Description
本発明は、医薬の合成中間体として有用な新規
ハロゲン置換β−ラクタム化合物及びその製造法
に関する。さらに詳しくいえば、本発明は、式
()
(式中、X、YはX=ハロゲン、Y=水素または
X=Y=ハロゲンを表わす)
で表わされるハロゲン置換β−ラクタム化合物及
びその製造法に関するものである。
式()で表わされるハロゲン置換β−ラクタ
ム化合物は、文献未記載の新規化合物であり、医
薬の合成中間体として重要なものである。
例えば、式
で表わされるアズスレオナム(Azthreonam、
SQ26776)、はグラム陰性菌に対して強い抗菌活
性を示す新しい抗性物質とし注目されるβ−ラク
タム化合物であが、この化合物の多くの類縁体が
本発明の化合物()から合成できる。
例えば、以下に示す反応で、C3、C4の立体配
置がシスあるいはトランスである各種の誘導体が
合成できる(本出願人による特願昭57−193239参
照)。
本発明の化合物()において、X、Yによつ
て表わされるハロゲンは塩素、臭素あるいは沃素
を含む。更に、本発明の化合物()は、X、Y
がX=ハロゲン、Y=水素の場合、C6、C7位が、
あるいはX=Y=ハロゲンの場合にはC6位が不
斉炭素となり、したがつて各場合に対応した光学
異性体が存在するが、それらのいずれも本発明に
含まれる。

