マグネシウムアルミニウムシリケート及び複数の活性成分
を含有した速溶解性剤形
技術分野
本発明は、マグネシウムアルミニウムシリケートと2種以上の薬学上許容され
る活性成分からなる、良好な味の速溶解性吸着質組成物に関する。
発明の背景
吸着質(adsorbates)は多くの化合物の望ましくない味を遮蔽するために長年に
わたり用いられてきた。この能力に関するマグネシウムアルミニウムシリケート
の使用は、例えば1987年12月8日付の米国特許第4,711,774号、
1987年12月29日付の第4,716,033号、1988年7月19日付
の第4,758,424号、1988年7月19日付の第4,758,425号
、1988年8月2日付Denick,Jrらの第4,761,274号、1988年6
月28日付Mozdaの米国特許第4,753,800号及び1964年7月14日
付Zentnerの米国特許第3,140,978号明細書で開示されている。
先行技術では薬理学上活性な化合物の味を遮蔽する上で有用としてマグネシウ
ムアルミニウムシリケートを開示しているが、複数の薬理活性剤を配合したこの
種類の速溶解性味遮蔽処方物に関する必要性がなお存在している。味遮蔽剤とし
てマグネシウムアルミニウムシリケートを含有した即時放出剤形の従来の開示は
、単一の医薬活性成分の味を遮蔽することに一般的に集中していた。これは、用
いられる苦味成分の量が増加すると(例えば、初期化合物の高い用量又は追加化
合物の混合)、用いられるマグネシウムアルミニウムシリケートの量もそれに対
応した増加を要する、という仮定に基づいている。この仮定と先行技術の集中化
と
の因果関係は、マグネシウムアルミニウムシリケートの量が増加すると起きる問
題を正しく認識すると、明らかになってくる。これらの問題には崩壊時間減少、
医薬活性成分の放出減少及び嗜好性減少(即ち、望ましくない土(clay)の味を感
じる)がある。しかしながら、本発明者らは、複数の医薬活性化合物の存在にも
かかわらず、単独の味遮蔽剤マグネシウムアルミニウムシリケートを配合した速
溶解性剤形が上記問題を避けるために十分に低いレベルであってもそれらの味遮
蔽能力をなお留めていること発見した。したがって、本発明の目的は2種以上の
薬理活性剤を含有した良味吸着質組成物を提供することである。本発明の別の目
的は、上記医薬活性剤を配合した良味速溶解性組成物を提供することである。更
に別の目的は、複数の医薬活性剤を含有した良味速溶解性薬剤の製造方法を提供
することである。
これらの目的及び追加目的は、以下の詳細な記載から容易に明らかとなるであ
ろう。
発明の要旨
本発明は:
(a)マグネシウムアルミニウムシリケートからなる土(clay)成分;
(b)鎮咳剤、抗悪心剤、栄養補給剤、緩下剤、食欲抑制剤、鎮痛剤、抗喘息
剤、抗ヒスタミン剤、うっ血除去剤、去痰剤、制酸剤、止瀉剤、H2レセプター
拮抗剤及びそれらの混合物からなる群より選択される2種以上の薬学上許容され
る活性成分の安全有効量;及び
(c)製薬上許容されるキャリア(製薬上許容されるキャリアは水溶液中で速
やかに崩壊できる);
を含んでなる医薬組成物に関する。
本発明は、本発明の組成物の安全有効量の投与からなる、感冒、呼吸障害、咳
、かぜ、感冒に伴うかぜ様及び/又はインフルエンザ症状、胃腸障害とアレルギ
ー
と関連するような症状の治療方法にも更に関する。
本明細書のすべてのパーセンテージ及び比率は他で指摘されないかぎり重量に
よる。加えて、すべての測定は他で指摘されないかぎり25℃で行われる。
本明細書で用いられる“安全有効量”とは、活性成分がヒトで過度の有害な副
作用なく妥当な利益/危険比で釣り合って指示される症状を緩和及び/又は治療
する上で有効な量である。
発明の具体的な説明
本発明の組成物は、下記のような必須成分と様々な非必須成分を含有している
。
必須成分 マグネシウムアルミニウムシリケート
本発明の第一必須成分は、安全有効量の式Al2MgO8Si2のマグネシウム
アルミニウムシリケートである。マグネシウムアルミニウムシリケートはコレラ
イナイト、サポナイト、サファイアリン、シェリダナイト、ゼベダスサイトのよ
うなスメクタイト鉱物に天然で存在し、様々な化粧品及び医薬処方剤で広く用い
られている。本例で用いられる精練されたマグネシウムアルミニウムシリケー
的化学分析は以下のとおりである:
二酸化ケイ素 63.0
酸化マグネシウム 10.5
酸化アルミニウム 10.5
酸化第二鉄 0.9
酸化カルシウム 2.3
酸化ナトリウム 2.4
酸化カリウム 1.2
強熱減量 7.5
2.6mg/m2の混合密度を有する。
本発明の組成物は、典型的には約0.01〜約50重量%、好ましくは約0.
