Description
本発明は(ω−ピペラジニルアルコキシ)ポリ
アルコキシベンゼン誘導体およびその酸付加塩に
関するものである。
さらに詳しくは血圧降下剤として有効な(ω−
ピペラジニルアルコキシ)ポリアルコキシベンゼ
ン誘導体に関するものである。
本発明化合物は下記一般式()で示される。
上記一般式中mは2または3の整数を示し、n
は3または4の整数を示し、R1は炭素数1〜3
のアルキル基を示し、R2はピリジル基、ピラジ
ニル基、ピリミジル基またはハロゲン原子、トリ
フルオロメチル基、アルコキシ基およびアルキル
基から選択される1種以上の基で置換されていて
もよいフエニル基を示す。
ハロゲン原子としてはフツ素原子、塩素原子、
シユウ素原子等が挙げられ、アルコキシ基として
は、メトキシ基、エトキシ基等炭素数1〜5の低
級アルコキシ基が挙げられる。
R2のアルキル基としてはメチル基、エチル基
等の炭素数1〜5の低級アルキル基が挙げられ
る。フエニル基上の置換基(R1O)の位置は限定
されないが2,6−、2,3−、3,4−、3,
5−、2,3,4−または3,4,5−等が挙げ
られる。
以下本発明化合物の製造法を説明する。
本発明の化合物は下記一般式()
(上記一般式中R1、m、nは一般式()にお
けるR1、m、nと同義であり、Xはハロゲン原
子を示す。)で示されるハロゲノアルコキシポリ
アルコキシベンゼンと下記一般式()
(上記一般式中R2は一般式()におけるR2と
同義である。)で表わされるピペラジン誘導体と
の反応により得られる。
ハロゲノアルコキシポリアルコキシベンゼンと
ピペラジン誘導体とはそれぞれ1:1で反応する
が、通常ピペラジン誘導体を過剰に使用する方が
反応が円滑に進行する。従つてピペラジン誘導体
はハロゲノアルコキシポリアルコキシベンゼン1
モルに対し1〜10モル使用される。
反応は無溶媒でも十分進行するが、反応を円滑
に進めるために、不活性溶媒を用いてもよい。溶
媒としては水、ジオキサン、テトラヒドロフラ
ン、ジメチルホルムアミド、ジメチルスルホキシ
ド、低級アルコールまたはこれらの二種以上の溶
媒の混合物が用いられる。
反応温度は特に限定されないが通常室温から
150℃である。
反応時間は反応温度及び原料の反応性、溶媒の
種類により異なるが通常10分から20時間の範囲で
ある。
また反応により生ずるハロゲン化水素を補集し
て反応を促進させるために、塩基類を添加しても
よい。塩基類としては、水酸化カリウム、炭酸カ
リウム、水酸化ナトリウム、炭酸水素ナトリウ
ム、炭酸ナトリウム等の無機塩類、ピリジン、ト
リエチルアミン等の第三級有機アミン類等であ
る。その使用量はピペラジン類1モルに対し通常
1〜5モルである。
望ましい酸付加塩を得るためには、反応終了後
過剰のアミン類及び溶媒を蒸留あるいは水洗によ
り除き、場合によつては水酸化ナトリウムあるい
は水酸化カリウム等の強塩基水溶液を加え、遊離
の(ω−ピペラジニルアルコキシ)ポリアルコキ
シベンゼン誘導体とし、その後エーテル、クロロ
ホルム、ベンゼン、トルエン等の溶媒で本化合物

以下本発明化合物の血圧降下作用について説明
する。
本発明化合物の血圧降下作用は以下の方法で検
討した。すなわち、動物は自然発症高血圧ラツト
(SHR)(300〜370g、5〜7月令)を用い、エ
ーテル麻酔下に尾動脈より挿入したカテーテルに
より、無麻酔下で観血的に血圧および心拍数を測
定し、薬物投与前の平均血圧および心拍数を求め
た、1時間ごとに薬物を1、3、10mg/Kgを経口
