分 割 方 法
本発明は、一般式(I)
(式中、RおよびR2はそれぞれ、ハロゲンまたはCのアルキル基を示す)
で表わされる 2.2−ジメチル−3−(2,2−ジ置換ビニル)シクロプロパ
ンカルボン酸の光学異性体の新規な製造方法に関し、前記方法は、相当するラセ
ミカルボン酸と一般式(It)(式中、R3はシアノ基またはカルボキシアミド
基を意味する)で表わされる光学的に活性な塩基性2−フェニルグリシン誘導体
とから生成したジアステレオマー塩を分割することからなる。
一般式(I)の2.2−ジメチル−3−(2,2−ジ置換ビニル)シクロプロパ
ンカルボン酸は、効力が強くかつ商業的に供しうる殺虫剤を合成するための中間
体である。一般式(1)の酸は、その2個の不斉中心によって4個の立体異性体
の形で存在している。個々の立体異性体から得られるエステルの生化学的活性ス
ペクトルはかなり相違すると言われており、従って、出発物質としてのラセミ酸
から鏡像異性体を分離することが重要視されている。
工業的な合成法[DE−^2.713.5311および2.80+1.922
、および−ラセミ化合物を分離するいくつかの方法がすでに成功をおさめている
。また、−E/X−ラセミ酸およびトランろ−ラセミ酸の光学異性体を分離する
いくつかの方法も知られている1例えば、[;B−PS 1,178.423に
おいてはトランス−菊酸をキニンを用いて分割し、再結晶および塩生成をそれぞ
れ数回くり返して(IR)異性体を得ている。
一般式(I)の酸の異性体を分離するために、光学的に活性なα−ナフチルエチ
ルアミン(DE−^2.300.325) 、 α−フェニルアミノ−1−(
4−ニトロフェニル)−プロパン−1,3−ジオール(FR−PS 79131
16)を使用することが記載されている。これらの方法では、殆どの場合に分割
剤をラセミ酸に対する計算量と等量で使用しているが、これらの分割剤は高価で
入手が困難な化合物であり、しかも低効率でしか再生できないという欠点がある
。DE−^2.826.952においては、一般式(1)の酸の光学異性体を分
離するためにエチル (−j −2−フェニルグリシネートが使用されている。
ジアステレオマー塩の生成は水性媒体中で行われ、析出した結晶性塩もしくは母
液から得られる一般式(I)の光学的に活性な酸を石油エーテルから再結晶する
。
しかしながら、塩の生成のために種々の光学純度の分割剤が使用されるため、一
般式(I)の酸の鏡像異性体の光学純度も種々であった。この方法の重大な欠点
は、ジアステレオマー塩の生成がラセミ酸に対する計算量の50倍以」二の水中
で行われ、装置の利用度や生産性が低いことである。塩生成の母液中に残存して
いる鏡像異性体の精製方法は上述の方法においては論じられていない。上記特許
明細書の製造例において、トランス−ペルメトリン酸(pe+melh+1ni
c acid)の旋光能の値 (36°)は文献[DE−PS 2,628.4
77 ]に記載された旋光能の最高値よりも約10%低い。使用される分割剤は
加水分解作用に対して鋭敏であり、低効率でしか再生することができない。
このように、容易に入手しうる分割剤を用い、簡単な方法で光学的に純粋な状態
の一般式(I)の酸の光学゛異性体の両方を生成し得る製造方法の開発が試みら
れた。
一般式(■)(式中、R3はシアノ基またはカルボキンアミド基を意味する)の
塩基性(R)−2−フェニルグリシン誘導体が、一般式(I)のシクロプロパン
カルボン酸と、極性溶媒または極性溶媒混合物中で良好な結晶性塩を生成するこ
とを見出した。一般式(II)の分解剤は出発化合物としてのベンズアルデヒド
から簡単な方法で製造することができる。(R)−2−フェニルグリシンニトリ
ルはFR−PS 2.141.354に従って製造するこきができ、(R) −
2−フェニルグリシンアミドは公知の方法に従ってニトリルの酸性水和反応によ
り非常に好収率で製造することができる。
析出したジアステレオマー塩中に存在する一般式(1)の酸の配置は、いずれの
場合においても、−・般式(■)の分解剤のR3基の性質に依存するものである
という我々の認識は予期せざるものであり、従って新規である。R3がシアノ基
である場合には、いずれの場合においても、結晶塩中に(1s)配置を有する一
般式(I)の酸が多く存在し、R3がカルボキシアミド基である場合には、結晶
性ジアステレオマー塩がら(IR)配置を有する一般式(1)の酸が常に得られ
る。
この認識によれば、一般式(I)のシクロプロパンカルボン酸の両方の鏡像異性
体は、同じ配置を有する一般式(n)の分割剤を用いることによって純粋で結晶
性のジアステレオマー塩として得ることができる。
また、本発明は、水中または水とCl−4のアルコールとの混合物中で一般式(
ff)の分割剤を使用することによって、良好な効率で塩が生成されつるという
我々の認識に基くものである。
方法の効率という観点からすれば、一般式(I)のラセミ酸を1当量よりも少な
い量、適当には 1/2当量の一般式(II)の分割剤と反応させ、一般式(I
)の光学活性鏡像異性体混合物は、好ましくは、過剰に存在する鏡像異性体に対
して当量の一般式(II)の分割剤で分割を繰り返すことによって精製しつると
いう我々の所見は重要なものである。塩生成の過程において、分割剤と反応しな
い一般式(I)の酸は、そのアルカリ金属塩の形で溶液中に保持されている。
(R,)−2−7エニルグリシンアミドを用いて分割を行う場合に過剰のアルカ
リを使用することは、塩中に存在する酸鏡像異性体の収率および光学純度が減少
するので不適当であると言われている。(R)−2−フェニルグリシンニトリル
を使用する際に過剰のアルカリを存在させると、ジアステレオマー塩の収率は低
下するが、光学純度は増加する。
このように、本発明による方法は、一般式(I)のラセミ化合物(式中、R1お
よびR2は上記に規定した通りである)またはその塩と、一般式(II)のフェ
ニルグリシノ誘導体(式中、R3はシアノ基またはカルボキンアミド基を意味す
る)またはその酸付加塩とを反応させ、結晶の形で析出した一対のジアステレオ
マー塩の一つをそれ自体公知の方法で分離し、使用した一般式(Ir)の化合物
におけるR3がシアノ基を意味する場合には(IS)配置を有する光学的に活性
な一般式(I)の酸、または使用(また一般式(IT)の化合物におけるR3が
カルボキンアミド基を意味する場合には(IR)配置を有する光学的に活性な一
般式(I)の酸をそれぞれ酸によって遊離し、必要に応じてこの操作を1〜5回
繰り返し、母液中に残存するジアステレオマー塩をそれ自体公知の方法で回収し
、そこから一般式(1)の光学的に活性な対掌体酸をそれ自体公知の方法で取得
することからなる。
好ましくは、一般式(I)のラセミ酸1モルをアルカリ金属水酸化物1モルさ共
に水に溶解し、次に水中または水と01−4アルコールとの混合物中の一般式(
IF)の分割剤の塩酸塩溶液を20℃〜60℃の温匣で添加する。反応混合物を
o℃〜25℃の温度に冷却し、結晶性のジアステレオマー塩を清適によって分離

