Description
〔技術分野〕
本発明はバシルス属細菌による新規なアミラー
ゼの製造法に関するものである。
〔従来技術〕
澱粉をグルコースあるいはマルトースに分解す
るグルコアミラーゼ、β−アミラーゼやα−アミ
ラーゼは自然界に広く存在することが知られてい
るが、より分子量の大きい、例えば、マルトトリ
オース(G3)、マルテトラオース(G4)、マルト
ペンタオース(G5)マルトヘキサオース(G6)
などのオリゴ糖単位で切断するアミラーゼについ
ての報告は少ない。
これらオリゴ糖は、食品の増量剤、賦形剤ある
いは甘味調整剤として注目されている。特に、
G4〜G7の各オリゴ糖は、診断用アミラーゼの活
性測定用基質としても注目されている。
〔目的及び効果〕
本発明者は、これらオリゴ糖の製法を確立する
ことを目的として、広く自然界より微生物の検索
を行つてきた結果、アミロース、アミロペクチ
ン、澱粉などのα−グルカンを特異的にマルトテ
トラオース(G4)に加水分解する新規なアミラ
ーゼがバシルス・サーキユランス(Bacillus
circulans)と同定した細菌により生産されるこ
とを認めた。
澱粉からマルトテトラオースを生成するアミラ
ーゼとしては、シユードモナス スツツエリ
(Pseudomonas stuzeri)の生産するアミラーゼ
が、アミロースやアミロペクチンの非還元性末端
からマルトテトラオース単位で加水分解すること
が知られている。
しかし、この酵素はアミロペクチンやグリコー
ゲンからは高分子量のリミツトデキストリンを残
すと報告されている。〔アーカイブ バイオケミ
ストリー アンド バイオフイジクス(Archive

(1971)〕。そして、この酵素による澱粉からのマ
ルトテトラオースの収量は約55%、アミロペクチ
ンからの収量は52%、と報告されている〔アメリ
カ合衆国特許USP3654082(1972)アミラーゼ
シンポジウム、6巻、31頁(1971)〕。このほか、
澱粉からマルトテトラオースを生成するアミラー
ゼとして、アミラーゼG4,5が知られている
(特公昭59−4119号公報)が、この酵素は、澱粉
からマルトテトラオースとマルトペンタオースの
両オリゴ糖を主成分として生成する酵素であつ
て、例えば、同公報1頁2欄34行〜2頁3欄4行
に、同酵素は澱粉から、マルトテトラオース15〜
35%、マルトペンタオース15〜30%含む糖化物を
生成すると記載されている。
しかるに、本発明のバシルス属細菌の生産する
アミラーゼは、澱粉から約65%〜75%の極めて高
い収量でマルトテトラオースを生成することが認
められた。しかし、マルトペンタオースの生成量
は第1表および第2図などから明らかなように、
極めて微量である。そして、マルトテトラオース
の収量は、使用する澱粉の種類、液化度(DE)、
酵素量などにより影響されるが、通常、グルコー
ス1〜5%、マルトース5〜15%

本発明は、バシルス属に属し、澱粉からマルト
テトラオースを主成分として生成する、エンド型
作用をもつα−アミラーゼであるアミラーゼG4
を生成する微生物を培養し、培養物からアミラー
ゼG4を採取することを特徴とするバシルス属ア
ミラーゼG4の製造法である。
以下に、本発明の内容を、更に具体的に説明す
る。
本発明により生産されるアミラーゼG4は、下
記の酵素的性質を有する。
(1) 作用;アミロース、アミロペクチン、グリコ
ーゲンなどのα−グルカンをマルトテトラオー
スを主成分とする分解物に分解する。本酵素は
エンド型の分解様式を持つα−アミラーゼの一
種であり、液化澱粉に作用させるとき約65〜75
%の収量でマルトテトラオースが得られる。
(2) 作用温度範囲及び最適作用温度;1%可溶性
澱粉、0.05Mリン酸緩衝液の下で作用させたと
き約75℃まで作用し、最適作用温度は約50℃で
ある。(第1図a)。
(3) 作用PH範囲及び最適作用PH;約4〜約12の広