1〜約25%、更に好ましくは約1.0〜約10%、最も好ましくは約1.0〜
約5%のマグネシウムアルミニウムシリケートを含んでいる。
マグネシウムアルミニウムシリケートは、例えば1988年8月2日付Denick
,Jrらの米国特許第4,761,274号;1988年6月28日Mozdaの米国特
許第4,753,800号;1964年7月14日Zentnerの米国特許第3,1
40,978号明細書で更に十分に記載されており、それらすべてが参考のため
本明細書に組み込まれる。薬学上許容される活性成分
本発明で有用な2種以上の薬学上許容される活性剤は、経口投与に適して薬理
作用を有する様々なグループの化合物又は物質の中から選択される。これらの薬
学上許容される活性化合物又は物質は他の必須成分と適合して、しかも他に含有
された活性物質又は化合物との組合せに適合しているべきである。薬学上許容さ
れる活性化合物又は物質は約0.01〜約75%、好ましくは約0.1〜約50
%、更に好ましくは約1.0〜約25%、最も好ましくは約1.0〜約10%の
レベルで存在する。有用な薬学上許容される活性物質又は化合物には気管支拡張
剤、食欲抑制剤、抗ヒスタミン剤、栄養補給剤(例えばビタミン、ミネラル、脂
肪酸、アミノ酸等)、緩下剤、鎮痛剤、制酸剤、H2レセプター拮抗剤、止瀉剤
、うっ血除去剤、鎮咳剤、抗悪心剤、抗微生物剤、抗真菌剤、抗ウイルス剤、去
痰剤、抗炎症剤、解熱剤、それらの薬学上許容される塩及びそれらの混合物があ
るが、それらに限定されない。
“薬学上許容される塩”という用語は、無機塩基及び有機塩基を含めた薬学上
許容される無毒性塩基から製造される塩に関する。無機塩基から誘導される塩に
はナトリウム、カリウム、リチウム、アンモニア、カルシウム、マグネシウム、
第一鉄、亜鉛、二価マンガン、アルミニウム、第二鉄、三価マンガン塩等がある
。
薬学上許容される有機無毒性塩基から誘導される塩には、一級、二級、三級及び
四級アミン、天然置換アミンを含めた置換アミン、環式アミンと塩基性イオン交
換樹脂、例えばトリエチルアミン、トリプロピルアミン、2−ジメチルアミノエ
タノール、2−ジエチルアミノエタノール、リジン、アルギニン、ヒスチジン、
カフェイン、プロカイン、n‐エチルピペリジン、ヒドラバミン、コリン、ベタ
イン、エチレンジアミン、グルコサミン、メチルグリカミン、テオブロミン、プ
リン類、ピペラジン、ピペリジン、ポリアミン樹脂等の塩がある。
本発明の組成物で有用なうっ血除去剤の例にはプソイドエフェドリン、フェニ
ルプロパノールアミン、フェニレフリン、エフェドリン、それらの薬学上許容さ
れる塩及びそれらの混合物がある。
本発明の組成物で有用な鎮咳剤の例にはデキストロメトルファン、クロフェジ
アノール、カルベタペンタン、カラミフェン、ノスカピン、ジフェンヒドラミン
、コデイン、ヒドロコドン、ヒドロモルホン、それらの薬学上許容される塩及び
それらの混合物がある。
本発明で有用な去痰剤(粘液溶解剤としても知られる)の例にはグアヤコール
酸グリセリル、抱水テルピン、塩化アンモニウム、N‐アセチルシステイン、ア
ンブロキソール、それらの薬学上許容される塩、それらの薬学上許容される塩及
びそれらの混合物がある。
本発明で有用な鎮痛剤の例にはモルヒネ、コデイン、メペリジン、ペンタゾシ
ン、プロポキシフェン、アセトアミノフェン、アロプリノール、アセチルサリチ
ル酸、コリンサリチル酸、ケトプロフェン、ケイ酸マグネシウム、フェノプロフ
ェン、イブプロフェン、インドメタシン、ナプロキセンとその他多く、更にそれ
らの薬学上許容される塩及びそれらの混合物がある。
本発明で有用な抗ヒスタミン剤の例にはブロムフェニラミン、クロルフェニラ
ミン、クレマスチン、d‐クロルフェニラミン、ジフェンヒドラミン、ドキシル
アミン、プロメタジン、テルフェナジン、トリプロリジンとその他多く、更にそ
れらの薬学上許容される塩及びそれらの混合物がある。
鎮痛剤、うっ血除去剤、抗ヒスタミン剤、去痰剤及び鎮咳剤とそれらの許容さ
れる投与量範囲は、1988年11月8日付で発行されたSunshineらの米国特許
