鋳型指向性環化
発明の分野
本発明は、環状ペプチド、固体物に結合した環状ペプチド、および環状ペプチ
ドライブラリーの調製方法に関する。
背景
ペプチドは、2つのチオール含有残基を必要とするジスルフィド結合形成、2
つの側鎖官能基の間のアミドまたはエステル結合形成、1つの側鎖官能基および
骨格αアミノまたはカルボキシ基の間のアミドまたはエステル結合形成、または
骨格αアミノまたはカルボキシ基の間のアミド結合形成といった多くの方法によ
って環状構造に変換できる(環状ホモデティックペプチド)。伝統的に、これら
の様々な環状化反応は高度に希釈された溶液中で行われた。例えば、ブラディ(
Brady)ら、ジェイ・オーグ・ケム(J.Org.Chem.)、1979年、第44巻、
3101−3105頁、およびマクマレー(McMurray)ら、テット・レット(Te
t.Lett.)、1993年、第34巻、8059−8062頁、参照。新規樹脂お
よび直交保護基法の出現とともに、ペプチドを固体支持体に付着させたまま、こ
れらの環化のうちいくつかを行うことができるようになった。ほとんどの固相ペ
プチド合成法はカルボキシ末端の固体支持体への付着を必要とするので、環化が
側鎖官能基および/またはαアミノ基を必要とする場合、これはもっとも適して
いる。例えば、側鎖アミド結合はアミン含有およびカルボキシル含有側鎖の間で
形成され、一方、ペプチドは樹脂に付着したままである。かくして、シラー(Sc

ている。同様に、ジスルフィド結合を介した環化ペプチドについて、プロックス
(Ploux)ら、イント・ペプチド・プロテイン・レス(Int.Peptide Protein Res.)
、1987年、第29巻、162−169頁、および、アルベリシオ(Alberici
o)ら、イント・ペプチド・プロテイン・レス(Int.Peptide Protein Res.)、
1991年、第37巻、402−413頁は、ペプチドが樹脂に付着したままの
ペプチドの環化に関する方法を記載している。
環状ホモデティックペプチドの固相合成は、環化反応に遊離アミノおよびカル
ボキシル末端の両方を必要とするので、より多くの試みが必要である。これに対
する一つのアプローチは、側鎖官能基を介したC末端アミノ酸の樹脂への付着、
および直交カルボキシル保護基を用いた脱保護ならびにアミノおよびカルボキシ
ル末端の環化であった。このアプローチは、マクマレー・ジェイ(McMurray,J.)
、テット・レット(Tet.Lett.)、1991年、第32巻、7679−7682
頁、トレチャック(Trzeciak)ら、テット・レット(Tet.Lett.)、1992年
、第33巻、4557−4560頁、およびトロメリン(Tromelin)ら、テット
・レット(Tet.Lett.)、1992年、第33巻、5197−5200頁により
記載されている。これは元来、側鎖が樹脂に付着できるアミノ酸のみがC末端に
位置できるので、調製できるペプチドの性質を制限する。
環状ホモデティックペプチド調製へのもう一つのアプローチは、オキシム樹脂
上での調製を包含し、それは、環化に付随して樹脂からペプチドを開裂させるた
めのαアミノ基によるペプチド−樹脂結合の分子内アミノ分解を可能にする。こ
れは、エサペイ(Osapay)ら、テット・レット(Tet.Lett.)、1990年、第
43巻、6121−6124頁、およびヒー(He)ら、ジェイ・アメリ・ケム・
ソサ(J.Amer.Chem.Soc.)、1993年、第115巻、8066−8072
頁によって記載されている。この方法は、調製された配列においてより大きな融
通性を可能にするが、低収量により妨害される可能性がある。そうでなければ、
この方法では、側鎖が保護された環状中間体を樹脂から除去し、溶液脱保護工程
を必要とし、樹脂結合環状ホモデティックペプチドの合成は不可能である。
ペプチドを環化する新しい方法、詳細には、樹脂上でペプチドを環化する新し
い方法が必要である。本発明は、樹脂上でのホモデティックペプチドの合成およ
び環化のための簡便で多目的に使用できる方法を提供する。さらに、本明細書中
に記載するごとく、ペプチド中に環化を指向または促進するアミノ酸残基を取り
入れることによって、環状ペプチドの収率が改善されるかもしれない。
発明の概要
本発明の目的は、環状ホモデティックペプチドの新規合成方法を提供すること
である。本発明のもう一つの目的は、樹脂結合環状ホモデティックペプチドの新
規合成方法を提供することである。さらにもう一つの本発明の目的は、生物活性
分子のスクリーニングに使用される環状ペプチドライブラリーを製造することで
ある。
本発明の特徴は、ペプチドおよび樹脂の間に選択的で分裂可能な結合を提供す
る最初の架橋基、および側鎖基を介したペプチドの樹脂への付着手段を提供する
2番目の架橋基との組み合わせである。本発明のさらにもう一つの特徴は、ペプ
