新規な非環式、硫黄含有ペプチド
本発明は、ナトリウム排泄増加作用をもち、低下された塩基度をもつ新規な非
環式、硫黄含有ペプチド、それらの製造方法、および薬物における、特に血圧降
下剤としてのそれらの使用に関する。
刊行物では、ANP受容体について2つの一般的クラス、グアニル酸シクラー
ゼに結合するもの(A/B受容体)およびグアニル酸シクラーゼに結合しないも
の(C受容体)を開示している。A受容体およびB受容体は、サイズが120〜
140kDAであり、貫膜領域をもち、そして特定のグアニル酸シクラーゼを通
してcGMP系(細胞内)に対するANP効果(細胞外)を仲介する一種のタン
パク質である。両受容体タイプは、高い親和力でANPに結合するのに対し、B
NPおよびCNPに対する親和力は異なる。
ANP A/B受容体は、例えば、内皮細胞、平滑血管筋細胞において、また
糸球体の脈管膜および上皮細胞において生じる。生物学的に無症候性のC受容体
、64kDAの貫膜タンパク質のホモダイマーは、腎皮質に、また血管、内皮お
よび平滑筋細胞に広がっている。
C受容体リガンドでは、もしそれらのコア配列が保持されるならば、親和力を
失わずに広範囲に亙ってサイズを低下させ得ることは、周知である。生体内での
所見によると、C受容体が、クリアランス機能を遂行していることが示された。
麻酔されたり、または意識のあるラットへのC−ANF(4−23)の注入は、
内因性ANP血漿レベルにおける可逆的増加と、血圧の低下を引き起こす(この
関連では、Maack et al.,
Science 238 (1987),675-678;Maack et al.In.:Bio1ogical and molecular
aspects of atrial factors,edited by P.Needleman,New York 1988,p.57-7
6,参照)。これらの効果は、ANP(1−28)の注入によって誘発される効果
に匹敵する。
15個のアミノ酸まで端を切り取られたANP分子を表すC受容体リガンドが
、存在することが、同様に知られている。
また、これと関連して、コア配列Arg−Ile−Asp−Arg−Ile(
RIDRI)が,受容体の親和力に必須であることも知られている(この関連で
は、Lewicki et al.米国特許第4,757,048号、参照)。
本発明によるペプチドにおいて、最初のアルギニンの位置は、非塩基性、環式
アミノ酸により占められており、それが、驚くべきことに、親和力における大き
な増大をもたらす。
本発明は、一般式(I)の、少なくとも1つの硫黄含有基をもつ新規な非環式
ペプチドおよびそれらの塩に関する。

一般式(I)の本発明による化合物は、数個の不斉炭素原子を有する。それら
は、互いに独立して、DもしくはL型で存在し得る。本発明は、光学的鏡像体、
並びに異性体もしくはラセミ体の混合物を包含する。基A,B,D,EおよびG
は、互いに独立して、光学的に純粋に、好ましくはL型で存在するのが好ましい
。本発明の目的のためのアミノ保護基は、ペプチド化学において使用される慣用
のアミノ保護基である。
それらは、好ましくは、ベンジルオキシカルボニル、3,4−ジメトキシベン
ジルオキシカルボニル、3,5−ジメトキシベンジルオキシカルボニル、2,4
−ジメトキシベンジルオキシカルボニル、4−メトキシベンジルオキシカルボニ
ル、4−ニトロベンジルオキシカルボニル、2−ニトロベンジルオキシカルボニ
ル、2−ニトロ−4,5−ジメトキシベンジルオキシカルボニル、メトキシカル
ボニル、エトキシカルボニル、プロポキシカルボニル、イソプロポキシカルボニ
ル、ブトキシカル
ボニル、イソブトキシカルボニル、tert−ブトキシカルボニル、アリルオキ
シカルボニル、ビニルオキシカルボニル、2−ニトロベンジルオキシカルボニル

