Abstract
る腸レベルで免疫応答の調節に対し形質転換成長因子ベ
ータファミリーの乳由来のポリペプチドを提供すること
を目的とする。 【構成】 ヒトまたは動物の胃腸管の免疫応答に関連す
るMHCのモジュレーションに対し哺乳動物の乳または
初乳由来のTGF−β−様 MGF(乳成長因子)の使
用および有効量の哺乳動物の乳または初乳由来のTGF
−β−様 MGFを含有する食品組成物または経腸調製
品。および医薬製剤。
Description
性複合体)と関連する腸レベルで免疫応答の調節に対し
形質転換成長因子ベータファミリーの乳由来のポリペプ
チドを使用することに関する。本発明は特に食品組成
物、経腸調製品または医薬組成物に対する哺乳動物の乳
または初乳由来のTGF−β−様 MGF(乳成長因
子)の使用および有効量の哺乳動物の乳または初乳由来
のTGF−β−様 MGFを含有する食品組成物または
経腸調製品に関する。
および牛の乳は成長因子を含む多数の生物学的活性ポリ
ペプチドを含有する(West,D.W.Exp.Cl

989)。これらの因子のうちの1つ、MGF(乳成長
因子)は最近形質転換ベータ(TGF−β)ファミリー
のメンバー、特にTGF−β2(Cox D.A.ら、

することが確認された。TGF−βはかなりの構造的お
よび生物学的相同性を有する、少なくとも5つの別個
の、しかし密接に関連する群から成るポリペプチドファ

スルフィド架橋により結合した同一の12.5kDaポ
リペプチド鎖から成る約25kDaの同一二量体タン白
である。これらは他のタン白と潜在的複合体を形成で
き、これらの複合体は酸処理または温和なタン白分解に
より活性化できる(Roberts,A.B.ら)。こ
れらは複数作用性であり、標的細胞のタイプ、その分化
状態および他の因子の存在により多くの生物学的活性を
有する。これらの活性は細胞増殖および分化の刺激また
は抑止、細胞外マトリックス沈着の調節、免疫モジュレ
ーション、ステロイドホルモン生合成および血管形成を
含む(Roberts,A.B.ら)。

獸の腸絨毛の上皮細胞は構造的にMHC−クラスIIを発

後では離乳期後まで腸嚢胞によるMHC−クラスIIの発
現はほとんど、または全く見られない。こうして乳に抑
圧因子が存在することを示している(Hughes、
A.ら)。
疫調節性を有し、炎症および免疫反応の数段階で作用す
る。例えばこれらはTおよびBリンパ球(Kerhl,
J.H.ら、J.Immunol、137、3855〜
3860、1986;Kerhl,J.H.ら、J.E
xp.Med.163、1037〜1050、198
6)および胸腺細胞(Ristow,H.J.Pro
c.Natl.Acad.Sci.USA 83、55
31〜5534、1986)の増殖を抑止する。これら
はIL−1、IL−2およびIL−3を含むインターロ
イキンおよび腫瘍壊死因子およびインターフェロンなど
の他の免疫調節剤の作用に拮抗する(Roberts,
A.B.ら)。免疫作用に及ぼす大部分のこれらの効果
は抑制されるが、TGF−βはIgGおよびIgM分泌
細胞をIgA分泌細胞にスイッチするアイソタイプに重
要な役割を演ずるようである(Lebman,D.A.

990)。特にMGFの報告された免疫抑圧効果に関
し、この因子は抗−CD3またはインターロイキンによ
り誘発されたヒトのリンパ球の増殖を減少させることが

3265、1989)。TGF−βは抗原の供与に重要
なある補助細胞の機能を妨害し、特に黒色腫、グリア細
胞およびアストログリアによるMHC−クラスIIの発現
を抑圧することが示された(Czarniecki,

胞に対するMHC−クラスII発現の調節は従来報告され
ていなかった。

基づく腸の管腔抗原に対する過剰反応性による粘膜の免
疫調節障害に帰せられる(Challenges in

nD.C.)。この2つの可能性は粘膜免疫応答に調節
の存在しないことを暗示し、炎症応答の開始および永続
性に免疫学的役割を強調している。
介免疫応答の調節、特に腸炎症病(例えば、クローン
病、大腸潰瘍)またはヒトまたは動物の移植組織対宿主
反応の治療、ヒトまたは動物の離乳時の下痢の予防、ま
たはヒトまたは動物の胃腸管のアレルギー反応の予防に
対する食品組成物、経腸調製品または医薬製剤を供する
ことである。