投与し、降圧作用を判定し、投与前値からの降下
率で表わした。最大降圧率は投与前の平均血圧よ
りの最大降下%を示し、SDBP10値は10%の降圧
を1時間維持する用量(mg/Kg.P.O.)を示す。
結果を表−1に示す。
また、急性毒性値(LD50)はマウスを用い、
リツチフイールド ウイルコクソン(Litchfield
−Wilcoxoh)法により算出し、その結果を表−
2に示す。
本発明化合物はは表−1に示す如く等しく、3
mg/Kg経口投与で十分な血圧降下作用を示し、薬
効の発現も速く、作用も持続的である。又、表−
2に示す如く急性毒性も弱く、薬効の発現量を考
慮すれば非常に安全性の高い薬物であると推定さ
れる。

本発明化合物はいかなる方法でも投与できる
が、好適には以下のような方法が実施される。
すなわち皮下注射、静脈内注射、筋肉注射、腹
腔内注射等の非経口投与もまた経口投与も可能で
ある。
投与量は患者の年令、健康状態、体重、同時処
理があるならばその種類、処理頻度、所望の効果
の性質等により決定される。
一般的に有効成分の1日投与量は0.1〜100mg/
Kg体重、通常1〜3mg/Kg体重であり、1回ある
いはそれ以上投与される。
本発明化合物を経口投与する場合は錠剤、カプ
セル剤、液剤、液剤、エリキシル剤の形体で、ま
た非経口投与の場合は液体あるいは懸濁等の殺菌
した液状の形体で用いられる。上述の様な形体で
用いられる場合、固体あるいは液体の毒性のない
製剤的担体が組成に含まれ得る。
固体担体の例としては通常のゼラチンタイプの
カプセルが用いられる。また有効成分を補助薬と
ともにあるいはそれなしに錠剤化、粉末包装され
る。
これらのカプセル、錠剤、粉末は一般的に5〜
95%、好ましくは25〜90%重量の有効成分を含
む。
すなわちこれらの投与形式では5〜500mg、好
ましくは25〜250mgの有効成分を含有するのがよ
い。
液体担体としては水あるいは石油、ピーナツ
油、大豆油、ミネラル油、ゴマ油等の動植物起原
の、または合成の油等が用いられる。
また、一般に生理食塩水、デキストロースある
いは類似のシヨ糖溶液、エチレングリコール、等
のグルコール類が液状担体として好ましく、とく
に生理食塩水を用いた注射液の場合には通常0.5
〜20%、好ましくは1〜10%重量の有効成分を含
むようにする。
経口投与の液剤の場合は、0.5〜10%重量の有
効成分を含む懸濁液あるいはシロツプがよい。
この場合の担体としては香料、シロツプ、製剤
学的ミセル体等の水様賦形剤を用いる。
以上説明したように本発明化合物は血圧降下剤
として有効に使用できる。
実施例 1
1−〔3−(4−フエニル−1−ピペラジニル)
プロポキシ〕−3,4−ジメトキシベンゼン
1−(3−ブロモプロポキシ)−3,4−ジメト
キシベンゼン5.0gに、N,N−ジメチルホルム
アミド20mlとフエニルピペラジン3.5g、トリエ
チルアミン3.8mlを加え、室温で5〜6時間撹拌
する。
反応溶媒を減圧留去し、残査にエチルエーテル
70mlと2N水酸化ナトリウム水溶液10mlを加え、
水層分離し、更に水、飽和食塩水で順次洗浄し、
エチルエーテル層を無水硫酸ナトリウムで乾燥
後、氷冷撹拌下20%塩酸−酢酸エチル8.3mlを加
える。析出結晶を取し、エタノールより再結晶
し、1−〔3−(4−フエニル−1−ピペラジニ
ル)プロポキシ〕−3,4−ジメトキシベンゼ
ン・2塩酸塩6.6gを得る。(収率85%)
上記化合物の物性は表3のNo.1の欄に記載され
ている。
同様の方法によつて他の化合物も製造しうる。
