合成原料として有用な物質である。たとえば(R)−
3−クロロ−1,2−プロパンジオールはL−カ
ルニチンの合成に利用されている(特開昭57−
165352)。
(従来の技術と問題点)
(R)−3−ハロゲノ−1,2−プロパンジオール
の製法に関しては、Hayden F.Jonesによりメチ
ル−5−クロロ−5−デオキシ−α−L−アラビ
ノフラノシドより得る方法〔ケミストリー・アン
ド・インダストリー(Chemistry and Industry)
P.538、15July、1978〕、又H.Jacksonらにより
1,2,5,6−ジアセトニル−D−マンニトー
ルより得る方法〔ケミストリー・アンド・バイオ
ロジカル・インターアクシヨンズ(Chem.−
Biol.Interactions)、13、193(1976)〕、同様にY.
Kawakamiらの方法〔ジヤーナル・オブ・オー
ガニツク・ケミストリー(Journal of Organic
Chemistry、47、3581(1982)〕などが知られてい
る。又(S)−3−ハロゲノ−1,2−プロパンジオ
ールの製法に関しては、H.F.Jonesによるメチル
−6−クロロ−6−デオキシ−α−D−グルコピ
ラノシドより得る方法(西独特許第2743858号)、
Porter K.E.らによる1,2,5,6−ジアセト
ニル−D−マンニトールより得る方法〔Chem.−
Biol.Interactions.41、95(1982)〕などが知られ
ている。
しかしながらこれらの方法は原料物質が入手し
にくかつたり、工程が複雑であつたりして工業的
製法とはいいがたく、工業的に有利な光学活性3
−ハロゲン−1,2−プロパンジオールの製造が
望まれていた。
本発明者らは従来高価な原料を用い複雑な反応
によつて得ていた光学活性3−ハロゲノ−1,2
−プロパンジオールの工業的生産を目指して研究
した結果、安価な(R,S)−3−ハロゲノ−1,
2−プロパンジオールに微生物を作用させること
により(S)−3−ハロゲノ−1,2−プロパンジオ
ールを選択的に代謝させ、(R)−3−ハロゲノ−
1,2−プロパンジオールを残存させうること
(特願昭60−264568、特願昭61−000409)、又は(R)
−3−ハロゲノ−1,2−プロパンジオールを選
択的に代謝させ、(S)−3−ハロゲノ−1,2−プ
ロパンジオールを残存させうること(特願昭60−
264567)を既に見い出している。
(問題点を解決するための手段)
本発明者らは、微生物反応利用による光学活性
3−ハロゲノ−1,2−プロパンジオールの生産
性をたかめるため鋭意研究した結果、SH基含有
化合物を反応液中に添加することにより基質濃度
を高め、生産性向上につなぎうることを見い出
し、本発明を完成するに至つた。
本発明に使用しうるSH基含有化合物としては
ジチオスレイトール、ジチオエリトリトール、グ
ルタチオン、システイン、メルカプトエタノー
ル、チオグリセリン、2,3−ジメルカプトプロ
パノール、チオ酢酸、ジメルカプトコハク酸、2
−メルカプトプロピオン酸、3−メルカプトプロ
ピオン酸、チオグリコール酸、tert−ブチルメル
カプタン、水硫化ナトリウム、水硫化カリウムな
どがある。
光学活性(S)−3−ハロゲノ−1,2−プロパン
ジオールのみを選択的に代謝し、(R)体を蓄積させ
る場合には、微生物としてブレタノミセス
(Brettanomyces)属、キヤンデイダ(Candida)
属、エンドミセス(Endomycea)属、ゲオトリ
カム(Geotrichum)属、ナドソニア
(Nadsonia)属、パシソレン(Pachysolen)属、

一方、光学活性(R)−3−ハロゲノ−1,2−プ
ロパンジオールのみを選択的に代謝し、(S)体を蓄
積させる場合には、微生物としてピキア
(Pichia)属、トリコスポロン(Trichosporom)
属、コリネバクテリウム(Corynebacteriun)属
に属する微生物を挙げることができる。更に詳し