チド中の、環化反応を促進するアミノ酸配列およびアミノ酸残基の使用である。
一つの具体例において、本発明の好ましい特徴は、2番目の架橋基と環化を促進
する鋳型残基との組み合わせである。
詳細な説明
本発明者らは、環状ホモデティックペプチドおよびペプチドライブラリーの調
製のための2つの新しいアプローチを発展させ、組み合わせた。かくして、本発
明者らは鋳型に基づく環化を固相合成に適用し、固体支持体上での環状ホモデテ
ィックペプチドの完全な合成および脱保護を可能にした。この方法の変法は、側
鎖脱保護環状ペプチドの樹脂からの遊離を可能にする。これらは、遊離または樹
脂結合環状ホモデティックペプチドのいずれかの合成、および環状ホモデティッ
ク
ペプチドライブラリー作成を可能にする。
一つの態様において、本発明は、構造:

Pg2は、所望により、ペプチドのアミノ末端の保護基であってもよく;
S1は、所望により、固体支持体に付着する保護官能基であってもよく、所望
により、リンカーL1を介してもよいが、ペプチドまたはL2に付着せず、
および、S1は共有結合を形成してS2と反応でき;
S2は、所望により、共有結合を形成してS1と反応できる、L2上の保護官
能基であってもよい]
で示される直鎖状樹脂結合ペプチドを調製すること;
2)必要ならば、S1およびS2からいずれかの保護基を除去し、および、式(
II):
で示される化合物を形成するために、S1およびS2の間に共有結合を形成する
こと;
させること、および必要ならば、遊離アミノ末端を提供するためにPg2を除去
し、式(III):
で示される化合物を形成すること;
4)アミノ末端およびカルボキシ末端を環化して、式(IV):
で示される化合物を形成すること;および
5)保護基Pg1を除去して、式(V):
で示される化合物を形成することからなる。
この方法は、上記工程を行い、次いで、樹脂からペプチドを開裂させることに
らのペプチドの開裂および側鎖保護基Pg1の除去は、所望の遊離環状ペプチド
を得るために行うことができる。そのペプチドは、リンカーL2およびそれが付
着しているアミノ酸の間の結合を開裂させることにより樹脂から除去される。
簡便さゆえに、本明細書中においていくつかの略号が使用される。かくして、
Alocはアリルオキシカルボニルを意味し、β−Alaはβ−アラニン(3−
アミノプロパン酸)を意味し、BHAはベンツヒドリルアミン−ポリスチレン樹
脂を意味し、Bocはt−ブチルオキシカルボニルを意味し、Bnはベンジルを
意味し、BOPはヘキサフルオロリン酸ベンゾトリアゾール−1−イルオキシ−
トリス(ジメチルアミノ)ホスホニウムを意味し、DBUは1.8ジアゾビシク
ロ[5.4.0]ウンデク−7−エンであり、DPPAはジフェニルホスホリルア
ジドであり、DCCはジシクロヘキシルカルボジイミドであり、DMAPはジメ
チルアミノピリジンであり、EDCはN−エチル−N’(ジメチルアミノプロピ
ル)−カルボジイミドであり、DMFはジメチルホルムアミドを意味し、Fmo
cは9−フルオレニルメチルオキシカルボニルを意味し、HBTUは(ヘキサフ
ルオロリン酸1−ヒドロキシベンツトリアゾリルテトラメチル−ウロニウム)を
言い、HMBAは4−(4−ヒドロキシメチル−3−メトキシ−フェニルオキシ
)酪酸を意味し、HOBTは1−ヒドロキシ−ベンゾトリアゾールを意味し、S
ucはサクシニルを意味し、Hypはヒドロキシプロリンを意味し、Zはベンジ
ルオキシカルボニルを意味する。一般に、アミノ酸の略号は、ユー・ジェイ・バ
イオケム(Eur.J.Biochem.)、第158巻、9頁、(1984年)において記
載されたごとく、IUPAC−IUBジョイント・コミッション・オン・バイオ
ケミカル・ノメンクリーチャー(Joint Commission On Biochemical Nomenclatu
re)に従う。
本明細書において使用される場合、アミノ酸はアミノ末端およびカルボキシ末
端を、好ましくは炭素骨格上に1,2−、1,3−、または1,4−置換パターン
で有する化合物を包含する。αアミノ酸がもっとも好ましく、タンパク質中に見
いだされる20の天然アミノ酸(グリシンを除くL−アミノ酸である)、対応す
るD−アミノ酸、タンパク質中に見いだされない、生合成的に入手できるアミノ
酸(例えば、4−ヒドロキシ−プロリン、5−ヒドロキシ−リジン、シトルリン
、オルニチン、カナバニン、デンコリン酸、β−シアノアラニン)、および、ア
ミノ−イソ酪酸、ノルロイシン、ノルバリン、Cαアルキル化およびNαアルキ
ル化アミノ酸、Dtc、Tpr、ホモシステインおよびホモセリンのごとき合成
によるαアミノ酸を包含する。βアラニンおよびγアミノ酪酸は1,3−および
1,4−アミノ酸の例であり、その他多数が当該分野においてよく知られている
。スタチン様同等物(CONH結合がCHOHによって置換されている、2つの
アミノ酸からなるジペプチド)、ヒドロキシエチレン同等物(CONH結合がC
