キレート化剤としてのN,N′−ビス(2−ヒドロキシベンジル)エチレンジア
ミン−N,N′−ジ酢酸誘導体
本発明は、下記に示す如き式(I)の〔N,N′−ビス(2−ヒドロキシベン
ジル)エチレンジアミン N,N′−ジ酢酸〕の誘導体を包含する新規かつ合成
的に有効なキレート化剤、このような化合物を含有する医薬組成物およびこれら
の誘導体の使用に関する。本発明はまた価値ある出発物質を表わしそして価値あ
る治療特性を示す〔N,N′−ビス(2−ヒドロキシベンジル)エチレンジアミ
ン N,N′−ジ酢酸〕誘導体のアルカリおよびアルカリ土類金属塩に関する。
本発明は特に次式(I):
{式中、Rは−(CO)−R1,−O−(CO)−R2,−O−(CO)−O−
であり;ここでR4およびR5は互いに独立にC1−C3アルキル又はC3−C7シク
ロアルキルであるか又は一緒になって基−(CH2)n−(nは3〜6の整数である)
を形成し;R2はC1−C6ア
ルキル、未置換フェニル又はハロゲンおよびヒドロキシから成る群から選ばれる
1〜4個の置換基により置換されたフェニルであり;R3はC1−C6アルキル又
はC3−C7シクロアルキルであり;R4はC1−C3アルキルである}
で表わされる〔N,N′−ビス(2−ヒドロキシベンジル)エチレンジアミン
N,N′−ジ酢酸〕誘導体、これらの化合物の製造方法、これらの化合物を含
有する医薬組成物およびこれらの化合物の使用に関する。
式Iの化合物は、HBEDの誘導体でありこのHBEDは〔N,N′−ビス−(2−ヒ
ドロキシベンジル)エチレンジアミン N,N′−ジ酢酸〕であり、次式II:
の生成物である。
HBEDは、米国特許 4258196において鉄オーバーロード(overload)の治療に対
するキレート化剤として記載されている。猿およびヒトにおける試験において示
されるように、HBED以上の式Iの化合物の主な利点は、遊離分子が重金属を全く
結合しないことである。従って、式Iの化合物は損失なしで腸管を通過する。遊
離分子としてそれらは重金属イオンを取り上げないけれども、それらが血流中で
循環するときそれらの吸収後驚くべき程にこの金属特性を展開させその結果それ
らは温血動物に全身的に、最も好ましくは経口的に又は非経口的に成功裏に投与
できる。経口的に投与すると、それらは
HBEDよりも著るしく高活性を示す。従って、本発明は、全身的に投与するとき有
効なキレーター(chelators)である試剤を与える。式(I)の遊離化合物は、重
金属イオンに結合しないけれども、それらは動物の血液中で強いキレート化作用
を発現する。
式(I)の範囲内の治療的に好ましいサブグループは、式I(式中、Rは式I
で定義された意味でありそしてR1は−NR4R5であり、ここでR4およびR5は互い
に独立にC1−C3アルキルであり;R2はC1−C3アルキルであり;R3はC1−
C3アルキル又はC3−C7シクロアルキルであり;ハロゲンはフッ素、塩素又は
臭素であり;シクロアルキルはシクロヘキシルであり;R4はメチルである)の
これらの代表的化合物およびその塩からなる。
しかし最も好ましい化合物は、式(I)(式中、Rは−O−CO−R2を表わしそし
てR2はC1−C6アルキルを表わしそして好ましくはC1−C3アルキルを表わす)
の化合物である。
本発明の範囲内において、以前および以後において用いられる定義は好ましく
は次の意味を有する:
C1−C6アルキルは与えられた数の炭素数によって特徴づけられた未分枝又は分
枝アルキルを表わす。典型的代表例はメチル、エチル、n−プロピル、イソプロ
ピル、n−ブチル、第二ブチル、n−ペンチル、イソペンチン、第三ペンチル、
ネオペンチル、n−ヘキシル、1−メチル−ペンチル、1,1−ジメチル−ブチ
ル、3,3−ジメチル−ブチル、2,2−ジメチル−ブチル、2−メチル−ペン
チル、3−メチル−ペンチル、および4−メチル−ペンチルである。ハロゲンは
