−アザビシクロ[2,2,2]オクト−3−イル]−4−アミノ−5
−クロロ−2,3−ジヒドロベンゾ[b]フラン−7−カルボキサミド ブロマ
イド−水塩(内部コードFCE 28773A);

を上記の方法にしたがって試験し、得られた結果を第1表に示す。

IC50=抑制剤の不存在下に得られる結合放射性リガンド濃度の50%を変位せ
しめる試験化合物の濃度;

Kd=放射性リガンド−リセプタ複合体の解離定数;
EC50=有効濃度:最大反応の50%(この場合は最大弛緩の50%)を誘発さ
せる試験化合物の濃度;
i.a.=固有活性:最大反応/天然アゴニストの最大反応(この場合は最大弛緩
/5−HT最大弛緩)。
表に示した結果は明かに、本発明の化合物が5−HT4リセプタ部位に対する
高い有効性を示すと同時に5−HT4リセプタ活性の促進に特に効果的であるこ
とを示す。
本発明の化合物は各種の投与剤形として、たとえば錠剤、カプセル、糖衣錠も
しくはフィルムコーティング錠、液状溶液もしくは懸濁液として経口的に;座薬
の形態で直腸内に;非経口的(たとえば筋肉内)に或いは静脈注射もしくは灌流
により投与することができる。
投与量は患者の年令、体重、状態および投与経路に依存する。たとえば成人に
経口投与すべく採用される投与量は、たとえば
本発明の代表的化合物FCE 29034Aの場合1回の投与当たり約1〜約5
00mgにて毎日1〜5回の範囲とすることができる。
本発明は、活性成分としての本発明の化合物を医薬上許容しうる賦形薬(キャ
リヤもしくは希釈剤とすることができる)と組合せてなる医薬組成物を包含する
。
本発明の化合物を含有する医薬組成物は一般に常法にしたがって作成され、医
薬上適する形態で投与される。
たとえば固体経口剤形は活性化合物と一緒に希釈剤、たとえば乳糖、デキスト
ロース、蔗糖、セルロース、コーンスターチもしくは馬鈴薯澱粉;滑剤、たとえ
ばシリカ、タルク、ステアリン酸、ステアリン酸マグネシウムもしくはカルシウ
ムおよび/またはポリエチレングリコール;結合剤、たとえば澱粉、アラビアゴ
ム、ゼラチン、メチルセルロース、カルボキシメチルセルロースもしくはポリビ
ニルピロリドン;崩壊剤、たとえば澱粉、アルギン酸、アルギネートもしくはグ
リコール酸ナトリウム澱粉;起泡性混合物;着色料;甘味料;湿潤剤、たとえば
レシチン、ポリソルベート、ラウリルサルフェート;並びに一般に医薬処方物で
使用される無毒性かつ薬理上不活性な物質
を含有することができる。前記医薬製剤は公知方法でたとえば混合、粒状化、錠
剤化、糖衣またはフィルムコーティングの各方法によって製造することができる
。
経口投与のための液体分散物はたとえばシロップ、乳液および懸濁液とするこ
とができる。
シロップはキャリヤとしてたとえば蔗糖または蔗糖とグリセリンおよび/また
はマニトールおよび/またはソルビトールとを含有することができる。
懸濁液および乳液はキャリヤとしてたとえば天然ガム、寒天、アルギン酸ナト
リウム、ペクチン、メチルセルロース、カルボキシメチルセルロースもしくはポ
リビニルアルコールを含有することができる。
筋肉内注射のための懸濁液もしくは溶液は活性化合物と一緒に医薬上許容しう
るキャリヤ、たとえば滅菌水、オリーブ油、オレイン酸エチル、グリコール(た
とえばプロピレングリコール)および所望に応じ適量のリドカイン塩酸塩を含有
することができる。静脈内注射もしくは灌流のための溶液はキャリヤとしてたと
えば滅菌水を含有することができ、好ましくは無菌の等張性塩水溶液の形態とす
ることができる。
座薬は活性化合物と一緒に医薬上許容しうるキャリヤ、たとえばココア脂、ポ
リエチレングリコール、ポリオキシエチレンソルビタン脂肪酸エステル表面活性