HOHCH2によって置換されている、2つのアミノ酸からなるジペプチド)、

の有用な残基である。
本明細書において使用される場合、ペプチドは、アミド結合によって連結され
たアミノ酸の配列を示す。本発明のペプチドは、3ないし10アミノ酸残基、好
ましくは5ないし7残基のアミノ酸配列からなり、それぞれの残基はアミノおよ
びカルボキシ末端を有することによって特徴付けされる。
本明細書において使用される場合、アミノ酸配列を囲んだ四角括弧の印は、環
状ペプチドを示し、そのアミノ末端はカルボキシ末端に連結されており、例えば
、αアミノ酸の場合は最初の残基のαアミノ基が最後の残基のカルボキシ基に連
結されている。
ピン、または固相ペプチド合成に適した他のかかる形態であってもよい。かかる
樹脂の例は、ベンツヒドリルアミンポリスチレン、アミノメチルポリスチレン、
ポリエチレングリコールーポリスチレン等である。かかる固体支持体は一般に市
販されている。かかる樹脂の典型は、テンタゲルR(TentaGelR)、PEG−PS
、メリフィールド(Merrifield)およびシェパード(Sheppard)樹脂である。ポ
リスチレン基盤樹脂は特に有用である。
本明細書において使用される場合、リンカーという語は、1原子または分子断
片(原子の集合)を共有結合によりもう一つの原子または分子断片に結合させる
いかなる分子断片も意味する。典型的に、リンカーは、断片中において、エーテ
ル、スルフィド、ジスルフィド、エステルまたはアミドのごとき介在機能部位で
所望により置換されたアルキレンまたはアリルアルキル基からなるであろう。か
かる機能部位は普通、リンカーを創造(または2つの分子断片を結合させる)、
または既存のリンカーを破壊する(および分子断片に分離する)ために使用され
るであろう。
ペプチド結合を形成するためのカップリング法は一般に当該分野においてよく
知られている。一般に、ボダンスキー(Bodansky)らによるザ・プラクティス・
オブ・ペプチド・シンセシス(The Practice of Peptide Synthesis)、スプリ
ンガー−バーラグ(Springer-Verlag)、ベルリン(Berlin)、1984年で発
表されているペプチド合成の方法は技術を説明しており、本明細書に参照により
取り込まれる。典型的なカップリング法はカルボジイミド、活性化無水物および
エステルおよびハロゲン化アシルを使用する。EDC、DCC、DPPA、PP
A、BOP試薬、HOBt、N−ヒドロキシサクシンイミド、および塩化オキザ
リルのごとき試薬は典型的である。アリ(Ali)ら、ジェイ・メド・ケム(J.Me

J.Med.Chem.)、第30巻、2291頁(1987年)。アミノ酸の側鎖の反
応性官能基、例えば、不適切に反応を起こすかまたは合成の間妨害するような、
ヒドロキシル、メルカプタン、カルボン酸、アミノ、グアニジノ、イミダゾール
、またはインドール基のごとき官能基は、一般に合成の間保護される。保護基は
、かかる官能基の反応性を覆い隠すような基であるが、合成後期のある時点で除
去され元来の官能基を回復させるものである。反応性官能基のための適切な保護
基およびこれらの官能基を脱保護するための試薬は、グリーン(Greene)らによ
るプロテクティブ・グループス・イン・オーガニック・シンセシス(PROTECTIVE
GROUPS IN ORGANIC SYNTEHSIS)、第2版、ジョン・ウィレイ・アンド・サンズ
(John Wiley and Sons)、ニューヨーク(New York)、1991年に開示され
ている。脱保護は保護基の除去および水素原子による置換を示す。
Pg1は、固相合成の間、アミノ酸残基上の側鎖官能基を保護するために使用
される選択的に除去可能な保護基である。
Pg2は、固相合成の間、ペプチドのアミノ酸残基のαアミノ基を保護するた
めに使用される選択的に除去可能な保護基である。
リンカーL2は、選択的に開裂できる部位によってペプチド中のアミノ酸に付
着し、付着部位S1と共有結合を形成する2番目の付着部位S2を含有するラジ
カル基である。好ましい具体例において、L2は、ペプチドの環化を引き起こす
ために鋳型として働く残基に付着している。
S1は、リンカーL2上で2番目の付着部位S2と共有結合を形成する付着部
位である。S1の官能基がS2の官能基と反応できるならば、S1は固体支持体
上または固体支持体に付着したリンカー上に存在しているであろう。L1および
ペプチドの間の結合が開裂される場合、S2部位の機能はペプチドが樹脂に結合
する2番目の経路を提供することであるので、S1はペプチドそれ自身の上に存
在しない必要がある。好ましい具体例において、S1はリンカーL1上に存在す
る。本明細書において使用される場合、リンカーLは、S1およびS2間の結合
形成の結果生じたペプチドおよび固体支持体間の連結であることは明かであろう
