フッ素化側鎖を有する3−カルボキシステロイド
本発明は、側鎖にフルオロ置換を有する新規の3−カルボキシステロイド、そ
の製造方法、該化合物を含む医薬組成物、及びテストステロン5α−還元酵素の
阻害によりアンドロゲン作用を阻害する物質としての前記化合物の使用に関する
。
あるアンドロゲン応答組織では、テストステロンの作用は主にその5α−還元
代謝物質であるジヒドロテストステロン(DHT)を介して発揮される(Bru
chowsky N.,Wilson J.D.;J.Biol.Chem.2 43
,5953,1968)。テストステロンからジヒドロテストステロンへの
変換は5α−還元酵素によって触媒され、5α−還元酵素が阻害されると、ジヒ
ドロテストステロンの生成が低下し、その特異的アンドロゲン効果が減衰するか
又は阻止される。
5α−還元酵素阻害剤は、アンドロゲン過剰状態、例えば良性前立腺肥大症及
び前立腺癌のような特定の前立腺疾患、並びにアクネ、脂漏症、女性多毛症及び
男性型禿頭症
のような特定の皮膚−毛状態を治療するための医学的用途で有用であり得る(S
iiteri P.K.,Wilson J.D.,J.Clin.Inves
t.49,1737,1970;Price V.H.,Arch.Derma
tol.III,1496,1975;Sandberg A.A.,Urolo
gy 17,34,1981)。乳癌の治療でも、5α−還元酵素阻害剤を有利
に使用し得る。なぜなら、前記腫瘍はアンドロゲンの存在によって悪化すること
が知られているからである。アンドロスト−4−エン−3−オン−17β−カル
ボン酸及びそのメチルエステル(Voigt及びHsia,Endocrino
logy,92,1216(1973);カナダ特許第970,692号)は、
5α−還元酵素阻害剤として開示された最初のステロイド化合物の一つである。
開裂環(expanded ring)に3−ケト−4,5−ジエン系を有す
る二つの5,10−セコステロイドは、マウス副睾丸5α−還元酵素の選択的阻
害剤であることが判明した(Robaireら,J.Steroid Bioc
hem.8,307−310(1977))。
(20R)−4−ジアゾ−21−ヒドロキシ−20−メ
チル−5α−プレグナン−3−オン及びその類似体は、テストステロン5α−還
元酵素の活性化酵素阻害剤(enzyme activated inhibi
tor)であると報告されている(Blohmら,Biochem.Bioph
ys.Res.Comm.95,273−80(1980);米国特許第4,3
17,817号)。基質タイプ3−ケト−D4−プロゲスチン及びアンドロゲン
に6−メチレン部分を導入することによって、別の一連の酵素制御型(enzy
me−directed)不可逆的5α−還元酵素阻害剤も製造された(Pet
rowら,Steroids 38,452−53(1981);米国特許第4
,396,615号)。
その後、3−カルボキシステロイドの不飽和誘導体が、テストステロンに対す
る不競合5α−還元酵素阻害剤として報告された(Biorg.Chem.17
,372−376(1989);欧州特許出願第0289327号;WO 92
20700号;欧州特許出願第0465123号;欧州特許出願第052848
5号;欧州特許出願第0567271号)。
4−アザステロイドは、最も研究の進んでいるステロイ
ド5α−還元酵素阻害剤である。当業界で知られているこの種の化合物は、極め
て多くの出版物及び特許で開示されている。特に、17β−アシルアミド及びそ
の代謝物質は、J.Med.Chem.27,1690−1701(1984)
,J.Med.Chem.29,2298−2315(1986),欧州特許出
願第0004949号;米国特許第4,377,584号;欧州特許出願第01

17β−アルカノイル誘導体は、J.Med.Chem.29,2298−2

本発明は、下記の式(I)
