ベンゾチオフェン誘導体の製造方法
本出願は、一般式(I)
[R1はNH-C1-C20-アルキル、NH-アリール、NH-ベンジルを表し、そし
て
R2はH、メチルまたはハロゲンを表す]
で表される置換されたベンゾチオフェン誘導体を製造する方法に関する。
式(I)のベンゾチオフェン誘導体は公知であり、そして公知の医薬品、植物保
護剤及び材料保護剤の製造中間体として使用される。
ベンゾチオフェン誘導体の製造ルートは、同様に公知であり、例えば、ヨーロ
ッパ特許第568 289号、Tetrahedron Lett.33,7499(1992)、Tetrahedron 39,4
153(1983)、J.Org.Chem.,37,3224(1972)、米国特許第5169 961号明細書
、ヨーロッパ特許第572 712号及びJ.Chem.Soc.Perkin.Trans.I,1984
,385に記載されている。それらのすべては、収率が不満足であり、及び/また
は出発材料の入手可能性に劣り、並びに装置に特別な要求がなされることに特徴
がある。
ここでそして以下において、アルキルは、好ましくは、場合により1〜2個の
酸素原子で遮られていてもよい直鎖、分枝または環式のアルキ
ル、例えばメチル、エチル、n-,i-プロピル、n-,i-,s-,t-ブチル、n-,i-,
s-,t-ペンチル、ヘキシル、ヘプチル、オクチル、ノニル、デシル、ウンデシル
、ドデシルまたはそれらの分枝構造異性体並びにシクロペンチル、シクロヘキシ
ル、シクロヘプチル、シクロオクチル、シクロドデシルを表す。
一般式(II)
で表されるケイ皮酸から、式(III)及び(IV)
で表される中間体を単離または精製することなく、式(III)及び(IV)で表される
中間体を経由して式(I)で表されるベンゾチオフェンカルボキシアミドをワンポ
ット法(one-pot process)で製造し、そして次に得られた式(I)で表される化
合物を、場合により中間体を単離することなく、ワンポット反応(one-pot reac
tion)の形式で酸化して、式(Ia)
で表される公知のスルホンを得るのが可能であることが見い出された。
式(III)で表される3−クロロ−ベンゾチオフェン−2−カルボニルクロリド
の製造法は一般に公知である[T.Higa,J.Org.Chem.40 3037(1975);
A.J.Krubsack,Tetrahedron Letters,5149(1968);S.Nakagawa,Te
trahedron Letters,3719(1970);T.Higa,J.Org.Chem.41 3399(1976
)]。この目的のためには、塩基の存在下または不存在下でそして希釈剤の存在
下または不存在下でケイ皮酸を塩化チオニルと反応させる。
一般に、塩化チオニルは過剰に使用され、好適には3〜10当量、非常に好適
には3〜6当量が使用される。
この場合、先に塩化チオニルを導入し、次にケイ皮酸を添加するか、或いは先
にケイ皮酸を導入し、次に塩化チオニルを滴下するかの方法のいずれも可能であ
る。しかし、更に、最初に塩化チオニルの一部を導入し、次にケイ皮酸を添加し
、引き続いて残りの塩化チオニルを添加することも可能である。好適には、最初
に塩化チオニルの一部を導入し、ケイ皮酸を添加し、次に残りの塩化チオニルを

反応温度は広い範囲で変えることができる。好適には、反応は0℃〜200℃
で、きわめて特に好適には70℃〜160℃で行われるが、し
かし、ケイ皮酸または塩化チオニルの添加の開始時には、好適には0℃〜70℃
に維持される。
場合により、塩基も本反応に添加することができる。ここで使用される塩基は
、好適には例えばピリジン、トリエチルアミン、キノリン、ジメチルアミノピリ
ジンまたは他の置換されたピリジンである。
反応は希釈剤なしで行うことができるが、場合により希釈剤の存在下で行うこ
ともできる。好適には、この方法は希釈剤なしで行うか、或いは希釈剤、例えば