、例えばフッ素、臭素又は塩素、好ましくはフッ素又は塩素である。C3−C7シ
クロアルキルは、3〜7個の炭素原子を有する炭素環式飽和環、例えばシクロプ
ロピル、シクロブチル、シクロペンチル、シクロヘキシル又はシクロヘプチル、
好ましく
はシクロペンチル又はシクロヘキシルを表わす。
式(I)の範囲内の最も好ましい代表化合物は次の化合物である:

ンジル)〕エチレンジアミン N,N′−ジアセタート。
式(I)の化合物は、塩を形成することができる。本発明に係る化合物の塩は
、特に医薬として許容され得る非毒性の塩である。そのような塩は特に金属塩お
よびアンモニウム塩、例えばアルカリ金属又はアルカリ土類金属塩、例えばナト
リウム、カリウム、マグネシウム又はカルシウム塩並びに関連した2価の塩、例
えば亜鉛塩、およびアンモニア又は適当な有機アミンとのアンモニウム塩であり
、特に脂肪族、脂環式、脂環式−脂肪族又は芳香脂肪族、第二又は第三モノ−、
ジ−又はポリ−アミン、および複素環式塩基との塩形成が考慮される。そのよう
なアミンは、例えば低級アルキルアミン、例えばトリエチルアミン、ヒドロキシ
−低級アルキルアミン、例えば2−ヒドロキシエチルアミン、ビス−(2−ヒド
ロキシエチル)−アミン又はトリス(2−ヒドロキシエチル)−アミン、カルボ
ン酸の塩基性脂肪族エステル、例えば4−アミノ安息香酸2−ジエチルアミノエ
チルエステル、低級アルキレンアミン、例えば1−エチルピペリジン、シクロア
ルキルアミン、例えばジシクロヘキシルアミン、又はベンジルアミン、例えばN
,N′−ジベンジル−エチレンジアミン、またピリジンタイプの塩基、例えばピ
リジン、コリジン又はキノリンである。式(I)の化合物はまた例えば、無機酸
、例えば塩酸、硫酸又はリン酸と、又は適当なカルボン酸又はスルホン酸、例え
ばメタンスルホン酸と、又はアミノ酸例えばアルギンおよびリシンと分子間(分
子内、すなわち双性イオンに反し)酸付
加塩を形成することができる。
単離および精製のため、医薬として許容し得ない塩も又使用できる。ただ、医
薬として許容できる非毒性塩が治療の適用に用いられ、そしてその理由でそれら
は好ましい。
温血動物に吸収されると、式Iの化合物は金属イオン、特に重金属イオンと安
定な錯体を形成せしめる形に変換される。重金属イオンの内、特に3+の酸化状
態のイオン、例えばAl3+又は、最も特にFe3+が言及される。
従って、式(I)の物質は全身的投与されるとき、特に経口又は経皮適用後に
、価値ある薬理特性を示す。
動物の体内で重金属イオン、特に3+の酸化状態のイオン、例えばAl3+又は最
も特にFe3+との安定な錯体を形成し得る式(I)の化合物の性質のため、式(I
)の物質は、例えば組織内の鉄−含有色素の沈着を防止し、そして鉄が生物体内
に着色した場合、例えば血色素症、血鉄症においておよびまた肝臓の硬変におけ
る鉄の除去をもたらす。式(I)の物質はまた生物体から他の重金属、例えはア
ルミナおよびクロミニウムおよび銅の除去に対して用いることができる。従って
、式(I)の化合物は透析脳障害、骨軟化症およびアルツハイマー症の場合にお
いて使用できる。
本発明に係る式(I)の化合物およびその塩は、自体公知の方法に従って化学
合成により製造できる。式(I)の化合物は、冷却しながら水素化ナトリウム又
は炭酸水素カリウムを、次式(II):
(式中、Xは水素又は基−CH2Jを表わす)
で表わされる化合物に添加し次いで生成溶液に、
(a)Xが水素を表わす場合に次式(III):
Y−CH2−R (III)
の化合物を加えるか、又は
(b)Xが基−CH2Jを表わす場合に次式(IV):
HO−C(O)−R (IV)
(ここで式(III)および式(IV)においてRは式(I)で定義された意味と
同じでありそして
Yはハロゲン、好ましくは塩素、臭素又はヨウ素を表わし、最も好ましくは臭
素又はヨウ素を表わす)