。S1がリンカーL1上に存在する場合、LはL1およびL2からなるであろう
;S1が固体支持体自身の上に存在する場合、LはL2のみからなるであろう。
付着部位S1がペプチドの固相合成の条件下で別の方法で反応する反応性の官能
基である場合、S2と結合を形成する前に除去される保護基によって保護される
であろう。かかる保護基は他の保護基と直交しているべきであり、例えば、それ
はPg1またはPg2のごとき分子中の他の保護基に影響しない条件下で除去でき
、さらにそれは、成長するペプチド鎖中の成分アミノ酸残基のαアミノ基のため
の保護基のごとき他の保護基を除去するために使用される条件に対して安定であ
るべきである。例えば、S1およびS2間の結合がペプチド結合である場合、付
着部位S1は適切にはAloc基のごとき適切な保護基Pg3によって保護され
るアミノ基であり;付着部位S2は適切には、アリルアルコールエステルのごと
き適切な保護基Pg4によって保護されるカルボキシル基である。Fmoc基は
成分アミノ酸のαアミノ基のための適切な保護基Pg2であろう。他の適切な直
交する保護方法が当該分野において知られている。
しかしながら、珍しい具体例において、付着部位S1は固体支持体上にあり、
付着部位は固体支持体に付着するリンカー上にある。好ましい具体例において、
付着部位はリンカーL1それ自身に付着している。
S2はS1と共有結合を形成する付着部位であり、リンカーL2上に存在する
。付着部位S2が、ペプチドの固相合成の条件下で別の方法で反応する反応性の
官能基である場合、それは、S1で反応性基と結合を形成する前に除去される保
護基Pg4によって保護されるであろう。かかる保護基は他の保護基、およびペ
プ
チド−樹脂複合体における開裂部位に直交するべきであるが、これらの基は合成
手順の同時点で活性化されるので、かかる保護基はPg3に対して直交する必要
はない。付着部位S1およびS2に適切な基を図示する:
これらはPd(O)によって開裂でき、Pg1のための適切な直交保護基は、酸
によって開裂できるt−ブチル基類(ペンタメチレンクロマン−6−スルホニル
(Pmc)基によって保護されるArgを除く)であり、アミノ酸のαアミノ基
のためのFmoc基は、塩基によって開裂される。反応性基および保護基の他の
組み合わせは当業者にとって明かであろう。
は、リンカーL1とカルボキシル基を介して最初のN保護アミノ酸をカップリン
グさせ、次いで、残りの残基を次々にカップリングさせることによって、固相ペ
プチド合成の通常の方法で合成される。そのペプチドは典型的には、N保護アミ
ノ酸、樹脂へ添加された最後の残基のアミノ基の連続的脱保護によって行われる
ペプチド合成、および次のN保護アミノ酸のカップリングから合成される。ある
例において、一つまたは複数のアミノ酸をリンカーL1に付着させ、次いで、支
持体にL1を付着させることはより効果的であろう。アミノ酸が反応性の官能基
を含有する側鎖を有する場合、これらの官能基は適切な保護基Pg1によって保
護される。アミノ酸のアミノ基は適切な保護基Pg2によって保護される。
環化を指向する鋳型のために、ペプチドQは、残基または残基の組み合わせが
ペプチドのアミノ末端およびカルボキシ末端が近接しているコンフォメーション
を好む二次構造をペプチドにとらせるような方法で合成される。かくして、鋳型
残基はアミノ酸残基または残基の組み合わせであり、コンフォメーション的に束
縛されて直鎖状ペプチドの環化を促進する。例えば、直鎖状ペプチドH−D−T
rp−D−Asp−Pro−D−Val−Leu−OHは、Pro残基がアミノ
末端およびカルボキシ末端を近接させ、環化反応を促進する低エネルギーのγタ
ーンコンフォメーションをとるので、非常に容易に、かつほとんど定量的収量で
環化する。適切な鋳型は、Pro、Hyp、Aib、還元型ジペプチド同等物ま
たはチオアミド同等物のごときアミノ酸を包含するγターンの誘導体、D−Pr
o−Pro(または、より一般的には、D−XがD形のアミノ酸であり、N−(
Alk)Yがアミノ基上でアルキル化されているアミノ酸Yである、D−X−N
−(Alk)Y)のごときジペプチドを包含するβターンの誘導体、または、非
ペプチドターン模倣物;および、環状ヘキサペプチドにおいて伸長した骨格コン
フォメーションを誘導するD−Pro−Gly−Proを包含するであろう。例
えば、かかる要素は、パイショフ(Peishoff)ら、ジェイ・メド・ケム(J.Med
.Chem.)、第35巻、3962頁(1992年)によって開示されている。こ
れらの要素は、それらが有する大きな塊(例えば、Aib、Cα−メチルアミノ
酸、Nα−メチルアミノ酸)による立体的障害、または、環構造(例えば、Pr
o)の炭素骨格の要素を取り込むこと、または、隣接する残基に水素結合を形成
すること(例えば、チオアミド同等物、ジペプチド同等物)によって、ある結合
の回転が制限されることによりその効果を有する。他のコンフォメーション的制
