Description
本発明は(ω−ピペラジニルアルコキシ)アル
キレンジオキシベンゼン類又はその酸付加塩を有
効成分とする血圧降下剤に関するものである。
本発明における上記アルキレンジオキシベンゼ
ン類は下記一般式()で示される。
上記一般式()中mは1〜3の整数を示し、
nは2〜10の整数を示す。

ゲン原子、トリフルオロメチル基、アルコキシ
基、アルキルカルボニル基、アルキル基及びニト
ロ基から選択される1種以上の基で置換されてい
てもよいフエニル基を示す。
ハロゲン原子としてはフツ素原子、塩素原子、
シユウ素原子等が挙げられ、アルコキシ基として
は、メトキシ基、エトキシ基、プロポキシ基、ブ
トキシ基等の炭素数1〜5の低級アルコキシ基が
挙げられる。アルキルカルボニル基としてはアセ
チル基、プロピオニル基、ブチリル基、イソブチ
リル基等の炭素数1〜5の低級アルキルカルボニ
ル基が挙げられる。アルキル基としてはメチル
基、エチル基、プロピル基、イソプロピル基、ブ
チル基等炭素数1〜5の低級アルキル基が挙げら
れる。フエニル基上の置換基の位置及び置換基の
数は限定されない。またピペラジニルアルコキシ
基のジオキシベンゼン環上の置換位置もとくに限
定されない。
以下これらの一般式()で示される化合物の
製造法を説明する。

るピペラジン類との反応により得られる。
ハロゲノアルコキシアルキレンジオキシベンゼ
ン類とピペラジン類とはそれぞれ1:1で反応す
るが、通常ピペラジン類を過剰に使用する方が反
応が円滑に進行する。従つてピペラジン類はハロ
ゲノアルコキシアルキレンジオキシベンゼン類1
モルに対し1〜10モル使用される。
反応は無溶媒でも十分進行するが、反応を円滑
に進めるために、不活性溶媒を用いてもよい。溶
媒としては水、ジオキサン、テトラヒドロフラ
ン、ジメチルホルムアミド、ジメチルスルホキシ
ド、低級アルコールまたはこれらの二種以上の溶
媒の混合物が用いられる。
反応温度は特に限定されないが通常室温から
150℃である。
反応時間は反応温度及び原料の反応性、溶媒の
種類により異なるが通常10分から20時間の範囲で
ある。
また反応により生ずるハロゲン化水素を捕集し
て反応を促進させるために、塩基類を添加しても
よい。塩基類としては、水酸化カリウム、炭酸カ
リウム、水酸化ナトリウム、炭酸水素ナトリウ
ム、炭酸ナトリウム等の無機塩類、ピリジン、ト
リエチルアミン等の第三級有機アミン類等であ
る。その使用量はピペラジン類1モルに対し通常
1〜5モルである。
望ましい酸付加塩を得るためには、反応終了後
過剰のアミン類及び溶媒を蒸留あるいは水洗によ
り除き、水酸化ナトリウムあるいは水酸化カリウ
ム等の強塩基水溶液を加え、遊離のピペラジニル
アルコキシアルキレンジオキシベンゼン類とし、
その後エーテル、クロロホルム、ベンゼン、トル
エン等の溶媒で本化合物を抽出する。さらに望ま
しい酸を加えて中和すると目的とする酸付加塩が
得られる。

また上記化合物の酸付加塩も本発明に係る血圧
降下剤の有効成分として使用される。付加塩とし
て用いられる酸としては、塩化水素酸、シユウ化
水素酸、硫酸、リン酸、硝酸等の無機酸、酢酸、
コハク酸、アジピン酸、プロピオン酸、酒石酸、
フマル酸、マレイン酸、シユウ酸、クエン酸、安
息香酸、トルエンスルホン酸、メタンスルホン酸
等の有機酸が挙げられる。

