【発明の詳細な説明】ヒストリチクス菌からのコラゲナーゼ産生に応答性の遺伝子の分子クローニング 背 景
本発明は、ヒストリチクス菌(Clostridium histolyticum)コラゲナーゼをコ
ードする遺伝情報の単離及びクローニング及び適する宿主内でのその遺伝情報の
発現に関する。特に、この発明は、ヒストリチクス菌ゲノミックコード配列の自
然発現により測定された翻訳産物よりも高い分子量を有する型を含むヒストリチ
クス菌コラゲナーゼの型をコードする遺伝情報の単離及びクローニングに向けら
れている。要 旨
本発明によれば、ヒストリチクス菌コラゲナーゼの酵素活性及び抗原性を有す
るポリペプチドをコードする組換えDNAセグメントが提供される。しかしなが
ら、このポリペプチドは、天然のヒストリチクス菌コラゲナーゼ、即ち、ヒスト
リチクス菌ゲノムの自然発現により産生され続いてヒストリチクス菌から精製さ
れるコラゲナーゼと区別可能である。ここでは非天然型と言うことにする本発明
のこのポリペプチドは、ヒストリチクス菌ゲノミックコード配列の自然発現によ
り測定された翻訳産物よりも高い分子量を有する。
特許請求する組換えDNAセグメントは、ヒストリチクス菌由来のプロモータ
ーを含む。このプロモーターは独立して作動し、
特許請求するDNAセグメントが前記プロモーターの制御の下に転写されるのを
可能にして、特許請求する組換えDNAセグメントの外部にあるプロモーターが
機能することなしに特許請求するポリペプチドを産生する。
特許請求する組換えDNAセグメントは、更に、特許請求する非天然の、つま
り高分子量のポリペプチドよりも低い分子量を有する、コラゲナーゼ活性を有す
る天然ポリペプチドを発現することができる。
本発明は、更に、特許請求する組換えDNAセグメントを含みかつ宿主細胞を
形質転換して特許請求する非天然ポリペプチドを産生することができるベクター
を提供する。
本発明者らは、形質転換された宿主細胞内の特許請求する組換えDNAセグメ
ントの発現がその宿主細胞の株によって決定されるということを見出した。従っ
て、本発明のベクターで形質転換された異なる大腸菌宿主細胞が提供される。宿
主細胞株の1つは、コラゲナーゼ活性と抗原性を有する本発明の非天然ポリペプ
チドを産生する。この宿主細胞によって産生されるコラゲナーゼ活性を有するポ
リペプチド全体のうち50重量%を超えるものが非天然高分子量ポリペプチドを
含む。
本発明は、更に、本発明のベクターで形質転換された他の大腸菌宿主細胞を提
供する。これら宿主細胞は、コラゲナーゼ活性と抗原性を有しかつ110,00
0の分子量を有するポリペプチドを産生する。これら細胞によって産生されるコ
ラゲナーゼ活性を有するポリペプチドの50重量%を超えるものが110kdコ
ラゲナーゼを含む。
本発明の他の側面は、ヒストリチクス菌コラゲナーゼの実質的に精製された調
製物に関連する。1つの試料は、コラゲナーゼの非天然型を含む。本発明の実質
的に精製された他の調製物は、110kdの分子量を有するコラゲナーゼを含む
。これら型のコラゲナーゼは、本発明のベクターで形質転換された異なる株の大
腸菌宿主細胞に由来する。
更に、本発明に従って遺伝子操作された大腸菌宿主細胞により産生されるコラ
ゲナーゼを用いる方法が提供される。これら方法は、結合組織の消化及び包埋細
胞の放出、膵臓組織から分散された膵島の単離、血管からの内皮細胞の単離、及
び分散した腫瘍細胞を単離するための腫瘍の解離などに適している。この方法は
、次の2工程を含む:
(1) 分散すべき組織を、本発明の実質的に精製された遺伝子操作コラゲナーゼ
を含有する緩衝溶液中で約25〜39℃で振とうしながらインキュベートし、包
埋細胞を放出及び分散させ;そして
(2) その分散した細胞を組織破片から分離する。組織破片から分散細胞を分離
する工程は、典型的には、密度勾配遠心分離法により行われる。
遺伝子操作した本発明のヒストリチクス菌コラゲナーゼは、髄核のヘルニア(
椎間板ヘルニア)の円板内(intradiscal)治療方法に用いてもよい。この方法
は、次の工程を含む:
(1) 特許請求する実質的に精製された遺伝子操作コラゲナーゼを含有する無菌
の緩衝溶液を調製し;そして
(2) その特許請求する実質的に精製されたコラゲナーゼを含
有する無菌の緩衝溶液を髄核に注入する。

本発明者らは、ヒストリチクス菌の遺伝子を大腸菌内にクローン化した。この
クローン化遺伝子、即ち、本発明の組換えDNAセグメントは、大腸菌内でポリ
ペプチドとして産物を発現できる。大腸菌内で発現されたその産物は、抗コラゲ
ナーゼ抗体と免疫反応性であるタンパク質の形でも検定可能なコラゲナーゼ活性
、即ち、コラーゲン消化の形でも検出可能である。
それが挿入される大腸菌宿主に依存して、4.9キロベースを有する同じ組換え
DNAセグメントが翻訳されると、以下の実施例3及び11に記載するように、
コラゲナーゼ活性を有する幾つかのポリペプチドを産することが見出された。特
許請求する本発明による組換えDNAセグメントで形質転換された大腸菌株DH
5αは、特に、コラゲナーゼ活性と抗原性を有する、天然のヒストリチクス菌コ
ラゲナーゼと区別可能なポリペプチドを産生した。この非天然ポリペプチドの明
確に異なる特徴は、それがヒストリチクス菌ゲノミックコード配列の自然配列に
よって測定された翻
訳産物よりも高い分子量を有することであった。自然発現によって測定されたヒ
ストリチクス菌コラゲナーゼは、コラゲナーゼをコードするDNAセグメントを
保持する遺伝子ベクターが導入されていないヒストリチクス菌により直接に産生
されるコラゲナーゼの型である。以下の実施例で記載するように、種々の商業的
供給業者から購入したヒストリチクス菌の天然コラゲナーゼは、本発明の非天然
コラゲナーゼと同じサイズの分子量を有しない。
実施例8に詳述するように、特許請求する組換えDNAセグメントは、ヒスト
リチクス菌由来のプロモーターを含むことが見出された。以下に記載するように