限要素は、これらの原理を使用する技術から企画または選択されるであろう。
環化を促進するために直鎖状ペプチド配列中にその鋳型が導入されなければな
らない。最初に樹脂への鋳型の付着、続いて鋳型から双方向へのペプチド鎖の伸
長はやっかいなアプローチである。C末端からの連続的伸長は、カップリングの
間にラセミ化をおこす危険性があり、カップリングの完了を容易に測定させない
。完全なC末端ペプチド部分を樹脂に連結した鋳型にカップリングすること、ま
たは、鋳型−C末端ペプチド部分を樹脂にカップリングさせることは、どちらも
断片カップリングを意味し、それは本質的に効率の悪い方法であり、さらにペプ
チドライブラリー作成には適さない。ペプチドの樹脂への付着、およびリンカー
L2の付着の点を操作するために、鋳型または配列中の二次構造が誘導する要素
の利点をとることは可能である。
好ましい具体例において、骨格リンカーL2を介したペプチドの樹脂への結合
に使用される反応性部位S1と同様、リンカーL1が樹脂への2つの不安定な連
結からなるような方式でその方法は行われる。さらに、リンカーL2は鋳型残基
に付着する。以下の具体例において、鋳型アミノ酸残基はCによって図示され、
式(I)に参照されるように、リンカーL1はB−Aによって表され、反応性の
官能基S2はB−A上に存在してPg4によって保護され;リンカーL2はDに
よって表され、反応性の官能基S1はD上に存在してPg3によって保護され;
ペプチドQは、アミノ酸の側鎖の官能基がペプチドの末端アミノ基の保護基を表
すPg1およびPg2によって保護される、[AA1−AA4]−C−[AA5−A
A8]によって表される。
その方法に従って、適切に保護されたリンカーB−Aはペプチド合成樹脂に付
着する。次いで、最初のアミノ酸がリンカーに付着し、次いで、ペプチド合成が
通常の固相ペプチド合成法によって行われる。適当な時点で、あらかじめ作成さ
れ保護された、Cがアミノ酸であり、Dがテザ(tether)(例えば、L2)であ
る、鋳型−テザPg2−C(D−Pg4)が伸長するペプチド鎖にカップリングさ
れる。直鎖状ペプチド合成の完了後、テザの反応性官能基上の保護基Pg4およ
びリンカー(L1)の反応性官能基上の保護基Pg3を除去する。次いで、テザ
およびリンカーをお互いにカップリングさせ、リンカーを介して樹脂上で環化し
たペプチドを作成する。次いで、C末端アミノ酸およびリンカーB−Aの間の結
合を開裂させ、鋳型−テザを介して樹脂に付着した直鎖状ペプチドを得る。アミ
ノ末端保護基Pg2の開裂により、両方の末端が遊離した樹脂結合直鎖状ペプチ
ドが得られ、環化が可能になる。次いで、環化が行われた後、側鎖保護基Pg1
の開裂により完全に脱保護された樹脂結合環状ホモデティックペプチドが得られ
る。この時点で、鋳型アミノ酸CおよびテザDの間の結合は、所望により、ヒド
ラジン分解、加水素分解、トリフルオロ酢酸またはHFのごとき通常の方法で選
択的に開裂し、遊離ペプチドを得ることができる。
このスキームの顕著な特徴は以下の通りである。樹脂はペプチド付着の2つの
部位を有するリンカーを取り入れており、そのうちの1つは一時的に覆い隠され
ている。ペプチドはペプチド合成の通常の方法で合成でき、高収率のカップリン
グ方法が行われ、ニンヒドリン試験によってカップリング完了の評価が可能であ
る。保護された直鎖状ペプチドは、リンカーおよびペプチドの鋳型部分に付着し
たテザの間に新たに合成された結合を介して樹脂に付着する。これにより、ペプ
チドのアミノ末端およびカルボキシル末端が遊離し、高度に効果的な鋳型指向性
環化を樹脂に結合したペプチド上で行うことができる。
本方法は多くの直交的な開裂可能な保護基およびリンカーを必要とする。アミ
ン保護基Pg2は、側鎖保護基Pg1、テザ保護基Pg4、リンカー保護基Pg3、
鋳型CおよびテザDの間の結合、およびカルボキシル末端アミノ酸およびリンカ
−B−Aの間の結合の存在下で開裂できなければならない。さらにそれは、Pg3
およびPg4の開裂条件、および、カルボキシル末端アミノ酸およびリンカーB
−Aの間の結合の開裂条件に対して安定でなければならない。同様に、Pg3お
よびPg4は、Pg2除去の条件に安定で、Pg2、Pg1、および鋳型Cおよびテ
ザDの間の結合、およびカルボキシル末端アミノ酸およびリンカーB−Aの間の
結合の存在下で選択的に開裂できなければならない。カルボキシル末端アミノ酸
およびリンカーB−Aの間の結合は、Pg2およびPg1および鋳型Cおよびテザ
Dの間の結合の存在下で選択的に開裂できなければならない。
特殊な具体例において、そのアプローチが以下のスキームI−IIIに図示さ
れている。スキームIは、酸に不安定なリンカー(例えば、L1)の部分合成、
および樹脂への付着のためのペプチドの最初の残基を図示する。スキームIIは
、ペプチドの環化を指向するためにペプチドに取り込まれた、リンカー(例えば
