薬理活性を有するアミド化合物
本発明は薬理活性を有する新規化合物、その製法およびその医薬としての使用
に関する。
ヨーロピアン・ジャーナル・オブ・ファーマコロジー(European Journal of
Pharmacology)146(1988)、187−188およびナウニン・シュミー
デベルグのアーカイブズ・オブ・ファーマコロジー(Naunyn−Shumiedeberg's A
rch.Pharmacol.)(1989)、340:403−410は、非古典的5−ヒド
ロキシトリプタミンレセプター(5−HT4レセプターと表わす)を、および5
−HT3レセプター拮抗物質でもあるICS205−930がこのレセプターの
拮抗物質として作用することを記載している。
WO91/16045(スミスクライン・アンド・フレンチ・ラボラトリーズ
・リミテッド(SmithKline and French Laboratories Limited))は心房性不整
脈および卒中の治療における強心薬5−HT4レセプター拮抗物質の使用を記載
している。
EP−A−501322(グラクソ・グループ・リミテッド(Glaxo Group Li

ター拮抗物質活性を有する化合物を記載している。
今回、5−HT4レセプター拮抗特性を有するある種の新規化合物が見いださ
れてた。
したがって、本発明は5−HT4レセプター拮抗活性を有する、式(I):

アルキルまたはアルキル含有基の例は、適宜、C1、C2、C3、C4、C5、C6
、C7、C8、C9、C10、C11またはC12分枝、直鎖または環状アルキルを包含
する。C1-4アルキル基は、メチル、エチル、n−およびiso−プロピル、n
−、iso−、sec−およびtert−ブチルを包含する。環状アルキルは、
4個までの炭素原子を有する1またはそれ以上のアルキル基で所望により置換さ
れていてもよいシクロプロピル、シクロブチル、シクロベンチル、シクロヘキシ
ル、シクロヘプチルおよびシクロオクチルを包含する。
アリールは、ハロ、C1-6アルキルおよびC1-6アルコキシから選択される1ま
たはそれ以上の置換基で所望により置換されていてもよいフェニルおよびナフチ
ルを包含する。
単環式ヘテロアリールの例は、ピリジル、ピリミジル、ピラジニル、ピリル、
イミダゾリル、チエニル、フラニル、オキサゾールまたはチアゾール(あらゆる
可能な異性体)を包含する。二環式ヘテロアリールは、ベンゾフラニル、ベンゾ
チオフェニル、インドリルおよびインダゾリル、キノリルおよびイソキノリル(
あらゆる可能な異性体)を包含する。
3〜8員複素環の例は、1または2個のN、OまたはSを含み、CまたはNで
結合した環状ポリメチレン、例えばN−結合ビペリジニルまたはピロリジニルを
包含する。
ハロは、フルオロ、クロロ、ブロモおよびヨードを包含し、好ましくはクロロ
である。
式(a)のLは有利にはC−H、C−CH3、C−ClまたはC−OCH3であ
る。
式(a)のQは有利にはNR1 aである。
R1 aは好ましくは水素またはメチルまたはエチル基である。
R1 bは好ましくはメトキシである。
R3 bは好ましくはアミノである。
R4 bは好ましくはハロである。
R5 bは好ましくは水素である。
ハロである置換基はフルオロ、クロロ、ブロモおよびヨードから選択される。
ハロであるR4 aは好ましくはヨードである。
X1−(CH2)x−X2部分の適当な例は、O−(CH2)2−O、O−(CH2
)3−O、O−CH2−O、O−(CH2)2−NR、O−(CH2)2−SまたはO
−CH2−CONR(ここに、メチレン結合のいずれも所望によりメチルなどの
C1-6アルキル基で一または二置換されていてもよい)を包含する。好ましくは
、かかるX1−(CH2)2−X2はO−(CH2)2−Oである。
X1−(CH2)x−X2部分のさらに適当な例としては、O−(CH2)2−CH2
、O−(CH2)3−CH2、O−CH2−CH2、またはX1=X2=CH2であり
、メチレン結合のいずれもメチルなどのC1-6アルキル基で所望により一または
二置換されていてもよい対応する基が挙げられる。好ましくは、かかるX1−(

