肝モデル発明の分野
本発明は、機能性毛細胆管網を有する一次肝細胞培養物を用い、肝毒性もしく
は胆汁分泌停止能、肝性抽出能、胆汁中への肝性放出、代謝物質の産生、または
肝細胞代謝を変性する能力に関し、化合物をスクリーニングするための有用且つ
新規な方法に関する。本発明の新規な方法の使用により、動物自体で毒性検査及
び医薬開発などを研究する必要が軽減し、また化学実験に必要な動物総数も減少
する。更に、このスクリーニング手順から得られた情報は、完全動物を用いて行
う試験において生じるような他の器官及び組織との間の反応に由来する干渉を含
むことがない。発明の背景
肝毒性もしくは胆汁分泌停止能、肝性抽出能、肝性放出、代謝物質の産生、及
び肝細胞代謝を変性する化合物の能力に関し、新薬候補をスクリーニングするた
めに完全動物を使用することに対しては科学的、経済的及び倫理的な面で制約が
あり、これらに対する配慮から化合物のin vivo動態を正確に反映するようなin
vitro/ex vivo系の研究が進
められている。
細胞外マトリックスの幾何学的形態が肝細胞機能に与える影響は、Dunnらによ
ってサンドイッチ構造を用いて報告されている(J.C.Y.Dunnら、The FASEB Jour
nal,Vol 3,174-177(1992);The Journal of Cell Biology,Vol 116,1043-10
53(1992);及びBiotechnol.Prog.,Vol 7,237-245(1991))。これらの論文に
よれば、コラーゲンサンドイッチ系で培養された成熟ラット肝細胞は、正常形態
、並びに、アルブミン、トランスフェリン、フィブリノーゲン、胆汁酸及び尿素
などの生理的分泌速度を少なくとも42日間維持していた。この期間中はこれらの
細胞が、正常肝臓中で検出されるレベルと同様のアルブミンmRNAを維持している
ことも判明した。
本発明は、肝細胞に対する化合物の作用及び化合物に対する肝細胞の作用を測
定するために肝細胞培養物が有用であることを実証した。ラット及びブタの肝細
胞培養物を硬質または軟質の基質、即ち非ゲル化またはゲル化コラーゲンの上で
増殖させ、次いでコラーゲンゲルで被覆した。これらの細胞は培養物全体に行き
渡る均一な毛細胆管網の形成を開始した。典型的には、コラーゲンゲルで被覆し
た24
時間後に、形成された肝細胞培養物の特徴はほぼ完全な連続的毛細胆管網を示す
ことであった。これらの「サンドイッチされた」細胞培養物を電子顕微鏡観察に
よって形態分析すると、大部分の隣接細胞間にチャンネルが形成され、これらの
チャンネルは、結合複合体、微絨毛及び頂膜直下の末端アクチン網のような生体
の毛細胆管の特性を多く有していることが確認された。これらの細管はまた、肝
細胞培養物に加えられた生体異物を濃縮する機能を有している。従って本発明に
おいては、このようにして培養された肝細胞が多様な化合物をスクリーニングす
るために使用でき、これに伴って実験動物の犠牲を極めて少なくできることが証
明された。発明の概要
機能性毛細胆管網を有する一次肝細胞培養物を用い、肝毒性もしくは胆汁分泌
停止能、肝性抽出能、肝性放出、代謝物質の産生、または肝細胞代謝を変性する
化合物の能力に関し、化合物をin vitroスクリーニングする方法がここに提供さ
れる。該方法は、
(a)コラゲナーゼ潅流によって肝細胞を単離し、
(b)単離した細胞を肝細胞培養培地によって洗浄し、肝細
胞培養培地と約90%の等張パーコール溶液との約50:50の混合物中に細胞を懸濁
させ、約350×重力で約5分間遠心し、
(c)肝細胞培養培地1mlあたり約1〜2×106細胞の細胞濃度に調整し、コラーゲ
ンをプレコートした培養皿に約6mlの細胞懸濁液を導入し、
(d)細胞を収容した皿を約37℃、約95%相対湿度及び約5%CO2の雰囲気中でイン
キュベートし、