剤またはレシチンを含有することができる。
以下、限定はしないが実施例により本発明をさらに説明する。実施例1
10mLの無水テトラヒドロフラン中の4−アミノ−5−クロロ−2,3−ジ
ヒドロベンゾ[b]フラン−7−カルボン酸(0.700g、3.28ミリモル
)とカルボニルジイミダゾール(0.580g、3.60ミリモル)との混合物
を40℃にて1.5時間にわたり加熱した。その後、5mLのテトラヒドロフラ
ン中の(1−ピペリジル)エチルアミン(0.427mL、4.92ミリモル)
を添加し、混合物をさらに2時間加熱し、次いで22℃にて16時間にわたり撹
拌した。揮発物を減圧下で蒸発させ、残留物を水と酢酸エチルとに取り、層を分
離させ、有機層を無水硫酸ナトリウムで脱水し、次いで蒸発させて1.0gの粗
製物質を得、これをシリカゲル上でのカラムクロマトグラフィー(溶出剤クロロ
ホルム/メチルアルコール/30%アンモニア溶液、46:4:0.1)に
より部分精製した。アセトン/イソプロピルアルコール中の遊離塩基にイソプロ
ピルアルコール中の1当量の塩酸を添加してカルボキサミドをその塩酸塩として
単離した。沈澱した固体を濾過し、ジエチルエーテルで洗浄し、乾燥させて74
6mg(60%)のN−[(1−ピペリジル)エチル]−4−アミノ−5−クロ
ロ−2,3−ジヒドロベンゾ[b]フラン−7−カルボキサミド塩酸塩水和物を
無色固体(m.p.=237−239℃)として得た。
同様に次の化合物を製造した:
N−[(1−ピペリジル)プロピル]−4−アミノ−5−クロロ−2,3−ジヒ
ドロベンゾ[b]フラン−7−カルボキサミド塩酸塩水和物(m.p.=192
−194℃);および
N−[(1−ブチル−ピペリド−4−イル)メチル]−4−アミノ−5−クロロ
−2,3−ジヒドロベンゾ[b]フラン−7−カルボキサミド塩酸塩(m.p.
=260−262℃)。実施例2
15mLの無水テトラヒドロフラン中の4−アミノ−5−クロロ−2,3−ジ
ヒドロベンゾ[b]フラン−7−カルボン酸(1.61g、7.56ミリモル)
とカルボニルジイミダゾー
ル(1.35g、8.32ミリモル)との混合物を40℃にて1.5時間加熱し
た。その後、10mLのテトラヒドロフラン中のN−(1−ブチル−ピペリド−
4−イル)メチルアルコール(2.59g、15.1ミリモル)と1,8−ジア
ザビシクロ[5,4,0]ウンデク−7−エン(DBU)(1.13mL、7.
56ミリモル)との溶液を添加し、この混合物をさらに2.5時間にわたり加熱
した。揮発物を減圧下で蒸発させ、残留物を水とジエチルエーテルとに取った。
層を分離させ、有機層を無水硫酸ナトリウムで脱水し、次いで蒸発させて3.5
gの粗製物質を得、これをシリカゲル上でのカラムクロマトグラフィー(溶出剤
クロロホルム/メチルアルコール、46:4)により精製して1.5g(54%
)の(1−ブチル−ピペリド−4−イル)メチル−4−アミノ−5−クロロ−2
,3−ジヒドロベンゾ[b]フラン−7−カルボキシレート(m.p.=124
−126℃)を無色固体として得た。
同様に次の化合物を製造した:
(1−ピペリジル)エチル−4−アミノ−5−クロロ−2,3−ジヒドロベンゾ
[b]フラン−7−カルボキシレート(m.p.=152−154℃);
(1−ピペリジル)プロピル−4−アミノ−5−クロロ−2,3−ジヒドロベン
ゾ[b]フラン−7−カルボキシレート;および
(S)−(+)−(1−アザビシクロ[2,2,2]オクト−3−イル)−4−
アミノ−5−クロロ−2,3−ジヒドロベンゾ[b]フラン−7−カルボキシレ
ート。実施例3
イソプロピルアルコール中の1当量の塩酸を、アセトン中の(1−ピペリジル