R1 dは好ましくは水素またはアミノである。

R4 dおよびR5 dは、しばしば、水素である。R4 dまたはR5 dがC1-6である場
合、該基は、しばしば、メチルである。
R1 eは好ましくはCF3またはエチル基である。
X5は好ましくはN、C−HまたはC−OCH3である。
R3 cは好ましくは水素である。
R4 eは好ましくは水素またはヨードなどのハロである。
CR1 fR2 f−CR3 fR4 fである場合のX6−X7の適当な例は、CH2−CH2お
よびCH=CHを包含する。しかし、X6−X7はNRz−CO、例えばNH−
COまたはNEt−COが好ましい。
Ra fは好ましくは水素である。
Rb fは好ましくは水素またはヨードなどのハロである。

R1 gおよびR2 gは、しばしば、水素であるか、またR1 gおよびR2 gはgem−ジ
メチルである。
rは、しはしば、1である。
R3 gは好ましくは水素である。
R4 gは好ましくは水素またはフルオロなどのハロである。

Zが式(h)である場合、アザ環が窒素原子で結合している場合はn1は好ま
しくは2、3または4であり、アザ環が、例えばqが2である場合の4位の炭素
原子で結合している場合はn1は1であることが好ましい。
Zが式(j)である場合、n2は好ましくはエステルまたはアミド結合間の炭
素原子数が2〜4個になるようにする。
pおよびmの適当な例は、p=m=1;p=0、m=1、p=1、m=2、p
=2、m=1である。
Zが式(k)である場合、n3は好ましくは2、3または4である。
R8およびR9は、好ましくは、共にアルキルであり、特にR8およびR9の一方
がC4またはそれ以上のアルキルである。

本発明はまた(i)、(ii)、(iii)、(iv)、(v)、(vi)または(vii
)の側鎖を有する式(I)の新規化合物を提供する。別の態様において、(i)
、(ii)または(iii)におけるピペリジン環はピロリジニルまたはアゼチジニ
ルで置換されていてもよく、および/または(i)または(ii)におけるN−置
換基はC3またはそれ以上のアルキルまたは所望により置換されてもよいベンジ
ルで置換されていてもよい。
別の態様において、式(i)または(ii)におけるN−置換基はEP−A−5
01322の式(I)およびEP−A−501322の実施例に関連して定義さ
れている(CH2)nR4で置換されていてもよく、または式(I)およびPCT
/EP93/03054(スミスクライン・ビーチャム・パブリック・リミテッ
ド・
カンパニ)の実施例に関連して定義されている置換基により置換されていてもよ
い。
式(I)の化合物の医薬上許容される塩は、塩酸、臭化水素酸、ホウ酸、リン
酸、硫酸などの通常の酸および酢酸、酒石酸、マレイン酸、クエン酸、コハク酸
、安息香酸、アスコルビン酸、メタンスルホン酸、α−ケトグルタール酸、α−
グリセロリン酸、およびグルコース−1−リン酸などの医薬上許容される有機酸
との酸付加塩を包含する。
医薬上許容される塩の例は、式(I)の化合物の第四級誘導体、例えば、化合
物Rx−T(ここに、RxはC1-6アルキル、フェニル−C1-6アルキルまたはC5- 7
シクロアルキルであり、Tは酸のアニオンに対応するラジカルである)により
四級化される化合物である。Rxの適当な例は、メチル、エチルおよびn−およ
びiso−プロピル;およびベンジルおよびフェネチルを包含する。Tの適当な
例は、クロリド、ブロミドおよびヨージドなどのハロゲン化物を包含する。
医薬上許容される塩の例は、N−オキシドなどの内部塩も含まれる。
式(I)の化合物、その医薬上許容される塩(第四級誘導体およびN−オキシ
ドを含む)はまた、水和物などの医薬上許容される溶媒和物を形成してもよく、
これらは本明細書において式(I)の化合物またはその塩について言及するいず
れの場合にも含まれる。
Zが(j)である式(I)の化合物における(CH2)n2部分は、縮合アザビ
シクロ基に関してαまたはβ配置であってもよい。
式(I)の化合物はアミド結合の形成に関する常法により調製される。
このように、さらに別の態様において、本発明は式(I)の化合物の製法であ
って、式(II):

