Claims (1)
- 【特許請求の範囲】 1.制御された、ほぼ均一な大きさでかつほぼ均一に分布した細孔を有する少な くとも一つの多孔領域を有する自立性薄膜構造体からなるマイクロカプセル。 2.無機材料から構成された外殻からなる封入カプセルであって、 前記外殻中の少なくとも一つの多孔領域が予め決められた、ほぼ均一な大きさ でかつほぼ均一に分布した細孔を有し、および 前記外殻が前記カプセルの内部と外部間の選択的分子バリヤーを提供する ことを特徴とする封入カプセル。 3.さらに、前記カプセルに対し追加の構造体支持を提供するリブ部材を有する 請求の範囲2記載の封入カプセル。 4.無機材料が生体適合性である請求の範囲2記載の封入カプセル。 5.無機材料が、コンフォーマル堆積を行いかつフッ化水素酸のエッチングに耐 えることができる請求の範囲2記載の封入カプセル。 6.無機材料が、多結晶シリコン、アモルファスシリコン、窒化ケイ素およびタ ングステンからなる群から選択される請求の範囲5記載の封入カプセル。 7.前記多孔領域が微小加工された薄膜のメンブレンフィルタを備えてなる請求 の範囲2記載の封入カプセル。 8.前記外殻が、環状壁と平坦な端面を有する直円筒である請求の範囲2記載の 封入カプセル。 9.前記環状壁が高い垂直アスペクト比の薄膜からなり、そして前記端面の少な くとも一つが微小加工された薄膜のメンブレンフィルタからなる請求の範囲8記 載の封入カプセル。 10.少なくとも一つの端面が、シーラントによって前記環状壁に結合されてい る請求の範囲9記載の封入カプセル。 11.前記シーラントが生体適合性の高分子材料である請求の範囲10記載の封 入カプセル。 12.前記シーラントがポリエチレンである請求の範囲11記載の封入カプセル 。 13.細孔の大きさが、前記カプセルの内部で産生される生物活性分子の自由拡 散を許容するが前記カプセルの外部に存在する免疫分子の通過を防止する請求の 範囲2記載の封入カプセル。 14.細孔の大きさが、約10オングストローム〜約1000オングストローム の範囲内である請求の範囲13記載の封入カプセル。 15.細孔の大きさが、約20オングストローム〜約50オングストロームの範 囲内である請求の範囲14記載の封入カプセル。 16.前記カプセルの内側をドナー細胞が粘着する第一有機層で処理し、そして 前記カプセルの外側を宿主細胞が粘着しない第二有機層で処理した請求の範囲1 3記載の封入カプセル。 17.前記カプセルが他の同じカプセルに結合されて複数の前記カプセルからな るシートが形成されている請求の範囲2記載の封入カプセル。 18.少なくとも一つの開口を有する外殻用のモールドを基板に形成させ、 前記モールドを犠牲層でコートし、 前記モールドが満たされるまで、前記犠牲層上に薄膜を成長させ、前記犠牲層を 除去して前記外殻を前記モールドから取り出し、 前記外殻によって画定された空間内に、入れるべき物体を入れ、次いで 前記外殻の開口を微小加工された薄膜の膜フィルターでカバーすることを含んで なる封入カプセルの加工方法。 19.前記物体が生物活性分子産生因子を含む請求の範囲18記載の方法。 20.カプセルの側壁構造体を示す個別のせん孔のパターンを基板上のマスキン グ層にパターン付けし、 前記パターンに従って、前記基板に孔を光電化学的にエッチングし、 前記孔を酸化しエッチングを行って前記孔を合体させてモールドを形成させ、 前記モールドを犠牲層でコートし、 前記犠牲層上に薄膜を成長させて前記モールドを満たし、 前記側壁構造体の一露出端に、第一の微小加工薄膜のメンブレンフィルタを形成 し、 前記犠牲層をエッチングして前記の側壁構造体とフィルタを前記モールドから取 り出し、 前記の側壁構造体とフィルタによって画定される空間内に、生物活性分子産生因 子を入れ、次いで 前記の側壁構造体の一開放端に、第二の微小加工薄膜の膜フィルターを形成する ことを含んでなる封入カプセルの加工方法。 21.カプセル内に入れるべき物体を含有する第一流体流を、前記物体の大きさ より大きい、前記カプセルの開口を通じて生成させ、次いで 前記第一流体流から第二流体流を生成させて、前記第一流中の物体が前記第二流 によって前記カプセル中に導入される、 ことからなる多孔質封入カプセルに充填する方法。 22.複数の流体アクセスポートを具備する中空ハウジング、 部分的に完成した多孔質封入カプセルを保有し、底部が多孔質の空洞を具備する トランスファーウエハを、前記ハウジングの前面に固定して、前記ハウジングを 第一領域と第二領域に分割する手段、 前記ハウジングの第一流体アクセスポートによって、前記ハウジングの前記第一 領域に接続された第一ポンプとリザーバ、および 前記ハウジングの第二流体アクセスポートによって、前記ハウジングの前記第二 領域に接続された第二ポンプを備えてなり、 前記カプセル内に入れるべき物体を含有する溶液を前記リザーバ内に入れ、次い で前記第一ポンプによって前記第一領域に供給し、次いで 第二ポンプが、前記多孔質のカプセルとウエハを通じて前記第一領域から前記第 二領域へ流体流を生成させて、前記カプセルに前記溶液中の物体を充填する ことを特徴とする多孔質封入カプセルに充填する装置。 23.マイクロカプセル内の細胞、組織または医薬組成物が産生する生物活性分 子を生理的に望ましい速度で拡散できるが前記マイクロカプセルの外部に存在す る免疫分子の通過を防止するよう、制御された均一な細孔の大きさを有する無機 の生体適合性マイクロカプセルを提供し、 所望の生物活性分子を産性できる細胞、組織または医薬組成物を前記マイクロカ プセルに充填し、次いで 前記カプセル内に入れた前記細胞、組織または医薬組成物を宿主生物に投与する ことを含んでなる生物活性分子を宿主生物に与える方法。 24.前記生物が動物である請求の範囲23記載の方法。 25.前記動物が哺乳動物である請求の範囲24記載の方法。 26.前記哺乳動物がヒトである請求の範囲25記載の方法。 27.自立性薄膜構造体を示す個別のせん孔のパターンを基板上のマスキング層 にパターン付けし、 前記パターンに従って、前記基板に孔を光電化学的にエッチングし、前記孔を酸 化しエッチングを行って前記孔を合体させてモールドを形成させ、 前記モールドを犠牲層でコートし、 前記犠牲層上に薄膜を成長させて前記モールドを満たし、 前記犠牲層をエッチングして前記側壁構造体とフィルターを前記モールドから取 り外す ことを含んでなる自立性薄膜構造体の加工方法。 28.請求の範囲27の方法に従って加工された自立性薄膜構造体。