(e)未付着細胞を完全に除去するために約2時間後に肝細胞培養培地を交換及び
補充し、
(f)細胞が拡散し細胞間接触が成立した後に肝細胞を収容した皿を約1mlの低温
中和コラーゲン溶液で被覆し、添加した中和コラーゲンをゲル化させるべく段階
(d)の条件で再度インキュベートし、
(g)更に約5mlの肝細胞培養培地を肝細胞に添加し、
(h)光学顕微鏡によって細管網の発達が観察されるまで段階(d)の条件でインキュ
ベートし、
(i)肝細胞培養物に適合性の溶媒中に約1fMから約50mMの化合物を含有する溶液
を約1μlから約5mlの量で肝細胞培養物に添加し、段階(d)の条件で約1分間か
ら約2週間インキュベートし、
(j)細管網、細胞極性または細胞骨格構造の総体的な形態変化または構造変化を
評価するか、肝細胞内部に含まれているかもしくは培養溶液中に残留している化
合物の量を測定するか、細管網内部に取りこまれている胆汁中に分泌された化合
物を定量するか、または、肝細胞代謝の変性の程度を測定する段階を含む。発明の詳細な説明
本発明者らは、機能性毛細胆管網を有する一次肝細胞培養物を用い、肝毒性も
しくは胆汁分泌停止能、肝性抽出能、肝性放出、代謝物質の産生、または肝細胞
代謝を変性する化合物の能力をin vitro評価するための新規な化合物スクリーニ
ング手順を見出した。本発明の方法は、
(a)コラゲナーゼ潅流によって肝細胞を単離し、
(b)単離した細胞を肝細胞培養培地によって洗浄し、肝細胞培養培地と約90%の
等張パーコール溶液との約50:50の混合物中に細胞を懸濁させ、約350×重力で約
5分間遠心し、
(c)肝細胞培養培地1mlあたり約1〜2×106細胞の細胞濃度に調整し、コラーゲ
ンをプレコートした培養皿に約6mlの細胞懸濁液を導入し、
(d)細胞を収容した皿を約37℃、約95%相対湿度及び約5%
CO2の雰囲気中でインキュベートし、
(e)未付着細胞を完全に除去するために約2時間後に肝細胞培養培地を交換及び補
充し、
(f)細胞が拡散し細胞間接触が成立した後に肝細胞を収容した皿を約1mlの低温
中和コラーゲン溶液で被覆し、添加した中和コラーゲンをゲル化させるべく段階
(d)の条件で再度インキュベートし、
(g)更に約5mlの肝細胞培養培地を肝細胞に添加し、
(h)光学顕微鏡によって細管網の発達が観察されるまで段階(d)の条件でインキュ
ベートし、
(i)肝細胞培養物に適合性の溶媒中に約1fMから約50mMの化合物を含有する溶液
を約1μlから約5mlの量で肝細胞培養物に添加し、段階(d)の条件で約1分間か
ら約2週間インキュベートし、
(j)細管網、細胞極性または細胞骨格構造の総体的な形態変化または構造変化を
評価するか、肝細胞内部に含まれているかもしくは培養溶液中に残留している化
合物の量を測定するか、細管網内部に取りこまれている胆汁内に分泌された化合
物を定量するか、または、肝細胞代謝の変性の程度を測定する段階を含む。
本発明の実施によれば、硬質または軟質のコラーゲン基質上で増殖させ次いで
コラーゲンゲルで被覆した一次肝細胞は、培養物全体に行き渡る均一な毛細胆管
の形成を開始し、この培養物を化合物のスクリーニングに使用し得る。典型的に
は、被覆の24時間後に肝細胞培養物は複数の細胞を相互接続する吻合性毛細胆管
網を形成していた。これらの「サンドイッチされた」肝細胞培養物の形態分析に
よれば、大部分の隣接細胞間にチャンネルが形成されており、これらのチャンネ
ルは接合複合体、微絨毛及び頂膜直下の末端アクチン網のような生体の毛細胆管
の特徴を多く有していることが確認された。
「サンドイッチされた」肝細胞培養物を頂膜マーカーに対する抗体(アミノペ
プチダーゼN、ジペプチジルペプチダーゼIV)によって免疫染色すると、チャン
ネル構造と結合した原形質膜領域が強力な蛍光染色を示した。更に、チャンネル
の形成以前及び以後にアクチンミクロフィラメント及び微小管に対する肝細胞培