の化合物を加えることにより製造される。
式(II)(式中、Xは水素である)の化合物は、HBEDでありこれは前記の如く米国
特許 4,528,196において言及されている。式(II)(式中、Xは−CH2Jである)
の化合物は、HBEDをCH2ClJと適当な条件下で反応させることにより製造できる。
このタイプの反応は文献に記載されており、熟練した化学者に周知である。
反応は、不活性双極子非プロトン性溶剤、例えばアセトン、アセトニトリル、
ニトロメタン、ジメチルホルムアミド(DMF)、ジメチルアセタミド(DMA)、テトラ
メチル尿素、ジメチルスルホキシド(DMSO)、テトラヒドロチオフェン−1,1
−ジオキシド(スルホラン)又はエチレングリコールのジエーテル中、比較的低
温度で好ましくは約0℃で、例えば約−10°〜30℃でそして好ましくは不活性ガ
ス雰囲気下で行なわれる。
式(I)の化合物の塩は、自体公知の方法で製造できる。従って、式(I)化
合物の酸付加塩は常法により、例えば酸又は適当なアニオン交換剤で処理するこ
とにより得られる。式Iの化合物の分子
内塩(双性イオン形)は、例えば化合物又は塩、例えば酸付加塩を例えば弱塩基
を用い等電点に中和することにより、又は液体イオン交換剤で処理することによ
り形成できる。
塩は常法で遊離化合物に変換できる;金属およびアンモニウム塩は、例えば適
当な酸で処理することにより遊離化合物に変換でき、そして酸付加塩は適当な塩
基性試剤で処理することにより遊離化合物に変換できる。
出発物質、特に式(II)および式(III)の出発物質は、商業的に入手可能で
ありそして/又は公知方法により製造できる。
本発明の医薬として許容され得る化合物は、例えば医薬組成物の製造に対して
使用でき、この医薬組成物は無機又は有機の固体又は液体の医薬として許容され
得る担体と共に又は混合されて有効量の活性物質を含有する。
本発明に係る医薬組成物は、温血動物、特にヒトに対し経腸例えば経口投与お
よび非経口、例えば皮下投与に適しておりそしてそれ自体で又は医薬として許容
され得る担体と共に薬理的に活性な物質を含有する。活性物質の用量は、温血動
物の種および年令および個々の状態、治療すべき病気および投与方法に依存する
。
新規医薬製剤は、約10%〜約95%、好ましくは約20%〜約90%の活性物質を含
有する。本発明に係る医薬組成物は、例えば単位用量形態、例えば糖剤、カプセ
ル剤、坐剤又はアンプル剤であってよく、そして約 0.1g〜約3g、好ましくは
約 0.3g〜約 1.0gの活性成分を含有する。
本発明の医薬組成物は、自体公知の方法により、例えば通常の混合、造粒、溶
解又は凍結乾燥方法により製造できる。経口用の医薬組成物は、活性物質を1種
以上の固体担体を一緒にし、所望により生成混合物を造粒し次いで所望により又
は必要により、錠剤又は糖
剤コアを得るため適当な補助薬を添加した後混合物又は顆粒を加工することによ
って製造される。
適当な担体は、特に充てん剤、例えば糖、例えばラクトース、スクロース、マ
ニトールもしくはソルビトール、セルロース製品および/またはリン酸カルシウ
ム、例えばリン酸三カルシウムもしくはリン酸水素カルシウム、更に結合剤、例
えばデンプン、例えばとうもろこし、小麦、米を用いたデンプンペーストもしく
はポテトペースト、ゼラチン、トラガカント、メチルセルロース、ヒドロキシプ
ロピルメチルセルロース、ナトリウムカルボキシメチルセルロースおよび/また
はポリビニルピロリドン、所望により、崩壊剤、例えば上述のデンプン、更にカ
ルボキシメチルデンプン、架橋ポリビニルピロリドン、寒天、アルギン酸もしく
はその塩、例えばアルギン酸ナトリウムである。補助剤は、主に流動調整剤およ
び湿潤剤、例えばケイ酸、タルク、ステアリン酸、もしくはその塩、例えばステ
アリン酸マグネシウムもしくはカルシウムおよび/またはポリエチレングリコー