、コラゲナーゼをコードする本発明の4.9kb組換えDNAフラグメントは、大
腸菌内で独立に機能するクロストリジウムプロモーターを含んだ。
特許請求する組換えDNAセグメントは、本発明の非天然コラゲナーゼよりも
低いコラゲナーゼ活性と分子量を有するポリペプチドを発現することが見出され
た。実施例に記載するように、特許請求するDNAセグメントから発現される天
然及び非天然コラゲナーゼの相対比は、本発明のベクターを介して特許請求する
DNAセグメントで形質転換される大腸菌宿主細胞に従って変動する。
ヒストリチクス菌からDNAを単離した後、クロストリジウムDNAを含有す
るpRK290ライブラリーを構築して(実施例1)そのコラゲナーゼ遺伝子を
スクリーニングした。挿入したコラゲナーゼ遺伝子の特徴を制限酵素分析によっ
て明らかにした(実施例2)。実施例4でコラゲナーゼ遺伝子の更なる特徴付け
を行った。そこでは、その挿入物を順次欠失させたプラスミドp
CT6及びpCT7内に反対の挿入方向で配置した。2551塩基対を配列決定
した。完全ヌクレオチド配列及び推定アミノ酸配列を配列番号:1に示す。この
DNA配列がクロストリジウムコラゲナーゼ遺伝子を含むことの更なる確認を実
施例5及び6に示す。
大腸菌内でのクロストリジウムコラゲナーゼ遺伝子の発現には、クロストリジ
ウムDNAを保持する適するプラスミド又は他のベクターでの宿主大腸菌細胞の
形質転換又は形質移入及びその形質転換細胞によって産生するコラゲナーゼの検
出を要する。米国特許出願第07/498,919号に記載された形質転換及び
コラゲナーゼ検出を行う標準的方法を用いて、本発明者らは、実施例3、7、9
、10及び11において、形質転換宿主細胞内での特許請求した組換えDNAセ
グメントの発現を宿主細胞の株によって確認する。実施例3は、約2%の形質転
換大腸菌可溶性タンパク質が、抗コラゲナーゼ抗体でのイムノブロッティングに
より、ウェスタンブロット及びSDS−PAGEのクマシーブルー染色でコラゲ
ナーゼであったことを証明している。
実施例8に記載するように、大腸菌内でのコラゲナーゼ発現の制御が、ヒスト
リチクス菌由来の特許請求したDNAセグメントを含む独立プロモーターの制御
下のものであることが見出された。
本発明者らは、以下の実施例10に記載するように、形質転換大腸菌細胞から
及びこれら細胞を生育させた培養培地から天然及び非天然のコラゲナーゼを産生
させて精製した。細胞からのこれら酵素の精製は、当該技術分野で既知の標準的
方法を包含するものであった。本発明者らは、意外にも、これら形質転換細胞が
生育した発酵ブロス中に大量のコラゲナーゼを見出した。その観察
に基づいて、実質的に純粋な非天然コラゲナーゼ及び110kdコラゲナーゼの
調製物を得るためのコラゲナーゼ精製戦略を立てた。
形質転換細胞から精製した組換えコラゲナーゼのコラーゲン消化活性は、細胞
から精製した組換えコラゲナーゼの活性とほぼ等しいことが分かった(実施例1
0の表1)。精製組換え非天然コラゲナーゼのコラーゲン消化活性を、市販の天
然型精製コラゲナーゼと比較した(実施例11の表2)。組換えコラゲナーゼは
、ウォーシントン(Worthington)・ケミカル、シグマ・ケミカル、及びカルバ
イオケム(CalBiochem)から得た天然コラゲナーゼよりも活性が50%から10
0%高かった。
特許請求するDNAセグメントを含有する遺伝子操作大腸菌により産生された
精製組換え非天然又は組換え天然110kdコラゲナーゼは、コラーゲンを消化
することが望まれるあらゆる応用に用いることができる。組織から細胞を単離又
は放出する特定の応用には:(1)細胞膜及び他の必須の特徴を破壊することなく
結合組織を消化して包埋細胞を放出すること;(2)内皮細胞を単離すること;(3)
腫瘍を解離すること;及び(4)髄核のヘルニア(椎間板ヘルニア)の円板内治療
;が含まれる。実施例12に示すように、本発明の精製110kd組換えコラゲ
ナーゼは、トリプシンと組み合わせると、ヒト伏在静脈から内皮細胞を単離する
のに効果的であった。
標的組織及び動物に依存して、本発明の方法を用いて解離組織試料を得るため
の組換えコラゲナーゼ(配列番号:2)、又は天然クラスIIコラゲナーゼ、又は
中性プロテアーゼの量は変動して
もよい(ウォルターズ(G.H.J.Wolters),“An Analysis of theRole of Colla
genase and Protease in the EnzymaticDissociation of the Rat Pancreas for
Islet Isolation”,Diabetologia,35:735-742(1992))。本発明の組成物及び
方法において組換えコラゲナーゼと共に要する中性プロテアーゼのタイプ及び量
は、標的組織に依存して変動してもよい。本発明の方法を用いて組織を消化又は
解離するのに、粗製又は精製天然コラゲナーゼ以外のプロテアーゼを、組換えコ
ラゲナーゼ(配列番号:2)を含む組成物に用いてもよい。実施例
以下の実施例は説明のためだけであり、本発明を限定するものと解釈されるべ
きではない。実施例1 無傷125kdコラゲナーゼをコードする遺伝子のクローニング及びスクリーニ ング
無傷125kdコラゲナーゼをコードする遺伝子の方向は、米国特許出願第0
7/498,919号において確認された。以下の実施例4及び5に記載するよ
うに、遺伝子の終点をその遺伝子の配列決定によって確認し、Bgl II部位に
密接していることが分かった。上の情報によって規定されるように、2.5kbD
NAフラグメントが、最初のEcoRI部位から2番目のEcoRI部位を通過
してBgl II部位に伸びていた。2.5kbDNAフラグメントは、約125k
dの分子量を有するコラゲナーゼをコードするには十分なサイズではない。それ
には、約4.4kbを要する。従って、より多くのコラゲナーゼ遺伝子配列を同定
しよう