、L2)に付着した、コンフォメーション的に制限された鋳型残基の合成を図示
する。スキームIIIは、モデルペプチドのための樹脂上鋳型指向性環化の一般
的方法を図示する。
スキームIIIに示した特殊な具体例において、樹脂はベンツヒドリルアミン
樹脂である。リンカーB−A(例えば、L1)は2つの部分からなる:Aはε−
Aloc−Lysであり、そのLysの側鎖は樹脂への2番目の付着部位を提供
し、Lys残基のカルボキシルはBHA樹脂にアミド結合を介して付着し;およ
び、Bは4−(4’−ヒドロキシメチル−3−メトキシ)フェノキシ酪酸(HM
PB)であり、そのHMPBのカルボキシルがLys残基のαアミンに付着して
伸長するペプチドはヒドロキシメチル基に付着する。t−ブチルを基礎とする保
護基によって保護されている側鎖を有するFmocアミノ酸は、本方法、即ち、
Pg2がFmocであり、Pg1がt−ブチルを基礎とするか、またはPMCであ
る方法において使用される。鋳型残基は4−ヒドロキシプロリン(Hyp)であ
り、テザL2は、サクシニル−β−アラニンであり、それはβ−Alaカルボキ
シル基を介してHypのヒドロキシルに付着する。サクシニルカルボキシル基は
アリルエステル(Pg4)として保護される。Fmoc基は酸に安定であり、P
d(O)試薬に安定であるが、2級アミン類で開裂可能である。アリルエステル
およびAloc基は、2級アミンおよび酸に対して安定であるが、Pd(O)試
薬で開裂可能である。ペプチドへのHMPB連結は希酸(2%TFA/CH2C
l2)で開裂可能であり、2級アミンおよびPd(O)に対して安定である。t
−ブチルおよびPmc側鎖保護基は、2級アミンおよびPd(O)に対し
て安定であるが、強酸(95%TFA/CH2Cl2)によって開裂可能である。
Hyp鋳型およびテザのβ−Alaの間のエステル連結は、上記すべての条件に
安定であるが、ヒドラジン分解によって開裂可能である。当業者は、直交性のた
めに要求されるすべてことを満たすならば、同様に使用できる他の有用な保護基
スキームの可能性があることを認識するであろう。
液体中または固相支持体上で組み合わせ的なライブラリーを調製および試験す
るための方法が知られており、例えば、ラム(Lam)ら、ネイチャー(Nature)
、第354巻、82頁(1991年)およびPCT/US91/04666(W
O92/00091);ホートン(Houghton)ら、ネイチャー(Nature)、第3
54巻、84頁(1991年)およびPCT/US91/08694(WO92
/09300)に開示されている。かかる方法は、環状ホモデティックペプチド
のライブラリーを得るための本発明方法との組み合わせであってもよい。例えば
、ペプチド鎖が出来上がるとき、固体支持体を各々発生したペプチドカップリン
グでいくつかの同モルの基に分割し、異なるアミノ酸を各々の基と徹底的に反応
させ、その基を再び組み合わせ混合し、次いで、次のカップリングのために基を
再び分割してもよい。このようにして、得られた直鎖状ペプチド樹脂複合体は、
各々異なるアミノ酸配列を有するビーズからなるが、各々のビーズは特有のペプ
チド配列を有するであろう。
ライブラリーは、ペプチド−樹脂複合体または遊離環状ペプチドの溶液のライ
ブラリーであってもよい。当該分野において知られているごとく、これらのライ
ブラリーは、標的分子に結合するか、または第2の分子の標的分子への結合を阻
害するペプチドを同定するために、標的分子または表面をスクリーニングする目
的に使用されてもよい。PCT/US92/09345(WO94/08051
)に開示されているように、暗号化方法は、かかる組み合わせ的ライブラリーの
活性ペプチドの検出を改良した本発明方法と組み合わせてもよい。
ライブラリーは、本方法に従って、ゲイセン(Geysen)ら、米国特許第5,1
94,392号および米国特許第4,562,157号によって開示されているミ
モトープ(minotope)法を使用することにより、部分的に既知の組成のペプチド
のグループを調製することによって、構築およびスクリーニングされてもよい。
活性ペプチドを部分的に開裂することによって樹脂ビーズをアッセイするための
、当該分野において知られている他の方法を適用してもよい。
以下の実施例は、化合物を作り使用する方法、ライブラリー、および本発明方
法を示すものであって、いかようにも制限するものではない。参照の簡便性のた
めに、例えば、スキームIIIにおける化合物5はIII−5とするように、あ
る化合物はスキームI−IIIにおいて示される数字によってラベルされる。
実施例I
酸に不安定なリンカーを予め負荷されたFmoc−ロイシンの調製
4[4−Nα−(9−フルオレニルメトキシカルボニル)−ロイシニル−オキシ
メチル]−3−フェニルオキシ]酪酸(1)スキーム1
乾CH2Cl2(100ml)およびピリジン(2.3ml、28ミリモル)中