第4,783,465号及び1986年10月28日付で発行されたSunshineら
の米国特許第4,619,934号明細書で記載されており、これらは参考のた
め本明細書に組み込まれる。
本発明で使用に適した胃腸剤の例にはアトロピン、クリジニウム及びジシクロ
ミンを含めた抗コリン作動剤;水酸化アルミニウム、次サリチル酸ビスマス、シ
メチコン、炭酸カルシウム及びマガルドレートを含めた制酸剤;シメチジン、フ
ァモチジン、ニザチジン及びラニチジンを含めたH2レセプター拮抗剤;フェノ
ールフタレイン及びカサントロールを含めた緩下剤;ジフェノキシラート及びロ
ペラミドを含めた止瀉剤がある。
更に、適切な鎮痛剤、うっ血除去剤、鎮咳剤、去痰剤及び抗ヒスタミン剤と、
気管支拡張剤、食欲抑制剤、緩下剤、制吐剤、抗微生物剤、抗菌剤、抗真菌剤、
抗炎症剤、抗ウイルス剤、解熱剤、栄養補給剤、抗コリン作動剤、制酸剤、H2
レセプター拮抗剤、止瀉剤、他の様々な胃腸化合物と、それらの許容される投与
量範囲の例は、参考のため本明細書に組み込まれるRemington′s Pharmaceutica
l Sciences,pp.734-789,791-799,861-868,907-945,875-888,1002-1034,1098-112

非必須成分
当業者であれば、他の多くの成分が本発明で含有に適していることをすぐに理
解されるであろう。非必須成分には着色剤;バニラ、チェリー、グレープ、オレ
ンジ、ペパーミント、スペアミント、アニス、ブルーベリー、ラズベリー、バナ
ナ、チョコレート、カラメル、ストロベリー、レモン、メントール及びプロスウ
ィート(ProsweetTM)MM50(天然及び人工フレーバーとプロピレングリコー
ルの組合せ、バージニア・デア・エクストラクト社(Virginia Dare Extract Co.
,Inc.),brooklyn,NYから市販)を含めた香味剤;サッカリン、デキストロース
、レブロース、スクロース、シクラメート、マンニトール及びアスパルテートと
その他多くを含めた甘味剤;キサンタンガム、アラビアガム、カルボキシメチル
セルロース、デンプン及びメチルセルロースを含めた懸濁剤;保存剤;ポリソル
ベート80、ラウリル硫酸ナトリウム、植物油及びステアリン酸マグネシウムを
含めた放出剤;及び水があるが、それらに限定されない。
本発明のもう1つの好ましい非必須成分は冷却剤又は冷却剤の組合せである。
適切な冷却剤は1979年1月23日付Watsonらの米国特許第4,136,16
3号、1980年10月28日付Rowsellらの米国特許第4,230,668号
及びRowsellらの米国特許第4,032,661号明細書で記載されたものであ
り、それらすべてが参考のため本明細書に組み込まれる。特に好ましい冷却剤は
、上記で組み込まれた米国特許第4,136,163号明細書で示されたN−エ
チル‐p‐メンタン‐3‐カルボキサミド(ステアリング・オーガニクス(Sterl
ing Organics)供給のWS‐3)である。もう1つの特に好ましい冷却剤は、3
‐1‐メントキシプロパン‐1,2‐ジオール(日本、東京の高砂香料社(Takas
ago Perfumery Co.,Ltd.,)供給のTK‐10)である。この物質は1984年7
月10日付Amanoらの米国特許第4,459,425号明細書で詳細に記載され
ており、参考のため本明細書に組み込まれる。
製造方法
本発明の良味医薬吸着質組成物は、諸成分を一緒に混合して、用いられる混合
装置のタイプを選択する上で、業界認識原理及び方法を用いて製造される。しか
しながら、マグネシウムアルミニウムシリケートと様々な多電荷カチオン系薬物
(例えば、クロルフェニラミン)との有害な相互作用‐乏しい薬物溶解性を生じ
る‐可能性のために、有効な薬物溶解性を保証する上である尺度に従うことが重
要である。このような状況はMcGinty,J.W.及びLach,J.L.,In Vitro Adsorption
of Various Pharmaceuticals to Montmorillonite,Jour.of Pharm.Sci.,65,896-
902(1976)で記載されており、参考のため本明細書に組み込まれる。本明細書で