[式中、Aは結合か又は直鎖もしくは分枝鎖C1−C6アルキレン鎖を表し、Wは
基−R1、−OR2、−NR3R4、又は
を表し、但し前記式中Yは酸素又は硫黄であり、R1、R2及びR4は1個以上の
フッ素原子で置換されたC1−C10アルキル基、又は1個以上のフッ素原子で置
換されたC6−C15シクロアルキルアルキル基、又は1個以上のフッ素原子で置
換されたアリール基、又は1個以上のフッ素原子で置換されたC7−C15アリー
ルアルキル基であり、R3並びにR5、R6及びR7の各々は、独立して、水素又は
未置換もしくは1個以上のフッ素原子で置換されたC1−C10ア
ルキル基、又は未置換もしくは1個以上のフッ素原子で置換されたC6−C15シ
クロアルキルアルキル基、又は未置換もしくは1個以上のフッ素原子で置換され
たアリール基、又は未置換もしくは1個以上のフッ素原子で置換されたC7−C1 5
アリールアルキル基であり、但しWが基

はβ配置の、即ち環の平面の上方にある置換基を示す。
側鎖のキラル中心の配置は不特定である。本発明は、単一「R」及び「S」エ
ピマー、並びにこれらの「RS」混合物を包含するものとする。
本発明は、式(I)の可能な異性体及びその混合物を総て包含する。
式(I)の化合物の代謝物質及び代謝前駆体も本発明の範囲内に含まれる。
本明細書では、アルキル基並びにアリールアルキル及びシクロアルキルアルキ
ル基の脂肪族部分は、直鎖又は分枝鎖であり得る。
直鎖又は分枝鎖C1−C6アルキレン鎖は、例えば直鎖又は分枝鎖C1−C4アル
キレン鎖、例えば−CH(CH3)−、−CH(CH3)CH2CH2−、−CH2
−、−CH2CH2−であってよい。
未置換C1−C10アルキル基は、例えば直鎖又は分枝鎖C1−C5アルキル、例
えばメチル、エチル、n−プロピル、イソ−プロピル、n−ブチル、イソ−ブチ
ル、sec−ブチル、tert−ブチルもしくはネオ−ペンチル、又は直鎖C6
−C10アルキル、例えばn−オクチル又はn−ノニルであり得る。
1個以上のフッ素原子で置換されたC1−C10アルキル基は、例えば
であり得る。
未置換C6−C15シクロアルキルアルキル基は、例えばC1−C10−シクロペン
チルアルキル基又はC1−C9−シクロヘキシルアルキル基、例えば
であり得る。
1個以上のフッ素原子で置換されたC6−C15シクロアルキルアルキル基は、

本発明の化合物の医薬的に許容し得る塩は、例えばナトリウムもしくはカリウ
ムのようなアルカリ金属、カルシウムもしくはマグネシウムのようなアルカリ土
類金属、または水酸化亜鉛もしくは水酸化アルミニウムといったような無機塩基
、あるいは例えばメチルアミン、ジエチルアミン、トリメチルアミン、エチルア
ミンのような脂肪族アミン、またはピペリジンのような複素環式アミンといった
ような有機塩基のいずれかである医薬的に許容し得る塩基との塩である。
R1、R2、R3、R4、R5、R6及びR7が1個以上のフッ素原子を含む基であ
る場合は、これらのフッ素原子が同一炭素原子と結合して−CF2−(ジフルオ
ロメチレン)又は−CF3(トリフルオロメチル)基を形成するのが好ましい。
特に、好ましい化合物は、CF3(トリフルオロメチル)基を含む化合物である
。
本発明の好ましい化合物類は、Aが結合か又は基

前掲の好ましい化合物を、式(I)に関して定義した置換基について表にまと
めて示す。
式(I)の化合物又はその医薬的に許容し得る塩は、下記の式(II)
[式中、Rは直鎖又は分枝鎖C1−C6アルキル基であり、A及びWは前述の意味
を表す]で示される化合物を加水分解して、A及びWが前述の意味を表す式(I
)の化合物を生成し、所望であれば式(I)の化合物をその医薬的に許容し得る
塩に変換し、及び/又は、所望であれば式(I)の異性体の混合物を単一異性体
に分離することによって生成し得る。式(II)の化合物の加水分解は例えば、メ
タノール、エタノール、テトラヒドロフラン、ジオキサンといったような適当な
