Description
本発明はベンゾチアジンカルボキシアミド誘導
体およびそれを含有する医薬組成物に関する。更
に詳しくは、優れた抗炎症作用を有するベンゾチ
アジンカルボキシアミド誘導体およびそれを含有
する抗炎症剤組成物に関する。そのような誘導体
として、本発明により一般式(1)
(式中、Xは水素原子またはハロゲン原子、
R1は低級アルキル基、R2は場合により低級アル
キル基、シクロアルキル基、トリフルオロメチル
基、低級アルコキシ基、低級アルキルチオ基、ハ
ロゲン原子、ジ−低級アルキルアミノ基、フエニ
ル基及びフエノキシ基から選ばれた1種又は2種
以上の置換基で置換されていることもあるフエニ
ル基、ヘテロ芳香族基、シクロアルキル基または
ナフチル基、R3は低級アルキル基、シクロアル
キル基、ハロゲノアルキル基、アシル基、ヒドロ
キシメチル基、カルボキシ基、低級アルコキシカ
ルボニル基、ハロゲノ低級アルコキシカルボニル
基、フエニル基、ベンジル基、カルバモイル基、
シアノ基、ジ−低級アルキルホスホリル基を表わ
す)で表わされる誘導体が提起される。
本発明において低級とは炭素数1から4迄の有
機基に対し用いられ、シクロアルキル基は主にシ
クロヘキシル基を意味し、更にヘテロ芳香族基は
主にフリル、チエニル、ピリジルおよびチアゾリ
ル基を表わす。
一般式(1)のベンゾチアジンカルボキシアミド誘
誘体は、下記一般式(2)
(式中、X、R1の定義は一般式(1)の場合と同
じ)で表わされるベンゾチアジンカルボン酸の活
性誘導体、例えばメチルエステルのような低級ア
ルキルエステル、酸クロライド、酸ブロマイドの
ようなカルボニルハライド、p−ニトロフエノー
ルとの活性エステル及びクロルギ酸エステルとの
活性酸無水物などと、一般式(3)
(式中、R2、R3の定義は一般式(1)の場合と同
じ)で表わされるアミン成分とを反応させること
により製造される。この場合、アミノは適当な酸
付加塩として用いられることもある。
このアミド化反応は普通のアミド製造条件下で
実施することが出来る。すなわち、通常、カルボ
ン酸(2)の活性誘導体に少なくとも等モルのアミン
(3)を用い、好ましくは溶剤の存在下に、そして場
合により酸捕集剤として作用する補助塩基の左右
下に、そして必要ならば冷却または加熱下で、例
えば−10℃〜200℃の温度範囲で反応を行うこと
が出来る。
溶剤または希釈剤としては、反応条件下で不活
性なもの、例えば芳香族炭化水素であるベンゼン
やトルエン、キシレン、エーテル類であるジオキ
サンやテトラヒドロフラン、ジメトキシエタン、
ハロゲン化炭化水素である塩化メチレンや四塩化
炭素、トリクロロエタンあるいはジアルキルアミ
ドであるN,N−ジメチルホルムアミドやN,N
−ジメチルアセトアミドなどが用いられる。
補助塩基として用いられる化合物には、例え
ば、炭酸ナトリウム、炭酸カリウム、炭酸水素ナ
トリウム、炭酸水素カリウムまたは第3級アミ
ン、例えばトリエチルアミンがあげられる。特に
補助塩基は、一般式(2)で表わされる出発物質をカ
ルボニルハライドの形で使用する場合や、一般式
(3)で表わされるアミン成分が酸付加塩の形で使用
される場合に要求される。
出発物質として用いられる式(2)のカルボン酸
は、既知化合物であり、例えば日特開昭52−
151180に開示の方法で製造出来る。しかし本発明
者らは、同様に公知である式(2)の化合物のメチル
エステルを効率良く加水分解して式(2)のカルボン
酸を製造する技術を開発したので参考例として後
述する。さた式(3)のアミン成分には、それ自体新
規な構造の化合物も含まれるが、市販品として購
入出来るものもある。
本発明の化合物は、価値ある薬理的特質である
抗炎症作用を示す。次表はWistar系雄性ラツト
を用いたカラゲニン浮腫法による抗炎症作用の測
定をウインターらの方法(Proc.Soc.EXP.&
Med.,111,544〜547(1962))に従い実施した場
合の本発明の種々の化合物を経口投与した際の浮
腫抑制率の測定結果を示したものである。