上記の如き微生物を培養する為の培地組成とし
ては、通常これらの微生物が生育しうる培地なら
何でも使用しうる。たとえば炭素源としてグルコ
ース、シユークロース、マルトースなどの糖類;
エタノール、グリセロール、1,2−プロパンジ
オールなどのアルコール類;酢酸、乳酸などの有
機酸類、又はこれらの混合物、窒素源として硫酸
アンモニウム、リン酸アンモニウム、尿素、イー
ストエキス、肉エキス、ペプトンなど、他に無機
塩、微量金属塩、ビタミン類など通常の培養に用
いられる栄養源を適宜混合して用いることができ
る。
上記微生物の培養は常法によればよく、例えば
培地PHを4.0〜9.5の範囲とし、培養温度を20〜45
℃の範囲にて好気的に10〜96時間培養するのが好
ましい。
(R,S)−3−ハロゲノ−1,2−プロパン
ジオールに微生物を作用させて光学活性3−ハロ
ゲノ−1,2−プロパンジオールを得る方法とし
て、前記の如く培養した培養液あるいはこの培養
液から遠心分離なとによつて得られる菌体の懸濁
液に基質を添加する方法、あるいは培地に基質を
添加し、培養と反応を同時に行なう方法、常法に
より固定化した微生物を適当な緩衝液に懸濁した
ものに基質を添加する方法などがある。
反応温度は15〜50℃、反応PHは4.0〜10.0の範
囲で行なうことが好ましく、PHを保持する為に適
宜緩衝液などを用いることができる。反応液中の
基質濃度は0.1〜10(W/V)%が好ましく、基質
は初期に一括して加えてもよいし分割添加しても
よい。その際、反応液中にSH基含有化合物を添
加する。添加するSH基含有化合物の濃度は0.05
〜10(w/v)%が好ましく、添加方法としては
基質と一緒に初期に一括して加えてもよいし分割
添加してもよい。
反応は通常振盪あるいは撹拌しながら行ない、
反応時間は基質濃度・酵素量その他の反応条件な
どによつて変わるが、72時間以内で終了するよう
に条件を選択するのが好ましい。反応の停止は残
存基質をガスクロマトグラフイーなどで分析し、
基質が約50%消費された付近で止めるのが収率の
面で好ましい。
このようにして得られた光学活性3−ハロゲノ
−1,2−プロパンジオールを反応液から採取す
るには、一般な光学不活性な3−ハロゲノ−1,
2−プロパンジオールを採取する方法を用いるこ
とができる。たとえば反応液から菌体を遠心分離
などで除いた後、上清を適当に濃縮し、酢酸エチ
ルなどの溶媒で抽出する。抽出液を無水硫酸ナト
リウムなどで脱水後、減圧で溶媒を除去すると光
学活性3−ハロゲノ−1,2−プロパンジオール
のシロツプを得ることができる。また蒸留により
更に精製してもよい。
(実施例)
以下実施例により本発明を具体的に説明する。
実施例 1
グリセロール 4%、(NH4)2HPO4 1.3%、
KH2PO4 0.7%、MgSO4・7H2O 800ppm、
ZnSO4・7H2O 60ppm、FeSO4・7H2O 90ppm、
CuSO4・5H2O 5ppm、MnSO4・4H2O 10ppm、
NaCl 100ppm、イーストエキス 0.3%からなる
培地を脱イオン水で作製し(PH7.0)、500ml坂口
フラスコに50mlずつ分注し、120℃で20分殺菌し
た。
上記培地にサツカロミコプシス・リポリテイカ
IFO 0717を接種し、30℃にて48時間振盪培養し
た。この培養液計150mlを遠心分離して菌体を進
め、水洗後、菌体を0.3Mリン酸緩衝液(PH7.0)
50mlに懸濁した液に(R,S)−3−クロロ−1,
2−プロパンジオールを0.5g及び表1に示した
SH基含有化合物を0.15g添加して30℃24時間振
盪しながら反応させた。
基質の分解率は反応液1mlを2mlの酢酸エチル
で抽出後、ガスクロマトグラフイーにて分析し、
測定した。

特に効果のみられたチオグリセリン及び水硫化
カリウム添加区について、以下のフローにて精製
を行つた。
反応液を遠心分離により除菌し、上清を約10ml
まで濃縮し、30mlの酢酸エチルで3回(計90ml)
抽出した。抽出液を無水硫酸ナトリウムで脱水
し、減圧下で溶媒を除去したところシロツプが得
られた。このものの蒸留後、比旋光度を測定した
ところ以下の値が得られた。

実施例 2〜40(ただし、6は欠番)
菌株を表2に示したものに変更した以外は実施
例1と同様の方法で実施し、ただし、17〜37は炭
素源としてグルコースを用いた。表2に示す結果
を得た(但し、SH基含有化合物としてチオグリ
