Abstract

PURPOSE:To solve difficulty in acquisition of sawdust and to culture Lentinus edodes in an mushroom bed by packing a medium using bagasse as a medium base and bran as a nutrient additive into a container, sterilizing, inoculating spawn, culturing a mushroom bed, spreading the mycelia, aging and immersing the mushroom bed in water. CONSTITUTION:Bagasse is used as a medium base and bran as a nutrient additive, 36-45wt.% bagasse is mixed with 12-18wt.% bran and 43-46wt.% water to prepare a medium. The medium is packed into a container made of polypropylene, sterilized under high pressure at 120 deg.C for 90 minutes, allowed to stand, chilled to about 18 deg.C by a cooler, spawn of Lentinus edodes is inoculated to the medium in a clear inoculating chamber free from various germs and the cells are cultured in a culture chamber in a darkroom at 18 deg.C room temperature at 70% humidity for 30 days. The mycelia are spread in the whole medium, then the mycelia are cultured at 22 deg.C room temperature and at 70% humidity under 600 lux for 60 days, the medium is aged, fruit body primordial are formed in a coating film of the mushroom bed, the mushroom bed is immersed in water at 28 deg.C for 24 hours and fruit bodies are produced and grown to culture Lentinus edodes in the mushroom bed.

2. Description of the Related Art Shiitake mushrooms are cultivated by using broad-leaved trees such as oak and cypress, which are cut into 1m-long pieces that are convenient for handling. It is placed under an artificial shade or under the artificial shade, and the hyphae of the shiitake mushroom are grown on the log. By doing this, in the second year, shiitake mushrooms would grow on the logs and be harvested. Recently, in order to eliminate the depletion of raw wood for sardines, reduce the amount of work due to the aging of producers, shorten the cultivation period, etc., instead of raw wood cultivation, a container using a fungal bed mainly composed of hardwood sawdust Cultivation (bag cultivation, bottle cultivation) has come to be carried out, and the production amount thereof is increasing. That is, a medium is prepared by mixing hardwood sawdust such as oak and syrup, a nutrient such as rice bran, and water, and the medium is filled in a heat-resistant bag or polypropylene bottle and sealed with a cap using a filter. The bacteria in the medium are subjected to high-pressure sterilization treatment, the inoculum is inoculated into the medium in a clean inoculation chamber, and the medium inoculated with the inoculum, that is, the bed is cultured in the culture chamber.

1 kg of this medium is packed in a polypropylene bag, sealed with a filter cap (step 2), and high-pressure sterilized at 120 ° C. for 90 minutes (step 3). The sterilized medium is placed in a cooling chamber, and the medium is cooled to about 18 ° C. using a refrigerator (step 4). After cooling, the inoculum is inoculated in a clean inoculation room free of germs (step 5). The medium inoculated with the inoculum is cultured in the culture room (step 6). In the first half of the culture, the cells are cultured for 30 days in a dark room with a room temperature of 18 ° C. and a humidity of 70%, and mycelia are spread over the entire medium (step 6-1). Room temperature 2 in the latter half of culture
It is cultured for 60 days at 2 ° C., 70% humidity and 600 lux illuminance, and the medium is aged to form the primordia of the fruiting bodies in the membrane of the bacterial bed (step 6-2).

Immerse in 8 ° C. water for 24 hours (step 7). After the water immersion treatment, fruiting bodies are generated in a generation chamber having an indoor temperature of 18 ° C., a humidity of 85% and an illuminance of 500lux (step 8). It is harvested about 10 days after it is placed in the generating room (step 9). As shown in FIG. 2, the yield is 140 g per 1 kg of the medium, that is, 14% based on the weight of the medium. This ratio is almost the same as the value when hardwood sawdust is cultivated as a medium substrate.