養物の二重蛍光標識を行うと、チャンネルの形成中に細胞性アクチン及びチュー
ブリンの顕著な編成替えが生じることが判明した。アクチン染色のパターンは、
張力繊維の散在性分布からチャンネル膜直下
の強力な周辺性染色に変化した。アクチン染色のこのような転位は、新しく形成
されたチャンネルを包囲して細管周囲アクチン網が形成されることを示す電子顕
微鏡写真の観察と一致した。チューブリン染色は、新しく形成された細管の前縁
に付着しこの前縁から放射状に伸びる微細管束列の存在を示していた。
ゲル化または非ゲル化のコラーゲンから成る基底マトリックスをコートした組
織培養皿上で一次肝細胞を平板培養し、細胞を拡散させて細胞間接触を生じさせ
、次いで培養物の上部を細胞外マトリックスで被覆しゲル化することによって肝
細胞培養物を調製する。肝細胞調製の少なくとも1日前に組織培養皿にコラーゲ
ンをコートする。硬質の非ゲル化増殖基質を得るために、3mg/mlのVitrogen(
登録商標)を含む200〜250μlの溶液(またはその他のコラーゲン溶液)を各100
mMの組織培養皿に添加し、テフロンポリスマンによって均等に拡散させる。コー
トされた皿を37℃で1夜維持し、次にコラーゲンを中和するために5mlの新しい
組織培養培地を添加する。ゲル化した基底マトリックスを得るためには、中和Vi
trogen(8:1:1のVitrogen(登録商標):10×ダルベッコの改質イーグル培地:0.1
NのNa0H)を調製し、上記と
同様にペトリ皿に拡散させる。プレートを37℃に30〜40分間維持してコラーゲン
ゲルを形成させ、次に新しい培地を皿に添加して37℃で保存する。使用直前に、
プレコートした培養皿から培地を吸引する。
標準コラゲナーゼ潅流法(Selgen,P.O.METHODSBIOL.,1976 13,29-83)を
用いて肝細胞を単離する。肝臓をカルシウム非含有緩衝液で8〜9分間潅流し、
次いでカルシウム及びコラゲナーゼ(0.3mg/ml)を含有する緩衝液で約10〜12分間
潅流する。次に肝臓を切開し、滅菌ナイロン網によって遊離肝細胞を未消化組織
から引き離す。次に、遊離細胞を2つの50ml遠心管に分割し、ホルモン非追加の
5%ウシ胎仔血清含有のダルベッコの改質イーグル培地で1回洗浄する。細胞ペ
レットを90%等張パーコールと培地との1:1混合物に再懸濁させ、350×gで5分間
遠心する。生存肝細胞を含有しているペレットを新しい培地に再懸濁させ、1回
洗浄し、プレコートした皿に肝細胞を9×106細胞/皿の密度で添加し、5%CO2の
インキュベーター内で37℃でインキュベートする。2〜3時間後、培地を、0.4
μg/mlのデキサメタゾン、4μg/mlのインシュリン及び20ng/mlのEGFを含む新し
い温培地に交換する。引き続いて培地を、ホルモン添加し
た新しい温培地に1日1回の割合で交換する。平板培養の24〜48時間後に、培養
培地を吸引除去し1mlの中和コラーゲン(8:1:1のVitrogen:10×ダルベッコの改
質イーグル培地:0.1NのNaOH)を添加することによって培養物をコラーゲンゲル
で被覆する。次に培養物を37℃で30〜40分間インキュベートしてマトリックスを
ゲル化させ、その後に新しい培地を皿に添加する。次に倒立顕微鏡を用いた位相
差顕微鏡観察によって毛細胆管の形成をモニターする。
本発明の1つの実施態様においては、肝細胞培養物に適合性の溶媒中に約1fM
から約1mMの化合物を含有する溶液を約1μlから約5mlの量で添加し、細胞を
収容した皿を約37℃、約95%相対湿度及び約5%CO2の雰囲気中で約5分間から
約96時間インキュベートすることによって、化合物の肝毒性または胆汁分泌停止
能を評価し得る。次いで肝細胞培養物を試験し、化合物によって肝細胞培養物に
何らかの毒作用が生じていればその程度を測定する。
