試験管内受精を改良するための二成分フィブリングルー組成物
本発明は、試験管内受精を改良するための成分AおよびBからなる二成分フィ
ブリングルー組成物に関する。
1987年、最初の試験管内受精(in vitro fertilization)ベビーがイング
ランドに誕生した。それから後、試験管内受精は、広く世界中で行われるように
なった。試験管内受精の適用症には、ほとんど全ての不妊症が含まれる。
試験管内受精が導入されてから、この方法のあらゆる面について、例えば排卵
誘発や卵採取のような技術が改良されてきた。ただし、一つの領域のみ、例外的
に深刻な問題と複雑な要因とを残している。その領域とは、胎芽の子宮への着床
である。子宮内への胚芽の着床の成功率は、胎芽当り8乃至9%、あるいは妊娠
率で20乃至30%(一度の試験管内受精で、一般に3または4胎芽を移植する
ため)程度に留まっている。高い着床率を得るために考えられるのは、この失敗
例において若い胎芽(2乃至8細胞)の子宮への移植が重要な役割をしうること
である。このような胎芽は、その段階で着床するには充分に成熟していない。他
方、子宮外で長時間胎芽を培養すると、胎芽を損傷し、それらの30%程度しか
通常の着床が起こる発生段階である未分化胚芽細胞期へ発達しない。従来の技術
で知られているフィブリングルーを用いて着床の改良する試みは、失敗に終わっ
ている。その考え方は、このフィブリングルーを用いて胎芽を子宮の壁(子宮内
膜)に付着させるというものである。
本発明の目的は、試験管受精を改良する組成物および方法を提供することであ
る。
本発明に従うと、上記の問題は、下記成分AおよびBからなる二成分フィブリ
ングルー組成物により解決することができる。
−成分Aは、フィブリノーゲンおよびプロテアーゼ阻害剤からなり、
−成分Bは、フィブリノーゲンを特異的に切断し、フィブリンポリマーの形成
を起こすことができる蛋白分解酵素からなり、そして
−哺乳類の胎芽細胞を培養するための因子が、成分AおよびBの一方または両
方に含まれている。
好ましくは、全血または血漿の寒冷沈降物が、成分Aのフィブリノーゲン含有
分画として用いられる。なお、市販されている寒冷沈降物の製品も利用できる。
寒冷沈降物を、第2因子と第5因子の間に濃縮すると都合がよい。
寒冷沈降物は、好ましくはウイルス不活性化されている。ウイルス不活性化の
操作は、例えば、PCT/EP91/00503号明細書に記載されている。こ
の方法の基本原理は、寒冷沈降物を特定の洗浄剤で処理して、その後、洗浄剤を
寒冷沈降物から除去することである。
成分Aに存在するプロテアーゼ阻害剤は、好ましくはアプロチニンである。そ
の濃度は、成分Aの総量に対して、10000U/ml以下であることが好まし
い。アプロチニンは市販れており、TrasylolRあるいはAntagosanRとの商標の製
品が利用できる。また、トラネキサム酸(4−(アミノメチル)シクロヘキサン
カルボン酸)やその許容できる塩も、アプロチニンの代わりに、あるいはそれと
組み合わせて用いるのに適当な薬剤である。
本発明の二成分フィブリングルー組成物の第二成分Bは、フィブリノーゲンを
特異的に切断できる蛋白分解酵素の溶液で調製される。好ましくは、トロンビン
が使用される。好ましくはトロンビンは、ヒトまたはウシのような哺乳類の血漿
から単離される。トロンビンは、凍結乾燥状態での扱いが可能である。トロンビ
ンの再構成は、塩化カルシウムを含む溶液中で起こる。フィブリノーゲンの切断
に特異的なプロテアーゼの濃度は、胎芽を子宮に移植する方法により決定する。
基本的には、遅く作用する二成分フィブリングルー組成物が必要とされる場合に
は、トロンビンの濃度を低くしなければならない。これに対して、速く作用する
フィブリングルーが必要である場合には、トロンビンの濃度を高くする(速効性
フィブリングルー)。トロンビンの典型的な濃度範囲は、遅い作用のフィブリン
グルーにおいて、0.4乃至4ユニット/mlである。
本発明のフィブリングルーの他の好ましい態様は、成分Bとして、蛇毒から単
離された蛋白分解酵素を含む。蛇毒酵素としては、南アメリカの毒蛇であるBoth
rpos moujiniから単離したバトロキソビン(batroxobin)を利用できる。この毒
素は化学的には、約36000の分子量を有する単鎖のグリコペプチドである。
この蛇毒は、デフィブラーゼ(DefibraseR)の名称で知られており、フィブリノ
ーゲンのala/16-arg/17 におけるグルー結合の切断を起こす。その結果、フィブ
