義歯安定化組成物
背景
通常の取外しできる義歯、デンタルプレート等は、適切なプレート又はベース
に取り付けられた歯からなる。義歯は通常技巧的に作製されるが、それらは完全
には合わないことがある。更に、たとえいかに最初満足できたとしても、しばら
くすると義歯の適合性は自然収縮と歯肉、粘膜組織等における変化のせいでゆる
んで不完全になる。ゆるんで不完全に合わせた義歯は通常矯正されて、義歯安定
剤の使用で安定化される。義歯安定剤は義歯と歯肉又は組織との隙間を埋めるた
めに用いられる。口腔内に義歯をいれる前に義歯安定剤が義歯プレート表面に適
用されるが、これは完全な適合のために歯肉及び粘膜組織と均一に接触するべき
である。義歯安定剤はその接着性のためだけではなく、義歯と歯肉又は組織との
間でクッション又はガスケットを設けるためにも処方され、それにより口腔内で
義歯を確実に取り付けている。
義歯安定化又は接着組成物に通常存在するいくつかの欠点は、相分離、1日2
回以上の接着剤適用の必要性と、デンタルプレート下から接着剤の滲出である。
滲出は生じる不快な味、不快な口内感と、デンタルプレート下から接着剤の喪失
のせいで、特に望ましくない。
改良された義歯接着組成物を開発するため長年にわたり相当な努力が行われて
きた。合成及び天然双方のポリマー及びゴムが、上記欠点を少なくする試みから
、単独で、組合せで、及び様々な接着剤と組合せて用いられてきた。しかしなが
ら、確実な固着を行い、ユーザーにとり美観上快く、現在入手しうる製品よりも
滲出が少なく、口及び/又は義歯からクリーニングすることが容易である、改良
され
た義歯安定化組成物に関する必要性はなお存在している。
優れた接着性を有しながら、滲出が少なく、ユーザーに快い美観を与える義歯
安定剤が処方できることが、本発明により発見された。本発明の接着組成物は、
創傷用包帯、水中接着剤、バイオ接着剤及び/又は他の活性剤用のデリバリービ
ヒクルとしても有効に使える。
本発明の目的は、長期間にわたり有効に義歯を適所に固着できて、なおも要求
があれば義歯を容易に取り外せる、義歯安定化組成物を提供することである。も
う1つの目的は、現在入手しうる安定剤よりも滲出が少なく、ユーザーにとり美
観上快い、改良された義歯安定化組成物を提供することである。もう1つの目的
は、本発明組成物の親水性が現在入手しうる製品よりも口及び/又は義歯からク
リーニングすることが容易である接着組成物を提供することである。
本発明のこれら及び他の目的は、以下の詳細な記載から容易に明らかとなるで
あろう。
発明の要旨
本発明は、a)下記繰返構造単位から本質的になる低級アルキルビニルエーテ
ル‐マレイン酸コポリマーの非混合部分塩:
(上記式中RはC1‐C4アルキル基を表す;nはコポリマーの分子中における
構造単位の繰返出現数を表す2以上の整数であって、nは1.2以上の比粘度を
有するとしてコポリマーを特徴付けうるほど十分に大きい;比粘度は25℃でメ
チルエチルケトン中で測定される;部分塩は反応しうる全初期カルボキシル基の
うち約0.1〜約60%で亜鉛又はストロンチウムカチオンのカチオン塩官能基
を含んでいる)約10〜約80%;及びb)約100以上で約600以下の平均
分子量を有するポリエチレングリコール約20〜約90%を含んでなる接着組成
物に関する。
発明の具体的な説明
本発明の接着組成物は、非混合亜鉛又はストロンチウム塩からなる低級アルキ
ルビニルエーテル‐マレイン酸コポリマーと、ポリエチレングリコールを含んで
なる。約400の平均分子量を有するポリエチレングリコールが、本発明組成物
で使用上最も好ましい。本発明組成物はクリーム、ペースト、粉末、液体、軟膏
及びローションとして処方される。本発明の必須及び任意成分の詳細な記載は以
下でなされている。低級アルキルビニルエーテル‐マレイン酸コポリマー塩