以上の例示は、一般式()において、nが2
の場合の好ましい化合物の例であるが、例示した
化合物のそれぞれに対応するnが3から5までの
化合物もすべて好ましい化合物として例示され
る。
本発明に係る血圧降下剤はいかなる方法でも投
与できるが、好適には以下のような方法が実施さ
れる。
すなわち皮下注射、静脈内注射、筋肉注射、腹
腔内注射等の非経口投与もまた経口投与も可能で
ある。
投与量は患者の年令、健康状態、体重、同時処
理があるならばその種類、処置頻度、所望の効果
の性質等により決定される。
一般的に有効成分の1日投与量は0.1〜100mg/
Kg体重、通常1〜30mg/Kg体重であり、1回ある
いはそれ以上投与される。
経口投与する場合は錠剤、カプセル剤、粉剤、
液剤、エリキシル剤等の形体で、また非経口投与
の場合は液体あるいは懸濁等の殺菌した液状の形
体で用いられる。上述の様な形体で用いられる場
合、固体あるいは液体の毒性のない製剤的担体が
組成に含まれ得る。
固体担体の例としては通常のゼラチンタイプの
カプセルが用いられる。また有効成分を補助薬と
ともにあるいはそれなしに錠剤化、粉末包装され
る。
これらのカプセル、錠剤、粉末は一般的に5〜
95%、好ましくは25〜90%重量の有効成分を含
む。
すなわちこれらの投与形式では5〜500mg、好
ましくは25〜250mgの有効成分を含有するのがよ
い。
液状担体としては水あるいは石油、ピーナツ
油、大豆油、ミネラル油、ゴマ油等の動植物起原
の、または合成の油等が用いられる。
また、一般に生理食塩水、デキストロースある
いは類似のシヨ糖溶液、エチレングリコール、プ
ロピレングリコール、ポリエチレングリコール等
のグリコール類が液状担体として好ましく、とく
に生理食塩水を用いた注射液の場合には通常0.5
〜20%、好ましくは1〜10%重量の有効成分を含
むようにする。
経口投与の液剤の場合、0.5〜10%重量の有効
成分を含む懸濁液あるいはシロツプがよい。
この場合の担体としては香料、シロツプ、製剤
学的ミセル体等の水様賦形剤を用いる。
実施例
5−{4−(4−フエニル−1−ピペラジニル)
ブトキシ}−1,3−ベンゾジオキソール:KOH
(11g)を水(10ml)に溶解し、t−ブタノール
(150ml)を加えた溶液に3,4−メチレンジオキ
シフエノール(25g)およびジブロモブタン(90
ml)を加え、加熱還流下1時間撹拌する。反応終
了後、溶媒を留去し、残渣を酢酸エチルで抽出す
る。抽出液を飽和食塩水で洗浄後、無水硫酸ソー
ダで乾燥し、溶媒を留去後減圧蒸留して、5−
(4−ブロモブトキシ)−1,3−ベンゾジオキソ
ール(45g、収率91%、沸点135゜/1mmHg)を
得る。
5−(4−ブロモブトキシ)−1,3−ベンゾジ
オキソール(3g)をDMF(20ml)に溶解し、N
−フエニルピペラジン(1.8g)およびトリエチ
ルアミン(1.4g)を加え、室温下一夜放置する。
反応終了後、水(50ml)を加え、酢酸エチルで抽
出する。抽出液は一度飽和食塩水で洗浄し、無水
硫酸ソーダで乾燥する。溶媒を留去後、残渣をア
ルコール−エーテルに溶解し、20%塩化水素/エ
タノールを加え、析出した結晶を口取し、さらに
エタノールより再結晶することによつて、目的と
する5−{4−(4−フエニル−1−ピペラジニ
ル)ブトキシ}−1,3−ベンゾジオキソール塩
酸塩(3.8g、81%収率)が得られる。本化合物
の物性を表1のNo.3の欄に示す。
以下同様な方法で化合物を製造し、得られた化

試験例
本発明の血圧降下剤の血圧降下作用を以下の方
法で検討した。すなわち、動物は自然発症高血圧
ラツト(SHR)(300〜370g、5〜7月令)を用
い、エーテル麻酔下に尾動脈より挿入したカテー
テルにより、無麻酔下で観血的に血圧および心拍
数を測定し、薬物投与前の平均血圧および心拍数
を求めた後、1時間ごとに薬物を1、3、10、30
mg/Kgを経口投与し、降圧作用を判定し、投与前
値からの降下率で表わした。結果を表2に示す。
また、急性毒性値(LD50)はマウスを用い、

に示す。
本発明の血圧降下剤は表2に示す如く等しく、
1mg/Kg経口投与で十分な血圧降下作用を示し、
薬効の発現も速く、作用も持続的である。又、急
性毒性も弱く、薬効の発現量を考慮すれば非常に