と、新たなライブラリーを調製した。これは、Bgl IIライブラリーの構築を
包含した。
ゲノミックDNAからpBluescriptのような小さなプラスミドに直
接にBgl IIフラグメントをクローン化しようと試みた。本発明者らは、12
5kdの分子量においてコラゲナーゼバンドを同定したが、独立の2実験を包含
する第2スクリーニングでは、その分子量は68kdのようであった。
本発明者らは、pRK290(ハース(Haas,D.),Experientia,39:1199(
1983))の如き低コピー数(細胞当たり1〜10コピー)の他のプラスミドを用
いた。これにより、全縁コラゲナーゼ遺伝子をクローニングすることができた。
しかしながら、Bgl IIフラグメントをpUC8の如き小さなプラスミドにク
ローニングする困難さが残った。にも拘らず、本発明者らは、全縁コラゲナーゼ
遺伝子を組み込んだそのようなpUC8プラスミドの構築を達成した。
A.ヒストリチクス菌からのDNA単離
ヒストリチクス菌ATCC21000をアメリカン・タイプ・カルチャー ・
コレクションから得た。この細菌を含有する紙製の錠剤をまずTYEブロス(培
養培地1リッター当たり15gトリプシン、10g酵母エキス、及び5gNaC
l)中に4℃で30分間時々振とうさせながら浸した。
この細胞懸濁液をTYEアガープレート上に塗って嫌気性条件下で37℃で生
育させた。単一のコロニーを拾って50mlTYEブロス中で生育させ、嫌気性
条件下で37℃で生育させた。遠心分離によって細胞を採集して、TES(0.
1Mトリス(pH8.
0)、0.1mM EDTA、及び0.15M NaCl)中に再懸濁した。こ
の細胞懸濁液を4回凍結及び融解することによって細胞を部分的に溶解した。次
いで、SDS及びプロナーゼKを添加してそれぞれ0.5%及び200μg/m
Lの最終濃度にした。ベックマンJ2−21遠心分離機での10,000rpm
で4℃30分間の遠心分離によって細胞破片を除去した。上澄み液を等容量のフ
ェノール−クロロホルムで3回抽出して、DNAをイソプロパノールで沈殿させ
た。このDNAをアガロースゲルで電気泳動に付し、臭化エチジウムの添加後に
蛍光を既知の濃度スタンダードの蛍光と比較することによって、DNA濃度を測
定した。精製クロストリジウムDNAの平均サイズは、0.6%アガロースゲル
での電気泳動によって測定して30kb及び40kbであった。
B.クロストリジウムDNAを含有するpRK290ライブラリーの構築
精製ヒストリチクス菌染色体DNAを制限酵素Bgl IIで消化した。エタノ
ール沈殿の後、消化DNAをTE緩衝液(10mMトリス(pH7.4)、0.
1mM EDTA)中に再懸濁した。適当量の消化DNAを、前もってBgl
IIで開裂してアルカリ性ホスファターゼで処理したpRK290と連結した。連
結は4℃で16時間行った。この連結DNAを記載されている通りに大腸菌株D
H5α内に形質転換した。形質転換細胞をTYE−Tc(1リッター当たり15
gトリプトン、10g酵母エキス、5gNaCl、及び15mgのテトラサイク
リン)プレート上にプレートして35℃一晩インキュベートした。
C.コラゲナーゼ遺伝子のスクリーニング
プレート当たり約1,000コロニーを得た。約10プレートを無傷コラゲナ
ーゼ遺伝子についてスクリーニングした。簡単に説明すると、35℃で16時間
のインキュベーションを行った後、乾燥したニトロセルロースペーパー(S&S
)をペトリ皿の上に置いてそれらコロニーを吸収した。そのニトロセルロースペ
ーパーを注意してプレートから離して、調製したばかりのTYE−Tcアガープ
レートに移し、そして35℃で更に4時間インキュベートした。もとのプレート
をマスタープレートとして4℃で貯蔵した。ニトロセルロースペーパーを注意し
てそのプレートから取り去って、0.1Mトリス(pH8.0)、0.2M N
aCl、及び5%スキムミルク中に浸し、そして静かに振とうしてそのニトロセ
ルロースペーパーから細菌細胞を除去した。このニトロセルロースペーパーをウ
サギ抗コラゲナーゼ抗体(米国特許出願第07/498,919号の実施例2に
詳述されている)と共に1:1000の希釈度で5%スキムミルクと共に室温で
1時間インキュベートし、0.2M NaCl及び1%トリトン(商標)X−1
00を含有する0.1Mトリス−HCl緩衝液(pH8.0)で洗浄し、次いで
プロテインA−ホースラディッシュペルオキシダーゼ複合体と共に5%スキムミ
ルクを含むPBS中1:1000の希釈度で室温で1時間インキュベートした。
ヤングとディビス(R.A.Young & R.W. Davis),“Efficient Isolation of Gen
es byUsing Antibody Probes”,Proc. Natl. Acad. Sci. U.S.A., 80,1194-119
8 (1983)に記載された通りに、洗浄操作を繰り返してこのニトロセルロースペー
パーをPBS中で4−クロロ−1−ナフトールとH2O2で発色させた。
このニトロセルロース上の陽性シグナルの位置に対応するプレート上のアガロ
ースの部分を取ってTYE−Tcブロス中に再懸濁させた。低細胞密度に段階的
に希釈した細胞懸濁液(プレート当たり30〜100細胞)をTYE−Tcプレ
ート上に再プレートし、第2スクリーニングを行って単一のコロニーを同定した
。P70と名付けられたこの1コロニーは強いシグナルを示した。これを採集し
てTYE−Tcブロス中で生育させた。このコロニーの特徴を以下の実施例2及
び3のように更に明らかにした。実施例2 挿入DNAの制限酵素分析
プラスミドP70内のクロストリジウムDNAの構成をより正確に特徴付けす
るために、P70を更なる制限酵素分析に付した。プラスミドP70からのDN
Aを、記載された通りに調製した(マニアチス(Maniatis))。精製DNAをBg
l IIで消化してクローン化DNA挿入物を放出させた。見掛けのサイズが4.