用いられる“多電荷カチオン系薬物”とは、2以上の正電荷置換基を有する化合
物を意味する。重要なステップとして、多電荷カチオン系薬物のpKaに等しい
か又はそれ以上の範囲にpHを維持し、しかも少くとも1種の他のカチオン系化
合物の添加後まで上記多電荷カチオン系薬物の添加を保留しておく。
本発明の成分が混合された後、混合物は発泡性又は他の水分散性物質と混和さ
れ、水性液体と接触すると速やかに崩壊する剤形に乾燥される。適切な沸騰技術
は、参考のため本明細書に組み込まれるpharmaceutical Dosage Form: tablets,
Vol.I,2nd ed.,A Lieberman ed.,1989,marcel Dekker,Inc.の第6章に記載され
ている。上記混和及び乾燥プロセスは、多くの固体調剤製造技術及び装置のうち
いずれかを用いることで行われる。固体調剤処方方法は当業界で周知であり、あ
らゆる適切な方法が利用できる。更に固体調剤処方に関する情報はRemington′s
Pharmaceutical Sciences,pp.1633-1664(Alfonso R.Gennaro,editor)(18th ed.
,1990)でみられる。
一方、得られる速溶解性剤形はフリーズドライで作ってもよい。フリーズドラ
イ又は凍結乾燥は、乾燥された組成物をオープンマトリックス網状構造にするこ
とで組成物の崩壊を容易にする。ほとんどの場合、これにより水性媒体で急速に
浸透させ、製品の活性成分のデリバリーを時間的に促進する。適切なフリーズド
ライ方法は当業界で周知であり、常用されている。フリーズドライ又は真空ドラ
イのいずれか適切な慣用的方法が利用される。フリーズ及びドライの好ましい方
法では組成物をファーストフリーズし、その後約2〜約5%の最終水分まで組成
物をドライする。フリーズドライ及び生産の適切な方法は、1987年2月17
日付Daviesの米国特許第4,642,903号、1990年8月7日付Blankら
の米国特許第4,946,684号、各々1981年12月15日付及び198
3年2月1日付でGregoryらに発行された米国特許第4,305,502号及び
第4,371,516号、1993年2月23日付Ilesらの米国特許第5,18
8,825号明細書で示されており、すべて参考のため本明細書に組み込まれる
。
本発明に適用しうる速溶解性技術の1つの他の方式は、ジャンセン・ファーマ
スーティカ社(Janssen Pharmaceutica Inc.)により開発されたものであり、商品
名クイックゾルブ(QuicksolvTM)で特定される。この技術は、参考のため本明細
書に組み込まれる米国特許第5,215,756号明細書で十分に記載されてい
る。

適切なサイズの容器で、約250〜1000RPMで混和するライトニン(Li
ghtninTM)ミキサー(モデル#TS2010又は30〜50RPMにセットされ
た高剪断ホモゲナイザー)により、下記成分を加えて各々溶解させてから次
レートを用いて加熱し、溶液を77℃に保ちながら、激しく(250〜1000
RPM)30分間混和する。加熱を止め、最少で更に15分間混和しながら混合
液を室温(25℃)まで冷却させる。グリシルリチン酸モノアンモニウム、プロ
スウィートTMMM24、ソルビン酸カリウム、アスパルテーム、サッカリンナト
リウム、WS‐3及びペパーミントを加える。
別に、マンニトール、スクロース及びキサンタンガムをドライブレンドし、元
の混合液にゆっくり加える。別にマレイン酸クロルフェニラミンを十分量の水で
溶解させ、元の混合液に加える。容量を1.5000lに調整し、15分間混和
する。上記混合液とは別に、適切な容器にポリソルベート80、TK‐10及び
メントールを加える。連続的に混和しながら、この混合液を元の混合液に加える
。混和を続けながら、溶液1.5mlを型に入れ、組成物をフリーズドライ又は
真空ドライする。このプロセスでは改善された味を有する医薬吸着質組成物を与
える。成人に2錠の投与が、呼吸器病又はアレルギーの症状を治療する上で使用
上通例の慣習的な投与量である。