炭化水素、例えばベンゼン、トルエンまたはヘキサン、ハロゲン化炭化水素、例
えばテトラクロロ炭化水素、クロロホルム、塩化メチレン、エーテル、例えばジ

するか或いは過剰の塩化チオニルを減圧下で除去する。
好適な飛沫同伴剤は、蒸留の際に塩化チオニルを飛沫同伴し得るあらゆる溶媒
である。好適には、この目的には高沸点の有機溶媒が使用され、きわめて特に好
適には、炭化水素、例えばベンゼン、トルエン、キシレン、クロロベンゼン、ヘ
キサン、ヘプタン、オクタン、クロロホルム、テトラクロロ炭化水素、エーテル
、例えばジブチルエーテル、メチルt-ブチルエーテル及びニトロメタン、ジメチ
ルホルムアミドまたはDMSOが使用される。しかし、次の段階一部分の溶媒を
飛沫同伴剤として使用することも可能である。
酸受容体の存在下または不存在下でそして希釈剤の存在下または不存在下で上
述のように製造した3−クロロ−ベンゾチオフェン−2−カルボニルクロリドを
式(IV)で表されるアミンR1-NH2と反応させると、中間体状態の式(V)で表
される3−クロロ−ベンゾチオフェン−2−カルボキシアミドが得られる。
方法を実施する際に、場合により希釈剤に溶解したアミン(IV)及び場合により
使用されるいずれかの酸受容体を、場合により希釈剤と混合した式(III)で表さ
れる“粗”3−クロロベンゾチオフェン−2−カルボニルクロリドに添加する。
この段階-部分において、反応温度は比較的広い温度範囲で変えることができる
。一般に、−10℃〜+100℃、好適には0℃〜60℃の温度を採用できる。
方法を実施する際に、添加したアミンの量及び場合により添加した酸受容体の量
は、存在する3−クロロベンゾチオフェン−2−カルボニルクロリドの量と約等
モルである。過剰のアミン及び酸受容体は一般に有害とはならない。
使用できる酸受容体は通常のあらゆる酸受容体である。これらには、好適には
第3級アミン、例えばトリエチルアミン及びピリジン;アルカリ金属水酸化物、

場合により使用される好適な希釈剤は、それ自体3−クロロベンゾチオフェン
−2−カルボン酸と反応しないあらゆる溶媒であり、好適には炭化水素、例えば
ベンゼン、トルエンまたはヘキサン、ハロゲン化炭化水素、例えば四塩化炭素、
クロロホルム、塩化メチレン、エーテル、例えばジエチルエーテル、ジプロピル
エーテル、ジブチルエーテル、メチ
ルt-ブチルエーテル、ジオキサン、テトラヒドロフラン、更に酢酸エチル、ジメ
チルホルムアミド、アセトニトリルまたはDMSOである。
次に、場合により希釈剤を変えそして場合によりHCl捕捉剤の存在下で先に
水で抽出した後に、この様式で得られる反応混合物を加圧下にまたは大気圧下に
触媒の存在下に水素と反応させて、式(I)のベンゾチオフェン−2−カルボキ
シアミドを形成する。
この方法において、高い空間-時間収率、ひいては特別な経済効率を達成する
ためには、高い圧力と温度を採用するのが有利である。本発明において有利な温
度は500℃〜200℃であり、70℃〜150℃の温度が特に有利であり、8
0℃〜120℃の温度がきわめて特に有利である。一般に5〜300バールの高
圧力が採用されるが、50〜200バールの圧力が特に有利であり、80〜12
0バールの圧力がきわめて特に有利である。
場合により使用される好適な希釈剤は、水素と3−クロロベンゾチオフェン−
2−カルボキシアミドに対して十分な溶解度を有するあらゆる溶媒であり、例え
ばアルコール、エーテル、芳香族炭化水素、ピロリドン類である。好適なアルコ
ールは、直鎖または分枝のC10までの第1級、第2級または第3級アルコールで
ある。特に好適なアルコールは、例えばメタノール、エタノール、プロパノール
、イソプロパノール、n-ブタノール、i-ブタノール、n-及びi-ペンタノール並び
にこれらの混合物である。きわめて特に好適なアルコールはメタノール、エタノ