ニルを含む。
本発明の目的のために、生理学的に受容できる塩が好適である。本発明による
化合物の生理学的に受容できる塩は、鉱酸、カルボン酸もしくはスルホン酸によ
る本発明による物質の塩であってもよい。特に好適な例は、塩酸、臭化水素酸、
硫酸、リン酸、メタンスルホン酸、エタンスルホン酸、トルエンスルホン酸、ベ
ンゼンスルホン酸、ナフタレンジスルホン酸、酢酸、プロピオン酸、乳酸、酒石
酸、クエン酸、フマール酸、マレイン酸もしくは安息香酸による塩である。
本発明の目的のための塩は、付加的に、金属塩、好ましくは一価の金属塩およ
びアンモニウム塩である。例えば、ナトリウム、カリウム、リチウムのようなア
ルカリ金属塩およびアンモニウム塩が、好適である。
上記定義の範囲にはいるヒドロキシル保護基は、tert−ブトキシジフェニ
ルシリル、トリメチルシリル、トリエチルシリル、トリイソプロピルシリル、t
ert−ブチルージメチルシリル、tert−ブチルジフェニルシリル、トリフ
ェニルシリル、トリメチルシリルエトキシカルボニル、ベンジル、2−ニトロベ
ンジル、4−ニトロベンジル、2−ニトロベンジルオキシカルボニル、4−メト
キシベンジル、ホルミル、アセチル、トリクロロアセチル、2,4−ジメトキシ
ベンジル、メチルチオメチル、メトキシエトキシメチル、[2−(トリメチルシ
リル)エトキシ]メチル、ベンゾイル、4−メチルベンゾイル、4−ニトロベン
ゾイル、4−フルオロベンゾイル、4−クロロベンゾイルもしくは4−メトキシ
ベンゾイルを含むシリーズからの保護基を、一般に表す。アセチル、ベンゾイル
、ベンジルもしくはメチルベンジルが、好適である。
上記定義の範囲にはいる硫黄保護基は、一般に、アセトアミドメチル、トリフ
ェニルメチル(トリチル)、tert−ブチル、4−メトキシべンジル、n−ブ
チルチオ、tert−ブチルチオ、ジフェニルメチル、ジフェニル−4−ピリジ
ルメチル、4−メチルベンジル、3,4−ジメチルベンジル、ベンジルもしくは
4−ピコリルを含むシリーズからの保護基を表す。トリフェニルメチル、ter
t−ブチルおよびアセトアミドメチルが、好適である。

適切な溶媒は、反応条件下で変化しない通常の有機溶媒である。これらは、好
ましくは、1,3−ジメチルテトラヒドロ−2(1H)−ピリミドン(DMPH
)、1−メチルピロリドン、エーテル類、例えばジエチルエーテル、グリコール
モノメチルもしくはジメチルエーテル、ジオキサンもしくはテトラヒドロフラン
、またはベンゼン、トルエン、キシレン、シクロヘキサンもしくは石油フラクシ
ョンのような炭化水素類、または塩化メチレン、ジクロロエタン(DCE)、ク
ロロホルム、四塩
化炭素のようなハロゲン化炭化水素類、または、ジメチルスルホキシド、ジメチ
ルホルムアミド、ヘキサメチルリン酸トリアミド、酢酸エチル、ピリジン、トリ
エチルアミンまたはピコリン、のような有機溶媒を包含する。同様に、該溶媒の
混合物を使用することも可能である。ジクロロメタン、ジクロロエタン、ジメチ
ルホルムアミドもしくは1−メチルピロリドンが、特に好適である。
方法[A]における個々の活性化および結合段階のために好適なものは、ジメ
チルホルムアミド、メタン、ジエチルエーテルおよび氷酢酸である。
洗浄(方法[A])および結晶化(方法[B])に好適に使用される溶剤は、
アルコール類、例えばメタノール、エタノールもしくはn−プロパノール、塩化
メチレンのようなハロゲン化炭化水素類、酢酸エチル、氷酢酸、ギ酸、およびエ
ーテル類、例えばジエチルエーテル、ジイソプロピルエーテルである。
メタノール、氷酢酸およびジエチルエーテルは、樹脂を洗浄するために好適で
ある。
方法[A]において使用される塩基および/または補助剤は、ピペリジン、ジ
イソプロピルエチルアミンおよびテトラフルオロホウ酸 2−(1H−ベンゾト
リアゾール−1−イル)−1,1,3,3−テトラメチルウロニウムである。
方法[B]は、一般に、場合により活性化された形で存在する遊離カルボキシ
ル基をもつ1個または複数個のアミノ酸グルーピングからなる適切な断片と、遊
離アミノ基をもつ1個または複数個のアミノ酸グルーピングからなる補足的断片
との反応による、適切なより大きい断片の調
製によって起きる;続いて、保護基について適切である場合には、除去されたり

