Abstract

PURPOSE:To allow easy detection with a high sensitivity by forming the analysis piece consisting of a filter medium of a porous film which absorbs, holds or immobilizes a reagent for inspecting a specimen and a water4 absorptive film which is brought into tight contact with this filter medium. CONSTITUTION:The filter medium 1 of the porous film refers to a membrane filter of about 0.1 to 10 mum pore size, ultrafilter of about 10 to 1000Angstrom pore size, etc., and is formed in a network structure by using nitrocellulose, nylon, etc., for its base material. The analysis piece is obtd. by superposing the filter medium 1 which previously absorbs, holds or immobilizes the reagent for inspecting the specimen and the water absorptive film 2 which absorbs the moisture in the specimen, heating the laminate to about 70 to 80 deg.C to tightly adhere and integrate the medium and the sheet, and cutting the laminate to a required size. The spotted specimen first reacts with the reagent for inspecting the specimen on the filter medium 1 and the product remains on the filter medium 1. The excess moisture is then expelled onto the lower sheet 2 and, therefore, the moisture is practically condensed. Consequently, high-sensitivity detection is relatively easily executed.

Conventionally, test specimens for such liquid test tests have been made by impregnating the surface of a support made of plastic such as vinyl chloride with reagents, but test paper-type specimens made by adhering and molding using double-sided adhesive tape or the like have been used. It is widely used because of its simplicity. However, even with this type of test paper,
Since there is a detection limit naturally, when performing even more sensitive detection, RIA method, latex agglutination reaction method, etc.
More complicated tests are needed.

(Means for Solving the Problems) As a result of intensive research, the present inventors have found that the reaction product between a test liquid test reagent and a test liquid is poorly water-soluble under certain conditions, for example, under a certain pH range. In some cases, by closely integrating the porous film filter medium 1, which has previously adsorbed and retained or immobilized the test reagent, and the water-absorbing sheet 2 as shown in Fig. 1, the spotted test liquid can be absorbed. First, it reacts with the test liquid test reagent on the porous film filter medium 1, and the reaction product remains on the porous film filter medium, and the excess moisture is removed to the water absorbent sheet below, so that it is effectively concentrated. We have found that highly sensitive detection is relatively easy to perform.

That is, the gist of the present invention is that a porous film filter material that absorbs and retains or immobilizes a reagent for test liquid and a water-absorbing sheet that absorbs moisture in the test liquid are brought into close contact with each other. The feature lies in the highly sensitive dry analysis piece.

As a result, when compared with the conventional test strip for urine protein testing (Toyoga Paper KK Utsubis protein measuring section), the analytical strip of Example 1.2 has much higher sensitivity as shown in the table below. Therefore, it became possible to determine the presence or absence of an albumin concentration of approximately 3 mg/di.