比較例として示した化合物N−ベンジル−4−
ヒドロキシ−2−メチル−2H−1,2−ベンゾ
チアジン−3−カルボキシアミド1,1−ジオキ
シドは特公昭51−42114の例2に記載の化合物で
ある。その抗炎症作用は本発明の化合物に比して
著しく弱いことが判つた。このことはまたJ.
Med.Chem.,14,(12),1171(1971)に記載のデ
ーターから明らかである。
本発明の化合物及びその塩は医薬品として、例
えば通常使用される担体物質との配合物の形態で
使用することが出来る。担体物質としては有機ま
たは無機の水、澱粉、ステアリン酸マグネシウ
ム、ゼラチン、アラビヤゴム、ラクトース、タル
ク、黄色ワセリン等をあげることが出来る。得ら
れる薬剤調製物は、固体の状態として、例えば錠
剤、カプセル剤、顆粒剤、丸剤、細粒剤、半固体
の状態として、例えば軟膏、または液体の状態と
して、例えば溶液、懸濁液もしくは乳剤として用
いることが出来る。薬剤調製物には必要に応じて
適当な補助剤、例えば保存剤、安定剤、乳化剤や
他の治療上価値ある物質を含ませることが出来
る。
本発明の化合物からなる医薬品の投与方法は疾
患の種類、患者の状態に応じて適宜選択すること
が望ましい。また一般にこれらの化合物は1日当
り10mgから1500mg、好ましくは20mgから1000mgの
範囲で投与されるのが適当である。
このように使用される本発明の化合物として好

次に本発明を実施例により更に説明する。
実施例 1
N−(1−フエニルエチル)−4−ヒドロキシ−
2−メチル−2H−1,2−ベンゾチアジン−3
−カルボキシアミド 1,1−ジオキシドの合
成:
4−ヒドロキシ−2−メチル−2H−1,2−
ベンゾチアジン−3−カルボン酸 1,1−ジオ
キシド5.1gをジイソプロピルエーテル51ml中に
加え、次いで塩化チオニル7.3mlを添加し加熱下
に撹拌した。還流温度で5時間反応後、減圧下に
溶媒及び未反応試剤を蒸発除去した。残留物に乾
燥塩化メチレン50mlを加え、更らにトリエチルア
ミン4.0g、1−フエニルエチルアミン2.4gを順
次加え室温で1夜撹拌した。反応液を水洗、乾燥
後溶媒を除去した。得られた粗生成物をシリカゲ
ルカラムクロマトグラフイーに付し、塩化メチレ
ンで溶出し得られた生成物を更に塩化メチレン−
n−ヘキサンの混合溶媒から再結晶し標題化合物
3.5gを得た。
融点:86〜88℃
実施例 2
実施例1の塩化メチレン溶媒の代りに、乾燥ジ
メチルホルムアミド50mlを溶媒とし、最初永水溶
中で30分撹拌し、次いで室温で一夜反応させた。
生成したトリエチルアミン塩酸塩を別し去り、
次いで減圧下に溶媒を除去した。残留物を塩化メ
チレン中に移しN−HCl次いで水で洗浄、乾燥後
得られた粗生成物を実施例1と同様の方法で精製
しN−(1−フエニルエチル)−4−ヒドロキシ−
2−メチル−2H−1,2−ベンゾチアジン−3
−カルボキシアミド 1,1−ジオキシド 5.3
gを得た。

The title compound was recrystallized from a mixed solvent of n-hexane.