9kbのDNAフラグメントをBgl II消化によってベクターpRK290か
ら放出させた。Bgl IIとEcoRIを組み合わせてp70を更に消化し、同
じ酵素で消化したpBB1及びpRS21と比較した。3つ全てのプラスミドは
、同一の1.7kbEcoRI−Bgl IIフラグメントを放出した。P70及
びpBBlのいずれも同一の0.8kbEcoRI−EcoRIフラグメントを
放出した。これら制限酵素分析は、3つ全てのプラスミドが共通のDNAフラグ
メントを有し、P70は2.4kbBgl II−EcoRIフラグメントの余剰
片を有したことを示唆した。実施例3 大腸菌株DH5α内で産生されたコラゲナーゼの特徴付け
免疫反応性タンパク質のサイズを調べるために、P70を含有する細胞を遠心
分離によって採集し、ゲル泳動用緩衝液中に再懸濁し、加熱し、ラエムリ(Laem
mli, U.K.),“Cleavage ofStructural Proteins During the Assembly of the
Head ofBacteriophage T4”,Nature, 227:680-685(1970)に記載された通りに
7.5%SDS−ポリアクリルアミドゲルにかけた。それらタンパク質をニトロ
セルロースペーパー上にエレクトロブロットして、米国特許出願第07/498
,919号(バーネット(W.H.Burnette),“Western Blotting: Electrophore
tic Transfer ofProteins form SDS-polyacrylamide Gels to UnmodifiedNitroc
ellulose and Radiographic Detection with Antibodyand Radioiodinated Prot
ein A”,Anal. Biochem., 112, 195-203(1981))のウサギ抗コラゲナーゼ抗体
で検出した。P70クローンからのコラゲナーゼの分子量は、予備染色したタン
パク質スタンダードとの比較によって決定した。
約125kd及びそれ未満の分子量を有する免疫反応性タンパク質バンドがP
70クローンから発現した。68〜100kdの分子量を有する幾つかのバンド
は、本発明者らが米国特許出願第07/498,919号に記載したものと類似
している。
大腸菌株DH5αをプラスミドpUC8又はP70でそれぞれ形質転換して2
50mlフラスコ内の50mlのTYEブロス中で生育させた。細胞を採取して
、10mMトリス(pH7.4)及び1mM PMSF中に再懸濁し、そして音
波処理した。遠心分
離によって細胞破片を除去した後、タンパク質濃度を測定して同量のタンパク質
を7.5%SDS−ポリアクリルアミドゲル上にのせて泳動させた。
本発明の組換えDNAセグメントの発現を図2に示す。パネルAは、クマシー
ブルー染色SDS−PAGEである。レーン1はプラスミドpUC8を保持する
大腸菌株DH5αであり、レーン2はプラスミドP70を保持する大腸菌株DH
5αである。パネルBは、P70を保持する大腸菌株から調製した細胞抽出物の
クマシーブルー染色(レーン1)及びイムノブロット(レーン2)SDS−PA
GEである。
pUC8を保持する細胞ではなくプラスミドP70を保持する細胞から調製し
た細胞抽出物では、非常に強いクマシーブルー染色タンパク質バンドが分子量2
00キロダルトンと97キロダルトンの間に可視化された。プラスミドP70形
質転換大腸菌株DH5αを5リッターバイオフロII発酵器(BioFloII Fermentor
)内で生育させてコラゲナーゼを産生させた。遠心分離によってそれら細胞を採
取した後、細胞ペーストを−70℃で貯蔵した。少量の細胞について更に処理し
て精製条件を決めた。音波処理及び澄明化した上澄み液をクマシーブルー染色及
び抗コラゲナーゼ抗体でのイムノブロッティングによって試験し、図2のパネル
Bに示した。クマシー染色バンドの位置は、免疫反応性タンパク質バンドと同じ
であった。従って、それは、200kdと97kdの間に存するクマシーブルー
染色性バンドがコラゲナーゼであることを確認するものであった。いずれの生育
条件の下でも、約2%の大腸菌可溶性タンパク質がコラゲナーゼであると判断さ
れた。実施例4 コラゲナーゼ遺伝子の特徴付け
A.DNA配列決定戦略
ヘニコフ(Henikoff, S.)(Methods in Enzymology,Unidirectional Digesti
on With Exonuclease III in DNASequence Analysis, 155:156(1987))により公
表された非ランダムDNA配列決定戦略に僅かに手を加えて、pBBlによりコ
ードされる2.5kb EcoRI−Bgl II DNAフラグメントの配列決
定に用いた。2.5kb BamHI/Bgl IIフラグメントをプラスミドp
Bluescript SK(−)のBamHI部位に反対方向に挿入すること
によってプラスミドpCT6とpCT7とを構築した。
プラスミドpCT6とpCT7とを、P42からの2.5kbBamHI−B
gl IIフラグメントをプラスミドpBluescriptのBamHI部位に
挿入することによって構築した。プラスミドpCT6とpCT7は、図3に示す
ように反対の挿入方向を表わしている。一方向性欠失体 (undirectionaldelet
ion)を作るために、両プラスミドをまずXhoI及びKpnIで消化して、二
本鎖DNAの凹んだ3’−ヒドロキシル末端と他端において突き出た3’−末端
を作った。XhoIにより作られたこの凹んだ3’−ヒドロキシル末端は、エン
ドヌクレアーゼIIIに感受性であり段階的に除去されたが、KpnIにより作ら
れた突き出た3’−末端は無傷のままであった。消化は、開裂部位から離れて標
的DNA配列の中に一方向で進行する。消化の度合いは時間によって制御した。