本明細書で用いられる“非混合コポリマー塩”という用語は、亜鉛又はストロ
ンチウムカチオンが同コポリマー上でいかなる他のエステル官能基又は非同一カ
チオンとも混合されずに、残りのカルボキシル基が未反応のままである、低級ア
ルキルビニルエーテルーマレイン酸コポリマーの亜鉛及びストロンチウム部分塩
に関する。
本明細書で用いられる“混合コポリマー塩”という用語は、亜鉛及びストロン
チウムが互いに、又は他のエステル官能基、又はカルシウム、ナトリウム、マグ
ネシウム、カリウム、アンモニウム及びそれらの混合物からなる群より選択され
る非同一カチオンと同コポリマー上で混合されている、低級アルキルビニルエー
テル‐マレイン酸コポリマーの亜鉛及び/又はストロンチウム部分塩に関する。
本組成物は、部分塩がカルシウム、ナトリウム、マグネシウム、カリウム、ア
ンモニウム、亜鉛、ストロンチウム及びそれらの混合物からなる群より選択され
るカチオン塩官能基を含む低級アルキルビニルエーテル‐マレイン酸コポリマー
の1種以上の混合部分塩約0.1〜約75%を更に含んでいることが好ましい。
亜鉛、カルシウムもしくはナトリウムカチオン又はそれらの混合を含む混合コポ
リマー塩が最も好ましい。亜鉛カチオンは、反応しうる全初期カルボキシル基の
うち約0.1〜約65%、好ましくは約10〜約45%、最も好ましくは約15
〜約30%のレベルで存在する。カルシウムイオンは、反応しうる全初期カルボ
キシル基のうち約10〜約75%、好ましくは約25〜約60%、最も好ましく
は約40〜約60%のレベルで存在する。ナトリウムカチオンは、反応しうる全
初期カルボキシル基のうち約1〜約20%、好ましくは約1〜約15%、最も好
ましくは約1〜約10%のレベルで存在する。
本ポリマー塩は、水性媒体中においてAVE/MAコポリマー(I)と、例え
ばヒドロキシド、アセテート、ハライド、ラクテート等のような、カルボン酸の
反応剤に典型的な官能基を有するカチオン性亜鉛又はストロンチウム化合物との
相互作用により製造されることが有利である。好ましい態様において、混合AV
E/MAコポリマー塩は、AVE/MAコポリマー(I)と、カチオン性亜鉛又
はストロンチウム化合物について上記されたような官能基を有するカチオン性カ
ルシウム、ナトリウム、マグネシウム、カリウム、アンモニウム、亜鉛、ストロ
ンチウム(及びそれらの混合)化合物との相互作用により製造される。好ましい
混合AVE/MAコポリマー態様では、亜鉛の酸化物とカルシウムの水酸化物が
利用される。
水酸化亜鉛は市販されていないため、反応剤としてその使用は、実際上水に不
溶性だが粒子表面上で水酸化亜鉛に水和される粒状酸化亜鉛の水性スラリーを用
いることにより、容易でより経済的に行われる。他方、水酸化ストロンチウムは
結晶又は粉末形のいずれかで入手でき、水約50部に可溶である。しかしながら
、水で処理された場合(注意:発熱)に水酸化物を形成する酸化ストロンチウム
の水溶液も用いてよい。
毒性、刺激性又は汚染副生成物を形成するアニオンは回避されるか、あるいは
特別な用心及び処置がポリマー塩最終生成物からのこのような副生成物の除去及
び不存を保証するために行われるべきである。用いられる具体的な化合物は、実
質上純粋な実質上灰白色(off-white)のポリマー塩最終生成物を確実に得られ
るよう、実質上純粋であるべきである。
低級アルキルビニルエーテルマレイン酸(AVE/MA)コポリマーは、酸コ
ポリマー(I)に容易に加水分解しうる対応低級アルキルビニルエーテル‐無水
マレイン酸コポリマーを作るために、メチルビニルエーテル、エチルビニルエー
テル、ジビニルエーテル、プロピルビニルエーテル及びイソブチルビニルエーテ
ルのような低級アルキルビニルエーテルモノマーを無水マレイン酸と共重合させ
ることにより容易に得られる。無水物及び酸形は双方とも市販供給業者から入手