α−トリフルオロメチルベンジルアミン塩酸塩
1.1gを無水エーテル20ml中に懸濁し、これにト
リエチルアミン0.5gを加え室温で30分間撹拌し
た。生成したトリエチルアミン塩酸塩を別し去
り、更に液からエーテルを除いて得られた残留
物に4−ヒドロキシ−2−メチル−2H−1,2
−ベンゾチアジン−3−カルボン酸メチルエステ
ル 1,1−ジオキシド1.4g及びキシレン30ml
を加え還流下に25時間、撹拌しながら反応を行つ
た。反応液からキシレンを除去し残留物をシリカ
ゲルカラムクロマトグラフイーに対し塩化メチレ
ンを用い溶出した。得られた混合物を更らにn−
ヘキサン−塩化メチレンの混合溶媒から再結晶し
標題化合物1.0gを得た。

(1) 4−クロルアントラニル酸エチルエステル塩
酸塩61.4gを濃塩酸(36.5%)500ml中に懸濁
し−5〜0℃に冷却した。この液に亜硝酸ナト
リウム20.4gを含有する水溶液70mlを滴下し
た。そのまま更に40分間撹拌した。次いで別に
二酸化イオウ166gを溶かした酢酸溶液600mlと
塩化第二銅・2水和物28.6gを溶かした水溶液
70mlとを混合した溶液を準備し、これを上記反
応液に30分間で加えた。その後−5〜−3℃で
1時間撹拌したあと冷却浴を取り外し、3時間
室温になる迄撹拌を続けた。この溶液を永水4
中に注加し、析出した生成物を取し、永水
で洗浄し4−クロル−2−クロルスルホニル−
安息香酸エチルエステル64.2gを得た。
融点:34〜35℃
(2) ピリジン1中にグリシンエチルエステル塩
酸塩31.6gを含有する懸濁液に4−クロル−2
−クロルスルホニル−安息香酸エチルエステル
64.2gを加え室温で7時間撹拌し、その後一夜
放置した。析出した固体を別し去り、液を
減圧下で濃縮し、残留物を酢酸エチル中で希塩
酸、次いで水で洗浄し、乾燥した。溶媒を減圧
下で留去し去り、しばらくすると残渣は固化し
4−クロル−2−(N−カルボエトキシメチル)
スルフアモイル−安息香酸エチルエステル62g
を得た。
融点:68〜70℃
(3) 乾燥トルエン1.2中にナトリウムエトキシ
ド16.1gを含有する懸濁液に4−クロル−2−
(N−カルボエトキシメチル)スルフアモイル
−安息香酸エチルエステル51.9gを加え60〜65
℃で6時間撹拌し、その後一夜放置した。次い
で反応液を永水1.5に注ぎ込み、希塩酸を加
え酸性とした。生成した結晶を取し、水洗し
7−クロロ−4−ヒドロキシ−2H−1,2−
ベンゾチアジン−3−カルボン酸エチルエステ
ル1,1−ジオキシド33.3gを得た。
融点:154〜156℃
(4) 水酸化ナトリウム5.3gを含む水溶液300mlに
エタノール450mlを加え、この液に7−クロロ
−4−ヒドロキシ−2H−1,2−ベンゾチア
ジン−3−カルボン酸エチルエステル1,1−
ジオキシド39.2gを添加した。得られた溶液に
室温でヨウ化メチル55gを1時に加えた。一夜
放置後、析出結晶を取し、水洗して7−クロ
ロ−4−ヒドロキシ−2−メチル−2H−1,
2−ベンゾチアジン−3−カルボン酸エチルエ
ステル1,1−ジオキシド36.1gを得た。
融点:126〜128℃
(5) キシレン10ml中に7−クロロ−4−ヒドロキ
シ−2−メチル−2H−1,2−ベンゾチアジ
ン−3−カルボン酸エチルエステル1,1−ジ
オキシド0.8gおよび1−フエニルエチルアミ
ン0.45gとを加え還流温度で8時間撹拌した。
一夜室温で放置して析出した結晶を取し、更
らにキシレンより再結晶して標題のN−(1−
フエニルエチル)−7−クロロ−4−ヒドロキ
シ−2−メチル−2H−1,2−ベンゾチアジ
ン−3−カルボキシアシド 1,1−ジオキシ
ド0.6gを得た。