異なる時間に採集したアリコー
トをS1ヌクレアーゼ消化に付した。この消化DNAサンプルをアガロースゲル
電気泳動によって分析し、所望のサイズを有するDNAフラグメントを含有する
サンプルを同定した。クレノウDNAポリメラーゼを添加して両DNA末端を平
滑にした。T4DNAリガーゼを添加してこのプラスミドを再環状化した。次い
で、連結DNAを大腸菌株DH5α内に形質転換した。
形質転換後、それら細胞をTYE−Apプレート上にプレートして、35℃で
一晩生育させた。欠失の検査及び配列決定用DNAの調製のために、コロニーを
ランダムに拾い上げて5mlのTYE−Apブロス中で一晩生育させた。遠心分
離によって一晩培養液から細胞を採集した。細胞ペレットをトリ−EDTA緩衝
液中に再懸濁してマニアチスに記載されたアルカリ法を用いることによって溶解
した。エタノール沈殿後、DNAペレットを手短に乾燥してTE(10mMトリ
ス(pH8.2)、0.1mM EDTA)中に再懸濁した。欠失のサイズは制
限酵素分析によって調べた。適当な欠失を有するクローンを同定してRNアーゼ
Aで更に処理した。RNアーゼ消化後、そのDNAを10PEG及び1.25M
NaClで沈殿させた。DNAペレットを100%エタノールで洗浄しTE緩衝
液中に再懸濁した。この方法により調製したDNAは、サンガーのジデオキシチ
ェインターミネーション法(サンガー(Sanger, F.)及びカールソン(A.R. Cou
lson),J.Molec. Biol. 94:441 (1975))を用いるDNA配列決定に適していた
。DNA配列決定に用いたクローン及び得られた配列情報を図3に纏めた。ある
場合には、プラスミドpCT12.8を系統的に欠失させ、サブクローンを用い
てこの配列の部分を決定した。
プラスミドpCT12.8は、P70の4.9kbBgl IIフラグメントをB
amHIで前処理したpBluescript (SK−)内に挿入することに
よって構築した。
B.DNA配列及び分析
全部で2808塩基対の完全配列を配列番号:1に示す。このDNA配列は、
通常、高比率のA及びTヌクレオチド(69%)と31%に過ぎないG及びCヌ
クレオチドを含有する。最初のEcoRI部位から2番目のEcoRI部位を通
過して塩基対2808に位置するTAA終結コドンで終わるオープンリーディン
グフレームを同定した。936アミノ酸を含有する推定タンパク質配列を特定し
て配列番号:2に示した。この部分は、通常、殆どの他のタンパク質と比較して
高度に荷電したアミノ酸(30%)を含有する。
図5に示すように、典型的な細菌性転写終結シグナル(GeneFunction in Prok
aryotes,ベックウイズ(J. Beckwith)ら編,p.123, Cold Spring Harbor Labo
ratory, Cold Spring Harbor,NY(1983)中のプラット(Platt, T.)及びベア(D.
G. Bear)の報文)を転写終結シグナル(TAA)から11塩基対下流に同定し
た。このシグナルは、ステム−ロープ構造に続くA−Tに富む配列からなる。こ
のデータは、このコラゲナーゼ遺伝子がこのステム−ロープ構造の下流のA−T
に富む域内で終了することを示唆した。実施例5 従来のクローンのDNA配列比較
従来の全てのクローンが、イムノブロット上でのそれらの移動
度から判断して、同一の免疫反応性タンパク質バンドを産生したことは明らかで
あるが、微変異性の可能性を排除するものではなかった。もしも従来得られてい
るクローンの間に微変異性が存在するとすれば、それは、ファン・バート(Van
Wart)ら[REF]によって提案された、遺伝子重複及びその後の突然変異が異な
るコラゲナーゼをもたらしたという概念を支持していたかも知れない。
それらの同一性を確認するために、次の2つのDNA配列プロジェクトを行っ
た:1)P6、P9、P41、P42、及びP51の如き異なる挿入物を有する
クローン間の配列の比較及び2)見たところ全てが同じ挿入サイズを有する従来
得られた異なるもとの単離物間の配列の比較。多クローニング部位を両端に配置
した(flanked)DNA配列をフランキングユニバーサルプライマーを用いて配
列決定した。平均150〜200塩基対のクロストリジウムDNA配列を両末端
から得た。配列を比較した後、1塩基対の相違も同定されなかった。この結果は
、同じクラスのコラゲナーゼ中に相違点をもたらすことを司る手段としての遺伝
子重複を支持しなかった。
実施例6P70由来のクローンのDNA配列と従来得られているクローンからの配列との 比較
P70と従来得られている他のクローンの同一性を調べるため、多くのクロー
ンを部分的に配列決定して比較した。クローンP70からの0.7kbEcoR
I及び1.8kbEcoRI−BglIIフラグメントをpUC13又はpBlu
escript内に
サブクローン化した。多クローニング部位を両端に配置したDNA配列をフラン
キングユニバーサルプライマーを用いて配列決定した。プラスミドpBB1、p
RS21、及びP41のフランキング配列を決定してP70のサブクローンから
得られた配列と比較した。相違点は認められなかった。この結果は、P70がp
BB1と同じDNAフラグメントを有しかつ染色体上の同じところに位置するこ
とを示唆した。
実施例7P70のサブクローンから産生したコラゲナーゼとpRS21から産生したコラ ゲナーゼとの比較
P70と先に記載した他のクローン間の同一性の特徴を更に明らかにするため
、見たところ同じ挿入物を有するサブクローンから産生したコラゲナーゼ産物を
比較した。P70をBgl II及びEcoRIで消化して、BamHI及びEc
oRIで予備消化したpUC18と連結した。連結DNAを大腸菌株DH5αに
形質転換し、TYE−Apプレート上にプレートし、そして35℃で一晩インキ
ュベートした。1.7kbEcoRI−Bgl II挿入物を含有しかつpRS2