できる。例えば、GAF Corporation,Wayne,N.J.は“ガントレズ”(GANTREZ
)商標名で各々“ガントレズSシリーズ”及び“ガントレズANシリーズ”とし
てポリマー遊離酸形(I)及び対応無水物形の双方を供給する。前者の酸シリー
ズではガントレズS‐97(M.W.TM50,000)が特に適切であり、後
者の無水物シリーズではガントレズAN‐149(M.W.=50,000)、
ガントレズAN‐169(M.W.=67,000)及びガントレズAN‐17
9(M.W.=80,000)コポリマーが特に適切である。(2‐ブタノン1
〜10g/1000ml溶液中で膜浸透圧法により測定したところ)約50,0
00〜約80,000の平均分子量を有するAVE/MAコポリマーの酸及び無
水物形は、1.2以上の前記された比粘度パラメーターを有することでも特徴付
けられる。無水コポリマーが水に溶解すると無水結合鎖は開裂して、高極性ポリ
マー遊離酸(I)が形成される。したがって、酸形よりも比較的安価である無水
物形は、酸の便利かつ安価な前駆体として用いてよい。高温は無水物‐酸加水分
解の速度を高める上で有利に用いられる。
一般的に、低級アルキルビニルエーテルーマレイン酸コポリマー(I)又はそ
の対応無水物は、均一混合物を形成するために激しい攪拌下で約70〜80℃に
前加熱された水に加えられる。無水物前駆体が利用されるならば、水性混合物は
無水物から酸形への完全な加水分解を保証するために攪拌下で約90℃に更に加
熱されることが勧められる。その後加熱が中止されるが、ミキシングはバッチが
透明になって同時に粘度が減少するまで続けられる(約65〜75℃)。カチオ
ン性亜鉛又はストロンチウム塩形成化合物の水溶液、又は例えばスラリー形態で
粒状酸化亜鉛の水性分散物が、最終生成物で希望される望ましいカチオン含有率
を示すために十分な量で、環境温度で別に調製されて、カチオン性ポリマー塩の
局部沈澱を防ぐために連続的な激しいミキシング下で熱ポリマー酸溶液にゆっく
り加えられる。添加終了後、ミキシングはすべての塩形成化合物がコポリマーと
確かに反応するように続けられる。
一方、亜鉛又はストロンチウム源を含有した水溶液が、均一なスラリーを形成
するために激しい攪拌下で70〜80℃に前加熱される。次いで低級アルキルビ
ニルエーテル‐マレイン酸コポリマー(I)又はその対応無水物が、完全な加水
分解を保証するため更に90℃に加熱して攪拌しながらスラリーに加えられる。
次いで反応バッチは、水分を蒸発させて、乾燥形でポリマー塩生成物を回収す
るために、熱風循環で約70℃に維持された対流式オーブンで、例えば浅い乾燥
トレーにより乾燥される。一方、反応バッチは水分を蒸発させてフレーク形でポ
リマー塩を回収するために、熱スチームで80〜100PSIG(約5.6〜7kg/
cm2)に維持された5つのドラムドライヤーに移される。
得られたフレークは、望ましい物理的性質を示して、満足すべき義歯安定化性
質を与えるように、粉砕及び篩分けに付される。
上記塩はもろいために、適切な粒度及び嵩密度が得られる。最良結果のために
は、粒子は140〜200メッシュシーブ(U.S.B.S.シリーズ)を通ることがで
きて、好ましくはそれらの最大寸法が0.74mm以下である。
本亜鉛又はストロンチウムAVE/MAコポリマー塩は水又は唾液と接触した
とき例外的な接着性を有しているため、それらは義歯安定化組成物における義歯
接着物質として極度に有用である。このような使用の場合、粒子形の塩は少くと
も−140メッシュU.S.B.S.シーブの粒度;0.3g/cm3以上、好ましくは

本亜鉛又はストロンチウムAVE/MAコポリマー塩は、義歯安定化組成物で
唯一の接着成分として又は他の活性接着成分と併用される共接着剤として、有効