特定のペプチド結合に好適に使用される補助剤は、縮合剤であり、特に、カル
ボキシル基が、無水物として活性化されるならば、また塩基であってもよい。こ
のために使用される適切なものは、カルボジイミド類、例えば、N,N’−ジエ
チルー、N,N’−ジイソプロピルー、N,N’−ジシクロヘキシルカルボジイ
ミド、塩酸N−(3−ジメチルアミノイソプロピル)−N’−エチルカルボジイ

−オキサゾリウムもしくは過塩素酸2−tert−ブチル−5−メチルイソオキ
サゾリウムのような1,2−オキサゾリウム化合物類、または2−エトキシ−1
−エトキシカルボニル−1,2−ジヒドロキノリンのようなアシルアミノ化合物
類、または無水n−プロピルホスホン酸、またはクロロギ酸イソブチル、または
ヘキサフルオロリン酸ベンゾトリアゾリルオキシ−トリス(ジメチルアミノ)ホ
スホニウムもしくはテトラフルオロホウ酸2−(1−ベンゾトリアゾール−1−
イル)−1,1,3,3−テトラメチルウロニウム、のような慣用の縮合剤であ
る。
方法[A]は、一般に、温度範囲10℃〜35℃、好ましくは25℃で起きる
。方法[B]は、一般に、温度範囲−30℃〜35℃、好ましくは−20℃〜2
5℃で起きる。
一般式(II),(III),(IV),(V),(VI)、(VII)およ
び(VIIa)の化合物は、実質的に既知であり、そして場合
により活性化された形で存在する遊離カルボキシル基をもつ1個または複数個の
アミノ酸グルーピングからなる適切な断片と、遊離アミノ基をもつ1個または複
数個のアミノ酸グルーピングからなる補足的断片との反応によって、そして適切
なより大きい断片とのこの方法の繰り返しによって、調製することができる;続
いて、保護基について適切である場合には、除去されたり、他の保護基によって
置換されることが可能である。
特定のペプチド結合および式(VII)および(IX)の残基の導入に好適に
使用される補助剤は、縮合剤であり、特に、カルボキシル基が、無水物として活