物の胃腸管でMHC発現のモジュレーションに対し有効
量の哺乳動物の乳または初乳由来のTGF−β2−様
MGFを含有し、その量は腸炎症病(例えば、クローン
病、大腸潰瘍)またはヒトまたは動物の移植組織対宿主
反応、ヒトまたは動物の離乳時の下痢の予防、またはヒ
トまたは動物の胃腸管のアレルギー反応の予防に対し有
効であることが好ましい。

−β2−様 MGFと称する。

hnology Ltd.)に対し高めた抗体はTGF
−β同質形に特異的の阻害活性を測定するため、バイオ
アッセイ分析前に標準品または試料と共インキュベート
した。このアッセイを使用してRLE細胞DNA合成の
50%抑制が50pg/mlのTGF−β1またはTGF−
β2により得られる。

した。次いで稀釈物を一連のTGF−β標準溶液と同時
にRLE細胞バイオアッセイを使用して分析した。TG
F−β−様 活性量の評価はTGF−β標準曲線に対し
試料により得たDNA合成抑制度を比較して測定した。
TGF−βの特定同質形の同定は抑制活性に及ぼす抗体
の中和に特異的同質的の効果を試験して測定した。

ヒトの組み換えインターフェロン−ガンマ(IFN−
γ、100U/ml)単独、TGF−β2と組み合せたI
FN−γ、IFN−γに続いてTGF−β2、TGF−
β2単独続いてIFN−γ、または対照として培地の
み。細胞は洗浄し、最初の24時間後再処理した。TG
F−β2は0.05ng〜4ng/mlの範囲の用量で使用し
た。処理期間後、細胞は洗浄し、固定し、プレートは所
要時まで−20℃で凍結保存した。
オチン複合体方法(Cerf−Bensussan,
N.ら、J.Immunol.、130、2615、1
983)はマウスの単クローン性抗体L234(Bec
ton Dickinson)を使用して単層で行なっ
た。この抗体はヒトMHC−クラスII組織適合性 抗原
HLA−DRを認識する。マウスのミエローマIgGタ
ン白(Zymed)を対照として使用した。別の試験シ
リーズでは、正常のラット小腸細胞ライン、IEC−1
8(Quaroni,A.ら、J.Cell Biol
ogy、80、248、1979)は50%集密度まで
成長させ、上記組み合せでIFN−γおよび/またはT
GF−β2処理を行なった。次に細胞はVersene
(LifeTechnologies Ltd.)を使
用して培養皿から分離し、標準的直接免疫蛍光技術を使
用して懸濁液で染色した。要約すれば細胞を洗浄し、正
常血清と5分、次にラットのクラスII MHC抗原を認
識するFITC−抱合マウス単クローン性抗体MRC
OX−6(セロテック)とインキュベートした。次に細
胞は洗浄し、少なくとも1時間1%パラ ホルムアルデ
ヒドで固定後、FACScan(Becton Dic
kinson)中で分析した。
胞は少なくとも一部IFN−γのような炎症シトカイン
により仲介されると思われる高量のクラスII抗原を発現
する。上記アッセイに使用されるHT−29未分化細胞
は本質的にクラスII分子を発現しない。腸炎症に侵され
る間に起こる部分的にせのできごとに対し、細胞はIF
N−γに暴露された。この反応に及ぼすTGF−β2の
効果を次に試験した。この効果以外ではHT−29細胞
に対するIFN−γ誘発のクラスII発現に対する暴露
は、すべての試験用量でTGF−β2による予備処理に
より取り消された(表2)。対照的に試験シトカインの
他の組み合せは高量のクラスII発現の結果となった。大
部分のIEC−18細胞は既にクラスII分子を発現した
が、発現の増加後IFN−γにより処理した(表3)。
もう一度、TGF−β2はこの誘発を抑制した。こうし
て、腸炎症反応中に、TGF−β2はクラスII抗原の局
部的発現をモジュレートすることができる。
MHC−クラスII抗原の実証からこれらの細胞が抗原供
与細胞として作用できるとする見解を支持する(May