溶媒中で、水酸化カリウム、水酸化ナトリウム、水酸化リチウムのようなアルカ
リ金属水酸化物の水溶液の存在下で、数時間〜数日に渡り、約0℃〜反応混合物
還流温度の温度で、任意に窒素からなる不活性雰囲気下に実施し得る。
Rが直鎖又は分枝鎖C1−C6アルキル基であり、A及びWが前述の意味を表す
式(II)の化合物は、例えば、下記の式(III)
[式中、W及びAは前述の意味を表す]で示される化合物を、C1−C6アルキル
アルコールの存在下で一酸化炭素(CO)と反応させて、R、A及びWが前述の
意味を表す式(II)の化合物を得ることにより生成し得る。
前述の式(III)の化合物をC1−C6アルキルアルコールの存在下で一酸化炭
素(CO)と反応させる操作は、例えば、適当な有機溶媒、好ましくはジメチル
ホルムアミド(DMF)中の式(III)化合物の溶液を、トリエチルアミン(T
EA)のような有機塩基、ビス(トリフェニルホスフィン)パラジウム(II)ア
セテートもしくはビス(トリフェニルホスフィン)パラジウム(II)クロリドの
ような
パラジウム錯体、及びメタノールもしくはエタノールのようなC1−C6アルキル
アルコールで処理することにより実施し得る。
任意に、パラジウム錯体は、トリフェニルホスフィンのようなホスフィン及び
酢酸パラジウム(II)もしくは塩化パラジウム(II)のようなパラジウム塩を別
個に添加することによりその場で形成してもよい。
その後、反応混合物を一酸化炭素(CO)で数分間パージ(purge)し、
次いでCOバルーン(balloon)下で1時間〜48時間にわたり、例えば
0℃〜40℃の温度で撹拌する。類似の方法が、例えばTetr.Lett.2
6(8),1109−12(1985)に記載されている。
式(III)の化合物は例えば、下記の式(IV)
[式中、A及びWは前述の意味を表す]で示される化合物をトリフルオロメタン
スルホン酸無水物と反応させることによって生成し得る。
この反応は例えば、Synthesis 438−40(1979)に記載の
方法に従い、前述の式(IV)の化合物を含む溶液に、例えば塩化メチレン、トリ
フルオロスルホン酸無水物、好ましくはトリフルオロメタンスルホン酸無水物の
ような適当な有機溶媒中で、2,6−ジ−tert−ブチル−4−メチルピリジ
ンのような有機立体障害塩基(organic hindered base)
を加えることにより実施する。
Aが前述の意味を表し、Wが基
[式中、R5、R6、R7は前述の意味を有する]を表す式(IV)の化合物は、下
記の反応式Iで示される合成経路によって製造し得る。
反応(a)では、A及びYが前述の意味を表す式(V)の化合物を、R5及び
R6が前述の意味を表す式(VI)のカルボジイミドと反応させることにより、A
、Y、R5、R6
及びR7が前述の意味を表し、但しR5及びR6が水素ではなく、R7が水素である
式(IV)の化合物を生成し得る。
反応(b)では、A、Y、R6及びR7が前述の意味を表す式(VII)の化合物
を、R5が前述の意味を表すが水素ではなく、Xがハロゲン原子、例えばヨウ素
である式(VIII)の化合物でアルキル化することにより、A、Y、R5、R6、R7
が前述の意味を表し、但しR5が水素ではない式(IV)の化合物を生成し得る。
反応(c)では、A及びR5が前述の意味を表す式(IX)の化合物を、R6及び
Yが前述の意味を表し、但しR6は水素ではない式(X)の化合物と反応させる
ことにより、R7が水素であり、A、Y、R5、R6が前述の意味を表し、但しR6
は水素ではない式(IV)の化合物を生成し得る。
Y=Sである式(V)の化合物、式(VII)の化合物及び式(IX)の化合物は
、Zがカルボキシル官能基の任意の適当な活性化基(activating g
roup)であり得る式(XI)の化合物から生成し得る。前記基は、例えば下記
の基:
のうちのいずれかであってよい。
特に、Yが硫黄である式(V)の化合物は例えば、公知の方法に従い、式(XI
)の化合物から生成し得る。