このような肝細胞毒性に関する化合物のスクリーニング方法は、明瞭な輪郭の
細管が形成され従って細管の形態変化を直接観察し得るので特に好都合である。
第2の実施態様においては、「サンドイッチされた」肝
細胞培養物を用い、肝細胞抽出能力に関して、スクリーニングすべき化合物を約
1pMから約50mMの量で含有する溶液を約1μlから約5mlの量で培養物に添加し
、肝細胞を約37℃、約95%相対湿度及び約5%CO2の雰囲気中で約1分間から約4
時間インキュベートすることによって、化合物をスクリーニングする。薬剤含有
緩衝液を除去し、等張塩溶液(即ち、リン酸塩緩衝塩溶液またはハンクの平衡塩
溶液)で細胞を洗浄し、界面活性剤溶液(即ち、ドデシル硫酸ナトリウム)で細
胞を溶解し、シンチレーションカウンティング、HPLC、吸光度または蛍光定量法
のような分析手順を用いて溶解液及び出発インキュベーション溶液中の薬剤の量
を測定することによって、肝細胞内部に含まれた化合物の量を決定する。または
、蛍光化合物の場合には、細胞に結合した蛍光の直接測定によって化合物の直接
量を評価し得る。
このような肝細胞毒性に関して化合物をスクリーニングする方法は、化合物が
、I型カチオン性、II型カチオン性、アニオン性、胆汁塩及びアシアログリコプ
ロテイン受容体のような活性キャリアー系を用いて肝細胞に運搬される場合、ま
たは化合物が肝細胞に受動的に吸収される場合には、肝臓による化合物の抽出に
関する標準を示すので特に好都
合である。
本発明の第3の実施態様においては、肝細胞培養物と化合物との相互作用によ
る代謝物質産生のスクリーニングが行われる。このスクリーニング手順では、肝
細胞培養物に適合性の溶媒中に約1pMから約50mMの化合物を含有する溶液を約1
μlから約5mlの量で培養プレートに添加する。次に培養物を、約37℃、約95%
相対湿度及び約5%CO2の雰囲気中で約30分間から約2週間インキュベートする。
時々インキュベーション培地サンプルを採取し、インキュベーションが完了した
時点で親化合物の減量と新しい代謝物質の産生量とを、HPLC、TLC、質量分析法
のような化学的分析技術を用いて測定し得る。同様の方法で、細胞に結合した親
化合物と代謝物質とを界面活性剤細胞溶解物から測定し得る。
本発明の第4の実施態様において、以下のスクリーニング方法で測定される肝
細胞代謝を変性する化合物の能力も本発明の範囲に包含される。この方法では、
適合性の溶媒中に約1pMから約50mMの化合物を含有する溶液を約1μlから約5m
lの量で肝細胞培養物に添加する。代謝経路用基質を添加し、培養物を約37℃、
約95%相対湿度及び約5%CO2の
雰囲気中で約30分間から約2週間インキュベートする。化合物の存在下または非
存在下に基質から形成された産物を同定及び定量することによって肝細胞の代謝
を測定する。
本発明の第5の実施態様は、このコラーゲンサンドイッチ構造中で増殖した肝
細胞が細管内への生体異物の排泄に適応するという知見に依存する。化合物を胆
汁中に放出する肝性放出能はこのスクリーニング方法を用いて測定され得る。肝
細胞培養物に適合性の溶媒中にスクリーニングすべき化合物を約1pMから約50mM
の量で含有する溶液を約1μlから約5mlの量で肝細胞培養物に添加する。次に
培養物を、約37℃、約95%相対湿度及び約5%CO2の雰囲気中で約1分間から約
4時間インキュベートする。この時点でリン酸塩緩衝生理食塩水を用いて細胞を
洗浄し、次に細管をEDTAで破壊する。即ち、EDTAは細胞膜を破壊することなく細
管を封鎖している融合膜を破壊する。この方法では、細胞内に取込まれた物質に
妨害されることなく、細管網内に分泌された物質を測定し得る。1mg/mlのカル
ボキシフルオレセンジアセテートを含む5μlの溶液または1mg/mlのローダミン
Bを含む5μlの溶液を培地に導入したとき、これらの化合物は、「サンドイッチ