2−ジヒドロキノリンのようなアシルアミノ化合物類、または無水n−プロピル
ホスホン酸、またはクロロギ酸イソブチル、またはヘキサフルオロリン酸ベンゾ
トリアゾリルオキシートリス(ジメチルアミノ)ホスホニウムもしくはテトラフ
ルオロホウ酸2−(1−ベンゾトリアゾール−1−イル)−1,1,3,3−テ
トラメチルウロニ
ウム、のような慣用の縮合剤である。
反応は、大気圧下および加圧もしくは減圧下の両方で(例えば0.5〜5ba
r)、好ましくは大気圧下で実施され得る。
一般式(II),(IV),(V),(VI)および(VII)の化合物は、
実質的に既知であるか、常法によって調製することができる。
一般式(VII)および(IX)の化合物は、ある場合に既知であるか、また
は基WおよびYが、また前記置換基Bを含む場合には、ある場合において新規で
あるが、次に、特に方法[A]で詳述したように、ペプチド化学において常用さ
れる方法によって調製することができる。
本発明による化合物は、予期し得なかった価値ある薬理効果スペクトルを示す
。それらは、心臓の収縮性、平滑筋の緊張力および電解質と液のバランスに影響
を与える。
それ故、それらは、病的に変化した血圧および心不全の治療、および動脈硬化
性疾患のための薬物として使用することができる。
さらに、それらは、腎不全、肝硬変、腹水症、肺浮腫、脳浮腫、妊娠の浮腫も
しくは緑内障の治療のために使用することができる。
ANP受容体リガンド:生体内試験
試験は、水以外の給餌を、前の晩に停止した雄のウイスター(Wistar)
ラット(300〜350g)を用いて実施された。その動物は、チオペンタール
(100mg/kg i.p.)により麻酔された。気管切開の後、カテーテル
が、血圧の測定のために大腿動脈中に、また物質の投与のために大腿の静脈中に
挿入された。尿は、膀胱カテーテルを通して得られた。ラットの直腸温度は、加
温ランプによって37.5±1℃に保たれた。その処置の終わりに、生理食塩水
5mg/kgの注入
を受けたラットは、続いて、試験を通じて注入速度1.2ml/hの連続的注入
を受けた。手術的処置が、平衡状態まで1時間続けられて、その後ナトリウムの
定量のための尿の収集およびcGMP排泄が開始された。各20分間、2回の対
照採取の後、試験されるべき物質が、生理食塩水中の溶液として、60分間注入
された。その試験は、1群当たり6動物により実施された。
ANP C受容体試験の実験方法
約104hANP C受容体を導入されたCHO細胞が、実験開始2日前に、
ミクロタイタープレートの1穴当たり接種される。培地は、実験開始前に吸引に
よって除去され、結合培地(DMEM/0.1%BSA)100plで置換され
る。続いて、前希釈された試験物質(最大1.0%DMSO)50μlおよび12 5
I−ANP(最終濃度約3x10-11mol/l)25,000cpmを含む結
合培地100μlが添加される。37℃60分間インキュベーション後、インキ
ュベーション培地が、吸引によって除去され、そして細胞が、氷冷DMEMで2
回洗浄され、続いて、0.2MNaOH/1%TritonX100 50μl
中で、一夜溶解される。細胞溶解液は、水100μlで希釈され、ファーマシ−
Tトレー(pharmacy T tray)中に移され、そしてシンチレータ
ー250μlが、Betaplateカウンターにおいてシンチレーションカウ
ントするために添加される。
生データは、KiおよびIC50値計算のために、コンピュータープログラムを
用いて解析される(Ki:使用された放射活性に対して修正されたIC50値;
IC50:試験されるべき物質が、放射性リガンドの特異的結合の約50%阻害を
もたらす濃度)。
エンケファリナーゼ活性の測定
アッセイ混合液は、エンドペプチダーゼ24.11 110μlの容量で、ト
リス/HClバッファーpH7.4中KCl 25mmol/lを含有する。酵
素濃度は、ラットにおける基質ANP103−126(Bacham Biochemica)が
、実験期間中60%分解されるように選択される。37℃で5分間予備循環した
後、試験されるべき物質を、10%DMSO中10μlの容量で添加する。37
℃、30分間インキュベーション後、20%トリクロロ酢酸20μlおよびアセ
トニトリル50μlを添加して、反応を停止する。10分後、15000gで3
分間、遠心を行い、上澄液50μl中に残留するANP103−126の量を、
HPLCによって定量する。試験生成物の代わりに、10%DMSO10μlを
含むサンプルを、比較のために使用する。
HPLC分析は、HypersilODS 5μmを充填されたミクロボア(
microbore)カラムにおいて実施される。プレカラム(4.6x10m
m)は、LichrosorbRP−18(5μm)を含む。移動相A:0.1
%V/V H3PO4中NaClO4 0.1mol/l;移動相B:流速0.5
ml/minで4分間にアセトニトリル濃度勾配22−50%B、205nmに
おける検出。
新規な活性物質は、既知の方法で、錠剤、コーティング錠、丸剤、顆粒剤、エ
アゾル剤、シロップ剤、乳剤、懸濁剤および液剤のような通常の製剤に、不活性
な、無毒な薬学的に適切な添加剤もしくは溶媒を用いて変換することができる。
治療学的に有効な化合物は、これらの製剤中に、各場合、全混合物の重量で約0
.5〜90%の濃度において、すなわち指示された用量範囲に達するために十分
な量において、存在する必
要がある。
製剤は、例えば、適切であれば乳化剤および/または分散剤を用いて、溶媒お
よび/または担体により活性物質を増量することによって調製されるが、例えば
、もし水が希釈剤として使用される場合には、場合により補助溶媒として有機溶
媒を使用することができる。
投与は、慣用の方法、好ましくは経口的もしくは非経口的に、特別には舌下に
もしくは静脈内に行われる。
一般に、静脈内投与で有効な結果を達成するためには、約0,001〜1mg
/体重kg、好ましくは約0.01〜0.5mg/体重kgの量を投与すること
が有利であることが証明されており、また経口投与の場合には、用量は、約0.
01〜20mg/体重kg、好ましくは0.1〜10mg/体重kgである。
それにもかかわらず、場合により、特に体重もしくは用法、薬物に対する個人
の反応、剤形の性質および投与される時間もしくは間隔に関連して、特定された
量とは差がでることも必要になろう。かくして、ある場合には、上記の最小量未
満による投与で十分なこともあり、反対に、他の場合には、述べられた上限を超
えねばならないこともある。比較的多量の投与においては、1日にわたり数回の
分割投与をすることが望ましいであろう。