のFmoc−ロイシン(10g、28ミリモル)の撹拌した溶液に、フッ化シア
ヌル(3.3ml、28ミリモル)を滴下した。反応物をアルゴン存在下室温で
4時間撹拌し、不活性沈澱物を濾過除去し、冷H2O(2x250ml)で洗浄
し、乾燥し(MgSO4)、次いで、乾燥するまでエバポレートした。
0℃の乾CH2Cl2(100ml)およびピリジン中の4(4−ヒドロキシメ
チル-3−メトキシ−フェニルオキシ)酪酸(6.80g、28ミリモル)の撹拌
した溶液に、CH2Cl2(50ml)中のフッ化Fmoc−ロイシニルの溶液を
添加した。反応物を室温にまで温めて、次いで、16時間撹拌した。次いで、反
応物を冷0.5NHCl水溶液で酸性化し、ブラインで洗浄し、乾燥し(MgS

a)Nα−(t−ブトキシカルボニル)−ヒドロキシプロリンベンジルエステル
(II−1)
メタノール(100ml)および水(50ml)中のNα−t−ブトキシカル
ボニル−ヒドロキシプロリン(5g、21.6ミリモル)の撹拌した溶液に、C
sHCO3(4.2g、21.6ミリモル)を添加した。15分間撹拌後、溶液を
乾燥するまでエバポレートし、メタノール(2x100ml)から2回、次いで
、最終的にDMF(100ml)から再エバポレートした。残渣をDMF(10
0ml)中に入れて室温で撹拌しながら、臭化ベンジル(3.1ml、26ミリ
モル)を一度に添加した。次いで、反応物をアルゴン存在下50℃で16時間撹
拌した。得られた懸濁液を減圧下でエバポレートし、酢酸エチル(150ml)
中に入れてH2O(150ml)で洗浄し、乾燥し(MgSO4)、エバポレート

間水素化した。セライトパッドで触媒を濾過除去し、氷酢酸(4x50ml)で
洗浄した後、濾液を減圧下でエバポレートし、メタノール(2x50ml)から
再エバポレートしてNα−(t−ブトキシカルボニル)−O−(β−アラニニル
)ヒドロキシプロリンベンジルエステル(II−3)(2.36g、98%)を
白色の固体として得た。
無水コハク酸(24g、240ミリモル)にアリルアルコール(25ml、3
60ミリモル)を添加した。反応物をAr存在下で16時間還流し、室温に冷却
し、飽和NaHCO3水溶液(400ml)に溶解し、Et2O(200ml)で
洗浄し、冷3N HCl水溶液で酸性化して、酢酸エチル(300ml)で抽出
し、ブラインで洗浄し、乾燥し(MgSO4)、次いで、減圧下エバボレートし
てコハク酸モノアリルエステル(36.02g、95%)を得た。
トルエン(50ml)中のコハク酸モノアリルエステル(13.75g、87
ミリモル)の撹拌した溶液に、塩化オキザリル(20ml、230ミリモル)を
添加した。反応物を16時間室温で撹拌し、次いで、減圧下でエバボレートした
。トルエン(2x50ml)からの再エバポレーションにより、以下のカップリ
ング反応で使用するサクシニルクロリドモノアリルエステル(14.68g、9
6%)を澄明な液体として得た。
(4:1)ジオキサン/H2O(100ml)中のNα−(t−ブトキシカル
ボニル)−O−(β−アラニニル)ヒドロキシプロリンベンジルエステル(2.
36g、7.8ミリモル)の撹拌した0℃の溶液に、NaHCO3(1.6g、1

CHCl3(75ml)中の上記TFA塩の撹拌した溶液に、Et3N(1.1
5ml、8.2ミリモル)、引き続いてN−(9−フルオレニルメトキシカルボ
ニル)サクシンイミド(1.52g、4.5ミリモル)を添加した。6時間撹拌後
、その溶液を1N HCl水溶液(10ml)で酸性化して、ブラインで洗浄し
、乾燥し(MgSO4)、乾燥するまでエバポレートした。シリカゲル上でのフ

実施例3
モデル環状ペプチド(4)スキームIIIの調製
シクロ−(D−トリプトファニル−D−アスパルチル−ヒドロキシプロリル−D
−バリニル−ロイシン)(III−4)
リジン再付着部位(III−1)を有する保護された直鎖状ペプチド酸不安定
樹脂が、以下の一般的合成方法に従ったFmoc−Nε−(Aloc)−リジン
、Fmoc−ロイシル−HMBA、Fmoc−D−バリン、Fmoc−(O−β
Ala−Suc−Oアリル)−ヒドロキシプロリン(II−5)、Fmoc−(
O−t−Bu)−D−アスパラギン酸、およびFmoc−Nin−Boc−D−ト
リプトファンの連続したカップリングおよび脱保護によって市販のベンツヒドリ
ルアミン樹脂(1.18ミリモル/g)上に構築された。
一般的カップリング方法
HBTU(3モル等量)を小さいエルレンマイヤーフラスコ中で振り混ぜなが
らDMFに溶解した。この溶液にFmoc−アミノ酸(3モル等量)、引き続い
てN−メチルモルフォリン(8モル等量)を添加した。次に、この溶液をロッカ
ーベッセル中の樹脂(1モル等量)およびHOBt(3モル等量)に添加し、2
時間振盪した。反応をカイザー試験でチェックして、反応終了後、DMF(4x
)次いで、CH2Cl2(2x)で洗浄した。