ルである。糖被錠剤コアには、特に所望によりアラビアゴム、タルク、ポリビニ
ルピロリドン、ポリエチレングリコールおよび/又は酸化チタンを含有する濃厚
糖液、適当な有機溶剤もしくは溶剤混合物のラッカー溶液を用いて適当なコーチ
ング(これは、所望により胃酸に抵抗する)が設けられ、あるいはまた胃酸抵抗
コーチングに対しては、適当なセルロース製品、例えばアセチルセルロースフタ
レートもしくはヒドロキシプロピルメチルセルロースフタレートの溶液が用いら
れる。例えば異なる用量の有効成分を同一視もしくは示すための着色剤もしくは
顔料が、更に錠剤もしくは糖皮、錠剤コーチングに添加される。
他の経口投与医薬組成物は、ゼラチンから造られる乾燥充てんカプセル剤であ
り、更にまたゼラチンおよび可塑剤、例えばグリセロ
ールもしくはソルビトールから造られる軟密閉カプセルである。乾燥充てんカプ
セル剤は、顆粒の形態、例えば充てん剤、例えばとうもろこしデンプン、結合剤
、例えばデンプンおよび/または潤滑剤例えばタルクまたはステアリン酸マグネ
シウムおよび所望により安定化剤との混合物中に有効成分を含有しうる。軟カプ
セル剤中に、有効成分は好ましくは適当な液体又はワックス様物質例えば脂肪油
、パラフィン油もしくは液体ポリエチレングリコール中に懸濁もしくは溶解され
、また安定剤を加えることもできる。
他の経口投与の形態は、例えば常法で製造されたシロップであり、このシロッ
プは例えば懸濁形で、そして約5%〜20%、好ましくは10%の濃度で、又は例え
ば5ml又は10mlの量で投与されたとき適当な単一用量を与える類似の濃度で活性
成分を含有する。また、例えばミルク中のセーキを製造するため例えば粉末又は
液体コンセントレートも適当である。
非経口投与に対して特に適した用量形態は、水溶性塩、例えば水溶性塩中の有
効成分の殺菌水性溶液又は殺菌水性注入懸濁液であり、これらの溶液又は懸濁液
は粘度、例えばナトリウムカルボキシメチルセルロース、ソルビトールおよび/
又はデキストランおよび所望により安定化剤を含有する。加えて、活性成分は補
助薬と共に又はそれなしで凍結乾燥形とすることができそして適当な溶剤を加え
ることにより非経口投与前に溶液にすることができる。
本発明は診断の目的のための組成物にも関しこの組成物は好ましくは水性溶液
形で又は乾燥調製品の形で式(I)の化合物の適当な金属錯体に関する。
本発明は、体内の過剰の3価のカチオン、例えばアルミニウム又は特に鉄(II
I)に関係する、前記した哺乳動物、特にヒトにおける病的状態の治療方法にも
関し、この方法は好ましくは、予防的に又
は治療的に有効な量の式(I)の化合物又はその医薬として許容され得る塩を投
与することを含んでなる。この目的に対し前記医薬組成物が用いられ、温血動物
に投与すべき本発明の化合物の約25mg/kg〜約 200mg/kg、好ましくは約20mg/
kg〜約 150mg/kgの日用量が用いられる。用量は、数回例えば3回の個々の用量
で投与できる。
全身系、例えば皮下投与に対し、式(I)の化合物のより多くの水可溶性塩形
、例えばナトリウム塩が好ましい。最も好ましい投与形態は経口である。択一的
投与形式は皮下投与である。
本発明は、特に式(I)の化合物、前述の如きその製造方法、活性成分として
式(I)の化合物を含有する医薬組成物に関する。一般的におよび実施例で述べ
るように、全身投与用医薬組成物および特に経口用量形態が好ましい。本発明は
体内の3価の金属イオンと関連する哺乳動物における病的状態の治療方法に関し
、この方法は治療的に有効な量の式(I)の化合物又はその医薬として許容し得
る塩を哺乳動物に全身的に、最も好ましくは経口的に投与することを含んでなる
。
次の実施例は本発明を説明するのに役立つが本発明の範囲を制限するものでは
ない。温度は摂氏である。例1:ビス アセトキシメチル−〔N,N′−ビス(2−ヒドロキシベンジル) 〕エチレンジアミン N,N′−ジアセタート