1と同じ挿入方向を有するクローンを同定し、pCT8Bと名付けた。プラスミ
ドpCT8B、pRS21、又はpUC18のいずれかを含有する大腸菌株DH
5αをTYE−Apブロス中で35℃で一晩生育させた。細胞を遠心分離により
採集してSDS−PAGE泳動用緩衝液で崩壊させた。放出されたタンパク質を
7.5%SDS−ポリアクリルアミドゲルにかけ、ニトロセルロースペーパーに
エレクトロブロットした。免疫反応性バンドを先に記載した通りに検出して比較
した。図1
0のレーン1に示すように、pUC18を保持する大腸菌から産された溶解産物
は、免疫反応性コラゲナーゼバンドをもたらさなかった。しかしながら、pRS
21(レーン2)又はpCT8B(レーン3)を保持する大腸菌から産生したい
ずれの細胞溶解産物も、68kdの分子量を有する同一の免疫反応性タンパク質
バンドを示した。これは両方のものが同じタンパク質を産生していることのあら
われである(レーン10)。
実施例8ヒストリチクス菌由来の独立プロモーターによる大腸菌内でのコラゲナーゼ発現 の制御
BamHIで予備消化したプラスミドpUC13内にP70からの4.9kb
Bgl IIフラグメントをクローニングすることによってプラスミドpCT11
.10を構築し図8に示した。このコラゲナーゼ遺伝子の転写方向は、Lacプ
ロモーターと同じである。
プラスミドpCT11.10を保持する大腸菌DH5αを50mlのTYE−
Apブロス中で1mM IPTGと共に又はIPTGなしで32℃で一晩振とう
(130rpm)しながら生育させた。細胞と上澄み液を遠心分離により分離し
た。この細胞と上澄み液を泳動用緩衝液と共に95℃でインキュベートしてから
、7.5%SDS−ポリアクリルアミドゲルにかけた。上澄み液及び細胞のもと
の容量にそれぞれ等しい20μl及び80μlを各レーンにのせ、図9に示した
。クマシーブルー染色後の細胞抽出物のタンパク質プロフィールを図9のレーン
1(IPTG含)及び2(IPTG不含)に示す。培養培地のタンパク質プロフ
ィールを
図9のレーン3(IPTG含)及び4(IPTG不含)に示す。
IPTG含又はIPTG不含では、細胞抽出物及び培養培地のいずれも組換え
コラゲナーゼの発現にいかなる相違も示さなかった。強力なlacプロモーター
誘導物質であるIPTGは、コラゲナーゼの発現を増加させなかった。これで、
4.9kbDNAフラグメントが、大腸菌内で独立に機能できるクロストリジウ
ムプロモーターを含有することがさらに確認された。
意外にも、図9のレーン3及び4に示すように、大量のコラゲナーゼが上澄み
液中に発見された。のせた当量の上澄み液(20μl)は、のせた当量の細胞(
80μl)の4分の1であったので、染色ゲルから判断して、80%のコラゲナ
ーゼが上澄み液中に存すると見積もられた。培養培地中でのコラゲナーゼの蓄積
は、大腸菌細胞が培地の中にコラゲナーゼを分泌したことを示すものであった。
これら細胞は、コラゲナーゼをコードする本発明の組換え遺伝子セグメントを含
むこれら宿主細胞が生育した培養培地からコラゲナーゼを生産又は精製するのに
簡単でコスト的に有効な方法を提供するという利益を有する。実施例9 大腸菌内で発現されたコラゲナーゼの細胞内局在性
異なる大腸菌コンパートメント内でのコラゲナーゼの細胞内局在性を調べた。
プラスミドP70を含有する大腸菌株DH5αをTYE−Tcブロス中で35℃
で5時間振とうしながら生育させた。遠心分離によって細胞を採集した。細胞ペ
レットを30mMトリス−HCl(pH8.0)、20%スクロース緩衝液中に
再懸濁した。ライソザイム(70マイクログラム/ml)及びEDT
A(2mM)を添加して4℃で30分間インキュベートし、細胞壁を分解した。
遠心分離によって周縁質画分を細胞質及び膜画分(CM画分)から分離した。こ
れら周縁質画分とCM画分をSDS−PAGEで検査してクマシーブルー染色及
びイムノブロット法によって検出した。図11のパネルAに示すように、クマシ
ーブルー染色ゲル上に125kdの分子量を有する明瞭なタンパク質バンドが周
縁質画分中に見られ(レーン1)、そして細胞質画分中に僅かに見られた(レー
ン2)。
図11のパネルBに示すように、多くの免疫反応性バンドがイムノブロットに
よって見られた。殆どのバンドが周縁質領域に存在した(パネルBのレーン1)
。これは、これらバンドに相当するタンパク質が内膜を通って分泌され周縁質域
に蓄積することができることを示唆したものである。ゲルにのせた量に基づいて
(周縁質画分は、CM画分と比較して半分の原液当量でのせた)、80%の12
5kdコラゲナーゼが周縁質領域に局在すると見積もられる。周縁質領域及び細
胞質領域(レーン2)に存する免疫反応性バンドの比率は、各免疫反応性バンド
の間で相違しているが、これらバンドの殆どは、見たところ、68kdの分子量
を有するタンパク質バンドを除いては、周縁質領域にむしろ局在している。この
68kdタンパク質は、むしろ細胞質領域内に存しており、周縁質領域には存し
ていない。このことは、この68kdコラゲナーゼが内膜を通って効率的に輸送
され得ないことを示唆している。この68kdタンパク質がpRS21内で産生
された68kdタンパク質と同一であるかは不明であるが、pRS21内で産生
された68kdタンパク質は、周縁質領域内に分泌され
ることができなかった。これらデータは、大腸菌内で産生された125kdの分
子量を有するコラゲナーゼが、大腸菌の分泌メカニズムを利用して内膜を通って
輸送され、周縁質領域内に蓄積され得ることを示している。実施例10 大腸菌細胞及び培養培地からのコラゲナーゼ生産と精製
A.大腸菌からのコラゲナーゼの生産
プラスミドP70を保持する大腸菌株W3110をニュー ・ブルンスウィッ
ク(New Brunswick)・バイオフロ III発酵器中で生育させて、コラゲナーゼを