で示される酸またはその反応性誘導体と反応させることを特徴とする製法からな
る。
式(II)の化合物と式(III)の化合物の反応はアミド結合の形成に関して当
業界で公知の条件を用いて行う。式(III)の酸の反応性誘導体は、例えば、酸
塩化物、酸無水物または反応性エステルである。
X’基は最終生成物において必要な置換基に変換可能な置換基を有するX基に
対応する核を含む。芳香族置換基におけるこのような変換の例は、水素の塩素へ
の塩素化、ニトロのアミノへの還元、脱臭素化などの脱ハロゲン化、および/ま
たは2,3−ジヒドロキシ−安息香酸の二臭化エチレンとの反応によるベンゾジ
オキサンの形成を包含する。
Z含有基における変換の適当な例は、置換および/または脱保護によるN−置
換基の通常の修飾、または2−、3−または4−置換ピペリジル目的化合物の場
合には適当なピリジル誘導体の還元を包含する。
しかし、Xおよび/またはZの工夫は、通常、エステルまたはアミド結合の前
に行う。
式(II)の化合物は式X'COOHのカルボン酸から、例えばアシルアジドを
経由して調製される(カーチウス反応(Curtius reaction))。酸X'COOH
はそれ自体当業界で公知であり、例えばXが(a)または(b)である場合、こ
のような酸はGB2125398号に記載されている。
式(III)の酸は、常法に従って、例えば対応するケトンから調製される。
式(I)においてZが(j)である場合、アザニ環式側鎖の(CH2)n2−C
OOH中間体は公知化合物であるかまたは式(IV):
で示されるケトンから常法に従って調製される。
本発明の化合物は5−HT4レセプター拮抗物質であり、かくして、一般に胃
腸障害、心血管障害およびCNS障害の治療または予防において用いられると考
えられる。
これらは過敏性腸症候群(IBS)、特にIBSの下痢症状の治療において有
効であり、即ち、これらの化合物は5−HTの効力を遮断し、小腸のニューロン
の活性化を介して腸の運動性を刺激する。IBSの動物実験において、これは、
都合よくは、排便率の減少として測定できる。これらはまたしばしばIBSと関
連する尿失禁の治療においても有用である。
これらは、他の胃腸障害、例えば上部腸運動性に関連するものにおいて、およ
び制吐剤として有用である。特に、これらは胃食道逆流疾患などの治療において
、および消化不良に有用である。制吐活性は、細胞毒性−薬剤/照射誘発性嘔吐
の公知の動物実験において測定される。
5−HTに付随する心房細動および他の心房性不整脈を予防する特定の強心剤
5−HT4レセプター拮抗物質は卒中の発生率を減少させると考えられる(適当
な動物試験法に関しては、エイ・ジェイ・カウマン(A.J.Kaumann)、1990
、ナウミン−シュミーデベルクのアーカイブス・オブ・ファーマコロジー(Naum
yn-Schmiedeberg's Arch.Pharmacol.)342、619−622参照)。
不安解消活性は海馬を介して行われるようである(デュミス(Dumuis)ら、1
988、モレキュラー・ファーマコロジー(Mol Pharmacol.),34,880−
887)。活性は標準的動物実験、社会的相互作用試験およびX−迷路試験にお
いて測定できる。
片頭痛患者は、しばしば、頭痛の兆候に先立って不安および情緒的ストレスの
症状を経験する(ザックス(Sachs)、1985、マイグレイン(Migraine),
パン・ブックス(Pan Books)、ロンドン)。片頭痛発作中および48時間以内
に、脳脊髄液中の環状AMPレベルが著しく増加することが観察されている(ウ
ェルチ(Welch)ら、1976、ヘッデイク(Headache)16,160−167
)。前兆期およびそれに付随する環状AMPレベルの増加からなる片頭痛は、5
−HT4レセプターの刺激に関連し、したがって、5−HT4拮抗物質の投与は片
頭痛発作の軽減に有効であると考えられる。
問題となる他のCNS障害は、精神分裂病、パーキンソン病およびハンチント
ン舞踏病を包含する。
本発明はまた、式(I)の化合物、またはその医薬上許容される塩と、医薬上
許容される担体とからなる医薬組成物を提供する。
このような組成物は混合により調製され、通常、経口、経鼻または経直腸など
または非経口投与により適用され、したがって、錠剤、カプセル、または経口液
体製剤、散剤、顆粒、トローチ、復元可能な散剤、経鼻用スプレー、坐剤、注射
および注入可能な溶液または懸濁液の形態である。経口投与可能な組成物が一般