ある方法は、例えば、Synthesis,671−2(1985)に記載の
方法に従い、Zが塩素である式(XI)の化合物を、CH2Cl2のような溶媒中で
、ジメチルチオホルムアミドの存在下、室温で例えば10分〜数時間にわたり激
しく撹拌しながら気体硫化水素と反応させる操作を
含み得る。
別の方法は、例えば、Zが
である式(XI)の化合物、即ちS−2−ピリジルチオエート誘導体を、過剰量の
硫化水素ナトリウム一水和物と反応させる操作を含み得る。この反応は、塩化メ
チレン、テトラヒドロフラン、アセトニトリルといったような溶媒中で、例えば
約0℃〜約50℃の温度で、例えば約1時間〜約48時間にわたり実施し得る。
Yが硫黄である式(V)の化合物はまた、チオカルボン酸の合成に関して文献
に記載されている一般的な方法、例えばHouben Weyl,Bd E 5
,832〜842ページ、又はDuns F.,Barton and Oll
is,Comprehensive Organic Chemistry,V
ol.3,Pergamon Press,Oxford,1979,420〜
32ページに記載の方法と類似の方法で合成し得る。
前述のように定義される式(VII)の化合物は、前述の式(XI)の化合物を、
下記の式(XII)
[式中、R6及びR7は前述の意味を表す]で示される尿素又はチオ尿素と反応さ
せることにより生成し得る。
式(IX)の化合物は例えば、式(XI)の化合物を、下記の式(XIII)
R5−NH2 (XIII)
[式中、R5は前述の意味を表す]で示されるアミンと反応させることにより生
成し得る。
式(XI)の化合物は例えば、よく知られている方法で、Yが酸素である式(V
)の化合物から生成し得る。
Yが酸素である式(V)の化合物は市販の化合物であり、又は公知の化合物か
ら公知の方法で製造できる。
Wが基NR3R4又はNHR3[式中、R3及びR4は前述の意味を表す]である
式(IV)の化合物は、例えば、Zがアミド結合及びペプチド結合の形成に有用な
カルボキシル
官能基の活性化基、例えば
である式(XI)の化合物を、下記の式(XIV)又は(XV)
[式中、R3及びR4は前述の意味を表す]で示される化合物と反応させることに
より生成し得る。
この反応は、例えば塩化メチル、酢酸エチル、アセトニトリル、テトラヒドロ
フラン、ジメチルホルムアミド、ベンゼン、トルエンといったような溶媒中で、
約0℃〜反応混合物還流温度の温度で、約1時間〜約48時間実施し得る。
Wが基OR2[式中、R2は前述の意味を表す]である式
(IV)の化合物は、例えば下記の方法で生成し得る:
a) A及びYが前述の意味を表す式(V)の化合物を、下記の式(XVI)
R2OH (XVI)
[式中、R2は前述意味を表す]で示される化合物と反応させるか、又は
b) Zがエステル結合の形成に有用なカルボキシル基の活性化官能基、例えば
である式(XI)の化合物を、R2が前述の意味を表す式(XVI)の化合物と反応さ
せる。
反応a)による式(IV)の化合物と式(XVI)の化合物との反応は、例えば塩
化メチレン、ジメチルホルムアミド、アセトニトリル、ベンゼンといったような
溶媒中で、N,N’−ジシクロヘキシルカルボジイミド又はN,N’−ジイソプ
ロピルカルボジイミドのようなカルボジイミド、及び4−N,N−ジメチルアミ
ノピリジン又は4−ピロリジ
ノピリジンのようなアシル化触媒の存在下、室温〜約50℃の温度で数時間〜2
日間にわたり実施し得る。
反応b)による式(XI)の化合物と式(XVI)の化合物との反応は、塩化メチ
レン、酢酸エチル、アセトニトリル、テトラヒドロフラン、ジメチルホルムアミ
ド、ベンゼン、トルエン、クロロホルムのような溶媒中で、任意に金属塩、例え
ば臭化第二銅(CuBr2)、トリフルオロ酢酸水銀(II)(Hg(OCOCF3
)2)、トリフルオロ酢酸銀(CF3COOAg)を存在させて、約0℃〜反応混
合物還流温度の温度で、1時間〜約48時間実施し得る。