リノペプチドAの放出と、単量性フィブリンIの形成が起きる。
寒冷沈降物が自家組織由来で調製された場合には、デフィブラーゼやレプチラ
ーゼ(Reptilase)のようなフィブリノーゲンをフィブリンに変換する蛇毒の使
用が好ましい。また、蛇毒の使用により、ヒトの全血製品の使用を回避すること
もできる。
本発明に従う場合、本発明のフィブリングルー中には、哺乳類の胎芽細胞を培
養するための因子が存在していなければならない。この因子を存在させるために
は、成分AおよびBの一方または両方に、因子を混合すればよい。あるいは、因
子を液体中に溶かして、その液体を成分AおよびBの一方または両方と混合して
もよい。哺乳類の胎芽細胞を培養するための好ましい因子は、塩化カルシウム、
塩化カリウム、硫酸マグネシウム、塩化ナトリウム、重炭酸ナトリウム、リン酸
二水素ナトリウム、D−グルコースおよびフェノールを含む。溶液は、好ましく
は約7.2のpHを有する。ピルビン酸と抗生物質(例、ゲンタマイシン)をさ
らに添加することも有効である。
ウイルスが存在しない条件下で成分Aを調製するための代表的な方法について
は、PCT/EP91/01850号明細書に示唆されている。
典型的な方法においては、寒冷沈降物とトロンビンを胎芽を培養するための培
地中に溶解する。もちろん、両方の成分は凝血反応を防止するために、分離して
おく。培養のための培地の好ましい組成として、二種類を以下に示す。一つは、
EBSSと呼ばれるもので、Gibco社から市販されている。EBSSは、アール
の平衡塩類溶液(Earle's Balanced Salt Solutions)の略語である。これは、
液体または粉末として購入することができる。液体は、無水塩化カルシウムを0
.2g/l、塩化カリウムを0.4g/l、MgSO4・7H2Oを0.2g/l
、塩化ナトリウムを6.8g/l、重炭酸ナトリウムを2.2g/l、NaH2
PO4・2H2Oを0.158g/l、さらにD−グルコースを1.0g/lおよ
びフェノールレッドを0.01g/lの量で含む。粉末状のものを1リット
ルに溶解した溶液は、無水塩化カルシウムを0.20g/l、塩化ナトリウムを
0.4g/l、無水硫酸マグネシウムを0.0977g/l、塩化ナトリウムを
6.80g/l含み、NaH2PO4・2H2O、D−グルコースおよびフェノー
ルレッドの量は、液体と同じ含有量である。ピルビン酸を好ましくは0.015
g/lの量で、ゲンタマイシンを50mg/lの量で更に添加することも有効で
ある。
試験管内受精を実施する分野によって、特定の要求に従うように培地を変更す
ることができる。例えば、繁殖のような農業分野においては、要求が異なる。上
記の培養のための培地は、ヒトにおける試験管内受精においても利用できる。
試験管内受精の成果の向上に失敗していた従来のフィブリングルー製品と比較
すると、本発明のフィブリングルーの有利な点は、その抗繊維素溶解作用が高く
なっているため、一方では胎芽の子宮への固着を防止または遅延し、他方では凝
血を阻害する。本発明の二成分フィブリングルー組成物は、胎芽が子宮壁に留ま
り永久的に固着するまで、子宮内の環境下で胎芽が生存することを可能にする。
胎芽の生存は、胎芽細胞培養のための因子を含む培地によって維持される。驚く
べきことに、アプロチニンのようなプロテアーゼ阻害剤は、高濃度でも胎芽に影
響を与えずに、グルーの分解を防止する。さらに、本発明の他の好ましい態様で
は、二成分フィブリングルー組成物は、等浸透圧性である。このため、従来技術
のグルーが高浸透圧性であることによる不利な点が解消されている。高浸透圧性
であると、乾燥現象による胎芽の死が起こる。
本発明のフィブリングルーの有利な特徴は、試験管内での胎芽の成長に必須で
ある塩と栄養成分の存在である。また、本発明の組成物は、胎芽あるいは子宮に

推奨できる。
フィブリノーゲン源としては、寒冷沈降物の使用の方が、フィブリノーゲン濃
縮物よりも有利である。すなわち、前者の方は、接着に必要とされるフィブロネ
クチンやフォンビルブラント因子のような成分を含むからである。さらに、寒冷
沈降物は、胎芽の発達や定着を促進する蛋白質や低分子物質のような他の薬品も

本発明の二成分フィブリングルー組成物の調製のためには(上記の実験におい
て使用できるように)、寒冷沈降物を上記の培養用の培地中に溶解する。次に、
アプロチニンまたはトラネキサム酸を寒冷沈降物に加える。成分Aを含む寒冷沈
降物中のアプロチニンの濃度は、好ましくは6000乃至10000u/mlの