ール、n-プロパノール及びi-プロパノールである。好適なエーテルは対称及び非
対称のC10までのアルキルアルキルエーテル及びアルキルアリールエーテルであ
る。特に好適なエーテルはジメチルエーテル、ジエチルエーテル、
ジイソプロピルエーテル、メチルt-ブチルエーテル、テトラヒドロフランまたは
ジオキサンである。好適な芳香族炭化水素はベンゼン、トルエン及びキシレンで
ある。好適なピロリドン類は例えばN-メチルピロリドン、N-オクチルピロリド
ンである。
触媒には特定の要求はなされない。本発明においては、特に、従来技術の水素
脱塩素化触媒に対応するあらゆるものが使用できる。特に有利には、耐S系が使
用される。この意味で好適な触媒は、ラネーニッケル、ラネーコバルト、活性炭
素担体上のPd,Pt,Ru,Rh、アルカリ土類金属担体上のPd,Pt,Ru,Rh
である。特に有利なものは、ラネーニッケル、ラネーコバルト、活性炭素上に安
定化されたPd,Pt,Ru,Rh触媒である。
場合により、得られるHClに結合するかまたはHClをコンプレックス化す
るHCl捕捉剤を添加して、水素脱塩素化を行う。この場合に、HClに結合し
得るかまたはHClをコンプレックス化し得るあらゆる公知のHCl捕捉剤が使
用できる。本発明において特に好適な塩基はNaOH、KOH、Ca(OH)2、Ba
(OH)2及びピリジンである。
後処理として、混合物を触媒及び無機材料から濾別し、そして濾液を、蒸留、
クロマトグラフィーまたは結晶化による常法で精製する。
本発明によれば、場合により溶媒を蒸発させそして場合により別の希釈剤を添
加した後、上記粗濾液(ベンゾチオフェン−2−カルボキシアミドを含む)を、
精製することなく酸化し、一般式(VI)のベンゾチオフェン−2−カルボキシアミ
ド S,S’-ジオキシドを得る。
本発明の方法を実施するのに使用し得る酸化剤は、硫黄を酸化するために通常
使用できるあらゆる酸化剤である。好適には、過酸化水素または有機過酸、例え
ば過酢酸、4−ニトロ過安息香酸または3−クロロ過安息香酸、或いは無機酸化
剤、例えば過ヨウ素酸、過マンガン酸カリウム、クロム酸またはオキソン(oxon
e)である。
本発明の方法を実施するのに使用し得る希釈剤は、使用する酸化剤に応じて選
択される無機または有機溶媒である。好適には、メタノール、エタノール、プロ
パノール、イソプロパノールのようなアルコールまたはこれらと水との混合液ま
たは純水;酸、例えば酢酸、無水酢酸、プロピオン酸または双性非プロトン性溶
媒、例えばアセトニトリル、アセトン、酢酸エチルまたはジメチルホルムアミド
並びに非ハロゲン化またはハロゲン化炭化水素、例えばベンジン、ベンゼン、ト
ルエン、ヘキサン、シクロヘキサン、石油エーテル、ジクロロメタン、ジクロロ
エタン、クロロホルム、四塩化炭素またはクロロベンゼンが使用される。
本発明の方法は、反応助剤の存在下または不存在下に行うことができる。使用
できるのは、あらゆる使用可能な通常の有機または無機の酸受容体である。好適
には、アルカリ土類金属水酸化物もしくはアルカリ金属水酸化物、アルカリ土類
金属酢酸塩もしくはアルカリ金属酢酸塩、またはアルカリ土類金属炭酸塩もしく
はアルカリ金属炭酸塩、例えば水酸
化カルシウム、水酸化ナトリウム、酢酸ナトリウムまたは炭酸ナトリウムが使用
される。
本発明の方法は、好適な触媒の存在下または不存在下に行うことができる。使
用できるのは、硫黄を酸化するのに通常使用できるあらゆるこの種の触媒である
。好適には、重金属触媒が使用され、この意味で例示されるのはモリブデン酸ア
ンモニウムである。
本発明の方法を実施する場合に反応温度は比較的広い範囲で変えることができ
る。一般に、−30℃〜+200℃の温度、好適には0℃〜+140℃の温度が
採用できる。
一般に、2.0〜10モル、好適には2.0〜5.0モルの酸化剤、反応助剤
としての1.0〜1.5モル、好適には1.0〜1.3モルの塩基、並びに、場