Wが基R1[式中、R1は前述の意味を表す]である式(IV)の化合物は、Zが
基
である式(XI)の化合物を、下記の式(XVII)
R1−M(Hal)n (XVII)
[式中、Mは金属原子、特にアルカリ又はアルカリ土類金属原子、例えばリチウ
ム(Li)、マグネシウム(Mg)又はカドミウム(Cd)であり、Halはハ
ロゲン原子、好ましくは塩素、臭素又はヨウ素であり、nは0、1又は
2である]で示される化合物と反応させることにより生成し得る。
この反応は例えば、式(XI)の化合物を、例えばジエチルエーテル、テトラヒ
ドロフランのような無水溶媒中で、約−78℃〜約−30℃の温度で、約1時間
〜4時間にわたり、例えば窒素又はアルゴンのような不活性雰囲気下で、式(XV
II)の有機金属試薬、好ましくは有機マグネシウム化合物(グリニャール試薬)
又は有機リチウム化合物で処理することにより実施し得る。
式(XIV)、(XV)、(XVI)及び(XVII)の化合物は公知の化合物であり、又
は公知の化合物から公知の方法で合成し得る。
本発明の式(I)の化合物はテストステロン5α−還元酵素を特異的に阻害し
、従って効果的な抗アンドロゲン物質である。
例えば、本発明の化合物が5α−還元酵素に及ぼす阻害作用を、下記の手順に
従いin vitroで測定した。5α−還元酵素阻害のin vitroアッセイ
酵素源としての過形成ヒト前立腺のホモジェネートに由来する粒子フラクショ
ンを用いて、5α−還元酵素の阻害
を測定した。粒子フラクションは、前立腺ホモジェネートを140,000×g
で遠心分離することにより調製した。得られたペレットを数回洗浄したものを緩
衝液に再懸濁し、約10mgタンパク質/mlを含むアリコートに分けて−80
℃で貯蔵した。
5α−還元酵素のアッセイは、pH5.5の40mMトリス−HCl中で、1
mMジチオトレイトール、5mM NADPH、1mM[14C]テストステロン
、1アリコートの酵素調製物及び種々の濃度の阻害剤を含む最終量0.5mlで
実施した。37℃で30分間インキュベートした後、2mlの冷ジエチルエーテ
ルを加えて反応を終了させ、有機相を分離し、N2下で蒸発させ、酢酸エチルに
再懸濁した。
この抽出物中のテストステロン代謝物質を、展開溶媒系としてクロロホルム、
アセトン及びn−ヘキサン(2:1:2)を用いて、シリカゲルF254プレー
ト(Merck)上でTLCで分離した。プレート上の放射能を走査し、TLC
分析機(Berthold)により印字された定量プロットに基づいて分析した
。5α−還元代謝物質(5α−ジヒドロテストステロン、3α−及び3β−アン
ドロス
タンジオール)領域の14C−放射能を、テストステロン及び5α−還元代謝物質
領域の総放射能と対比させることにより、テストステロンのフラクション5α−
還元を計算した。
対照5α−還元酵素活性を50%低下させるのに必要な各化合物の濃度(IC50
)を、阻害剤濃度の対数に対して阻害率(%)をプロットすることにより決定
した。
前記検査方法で示された活性に基づいて言えば、本発明の化合物は、5α−還
元酵素の阻害によるアンドロゲン作用の低下が望まれる状態、例えば良性前立腺
肥大症、前立腺癌及び乳癌、並びに特定の皮膚−毛状態、例えばアクネ、脂漏症
、女性多毛症及び男性型禿頭症の治療に有用であり得る。従って、哺乳類、例え
ばヒト又は動物は、式(I)の化合物、又は該化合物の前述のような医薬的に許
容し得る塩を医薬的に効果的な量で投与することからなる方法により治療し得る
。
本発明の化合物の毒性はほとんど無視し得るものであるため、治療で安全に使
用できる。本発明の化合物は種々の投与形態で投与でき、例えば錠剤、カプセル
、糖衣錠又は被膜錠(film−coated tablet)、溶液
又は懸濁液の形態で経口投与するか、座薬形態で直腸投与するか、例えば静脈注
射もしくは点滴により非経口投与、例えば筋肉内投与するか、又は例えばクリー