い範囲に作用する。最適作用PHは6〜8.5であ
る(0.05M酢酸またはリン酸緩衝液、1%可溶
性澱粉下で作用、第1図b)。
(4) 熱安定性;0.1Mトリス緩衝液(PH7.0)の下
で加熱した場合、50℃、10分間の加熱で約40%
失活し、55℃、10分間の加熱で約85%失活した
〔第1図c〕。
(5) PH安定性;0.1M緩衝液の下で、室温(25℃)
で3時間放置後、残存活性を測定した。その結
果、PH6〜9の範囲で安定であつた〔第1図
d〕。
(6) 安定化;カルシウムイオンが存在するとき、
熱安定性の増加が認められた〔第1図cの破
線〕。
(7) 阻害剤;本酵素は、5×10-3MのHgCl2、
CuSO4、ZnSO4、AgNO3により、それぞれ、
約100%、約95%、約50%、約30%阻害された。
(8) 精製法;本酵素は、液体培養物の遠心上澄液
から、硫安分画、DEAE−セフアロース
(Sepharose)カラム クロマトグラフイーと
バイオゲル(Biogel)A0.5mカラム クロマト
グラフイー及び同ゲルによる再クロマトグラフ
イーにより電気泳動的に均一まで精製すること
ができる。
(9) 分子量;バイオゲル(Biogel)A0.5mを用い
たゲル濾過法により測定した分子量は約1万で
あつた。
(10) 力価測定法;0.1Mリン酸緩衝液(PH7.0)に
溶解させた2%可溶性澱粉0.5mlに適量の酵素
を加え、水で全量1mlとし、40℃で反応させ
る。この条件で1分間に1μモルのグルコース
に相当する還元力を生成する酵素量を1単位と
した。
以上の酵素的性質について、本発明以前に知ら
れているシユードモナス スツツエリのマルトテ
トラオース生成アミラーゼと比較すると(1)最適作
用PHは、アミラーゼG4はPH6〜8.5の広いPH範囲
にあるのに対し、シユードモナス スツツエリの
酵素はPH8付近にあること、(2)アミラーゼG4を
澱粉に作用させたとき、約65〜75%の収量でマル
トテトラオースが得られるのに対し、シユードモ
ナス スツツエリの酵素の場合は約55%であるこ
と、(3)アミラーゼG4の分子量は1万前後にある
のに対し、シユードモナス スツツエリの酵素
は、48000と58000にある〔バイオケミストリー
アンド バイオフイジクス アクタ
(Biochemistry and Biophysics Acta)566巻、
88頁(1979)〕など、アミラーゼG4とシユードモ
ナス スツツエリの酵素とは、澱粉からのマルト
テトラオースの収量、最適作用PH、分子量などの
性質において、顕著な違いのあるものであり、ア
ミラーゼG4は新規な酵素ということができる。
アミラーゼG4を生産する例示菌として、バシル
ス サーキユランス(Bacillus circulans)G−
4を挙げる。本菌の菌学的性質は下記に示す通り
であり、微工研条寄第820号として工業技術院微
生物工業技術研究所に寄託されている。
(1) 形態;桿菌、巾0.5〜1×5〜6μ、運動性な
し、グラム陰性。
(2) 胞子;通常、末端近くに1個、胞子嚢のふく
らみは認められない。
(3) 肉汁;混濁、沈降する。
(4) 肉汁寒天;生育良好、淡黄〜淡褐色、周辺及
び表面凹凸あり。
(5) グルコース肉汁寒天;生育良好、淡黄〜淡褐
色。
(6) グルコース硝酸寒天;生育悪い。
(7) ポテト;生育良好、乳白色に生育。
(8) グルコース・アスパラギン寒天;生育あまり
良くない。淡黄〜黄褐色。
(9) チロシン寒天;良く生育する、わずかに褐
色。
(10) ミルク;ゆつくり凝固、ペプトン化。
(11) インドール;生成しない。
(12) カタラーゼ;生成する。
(13) アセチルメチルカルビノール;生成しな
い。
(14) 硫化水素;生成しない。
(15) クエン酸;利用する。
(16) 硝酸塩の還元;陰性。
(17) クエン酸;利用する。
(18) 食塩肉汁;8%食塩含有培地までよく生育
し、10%食塩でも少し生育する。