er,L.ら、J.Exp.Med.,166、147
1〜1483、1987)。腸上皮細胞はIBDの疾病
原因論で主要な関係者と考えられている。MHC−クラ
スIIの発現の増加は上皮−T−ヘルパ−リンパ球の相互
作用を増加させ、これは順次IBDまたは永続性機作に
おいて主要なできごとでありうる。本研究は先ず第一に
腸上皮細胞に対しMHC−クラスII発現の抑制に対する
TGF−β2(およびTGF−β2−様 MGF)の作
用を実証する。これらの発見によれば、腸粘膜表面で局
所的に作用する免疫抑制剤の利用は、IBDおよび消化
管の他の炎症−免疫症状、すなわち小児脂肪便症および
移植組織対宿主反応に含まれる病原性機作を妨害する新
しい道具を供することができる。
2−様 MGFを、約10%の乾物を含む栄養的に均衡
のとれた経腸生成物に、こうして得た経腸生成物が約
0.1〜50、好ましくは0.5〜20μg TGF−
β2−様 MGF/g乾物量を含むような量で添加す
る。このように製造した経腸生成物は腸上皮細胞による
MHC−クラスII発現の抑制に有効であった。
2−様 MGFを液体または粉末形の均衡のとれた食品
に、こうして得た食品組成物が約0.1〜50、好まし
くは0.50〜20μgのTGF−β2−様 MGF/
g乾物量を含むような量で添加する。このように製造し
た食品組成物は腸上皮細胞によるMHC−クラスII発現
の抑制に有効であった。
Claims (14)
- 【請求項1】 ヒトまたは動物においてMHC発現のモ
ジュレーションに対し哺乳動物の乳または初乳由来のT
GF−β2−様 MGFの有効量を含有する食品組成
物。 - 【請求項2】 哺乳動物の乳または初乳由来のTGF−
β2−様 MGF量はヒトまたは動物の腸炎症病または
移植組織対宿主反応の治療に対し有効である、請求項1
記載の食品組成物。 - 【請求項3】 哺乳動物の乳または初乳由来のTGF−
β2−様 MGF量は離乳するヒトまたは動物の下痢予
防に対し有効である、請求項1記載の食品組成物。 - 【請求項4】 哺乳動物の乳または初乳由来のTGF−
β2−様 MGF量はヒトまたは動物の胃腸管のアレル
ギー反応の予防に対し有効である、請求項1記載の食品
組成物。 - 【請求項5】 請求項1〜4のいずれか1項に記載の食
品組成物の製造に対し哺乳動物の乳または初乳由来のT
GF−β2−様 MGFの使用。 - 【請求項6】 ヒトまたは動物のMHC発現のモジュレ
ーションに対し哺乳動物の乳または初乳由来のTGF−
β2−様 MGFの有効量を含有する経腸調製品。 - 【請求項7】 哺乳動物の乳または初乳由来のTGF−
β2−様 MGF量はヒトまたは動物の腸炎症病または
移植組織対宿主反応の治療に対し有効である、請求項6
記載の経腸調製品。 - 【請求項8】 哺乳動物の乳または初乳由来のTGF−
β2−様 MGF量は離乳するヒトまたは動物の下痢予
防に対し有効である、請求項6記載の経腸調製品。 - 【請求項9】 哺乳動物の乳または初乳由来のTGF−
β2−様 MGF量はヒトまたは動物の胃腸管のアレル
ギー反応の予防に対し有効である、請求項6記載の経腸
調製品。 - 【請求項10】 請求項6〜9のいずれか1項に記載の
経腸調製品の製造に対し哺乳動物の乳または初乳由来の
TGF−β2−様 MGFの使用。 - 【請求項11】 ヒトまたは動物のMHC発現のモジュ
レーションに対し哺乳動物の乳または初乳由来のTGF
−β2−様 MGFの有効量を含有する医薬製剤の製造
に対し哺乳動物の乳または初乳由来のTGF−β2−様
MGFの使用。 - 【請求項12】 医薬組成物はヒトまたは動物の腸炎症
病または移植組織対宿主反応の治療に対するものであ
る、請求項11記載の使用。 - 【請求項13】 医薬組成物は離乳するヒトまたは動物
の下痢予防に対するものである、請求項11記載の使
用。 - 【請求項14】 医薬組成物はヒトまたは動物の胃腸管