一般的Fmoc脱保護法
Fmoc保護基を、ペプチド樹脂をDMF中の20%ピペリジンで処理する(
1x5分および1x15分)ことによって除去した。次いで、樹脂をDMF(4
x)で洗浄した。
(1:1)DMSO/CH2Cl2(10ml)中の調製された樹脂ペプチド、
Fmoc−(Boc)D−Trp−(O−t−Bu)D−Asp−(O−β−A
la−Suc−Oアリル)Hyp−D−Val−Leu−HMBA−(Aloc
)Lys−BHA(III−1)(2g、0.8ミリモル)に、酢酸(460μ
l、8ミリモル)、(Ph3P)2PdCl2(24mg、34μモル)、引き続
いてBu3SnH(862μl、3.2ミリモル)をAr存在下室温で撹拌しなが
ら添加した。30分間撹拌後、樹脂を濾過し、(1:1)DMSO/CH2Cl2
(2x50ml)で洗浄した。反応を2回繰り返し、(1:1)DMSO/CH2
Cl2(2x50ml)、CH2Cl2(2x50ml)、CH2Cl2(1x50
ml)中のN−メチルモルフォリン、CH2Cl2(2x50ml)、次いで、n
−ヘキサン(2x50ml)で洗浄した。部分的に脱保護されたペプチド樹脂1
.97gを減圧乾燥後に得た。カイザー試験では遊離アミン陽性であった。
DMF(10ml)中の上記樹脂ペプチド(1.84g、0.75ミリモル)に
、Ar存在下、撹拌しながらHOBt(0.22g、1.6ミリモル)、N−メチ
ルモルフォリン(0.45ml、1.6ミリモル)、引き続いてHBTU(0.6
0g、1.6ミリモル)を添加した。16時間撹拌後、樹脂ペプチドを濾過し、
DMF(2x50ml)、CH2Cl2(2x50ml)、およびn−ヘキサン(
2x50ml)で洗浄した。カイザー試験では遊離アミン陰性であった。
DMF(50ml)中の20%ピペリジンで上記樹脂ペプチドを5分間処理し
、次いで、再びDMF(50ml)中の20%ピペリジンで15分間処理し、濾
過し、DMFで洗浄し、CH2Cl2(4x10ml)中の5%トリフルオロ酢酸
および5%アニソールの溶液で各々2分間処理した。次に、得られたヒドロキシ
プロリル側鎖付着直鎖状ペプチド樹脂を、CH2Cl2(2x50ml)、CH2
Cl2中の10%N−メチルモルフォリン、CH2Cl2(2x50ml)、およ
びn−ヘキサン(2x50ml)で洗浄し、減圧下乾燥後、1.22gの樹脂ペ
プチドを得た。カイザー試験では遊離アミン陽性であった。
DMF(10ml)中の上記樹脂ペプチド(1.20g、0.74ミリモル)に
、Ar存在下室温で撹拌しながら、HOBt(0.17g、1.2ミリモル)、N
−メチルモルフォリン(0.33ml、3.0ミリモル)、引き続いてHBTU(
0.45g、1.2ミリモル)を添加した。24時間撹拌後樹脂を濾過し、DMF
(2x50ml)、(1:1)CHCl3/MeOH(2x50ml)、CH2C
l2(2x50ml)、およびn−ヘキサン(2x50ml)で洗浄した。1.2
2gのペプチド樹脂(2)スキーム3を減圧下乾燥後に得た。カイザー試験では
遊離アミン陰性であった。
EtOH(5ml)中の上記ペプチド樹脂(2)スキーム3の一部に(0.5
1g、0.31ミリモル)、NH2NH21水和物(200μl、4.1ミリモル)
を添加した。48時間撹拌後、樹脂を濾過し、MeOH(2x20ml)で洗浄
した。粗保護環状ペプチド(66mg、29%)が、濾液のエバポレーションお
よび減圧下乾燥により得られた。シリカゲル上のフラッシュクロマトグラフィー
(CHCl3中の3%MeOH)による精製によって、分析学上純粋な物質(3

樹脂ペプチド(III−2)の残り(0.71g、0.44ミリモル)に、トリ
フルオロ酢酸(10ml)中の10%アニソールの溶液を添加した。15分間撹
拌後、樹脂を濾過し、トリフルオロ酢酸(10ml)中の10%アニソールでさ
らに15分間再処理した。完全に脱保護されたペプチド樹脂をCH2Cl2(2x
25ml)、CH2Cl2中のN−メチルモルフォリン(2x25ml)、CH2
Cl2(2x25ml)、およびn−ヘキサン(2x25ml)で洗浄した。
EtOH(5ml)中の上記ペプチド樹脂に、NH2NH2モノ水和物(300
μl)を添加した。48時間撹拌後、樹脂を濾過し、MeOH(2x20ml)
で洗浄した。濾液を減圧下エバポレートし、粗生成物をヒドラジン塩として得た
(77.0mg、29%)。YMC−ODS(S−50)カラム上てのフラッシ
ュクロマトグラフィーによる精製(30−50%CH3CN、0.1%TFA、H
2O)によって、標記化合物(III−4)を凍結乾燥された白色粉末として得
た(37mg)。さらに103mgの粗環状ペプチド(全68%)を、残った樹
脂をメタノール(2ml)中の無水ヒドラジン(2ml)に適用し、さらにAr
存在下60℃で48時間撹拌し、濾過し、メタノール(2x20ml)で洗浄し
、濾液を乾燥するまでエバポレートすることにより得た。HPLCK'=5.21