的に活性な酸は鉱酸を加えることによってシアステ17オマー塩から遊離し、清
適によって分離されるか、またはそれらを水−非混和性の有機溶媒中に抽出し、
次いで有機溶液を蒸発させて回収する。他の酸鏡像異性体は溶液中に最終的に存
在しているアルコール含有物を留去した後に、鉱酸で酸性化することによって塩
生成の濾液から回収される。

℃で残留水溶液[(R)−±−之赳(各−耐酸に富む物質を含有する]を加え、
次に反応混合物を5℃に冷却し、結晶性塩をが取し、それぞれ水5mlで2回洗
浄した。湿った塩を水15 mlに墾濁し、5モルの塩酸を加えてpHIまで酸
性にした。油状の析出物(IR)−1−ラ4スー耐酸をそれぞれクロロホルムI
Omlで3回抽出し、クロロホルム相を合わせ、硫酸ナトリウム上で乾燥し、

実施例 4
±−2?X−ラセミペルメトリン酸(一般式(I)においてRおよびR2が塩素
原子の化合物)4.18gをメタノール20 mlおよび5モルの水酸化ナトリ
ウム溶液2mlを含有する混合物中に溶解し、(R)−2−フェニルグリンンア
ミド152gを60”Cで撹拌上添加した。次に、水10m1を加え、室温まで
冷却し、ついで外部水冷によって0℃まで冷却し、析出物を消取し、水洗した。
湿った塩を水15+nlに懸濁し、10℃で撹拌しながら5モルの塩酸を加えて
plllまで酸性にした。結晶性の(IR)−九乏ンスーペルメトリン酸析出物
を消散し、水洗し、乾燥して生成物を得た。収量1.75go [α] D+
38.6° (c = 1. クロロホルム)。
塩生成の濾液を蒸発後、残渣を塩について上述したようにして後処理して(IS
)−トランス−ペルメトリン酸23gを得た。[α]D−20° (c=1.
クロロホルム)。
(IS)−トランス−ペルメトリン酸2.3gを水20m1と5モルの水酸化ナ
トリウム溶液1.2mlを含む混合物中に溶解し、次に水5mlに溶解した(R
)−2−フェニルグリシンニトリル塩酸塩を撹拌下25℃で添加した。混合物を
10℃に冷却し、析出物を消取し、それぞれ水3mlで3回洗浄した。湿った塩
を水10m1に懸濁し、5モルの塩酸を10℃で加えてpHlまで酸性にした。
結晶性の(13)−)ランス−ペルメトリン酸析出物をか取し、水洗、乾燥して
生成物 1.Igを得た。[α]D−38,6゜(c=1. クロロホルム)。
第2の塩生成の濾液に5モルの塩酸を10℃で加えてpHlまで酸性にし、はぼ
l・ランス−ラセミペルメトリン酸析出物を消取し、乾燥して生成物を得た。収
量1.15g、 [αコD−2.2° (C= 2. クロロホルム)。