(19) 炭水化物の利用;D−グルコース、D−フ
ラクトース、D−マンノース、L−アラビノー
ス、D−リボース、マルトース、シユークロー
ス、澱粉などから酸を生成するが、ガスの生成
はない。D−キシロース、L−ラムノース、L
−ソルボースの利用性は良くない。
(20) 最適生育温度;26℃前後。
(21) 最高生育温度;約60℃
(22) 死滅温度;100℃で30分間加熱しても死滅
しない。
以上の菌学的性質について、バージエイス マ
ニユアル オブ デタミネーテイブ バクテリオ
ロジー(Bergey's Mannual of Determinative
Bacteniology)の第7版及び第8版〔ザ ウイ
リアム アンド ウイルキンス カンパニー
(The Williams and Wilkins Company)、
(1957年及び1974年)を参照し、本菌をバシルス
サーキユランス(Bacillus circulans)の一種
と同定し、バシルス サーキユランスG−4と命
名した。
本発明によるアミラーゼG4を生産するための
培養は、窒素源として肉エキス、ペプトン、酵母
エキス、カゼイン、コーン・ステイープ・リカ
ー、大豆粕など、通常、微生物の培養に対し良く
用いられる有機窒素源が使用され、炭素源として
は、澱粉、デキストリン、マルトース、グルコー
ス、シユークロースなどが使用される。そして、
これに補足する栄養源として、無機窒素源、リン
酸塩、マグネシウム塩と各種金属塩を含む培地が
使用される。培養はPH5〜9、温度20〜60℃で好
気的に行われる。
アミラーゼG4は、菌体外に生産される酵素で
あるので、培養終了後、濾過または遠心分離によ
り除菌し、上澄液を回収する。必要により濃縮
し、硫安、硫酸ナトリウムによる塩析によるか、
または、アセトン、イソプロパノール、エタノー
ル、メタノールなどの有機溶媒を加えて、酵素を
沈澱物として収得し、乾燥、保存する。
アミラーゼG4を用いて、澱粉を糖化する反応
は次のようにして行う。澱粉は、酸または、α−
アミラーゼにより液化される。液化度はマルトテ
トラオースの収量に影響するので、望ましくは
DE20以下の液化澱粉が使用される(DEは固形分
中の還元力をグルコースとして表した百分率)。
基質濃度は、通常、5〜40%で行われる。反応PH
は、通常5〜9、温度は40〜60℃である。本酵素
は、カルシウムイオンの存在により、著しく熱安
定化されるので、糖化反応に際して、5×10-4〜
2×10-2M程度のカルシウム塩が添加される。
アミラーゼG4による、澱粉、アミロペクチン、
グリコーゲンなどα−1.6−グルコシド結合の分
岐をもつ基質を用いる糖化反応においては、イソ
アミラーゼ、プルナラーゼなどのα−1.6−グル
シダーゼの存在下で反応を行うと、糖化反応を促
進するため、アミラーゼG4を節減したり、また、
マルトテトラオースの収量を増加することができ
る。
次に、実施例により本発明の詳細を説明する。
実施例 1
ポリペプトン1%、リン酸2カリ0.3%、硫酸
マグネシウム(7水塩)0.1%、可溶性澱粉1%
からなる培地30mlを200ml容三角フラスコに入れ、
120℃で10分間殺菌したのち、バチルス・サーキ
ユランスG4(微工研条寄第820号)を接種し、30
℃で4日間振盪培養(160rpm)した。
培養後、遠心分離して得た上澄液について生産
されたアミラーゼG4を測定した結果、培地1ml
当たり6.4単位であつた。
実施例 2
実施例1で得られた酵素液を使用して、澱粉の
糖化を行つた。基質としては、DE4.2の液化澱粉
100mg(固形分)に、塩化カルシウム5×10-3M
とアミラーゼG4を第2表に記載の通り、基質1
g当たり0.5〜4単位加え、全量1mlとして、50
℃で44時間反応した。反応後、生成したマルトテ
トラオースを、高速液体クロマトグラフ法により
定量した結果は第2表に示す通りであつた。