された」肝細胞の細管網内に選択的に
分泌されて濃縮することが蛍光顕微鏡によって観察された。カルボキシフルオレ
セン蛍光はエチレンジアミン四酢酸(EDTA)で処理すると遊離し、これは融合膜に
よって結合されたマクロドメインの形成と一致する。
この方法はまた、多数の化合物の同時的スクリーニングに応用できる。適合性
溶媒によって可溶化され得る任意数の化合物を肝細胞培養物に同時に提供し得る
。肝細胞毒性が観察された場合には、どの化合物またはどの化合物群が効果を発
揮したかを決定するために、引き続いてこれらの化合物の各々を個別にスクリー
ニングするとよい。
「スクリーニング」なる用語は、一連の化合物またはその混合物の潜在的特定
活性に関する判定を意味する。
「化合物」なる用語は、毒性または治療特性を潜在的に有する任意の化学物質
またはその混合物を意味する。
「連続的な」という用語は、細管が細胞培養物全体に行き渡る多細胞チャンネ
ル網を形成し、この網が培養物内部の複数の細胞を統合し単離細胞の結合体では
なく多数細胞に連続していることを意味する。
「肝毒性」なる用語は、正常肝機能に有害な影響を与えるかまたは肝細胞の生
存率を低下させる活性を意味する。
「胆汁分泌停止能」なる用語は、正常な胆汁分泌量を減少させる傾向を意味す
る。
「一次肝細胞培養物」なる用語は、in vitroに維持された正常肝細胞を意味す
る。
「機能性毛細胆管網」なる用語は、化合物の長手方向への分泌能力を維持する
肝細胞間の吻合性相互接続管状路を意味する。
「コラゲナーゼ潅流」なる用語は、肝細胞を分離及び単離するために、コラゲ
ナーゼ溶液で肝臓をin situ潅流し細胞間接触及び細胞/基底膜接触の酵素的破
壊を生じさせることを意味する。
「肝細胞培養培地」なる用語は、肝細胞の生存を支持するために必要な栄養素
及び成長因子を含有する維持溶液を意味し、例えば、0.4μg/mlのデキサメタゾ
ン、4μg/mlのインシュリン及び2ng/mlの上皮増殖因子を補充した5%ウシ胎
仔血清、非必須アミノ酸、グルタミン、抗生物質、抗菌物質を含むダルベッコの
改質培地を意味する。
「パーコール溶液」なる用語は、細胞及び細胞性成分を密度差に基づいて遠心
分離するために使用されるポリビニルピロリドン皮膜をもつコロイド状シリカの
混合物を意味
する。
「コラーゲンをプレコートした培養皿」という表現は、コラーゲンゲルを表面
に吸着しているかまたは非ゲル化コラーゲンを表面で乾燥させたプラスチックの
ウエルまたはプレートを意味する。
「約1mlの低温中和コラーゲンで被覆する」という表現は、培養皿またはウエ
ルに予め付着させた細胞の上面にコラーゲン溶液を載せる処理を意味する。
「細胞間接触」なる用語は、隣接細胞間の付着を意味する。
「細管網が発達する」という表現は、多数細胞を相互接続する連続的細管の網
目構造が形成されることを意味する。
「肝細胞培養物に適合性の溶媒」という表現または「適合性溶媒」なる用語は
、実験手順及び肝細胞維持の正常な進行中を通じて、培養された肝細胞内に好ま
しくない生化学的または形態的変化を誘起しない溶液を意味する。
「細管網の総体的な形態変化または構造変化」という表現は、細管の物理的外
観の変化及び/または膜成分もしくは細胞骨格成分即ちアクチンもしくはチュー
ブリンの再分布を意味する。
「細胞極性」なる用語は、細胞に単側性(sideness)、即ち、細管に面する側及
び類洞間隙に面する側を与える、細胞性成分または膜成分の不均等な分布を意味
する。
「肝性抽出能」なる用語は、化合物が肝臓内部に吸収される傾向を意味する。
「胆汁中への化合物の肝性放出能」なる表現は、化合物が毛細胆管内に分泌さ
れる傾向を意味する。
「化合物からの代謝物質の産生をスクリーニングする」という表現は、肝臓内
の酵素的経路によって化学的に修飾され得る化合物の能力を判定することを意味
する。
