Description
【発明の詳細な説明】
本発明は、ピロリチジン類の新規な製造法に係
わる。さらに詳述すれば、本発明は、分子内に適
当な脱離基を有するオレフイン化合物とアチリジ
ンとを反応させ、ついで得られたピロリジン体を
塩基の存在下反応させることによりピロリチジン
類を製造する方法に係わる。
ピロリチジン化合物は、下記の基本構造を有す
る化合物である。
このようなピロリチジン骨格を有する化合物と
して、多数のアルカロイドが自然界に存在するこ
とはよく知られている。これらの化合物は各種の
生理作用および薬理作用を有するものであり、生
理作用および薬理作用について多くの報告があ
る。たとえば、以下のような報告がある。

これらの作用はほんの一例であり、非常に多岐
にわたる生理活性を与えている。それ故に、ピロ
リチジン骨格を有する化合物の合成方法もまた数
多く報告されている。たとえば以下の例がある。

これらの合成例は、ピロリチジン化合物の合成
方法の一部であり、この他多数の報告がある。
上記従来法による合成例1)については、工程
数は少ないが官能基を導入することが困難であ
り、例2)に至つては、特殊な試薬を用いなけれ
ばならない。また、ピロリチジンの7位に官能基
を有する化合物の合成例3)では、工程数が多く
なるばかりでなく、原料、試薬等の経済性につい
ても難点がある。また、他の報告についても同様
の欠点が指摘される。
発明者らは、従来法の欠点を解消するととも
に、各種のピロリチジン誘導体の合成に広く応用
できる合成法の開発を目的として研究を進め、本
発明の完成に至つた。
本発明の目的は、
一般式
[式中、R1およびR2は、水素またはヒドロキ
シ基(保護されていてもよい)であるか、または
R1およびR2は一緒になつて=Oまたはカルボニ
ル保護基を表してもよく;R3およびR4は、それ
ぞれ−CO2R、−COSR、−CN、−SO2R、−S(O)
R、−SR、−SeR、−Se(O)R、−NO2の中から選
ばれる電子吸引官能基(ここでRはアルキル基ま
たはアリール基である)または水素であり、ただ
しR3およびR4が共に水素である場合を除く]で
表されるピロリチジン化合物の製法において、
一般式()
(式中、Xはハロゲン原子またはp−トルエン
スルホニルオキシ基、メタンスルホニルオキシ基
から選ばれる脱離基であり、R1、R2、R3および
R4は前記と同意義である)で表されるオレフイ
ン化合物を、構造式()
を有するアチリジンと反応させ、得られた一般式
()
式中、XおよびR1、R2、R3およびR4は前記と
同意義である)で表されるピロリジン体を塩基の
存在下で処理することを特徴とする、ピロリチジ
ン化合物の製法を提供することにある。
本発明の方法によるピロリチジン類の合成を反
応式で示せば次の通りである。
本発明の方法によれば、7位に官能基を導入す
ることが容易で、しかも特殊な試薬や反応条件を
必要とせず、短工程で目的のピロリチジン化合物
が合成できる。
本発明の方法で得られるピロリチジン化合物と
しては、前記一般式において、R1およびR2は、
水素またはヒドロキシ基またはヒドロキシ保護基
(たとえば−OMe、−OAc、−OTHP(THP=テト
ラヒドロピラニル)など)であるか、またはR1
およびR2は一緒になつて=Oまたはカルボニル
保護基(たとえば【式】
【式】など)を表してもよく、R3および
R4が、電子吸引官能基(たとえば−CO2R、−
COSR、−CN、−SO2R、−S(O)R、−SR、−
SeR、−Se(O)R、−NO2など(R=アルキル基
またはアリール基)または水素である化合物(た
だしR3およびR4が共に水素である場合を除く)
であり、たとえばエチルピロリチジン−1−エク
ソーカルボキシレート、エチル−1−フエニルセ
レノピロリチジン−1−カルボキシレートなどで
ある。
前記反応式における原料化合物(1)は、従来より
公知のオレフイン体を合成する一般的な方法に従
つて容易に調製できる。一般()におけるXは
Cl、Br、Iなどのハロゲン原子またはp−トル
エンスルホニルオキシ基、メタンスルホニルオキ
シ基などの通常用いられる脱離基である。
アチリジン(2)は安価な化合物で、反応性に富
み、オレフイン化合物(1)との反応では一挙にピロ
リジン化合物(3)を与える。
ついで、得られたピロリジン化合物(3)を塩基の
存在下で処理することにより、目的のピロリチジ
ン化合物(4)が得られる。この際使用できる塩基と
しては、リチウムジイソプロピルアミド
(LDA)、t−BuOK、NaOMe、NaHなどがあ
る。
また、必要ならば塩基処理と同時に、φSeCl、
φSe・Seφ、φSCl、ハロゲン、アルキルハライド
などを反応させると種々の官能基を1位に導入す
ることができる。
さらに、本発明の原料となるオレフイン化合物
(1)を種々選択することにより、これまで知られて
いるピロリチジン化合物のみならず新規なピロリ
チジン化合物を短工程で、しかも経済的に合成す
ることが容易である。
また、本発明の方法によつて得られた化合物
は、種々の天然ピロリチジンアルカロイド化合物
への変換あるいは有用な生理活性を有する他のピ
ロリチジン誘導体を合成するため重要な中間体と
しても使用できる。
以下に本発明の製法の好適な1具体例である実
施例にて本発明を詳細に説明するが、本発明はこ
れに限定されるものではない。
実施例 1
(i) エチル−6−クロロ−2−ヘキセノエート
(前記一般式において、X=Cl,R1,R2,R4
=H,R3=CO2Et)の合成
まず原料のオレフイン化合物(1)として、エチ
ル−6−クロロ−2−ヘキセノエートを以下の
如くして調製した。
水素化ナトリウム2g(油性60%)を無水ベ
ンゼン(30ml)に懸濁させ、撹拌下にエチルジ
エチルホスホノアセテート11.25gを加え、1
時間撹拌した後、4−クロロブタナール5.32g
の無水ベンゼン溶液(10ml)を加え、さらに12
時間撹拌した。反応液を水洗し、無水硫酸ナト
リウムにて乾燥後溶媒を留去した。得られた油
状物を減圧蒸留(bp91〜92℃,3mmHg)に付
したところ、オレフイン化合物(1)7.9g(収率
89.6%)が得られた。
生成物についての各種分析の結果は次の通り
である。