以下、本発明の化合物()の製造法について
詳しく説明する。
本発明の化合物()は、X、YがX=ハロゲ
ン及びY=水素である化合物(a)と、X=Y
=ハロゲンである化合物(b)を含み、両者は
採用するハロゲン化試剤、反応条件等に依存し
て、一つの反応で生成する比率が変わる。従つ
て、いずれの化合物を所望するかによつて、その
所望する化合物を主生成物として与えるハロゲン
化試剤、反応条件等が任意に設定される。ハロゲ
ン化試剤を作用させる前段階の反応、すなわち、
式()
で示される既知化合物8−オキソ−2,2−ジメ
チル−3−オキサ−1−アザビシクロ〔4.2.O〕
オクタン(製法は後述)を塩基と反応させて活性
アニオン体と成す段階は、前記二化合物(a、
b)製造の共通必須反応であつて、用いる塩基
としては、ブチルリチウム、リチウムジイソプロ
ピルアミド、リチウムジシクロヘキシルアミド、
リチウムイソプロピルシクロヘキシルアミド、あ
るいはリチウムヘキサメチルジシラジドのような
有機リチウム化合物またはリチウムアミド化合物
が採用でき、必要に応じてヘキサメチルホスフア
ミド、ヘキサメチレンテトラミンあるいは2,
2,6,6−テトラメチルピペリジン等のカチオ
ン捕捉剤を共存させた系で生成する化合物()
のアニオン体の活性を高くする方法が採用され
る。
こうして発生させた化合物()のアニオン体
にハロゲン化試剤を作用させることによつて、目
的の化合物()に変換できるが、ハロゲン化試
剤として、プラスに荷電したハロゲンを放出しう
る陽性ハロゲン試薬、例えばN−ハロゲノスクシ
ンイミド、N−ハロゲノアセトアミド、ベンゼン
スルホニルハライド、p−トルエンスルホニルハ
ライド、N−ハロゲノサツカリン、N−ハロゲノ
−N,N−ジベンゼンスルホンアミド、t−ブチ
ルハイポハライドあるいは分子状ハロゲン、具体
的にはN−ヨードスクシンイミド、N−ブロモス
クシンイミド、N−クロロスクシンイミド、N−
ブロモアセトアミド、ベンゼンスルホニルヨー
ド、ベンゼンスルホニルブロミド、p−トルエン
スルホニルブロミド、p−トルエンスルホニルク
ロリド、N−ブロモスサツカリン、N−ブロモ−
N,N−ジベンゼンスルホンアミド、t−ブチル
ハイポブロミド、臭素、ヨウ素等をハロゲン化試
剤として用いた場合は、式()でX=ハロゲ
ン、Y=水素である化合物(a)が主生成物と
して得られ、ハロゲン化試剤としてポリハロアル
カン、例えば四塩化炭素、四臭化炭素あるいはク
ロロトリブロムメタン等を作用させた場合は、式
()でX=Y=ハロゲンである化合物(b)
が主生成物として得られることを見出した。化合
物(b)の生成する割合はハロゲン化試剤と化
合物()とのモル比が大きい程高くなり、通常
はいずれのハロゲン化試剤も化合物()に対し
て1.0〜3倍モル等量を使用し、a、bのい
ずれを所望するかによつてこのモル比を増減する
ことができる。aとbは、通常のシリカゲル
カラムクロマトグラフイーによつて容易に分離採
取できる。一連の反応は、テトラヒドロフラン、
1,2−ジメトキシエタン等の無水有機溶剤中
で、好ましくはアルゴン、窒素あるいはヘリウム
等の不活性ガス雰囲気下で、通常−80℃〜25℃の
範囲の温度で、好好ましくは−80℃〜0℃の冷却
下で実施される。
上記ハロゲン化反応で生成する、7−ジハロゲ
ン置換化合物(b)は、必要に応じて、触媒の
存在下、還元反応に付して7−モノハロゲン置換
化合物(a)に変換することができる。触媒と
しては、パラジウム触媒、ラネーニツケル等を用
いることができ、パラジウム触媒は5%パラジウ
ム−炭素、10%パラジウム−炭素、10%パラジウ
ム−BaSO4、30%パラジウム−BaCO3等の形で
用いられる。この時の反応溶剤としては、メタノ
ール、エタノール、ジオキサンあいは該溶剤と水
との混合溶剤が使用され、例えば触媒としてパラ
ジウムを用いる場合、化合物(b)に対して、
パラジウム金属として1〜10重量%、好ましくは
1〜2重量%使用し、反応過程で発生するハロゲ
ン化水素の捕捉剤として塩基、例えばリン酸水素
二ナトリウム、酢酸ナトリウムあるいは炭酸ナト
リウム等を1〜2.5倍モル当量共存させ、常圧か
ら数気圧の水素ガス雰囲気下に、20℃から100℃、
好ましくは20℃から40℃還元反応を実施すること
ができる。
本発明の方法でえられる7−モノハロゲン置換
β−ラクタム化合物(a)にあつては、C6、
C7位の立体化学がシス配置体とトランス配置体
の混合物として取得される。例えば、(6R)の立
体配置をもつ式()の化合物を出発物質とした
場合には、(6R、7R)のトランス体と(6R、7S)
のシス体の混合物して、また、(6S)の立体配置
をもつ式()の化合物の場合には、(6S、7S)
のトランス体と(6S、7R)のシス体の混合物と
してえられ、この混合比は主として熱力学的安定
性が支配する化学平衡によつて、通常シス:トラ
ンス=1:3〜4である。多くの場合、混合物の
ままで次の反応への中間化合物として特に支障な
く利用されるが、各異性体の単離を必要とする場
合には、高速液体クロマトグラフイーによつて、
シス体とトランス体各異性体を分離することがで
きる。
本発明の出発原料である化合物()は、
(6R)あるいは(6S)の光学活性体については大
野らの方法〔ジヤーナル・オブ・アメリカン・ケ
ミカル・ソサエテイー(J.Am.Chem.Soc.)、103
巻、2405頁、(1981年)〕でえられる(4R)また
は(4S)−〔(メトキシカルボニル)メチル〕−2
−アゼチジノンを本出願人開発の還元反応(特願
昭57−49527)に付して(4R)または(4S)−2
−ヒドロキシエチル−2−アゼチジノンとなし、
このアルコールをF.A.ブーフアードらの方法〔F.
A.ブーフアード(Bouffard)、D.B.ジヨンストン
(Johnston)、B.G.クリステンセン
(Christensen)、ジヤーナル・オブ・オルガニツ
ク・ケミストリー(J.Org.Chem)、45巻、1130頁
(1980)年)〕で()に導くことによつて製造で
き、6位のラセミ体はF.A.ブーフアードらの上
記文献に合成法が記載されている。
以下、実施例によつて本発明を具体的に説明す
る。
実施例 1
6(R)−7(R,S)−ブロモ−8−オキソ−
2,2−ジメチル−3−オキサ−1−アザビシ
クロ〔4.2.O〕オクタン()の合成
反応器を真空ポンプで吸引後、窒素ガス置換を
行ない、その後窒素気流下にジイソプロピルアミ
ン0.4gと窒素ガス雰囲気下蒸留精製した新鮮な
テトラヒドロフラン4mlを加え、混合溶液をドラ
イアイス−アセトン冷却する。次いで窒素気流、
攪拌下にn−ブチルリチウム(1.55Mヘキキサン
溶液2.3ml)を3分間で加えて、35分攪拌した。
こうして調製したリチウムジイソプロピルアミド
のテトラヒドロフラン溶液に、(6R)−8−オキ
ソ−2,2−ジメチル−3−オキサ−1−アザビ
シクロ〔4.2.O〕オクタン(化合物()、6R体)
0.5gをテトラヒドロフラン5mlに溶解した液を
6分間で加えて40分攪拌した。
次いでN−ブロモスクシンイミド0.6gをテト
ラヒドロフラン5mlとヘキサメチルホスホロアミ
ド0.5mlの混合液に溶解した液を6分間で加え、
ドライアイス−アセトン、窒素雰囲気下、5時間
攪拌反応させた。反応後、飽和塩化アンモニウム
水20mlを加えて反応を止め、混合物を酢酸エチル
で2回抽出する。有機層を無水硫酸マグネシウム
で乾燥後、過、濃縮して1gの油状物を得た。
この油状物をシリカゲル13g(トルエン中でカラ
ム調製)を用い、トルエン−アセトンを展開溶媒
として精製し、表記化合物の白色結晶0.3g(40
%)を得た。NMRスペクトルの積分値により、
6,7−トランス:6,7−シス体比は3:1で
あることを認めた。