HBED(1g、2.57mMol)を、15mlのジメチルホルムアミドに溶解し、生成溶液
を0−4℃に冷却し次いで不活性雰囲気下で保持した。水素化ナトリウム(280mg
、6.43mMol、オイル中55−60%懸濁液)を添加し次いで生成懸濁液を0−4℃で3
0分間撹拌した。
ブロモエチルアセタート(656ml、6.7mMol)を加え次いで反応混合物を室温で5
時間撹拌した。混合物を酢酸エチル(100ml)に注ぎそして水(3×50ml)、ブラ
イン(1×50ml)で洗い、Na2SO4で乾燥し、乾燥剤を濾過後溶剤を減圧下で蒸発
させた。
残留物を70gのシリカゲルを含有するクロマトグラフィーカラムで溶離剤とし
て酢酸エチル/ヘキサン1:1容量/容量を用いて精製した。純粋な化合物を含
有する分画を一緒にし次いで真空下で濃縮した。残留物を酢酸エチル/ヘキサン
から再結晶し表題化合物を白色結晶として得る、mp 100−101 ℃。

HBED(5.7g、14.6mMol)を、100mlのDMF(N,N−ジメチルホルムアミド)に溶
解し次いでKHCO3(5.9g、58.7mMol)を加えた。混合物を50分間撹拌した。
その期間後、ブロモメチルプロピオナート(6.9ml、61.67mMol)を加え次いで混
合物を18時間撹拌した。
次いで反応混合物を酢酸エチル(100ml)中に注ぎ次いで水(2×200ml)、ブライ
ン(1×100ml)で洗い次いでNa2SO4で乾燥した。濾過後溶液を真空下で濃縮した
。残留物を酢酸エチル/ヘキサンから再結晶し、融点91−92℃を有する目的化合
物を得る。

HBED(100mg、0.255mMol)を、0−4℃に冷却した5mlの DMFに溶解し次いでア
ルゴン雰囲気下で保持した。水素化ナトリウム(23mg、0.52mMol、オイル中55−
60%)を添加し次いで生成混合物を6−8℃で18時間撹拌した。
その時間後、反応物を酢酸エチル(30ml)で希釈し次いで水(2×10ml)、ブ
ライン(1×10ml)で洗い次いでNa2SO4で乾燥した。乾燥剤を濾過後、溶剤を真
空下で蒸発させた。
残留物をシリカクロマトグラフィーカラムで精製し次いで純粋な化合物を含有
する分画を一緒にし次いで真空下で蒸発させ無色オイルとして目的生成物を得た

HBED(200mg、0.51mMol)を無水 DMFに溶解し次いでアルゴン雰囲気下で0−4
℃に冷却した。水素化ナトリウム(52mg、1.2mMol、オイル中55−60%)を添加
し次いで残りの懸濁液を30分間撹拌した。次いで2−クロロ−N,N−ジエチル
アセトアミド(168ml、1.23mMol)およびヨウ化ナトリウム(18mg、0.12mMol)を
加え次いで真空下で濃縮し目的化合物を無色オイルとして得た。
その期間後、反応混合物を酢酸エチル(50ml)で希釈し次いで飽和NH4Cl(2×
20ml)、水(1×25ml)、ブライン(1×20ml)で洗い、Na2SO4で乾燥し次いで
濾過した。溶剤を真空下で蒸発させ次いで残留物をクロマトグラフィーで精製し
た。純粋な化合物を含有する分画を一緒にし次いで真空下で濃縮し無色オイルと

HBED(200mg、0.51mmol)を、無水DMF(5ml)に溶解し次いで0−4℃に冷却した
。NaH(50mg、1.12mmol)を添加し次いで懸濁液を1
時間撹拌した。次いで2−ブロモ N−モルホリノアセトアミドを添加し次いで
反応物を室温で7時間撹拌した。
その時間後、反応混合物を酢酸エチル(30ml)で希釈し、水(2×10ml)、ブ
ライン(1×10ml)で希釈しNa2SO2で乾燥した。乾燥剤を濾過後、溶剤を蒸発さ
せ次いで残留物をカラムでクロマトグラフィー処理した。純粋な化合物を含有す
る分画を一緒にし次いで溶剤を真空下で蒸発させ目的物をオイルとして得た。 1H-NMR(CDCl3,200MHz)