た。析出物を別し去り、液から溶媒を除去し
て得られたオイル状残留物にキシレン30mlおよび
7−クロロ−4−ヒドロキシ−2−メチル−2H
−1,2−ベンゾチアジン−3−カルボン酸エチ
ルエステル 1,1−ジオキシド3.2gを加え還
流温度で15時間撹拌した。次いで反応液からキシ
レンを減圧下に除去し、残渣をn−ヘキサン−酢
酸エチル中で結晶化し、得られた生成物を更にキ
シレンより再結晶し標題化合物2.5gを得た。

実施例 63
N−(α−トリフルオロメチルベンジル)−4−
ヒドロキシ−2−メチル−2H−1,2−ベンゾ
チアジン−3−カルボキシアミド 1,1−ジオ
キシドを含有する座剤
活性成分の微粉末20mgをO.D.O(日清製油(株)製、
中鎖脂肪酸トリグリセライド)5mlに懸濁し、ソ
フトゼラチンカプセル皮膜に充填し坐剤を得た。
参考例
4−ヒドロキシ−2−メチル−2H−1,2−
ベンゾチアジン−3−カルボン酸 1,1−ジオ
キシドの合成:
水酸化ナトリウム32.0g(0.8モル)を300mlの
メタノールに溶解し、この液を50〜60℃に保たれ
た1のキシレン中に加え撹拌した。次いでメチ
ル4−ヒドロキシ−2−メチル−2H−1,2−
ベンゾチアジン−3−カルボキシレート 1,1
−ジオキシド26.9g(0.1モル)を加えた。次い
で温度を上げメタノールを蒸留により系外け留去
しながら、最終的には約120℃迄昇温した。この
間約2時間を要した。得られた懸濁状の反応液を
約15℃迄冷却し、生成物を別した。過物をア
セトンで洗浄し、その後永水500ml中に溶解し、
少量の不溶物を過して除去した。水溶液に永冷
却で36%濃度の濃塩酸を少量ずつ加えpHを1付
近の酸性状態とし、析出した結晶を別し、冷水
を500mlで洗浄した。さらにこの結晶を35℃のメ
タノール120ml中に溶解し、活性炭1.5gを加え、
撹拌後、活性炭を別除去し、液のメタノール
溶液を永水中で冷却しながら、これに冷水を200
ml注いだ。充分冷却した後、リン片状の結晶を
過により単離し、冷水で洗浄した。得られた結晶
を風乾し、標題化合物を20.7g(収率81.1%)を
得た。
融点:141℃(分解)

ート 1,1−ジオキシドを参考例の方法に従
つて加水分解し4−ヒドロキシ−2−エチル−
2H−1,2−ベンゾチアジン−3−カルボン
酸 1,1−ジオキシドを得た。
収率:79%
融点:132〜133℃(分解)
(2) 4−ヒドロキシ−2−メチル−2H−1,2
−ベンゾチアジン−3−カルボン酸 1,1−
ジオキシドとα−トリフルオロメチル−4−メ
トキシベンジルアミン塩酸塩を用いて実施例1
の方法に従つてN−(α−トリフルオロメチル
−4−メトキシベンジル)−4−ヒドロキシ−
2−メチル−2H−1,2−ベンゾチアジン−
3−カルボキシアミド 1,1−ジオキシドを
得た。
融点:128〜130℃(n−ヘキサン−エタノー
ルから再結晶)

N−(α−クロロメチルベンジル)−4−ヒドロ
キシ−2−メチル−2H−1,2−ベンゾチアジ
ン−3−カルボキシアミド 1,1−ジオキシド
の合成:
N−(α−ヒドロキシメチルベンジル)−4−ヒ
ドロキシ−2−メチル−2H−1,2−ベンゾチ
アジン−3−カルボキシアミド 1,1−ジオキ
シド1.0gを6mlの塩化チオニル中に加え、室温
で2時間撹拌した。次いで減圧下に過剰の塩化チ
オニルを留去し、残留物をクロロホルム中に取出
し、水洗した。クロロホルム層を乾燥した後、溶
媒を留去し、得られたオイル状物をシリカゲルカ
ラムクロマトグラフイーに付し、クロロホルムで
溶出した。更にn−ヘキサン−ジクロルメタン中
で活性炭処理を行ない、溶媒を留去して、標題の
化合物0.3gを得た。