「肝細胞代謝を変性させる化合物の能力」という表現は、肝臓内の生化学的経
路と相互作用して代謝産物の定量的または定性的変化を生じさせる化合物の能力
を意味する。
「肝細胞代謝の変性度を測定する」という表現は、放射性標識基質もしくは蛍
光基質または化学分析のような分析技術を用い、生化学的経路による産物形成の
定量的または定性的変化を測定することを意味する。
このスクリーンに使用される化合物は、任意の標準分析技術を用いて定量でき
る。使用できる標準分析技術の非限定例としては、高性能液体クロマトグラフィ
ー(HPLC)、ガ
スクロマトグラフィー(GC)、質量分析法(MS)、電気泳動、光学吸収、蛍光定量、
放射性同位体の定量などがある。
細管網、細胞極性または細胞骨格構造の総体的な形態変化もしくは構造変化は
、光学顕微鏡観察、蛍光顕微鏡観察、電子顕微鏡観察、タンパク質の化学的修飾
、抗体標識などを用いて評価し得る。実施例 実施例1 ラット肝細胞に対するコラーゲン被覆の効果
乾燥した非ゲル化コラーゲンまたはゲル化したコラーゲンをコートした組織培
養プレート上で増殖させた一次ラット肝細胞の上面で、中和ウシ皮膚コラーゲン
(Vitrogen(登録商標))をゲル化させる実験を実施した。被覆コラーゲンマトリ
ックスのゲル化後、組織培養培地を皿に添加し、95%空気5%CO2の湿潤インキ
ュベータ内で37℃で細胞をインキュベートした。1日後の細胞を位相差顕微鏡で
観察すると、複数の細胞にわたって伸長しており細胞を包囲する屈折チャンネル
のように見える精密な細管チャンネル網が発達していた。電子顕微鏡及び蛍光顕
微鏡によって観察するとこれらのチャンネルが毛細胆管であることが確認された
。
被覆された肝細胞を電子顕微鏡で観察すると、両側に融合膜が結合し細管膜の直
下に緻密なアクチン網を有している隣接細胞間の微絨毛内包チャンネルが存在し
ていた。蛍光標識抗体を使用して蛍光顕微鏡で観察すると、天然の肝臓で観察さ
れるのと同様に細管チャンネル内に頂膜マーカーであるアミノペプチダーゼN及
びジペプチジルペプチダーゼが存在することが確認された。また、ファロイジン
の蛍光性接合体を用いると、天然の肝臓と同様に肝細胞中のアクチンが細管周縁
領域に高度に局在していることが判明した。電子顕微鏡観察によれば、コラーゲ
ンで被覆した細胞の形態は、ゲル化コラーゲンから成る基底膜上で増殖させたと
きのほうが乾燥コラーゲン上で増殖させたときよりも良好であることが判明した
。位相差顕微鏡観察によれば、ラット尾コラーゲン、ウシ鍵コラーゲン及びウシ
皮膚コラーゲンはいずれも基底膜及び被覆マトリックスの双方に有効なマトリッ
クスであることが判明し、Engelbreth-Holm-Swarmマウス腫瘍(MatrigelTM)由来
の細胞外マトリックスも被覆マトリックスとして有効であることが判明した。実施例2 ブタ肝細胞に対するコラーゲン被覆の効果
乾燥した非ゲル化コラーゲンまたはゲル化したコラーゲンをコートした組織培
養プレート上で増殖させた一次ブタ肝細胞の上面で、中和コラーゲン(Vitrogen
(登録商標))をゲル化させる実験を実施した。被覆コラーゲンマトリックスのゲ
ル化後、組織培養培地を皿に添加し、95%空気5%CO2の湿潤インキュベータ内
で細胞を37℃でインキュベートした。1日後の細胞を位相差顕微鏡で観察すると
、ラット肝細胞で観察されたものと同様に複数の細胞にわたって伸長した精密細
管チャンネル網が発達していることが判明した。実施例3 コラーゲンで被覆された肝細胞は生体異物を毛細胆管に濃縮する
コラーゲンでサンドイッチされた肝細胞が、ジカルボキシフルオレセンを細胞
内部から細管に運搬し化合物を細管に濃縮する能力を有していることを実証する
実験を実施した。コラーゲン被覆の5日後に、培養物を1〜10μg/mlのジカルボ
キシフルオレセンジアセテートを含有するハンクの平衡塩溶液中で37℃でインキ