p−アミノフェニル酢酸40g(265mmol)を、ジオキサン/H2O(

る。次に、二炭酸ビス(1,1−ジメチルエチル)116g(530mmol)
を導入し、そのpHを、さらに2NNaOHを添加して、pH10に保つ。20
時間後、ジオキサンをロータリーエバポレーターにおいて除去し、過剰の二炭酸

Nα−1,1−ジメチルエトキシカルボニル−S−プロリル−Nα−S−イソ
ロイシン ベンジルエステル
Nα−1,1−ジメチルエトキシカルボニルプロリン61.3g(285mm
ol)を、TBTU 100.8g(31.4mmol)と混合し、DMF10
0ml中に溶解する。活性化のために、DIEA 109ml(62.7mmo
l)を添加する。S−イソロイシン ベンジルエステル塩酸塩107g(415
mmol)を、DMF中に溶解する。30分後、2つの溶液を併せて、12時間
撹拌する。濃縮後、残渣を、
EAに採取し、洗浄(10%NaHCO3溶液で2回、H2Oで1回、飽和KHS
O4溶液で3回)する。Na2SO4で乾燥し、濃縮する。生成物を、堅い泡とし
て得る。その粗生成物を、移動相として、石油エーテル/EA混合液7:1→1
:1の1時間かけての濃度勾配、流速30ml/minを用いて、シリカゲルカ
ラム(60x10cm)で精製する。純粋な生成物98gを得る。

実施例IV
S−プロピル−Nα−S−イソロイシン ベンジルエステル、塩酸塩
実施例IIIからの化合物 76.1g(182mmol)を、氷酢酸中HC
l溶液(1.5N)の480ml(720mmol)と混合する。室温で1時間
後、その混合液を濃縮し、ジオキサンで採取し、さらに3回濃縮する。その残渣
を、H2Oから凍結乾燥すると、白色泡状物質77.2gを得る。BOCの脱離
の完了は、NMR分光法によって確認された。
実施例V
p−(1,1−ジメチルエトキシカルボニル)アミノフェニルアセチル−Nα
−S−プロピル−Nα−S−イソロイシン ベンジルエステル
実施例IIからの化合物 20.8g(82.8mmol)を、TBTU 2
7.7g(86.3mmol)と混合する。その混合物を、DIEA36ml(
207mmol)を添加したDMF200ml中に溶解する。30分後、DMF
中に溶解した実施例IVからの化合物2.45g(69mmol)を添加する。
12時間後、その混合液を、実施例IIIに記載したように精製する。表題の化
合物24.1gを得る。

4−チア−5,5,5−トリフェニルペンタノイル−Nα−S−フェニルアラ
ニン
実施例Xからの化合物5g(8.5mmol)を、ジオキサン30mlに溶解
し、H2O14mlを添加する。その混合液を、0℃に冷却し、0℃において、
H2O10ml中LiOHxH2O 530mg(12.8mmol)溶液を添加
する。その混合液を、RTまで放置し、48時間加水分解する。次いで、ジオキ
サンを蒸発させる。残渣を、pH10においてジエチルエーテルで2回、そして
pH2においてEAで3回抽
出する。Na2SO4で乾燥し、濃縮して、表題の化合物3.0gを得る。