生産した。発酵培地(FM,培地リッター当たりのグラム)は、KH2PO4,3
.5;K2HPO4,5.0;(NH4)2HPO4,3.5;MgSO4・7H2O
,3.5;酵母エキス,5;及びトリプトン,5からなる。50%のグルコース
を別々にオートクレーブに掛けリッター当たり10mlを添加した。CaCl2
及びZnCl2の1Mストック溶液を調製して別々にオートクレーブに掛けリッ
ター当たりそれぞれ20μlを添加した。P70を保持する大腸菌株をTYE−
Tc中で30℃で一晩生育させた。この一晩培養液を発酵器内に直接接種した。
OD600が16に等しい細胞密度に達した後、細胞を遠心分離によって採取し
て上澄み液と分離した。
B.細胞からの精製
細胞を遠心分離により採集して、1mM PMSFを含む10mMビス−トリ
ス緩衝液(pH6.5)中に再懸濁した。細胞を音波処理によって崩壊させ、破
片を遠心分離により除去した。タンパク質を逐次硫酸アンモニウム沈殿(sequen
tial ammonium
sulfate precipitation)によって分画した。コラゲナーゼを含有する画分を1
0mMビス−トリス緩衝液(緩衝液A)中に再懸濁して、同緩衝液に対して4℃
で一晩透析した。透析サンプルをまずDE52カラムクロマトグラフィーによっ
て分画した。コラゲナーゼを含有する画分をプールして緩衝液Aに対して透析し
た。透析サンプルをQセファロースカラムクロマトグラフィーによって分画した
。98%を超える純度を得るには、ゲル濾過が必要であるかも知れない。出発原
料中には多くの免疫反応性コラゲナーゼバンドが共存した。陰イオン交換カラム
クロマトグラフィーで異なる分子量のコラゲナーゼをある程度まで分離すること
ができた。以下の実施例11に記載する実験では、コラゲナーゼの最も大きな型
を用いた。
C.培地からの精製
意外にも、発酵ブロス中に大量のコラゲナーゼが検出された。培養培地からコ
ラゲナーゼを採集及び精製できる可能性を評価するために、精製戦略を立てた。
簡単に説明すると、遠心分離により細胞を除いた後に上澄み液を濃縮して、30
,000分子量カットオフの中空繊維(アミコン(Amicon))を用いてダイア濾
過(diafiltrate)した。濃縮した上澄み液を硫酸アンモニウム示差沈殿法(dif
ferential precipitation)により分画した。コラゲナーゼを含有する画分を採
集して、ビス−トリス緩衝液(pH6.5)中に再懸濁し、同緩衝液に対して透
析した。透析サンプルをDE52カラム(ワットマン)上にのせてNaClで溶
出させた。コラゲナーゼを含有する画分を採集してQセファロースカラム(ファ
ルマシア)で更に精製した。図7のレーン1は、プラス
ミドP70を保持する大腸菌株W31I0からの培養培地(レーン1)及びこの
培養培地から精製したコラゲナーゼ(レーン2)のクマシーブルー染色ゲルを示
す。細胞及び培養培地の両方から調製したコラゲナーゼのコラーゲン消化活性を
比較した。培地から調製したコラゲナーゼは、細胞から調製したものの約2倍の
活性を有した。
D.細胞及び培地から精製したコラゲナーゼのコラーゲン消化活性
細胞又は培養培地のいずれかから精製した本発明の組換えコラゲナーゼのコラ
ーゲン消化活性を測定した。以下の実施例11に記載する通りに活性を測定した
。以下の表1はこれら活性を纏めたものである。
この表の結果は、培地から精製したコラゲナーゼ活性が、細胞から精製した組
換えコラゲナーゼの活性にほぼ等しいことを示している。実施例11 組換えコラゲナーゼとヒストリチクス菌により産生された天然コラゲナーゼとの 比較
A.サイズ
組換えコラゲナーゼの同定物をヒストリチクス菌により産生された天然コラゲ
ナーゼと比較した。未精製及び精製天然型コラゲナーゼの両方を、シグマ、ベー
リンガー ・マンハイム・プロダクツ(コラゲナーゼA(CHSA)、コラゲナ
ーゼB(CHSB)、コラゲナーゼD(CHSD))、ウォーシントン、及びカ
ルバイオケムから購入した。異なる供給源のコラゲナーゼをSDS−PAGEに
かけ、それらの分子量を調べた。典型的なクマシーブルー染色ゲル及びイムノブ
ロットを図6に示す。図6は、組換えコラゲナーゼ(RCL)と天然コラゲナー
ゼ間の比較を示す;パネルAはクマシーブルー染色ゲルであり;パネルBはSD
S−PAGEのウェスタンブロットである。
図6のレーンの割り当ては次の通りである:
パネルA:レーン1,0.5μgのロット17RCL;レーン2,2.5μgの

本発明の精製組換えコラゲナーゼを、ベーリンガー ・マンハイム社から得た
市販のコラゲナーゼ(カタログ番号CHSA、CHSB、及びCHSD)と比較
した。異なる量のタンパク質を7.5%SDSポリアクリルアミドゲルにかけた
。ウェスタンブロット上のコラゲナーゼをウサギ抗コラゲナーゼ抗体で検出した
。抗コラゲナーゼ抗体の調製は、米国特許出願第07/498,919号に記載
した。
クマシーブルー染色によって多くのタンパク質が可視化できるが、染色性バン
ドの50%を超えるものがコラゲナーゼ特異性抗体によって検出できない。最大
型の組換えコラゲナーゼは、明らかに、ベーリンガー ・マンハイムから得たコ
ラゲナーゼのいかなる天然型よりも大きなサイズを示す。本発明の組換えコラゲ
ナーゼとカルバイオケム、ウォーシントン、及びシグマから得た天然コラゲナー
ゼとの比較で、同一の結果が得られた。
天然産物の最大コラゲナーゼ(110kd)に類似する組換えコラゲナーゼの
型は、特別の大腸菌宿主(JM105株)内で定量的にかつムラなく産生するこ
とができた。
B.活性
異なる販売業者から市販されている精製天然コラゲナーゼのコラーゲン消化活
性を、125kdの分子量を有する精製組換えコラゲナーゼと比較した。コラー
ゲン消化活性は、メンドル(Mendl, I)ら,J. Clin. Invest., 32:1323(1953)
に手を加えた方法に従って測定した。主要な変更は、1)用いた酵素の量を0.