(ii) エチル−N−(β−クロロエチル)−2−ピロ
リジンアセテート(前記一般式()において
X=Cl,R1,R2,R4=H,R3=CO2Et)の合
成
上記化合物(1)5gおよびアチリジン(2)20mlの
混合物を0℃にて3.5時時間撹拌し、ベンゼン
50mlを加えた後、過剰のアチリジンおよび溶媒
を10℃にて留去した。得られた油状物を減圧蒸
留(bp113〜115℃,3mmHg)に付したとこ
ろ、エチル−H−(β−クロロエチル)−2−ピ
ロリジンアセテート(3)4.51g(収率72.5%)が
無色油状物として得られた。
得られた生成物の各種分析の結果は次の通り
である。

(iii) エチル−ピロリチジン−1−エクソ−カルボ
キシレート(前記一般式()においてR1,
R2,R4=H,R3=CO2Et)の合成
ジイソプロピルアミン0.77ml、15%n−ブチ
ルリチウム−ヘキサン溶液3.5mlおよび無水
THF15mlの混合液に、上記の如くして得られ
た化合物(3)1gのTHF溶液15mlを−78℃にて
加えた。−78℃にて3時間撹拌した後、室温に
てさらに12時間撹拌し、水20mlを加え、エーテ
ルにて抽出した。抽出液を水洗し、無水硫酸ナ
トリウムにて乾燥した後、溶媒を留去した。さ
らに減圧蒸留(bp80〜82℃、3mmHg)に付し
たところ、エチル−ピロリチジン−1−エクソ
−カルボキシレート(4)0.715g(収率85.8%)
が無色油状物として得られた。
生成物についての各種分析の結果は次の通り
である。

実施例 2
エチル−1−フエニルセレノピロリチジン−1
−カルボキシレート(一般式()において
R1,R2=H,R3=COOEt,R4=−Seφ(化合
物4a)およびR1,R2=H,R3=Seφ,R4=
COOEt(化合物4b)の合成
ジイソプロピルアミン3.1ml、15%n−ブチル
リチウム−ヘキサン溶液14mlおよびTHF30mlの
混合液に、前記実施例1の如くして得られた化合
物(3)2gのTHF溶液30mlを−78℃にて加え、該
温度で1時間撹拌した後、徐々に−40℃まで温度
を上げ、その後再度−78℃まで冷却した。次い
で、−78℃にて塩化フエニルセレネニル(φ−
SeCl)2.1gのTHF溶液を加えて4時間撹拌した
後、水5mlを加え、エーテルにて抽出した。抽出
液を水洗し、無水硫酸ナトリウムにて乾燥後、溶
媒を留去した。得られた残留物をシリカゲルクロ
マトグラフイーに付し、塩化メチレン−メタノー
ル−水酸化アンモニウム(90:9:1v/v)の
溶出部より化合物(4a)または(4b)0.6g(収
率19.5%)が淡黄色油状物として得られた。
さらに溶出することにより化合物(4b)また
は(4a)0.5g(収率16.2%)が油状物として得
られた。
得られた生成物の各種分析の結果は次の通りで
ある。

本発明はこの特定の実施例に限定されるものでは
なく、本発明の精神を逸脱しないで幾多の変化変