調製実施例
実施例1(方法A)
S−(2−アセチルチオメチル−3−フェニル)プロパノイル−グリシニル−
グリシニル−S−プロリル−S−イソロイシニル−S−アスパルチル−S−アル
ギニル−S−イソロイシンアミド
合成は、Milligen 90 Continuous Flow Syn
thesizerにおいて実施される。合成用カラムは、乾燥Tentagel
−S−NH2樹脂1.6gを充填する。装填後、そのカラムに、流速20ml/
minで逆流してDMFを満たす。正常流に転換後、平衡は、10ml/min
で5分間実施される。アミノ酸単位の9バイアルが、次のように満たされる:
合成は、次のように実施される:
1.アミノ酸単位
Fmoc−アミノ酸を、DMF中0.6Mジイソプロピルエチルアミン溶液の
6.4ml中に、TBTUとともに溶解する。その溶媒をピペットで注入後、ピ
ペッティング針で、10秒間アルゴンを吹き込むことによって、22回の混合操
作を実施する。
2.自動合成
Fmoc保護基を除去するために、DMF中0.5Nピペリジン溶液を用いて
、7分間、流速7ml/minにおいて、その樹脂を洗浄する。ピペリジンを、
流速7ml/minでDMFを用いる洗浄によって洗い出す。ピペッティング針
の内外をDMFで洗浄し、活性化されたFmo
c−アミノ酸を、サンプルループ(loop)中にピペッティングする。サンプ
ル6.4ml全部を収集した後、サンプルループの内容物を、減速された流速(
6.4ml/min)でそのシステムにポンプで送る。次に、1.1分後、ポン
プによる循環を、流速7ml/minで60分間実施する。結合期間を完了後、
DMFでの洗浄除去を、流速7ml/minで10分間実施する。これと平行し
て、ピペッティング針の内外を洗浄し、合成の次の段階に備える。
合成サイクルの完了後、樹脂を、合成カラムから流出させ、1.メタノール
3x75ml、2.氷酢酸2x75ml、3.メタノール3x75ml、4.ジ
エチルエーテル3x75mlを用いて、フリット(frit)上で洗浄する。デ
シケーター中で乾燥して、ペプチド樹脂2.01gを得る。洗浄段階における容
量は、より大きいバッチに対しては、対応して増量される。
その樹脂を、TFA、チオアニソール、ジチオトレイトール、フェノールおよ
び水(5:5:2.5:5:2)の混合液20mlとともにインキュベートし、
室温で1時間切り離す。
次いで、TFA/ペプチド溶液を、吸引して濾取し、その樹脂を各回TFA5
0mlで3回洗浄する。
その濾液を、20倍量のジエチルエーテルと混合し、生成物を沈殿させる。遠
心(3000rpm、5分)後、遠心分離物を、ジエチルエーテルで洗浄し、再
び、遠心分離し、デシケーター中で乾燥する。
ポリペプチドを、アセトニトリル100mlおよびH2O 900mlの混合
液10ml中に溶解する。それを、調製用AbimedHPLCシステムにおい
て、250x32mmのカラムのLichrosor
bRP−18(10μm)で分離する。移動相は、A(アセトニトリル100m
lおよび水900ml/TFA0.5ml)およびB(アセトニトリル900m
lおよび水100ml/TFA0.5ml)である。濃度勾配液を、流速30m
l/minで流す(図1、参照)。
ペプチド178mgを得る(Rf=7.43)。
実施例2
S−(2−メルカプトメチル−3−フェニル)プロパノイル−グリシニル−グ
リシニル−S−プロリル−S−イソロイシニル−S−アスパルチル−S−アルギ
ニル−S−イソロイシンアミド
実施例Iからの化合物350mg(0.37mmol)を、メタノール14m
lに溶解する。エチルメルカプタン350μlの添加後、加水分解を、H2O
5ml中LiOHxH2O 20mg(0.48mmol)により実施する。R
Tで4時間後、その混合液を中和し、実施例1において述べた調製用HPLCシ
ステムで分離する。表題の化合物230mgを得る。

。実施例VIIIからの化合物6.05g(6.4mmol)を、DMF中に溶
解し、活性化された溶液に添加する。室温で24時間後、精製を、実施例III
に記載したように実施する。表題の化合物5.06gを得る。

3−メルカプトプロパノイル−Nα−S−フェニルアラニル−p−アミノフェ
ニルアセチル−Nα−S−プロピル−Nα−S−イソロイシニル−Nα−S−ア

実施例3からの化合物5gを、85%TFA、5%H2O、5%p−クレゾー
ルおよび5%ジチオトレイトールの混合液30ml中に溶解する。2時間後、そ
の溶液を、冷ジエチルエーテル2.5 lに添加する。ペプチドを、遠心によっ
て除去し、遠心分離物を、HV下でエーテル・
フリーにする。粗ペプチドを、20%アセトニトリル/H2Oに溶解し、30分
間に20%→60%アセトニトリルの濃度勾配により調製用HPLCシステムで