2又は0.4μgに減少したこと、及び2)基質(タイプ1コラーゲン)濃度を
10mg/mlに増加したことである。コラゲ
ナーゼ活性は次のように定義される:1ユニットのコラゲナーゼ活性は、37℃
で5時間インキュベーションした後にコラーゲンから放出されるL−ロイシンの
1μモル当量に等しい。異なる精製コラゲナーゼの酵素活性を以下の表2に示す
。
組換えコラゲナーゼのコラーゲン消化活性は、天然型の精製コラゲナーゼのも
のよりも約25〜100%高かった。この結果は、本発明の組換えコラゲナーゼ
が天然コラゲナーゼよりも優れていることを明確に示唆している。実施例12 組換えコラゲナーゼ+トリプシンでの内皮細胞単離
大腸菌内で産生した本発明の精製組換えコラゲナーゼを用いてヒト伏在静脈か
ら内皮細胞を採取した。ヒト伏在静脈を半分に分けて一方を0.1%タイプIIコ
ラゲナーゼ(ウォーシントン)、0.5%BSA、及びPBS/CMFで満たし
、他方を0.01%トリプシンを含むか又は含まない同緩衝液中の0.048%
組換えコラ
ゲナーゼで満たした。組換えコラゲナーゼ単独では、この静脈から内皮細胞を解
放しなかった。RCLとトリプシンを組み合わせることにより、内皮細胞の収率
は粗製コラゲナーゼに比較して2倍になった。これら結果は、本発明の組換えコ
ラゲナーゼを一定濃度のトリプシンと組み合わせることによってより多くの内皮
細胞が単離できることを証明した。これら結果は、異なる組織からの細胞単離に
、天然コラゲナーゼを本発明の組換えコラゲナーゼと置き換えることが可能であ
ることを更に示唆した。実施例13 組換えコラゲナーゼを含む酵素混合物を用いるラット膵島単離
組換えコラゲナーゼ(配列番号:2)、天然クラスIIコラゲナーゼ、及び中性
プロテアーゼを含む組成物を作り、膵臓組織を分散させて膵島を放出させた。
大腸菌から産生した約110,000ダルトンの分子量を有する精製組換えコ
ラゲナーゼ(配列番号:2)を用いて、本発明の方法に従ってラット膵臓からラ
ット膵島を採取した。天然クラスI及びIIコラゲナーゼ及び中性プロテアーゼは
、ウォルターズ,“An Analysis of the Role of Collagenase and Protease in
the Enzymatic Dissociation of the Rat Pancreas for IsletIsolation”,Dia
betologia, 35:735-742(1992)に従って調製した。
ラット膵臓の消化は、ウォルターズ(1992)の通りに行った。組換えコラゲナー
ゼ(配列番号:2)、天然クラスIIコラゲナーゼ及び中性プロテアーゼを含む混
合物は、粗製コラゲナーゼ調製物又は精製天然コラゲナーゼと中性プロテアーゼ
の組み合わせを
用いた場合と同じラット膵島収率をもたらした。この混合物は、10ml当たり
、KRH緩衝液(ウォルターズ,1992)2mgのクラスI組換えコラゲナーゼ(
配列番号:2)、0.8mgクラスII天然コラゲナーゼ、及び100ユニットの
中性プロテアーゼから構成された。同じ膵島収率を得るのに同じような消化時間
を要することも観察された。
これら結果は、実質的に精製された組換えコラゲナーゼ(配列番号:2)を含
む組成物は、膵臓組織を分散させて他の膵臓成分から膵島を放出させることを証
明した。更に、組換えコラゲナーゼ(配列番号:2)と他のプロテアーゼを含む
組成物を用いて膵臓を消化した場合は、ウォルターズの1992年の文献に記載され
た他の組み合わせに比較して、膵島当たりより大きな膵島質量を有する膵島がよ
り少なくなったことが観察された。本発明の方法を用いて膵臓を消化すると、本
発明の組換えコラゲナーゼ(配列番号:2)を含まない酵素組成物を用いた場合
に比較して、膵島の完全性への損傷がより少なくなるように思われた。
より大きな動物の膵臓消化から十分な膵島を得るには異なるロットの粗製コラ
ゲナーゼ調製物が必要であることは知られている。従って、他の動物の膵臓消化
から膵島調製物を得るには、組換えコラゲナーゼ(配列番号:2)、又は天然ク
ラスIIコラゲナーゼ、又は中性プロテアーゼの量は変動させることができる。組
織消化に有用なタンパク質分解酵素の濃度を決定する手順は、ウォルターズ,19
92に例示されている。高度に精製した組換えコラゲナーゼ(配列番号:2)を含
む組成物は、ヒトから膵島を単離する本発明の方法に用いることができる。
本発明をある種の好ましい態様に関してかなり詳しく説明してきたが、他の態
様も可能である。従って、補正した請求の範囲の精神及び範囲は、この好ましい
態様の説明に限定されるべきではない。