的使用に関してより便利であるので好ましい。
経口投与用錠剤およびカプセルは、通常、単位用量にて投与され、通常の賦形
剤、例えば結合剤、増量剤、希釈剤、錠剤化剤、滑沢剤、崩壊剤、着色剤、矯味
矯臭剤、および湿潤剤などを含有する。錠剤は当業界における周知の方法に従っ
て、例えば腸溶コーティングで被覆してもよい。
用いるのに適した増量剤は、セルロース、マンニトール、ラクトースおよび他
の同様な薬剤を包含する。適当な崩壊剤は、デンプン、ポリビニルポリピロリド
ンおよびデンプングリコール酸ナトリウムなどのデンプン誘導体を包含する。適
当な滑沢剤は、例えばステアリン酸マグネシウムを包含する。
適当な医薬上許容される湿潤剤は、ラウリル硫酸ナトリウムを包含する。経口
液体製剤は、例えば水性または油性懸濁液、溶液、乳液、シロップ、またはエリ
キシルの形態であってもよく、あるいは使用前に水または他の適当なビヒクルで
復元する乾燥粉末としてもよい。このような液体製剤は、通常の添加物、例えば
懸濁化剤、例えばソルビトール、シロップ、メチルセルロース、ゼラチン、ヒド
ロキシエチルセルロース、カルボキシメチルセルロース、ステアリン酸アルミニ
ウムゲルまたは水素化食用油脂、乳化剤、例えばレシチン、ソルビタンモノオレ
アート、またはアカシア;非水性ビヒクル(食用油を含んでもよい)、例えばア
ーモンド油、分別ココナッツ油、グリセリン、プロピレングリコール、またはエ
チルアルコールのエステルなどの油性エステル;p−ヒドロキシ安息香酸メチル
またはプロピルまたはソルビン酸などの防腐剤、および所望により通常の矯味矯
臭剤または着色剤等を含んでもよい。
経口液体製剤は通常水性または油性懸濁液、溶液、乳液、シロップ、またはエ
リキシルの形態であってもよく、あるいは使用前に水または他の適当なビヒクル
で復元する乾燥粉末としてもよい。このような液体製剤は、例えば懸濁化剤、乳
化剤、非水性ビヒクル(食用油を含んでもよい);防腐剤、および矯味矯臭剤ま
たは着色剤等の通常の添加剤を含んでもよい。
経口組成物は、混合、充填または錠剤化の通常の方法により調製される。繰り
返し混合操作を行い、大量の増量剤を用いる組成物全体に活性成分を分散させて
もよい。このような操作はもちろん当業界で通常のものである。
非経口投与の場合、本発明の化合物および滅菌ビヒクルを含有する液体単位投
与形態を調製する。化合物は、ビヒクルおよび濃度に応じて、ビヒクル中に懸濁
または溶解できる。非経口溶液は、通常該化合物をビヒクル中に溶解し、適当な
バイアルまたはアンプル中に充填する前に濾過滅菌し、密封することにより調製
する。有利には、局所麻酔薬、防腐剤および緩衝剤などのアジュバントをビヒク
ル中に溶かす。安定性を向上させるのに、組成物をバイアルに充填し、水を真空
下で除去した後、組成物を凍結させることができる。
非経口懸濁液は、化合物をビヒクル中に溶解せずに懸濁し、滅菌ビヒクル中に
懸濁する前に酸化エチレンに接触させることにより滅菌する以外は実質的に同様
にして調製する。有利には、界面活性剤または湿潤剤を組成物中に配合して、本
発明の化合物の均質な分散を促進する。
本発明はさらに、哺乳動物、例えばヒトにおける過敏性腸症候群、消化不良、
心房性不整脈および卒中、不安および/または片頭痛の治療または予防法であっ
て、有効量の式(I)の化合物またはその医薬上許容される塩を投与することか
らなる方法を提供する。
前記障害を治療するのに有効な量は本発明の化合物の相対的効力、治療する障
害の性質および重篤度ならびに哺乳動物の体重に依存する。しかし、70kgの
成人に関する単位投与量は通常、0.05〜1000mg、例えば0.5〜50
0mgの本発明の化合物を含有する。単位投与量を1日に1回またはそれ以上、
例
えば1日に2、3または4回、より一般的には1日に1〜3回、即ち、約0.0
001〜50mg/kg/日、より一般的には0.0002〜25mg/kg/
日の範囲で投与する。
前記投与量範囲内において有害な毒学的効果は起こらない。
本発明は、また、活性な治療物質として用いるための、特に、過敏性腸症候群
、胃食道逆流病、消化不良、心房性不整脈および卒中、不安および/または片頭
痛の治療において用いるための、式(I)の化合物またはその医薬上許容される
塩を提供する。
以下の実施例は式(I)の化合物の製造を説明し、以下の記載例は中間体の製
造に関する。
好ましい化合物は式(I)に示す安息香酸核の4−位にアミノ置換基および5
位に塩素置換基が存在する例に対応する。