実施例 2
実施例1で、ブロム化剤としてN−ブロモアセ
トアミドを用いた以外は全く同じ処方で6(R)−
7(R,S)−ブロモ−8−オキソ−2,2−ジメ
チル−3−オキサ−1−アザビシクロ〔4.2.O〕
オクタン()を得た。収率13%。(6,7−ト
ランス:6,7−シス体比=3:1)。
実施例 3
実施例1でブロム化剤としてp−トルエンスル
ホニルブロミドを用いた以外は全く同じ処方で6
(R)−7(R,S)−ブロモ−8−オキソ−2,2
−ジメチル−3−オキサ−1−アザビシクロ
〔4.2.O〕オクタン()をえた。収率41%。(6,
7−トランス:6,7−シス体比=2.74:1)。
また6(R)−7−ジブロモ−8−オキソ−2,2
−ジメチル−3−オキサ−1−アザビシクロ
〔4.2.O〕オクタン()は16.3%の収率で得られ
た。
実施例 4
実施例1でブロム化剤としてベンゼンスルホニ
ルブロミドを用いた以外は全く同じ処方で6(R)
−7(R,S)−ブロモ−8−オキソ−2,2−ジ
メチル−3−オキサ−1−アザビシクロ〔4.2.O〕
オクタン()をえた。収率36%。(6,7−ト
ランス:6,7−シス体比=2:1)。また6
(R)−7−ジブロモ−8−オキソ−2,2−ジメ
チル−3−オキサ−1−アザビシクロ〔4.2.O〕
オクタン()は11%の収率で得られた。
実施例 5
6(R)−7−ジブロモ−8−オキソ−2,2−
ジメチル−3−オキサ−1−アザビシクロ
〔4.2.O〕オクタン()の合成
反応器を真空ポンプで吸引後、窒素ガス置換を
行ない、その後、窒素気流下にジイソプロピルア
ミン6.6mlと窒素ガス雰囲気下蒸留精製した新鮮
なテトラヒドロフラン40mlを加え、混合溶液をド
ライアイス−アセトン冷却する。次いで窒素気
流、攪拌下にn−ブチルリチウム(1.55Mヘキサ
ン溶液28.5ml)を8分間で加えて100分攪拌した。
こうして調製したリチウムジイソプロピルアミド
のテトラヒドロフラン溶液に、化合物()(6R
体)3.25gをテトラヒドロフラン33mlに溶解した
液を13分間で加えて60分間攪拌した。次いで四臭
化炭素13.89gをテトラヒドロフラン35mlに溶解
した液を9分間で加えてドライアイス−アセトン
冷却、窒素雰囲気下で120分攪拌した後、氷−水
冷却にして更に20分攪拌反応させた。反応後PH
4.5のリン酸緩衝液(1M、100ml)を加え、酢酸
エチルで2回抽出を行なつた。有機層を無水硫酸
マグネシウムで乾燥後、過、濃縮、ポンプによ
る吸引を行なつて13.63gの固体物をえた。この
固体物をシリカゲル146g(トルエン中でカラム
調製)を用い、塩化メチレン、トルエン−アセト
ンを展開溶媒として精製し表記化合物の白色結晶
5.65g(86%)をえた。

素0.95gとリン酸水素二ナトリウム12水塩7.73g
の混合物にエタノール300mlと水(17滴)を加え、
1気圧の水素雰囲気下、室温で原料が消失するま
で攪拌反応させる。反応後、過により触媒を除
き、液よりエタノールを減圧下で留去し得られ
る混合物に水と塩化メチレンを加え分液抽出を3
回行なう。有機層を無水硫酸マグネシウムで乾燥
後、過、濃縮、ポンプによる吸引を行なつて
4.5gの6(R)−7(R、S)−ブロモ−8−オキ
ソ−2,2−ジメチル−3−オキサ−1−アザビ
シクロ〔4.2.O〕オクタン()を得た。収率89
%。(6,7−トランス:6,7−シス体比=
2.3:1)。

6(R)−7−ジクロロ−8−オキソ−2,2−

実施例5で四臭化炭素のかわりに四塩化炭素を

えた。収率58%。