な接着量、好ましくは少くとも20重量%で利用される。好ましくは、非混合亜
鉛又はストロンチウムAVE/MAコポリマー塩は、カルシウム、ナトリウム、
マグネシウム、カリウム、アンモニウム、亜鉛、ストロンチウム及びそれらの混
合物のようなカチオンを含む混合AVE/MAコポリマー塩と共に利用される。ポリエチレングリコール
本発明組成物はポリエチレングリコールも含む。一般的に、ポリエチレングリ
コールは一般式(OCH2CH2)nOH(nは4以上である)のポリマーである

ールが最も好ましい。本発明で使用に適したポリエチレングリコールは周知であ
り、Union Carbide Corporationにより商標名“カーボワックス”(Carbowax)
として販売されているように、市販されている。
本発明で有用なポリエチレングリコールのレベルは、少くとも−140メッシ
ュU.S.B.S.シーブの粒度により特徴付けられるAVE/MAコポリマー塩を含有
した組成物に基づいている。したがって、約100以上で約600以下の平均分
子量を有するポリエチレングリコールは、本発明組成物の約20〜約90重量%
、好ましくは約30〜約80%、最も好ましくは約35〜約75%のレベルで存
在する。任意成分
本発明組成物は、安全で接着上有効な量の共接着剤を含有してもよい。本明細
書で用いられる“安全で接着上有効な量”という用語は、口腔に義歯又は歯補綴
具を接着させるために十分な量を意味する。
好ましい共接着剤には、水への暴露で膨潤して粘滑物を形成する性質を有した
、水溶性親水コロイド又はポリマーがある。このような接着物質には天然ゴム、
合成ポリマーゴム、義歯安定化組成物で常用されて本AVE/MAコポリマー塩
と適合する接着物質、合成ポリマー、糖誘導体、セルロース誘導体とそれらの混
合物がある。このような物質の例には、カラヤゴム、グアーゴム、ゼラチン、ア
ルギン、アルギン酸ナトリウム、トラガカント、メチルセルロース、アクリルア
ミドポリマー、エチレンオキシドポリマー、ポリビニルピロリドン、カチオン系
ポリアクリルアミドポリマー、カルボキシメチルセルロース、ナトリウムカルボ
キシメチルセルロース及びポリ(ビニルメチル‐エーテルマレエート)の混合部
分塩がある。ナトリウムカルボキシメチルセルロースが本発明で使用上最も好ま
しい。一般的に、共接着剤は組成物の約5〜約70重量%のレベルで存在する。
他の適切な任意成分には、着色剤;メチル及びプロピルパラベンのような保存
剤;二酸化ケイ素及び8000の平均分子量を有するポリエチレングリコールの
ような増粘剤;流動パラフィン、ペトロラタム、鉱油及びグリセリンのようなビ
ヒクルがある。8000の平均分子量を有するポリエチレングリコール、鉱油及
びワセリンが好ましい。着色剤、保存剤、増粘剤及びビヒクルは、組成物の約0
〜約20重量%のレベルで存在する。
本発明の組成物は、フレーバーを含めた官能効果をユーザーに与える1種以上
の成分約0.01〜約5%を場合により含有してもよい。適切な成分には天然又
は人工甘味剤、メントール、乳酸メチル、冬緑油、ペパーミント油、スペアミン
ト油、リーフアルコールと、冷却剤3‐1‐メントキシプロパン‐1‐2‐ジオ
ール及びパラメンタンカルボキシアミド剤、例えば参考のためその全体で本明細
書に組み込まれるWatsonらの米国特許第4,136,163号明細書で記載され
たN‐エチル‐p‐メンタン‐3‐カルボキサミドがある。
本発明の組成物は粉末、クリーム、軟膏、液体又はペースト処方のような当業
者に知られる業界認識方法で製造される。このような処方の適切な例は、双方と
も参考のためそれら全体で本明細書に組み込まれる、双方ともDhabarらの198
5年5月21日付で発行された米国特許第4,518,721号及び1985年
4月30日付で発行された米国特許第4,514,528号明細書で開示されて
