Description
発明の分野
本発明はテトラサイクリン系抗生物質の1種で
あるミノサイクリンの医薬組成物への安定化配合
に関する。
発明の背景
テトラサイクリン系抗生物質にはテトラサイク
リンをはじめ、ミノサイクリンを含めて数種の類
縁化合物が包含されるが、これらはいずれも非常
に不安定な物質であり、従来からテトラサイクリ
ン系抗生物質を医薬組成物へ安定に配合するため
の検討が種々なされている。
例えば、特開昭52−90616号は2−ピロリドン
水溶液中でマグネシウム化合物のようなアルカリ
土類金属化合物を用いてオキシテトラサイクリ
ン、ドキシサイクリン、テトラサイクリン、クロ
ルテトラサイクリンまたはそれらの塩をキレート
化することにより、これらのテトラサイクリン系
抗生物質の安定化を図つた注射用の水溶液を開示
している。また、特開昭53−94028号はアルカリ
土類金属イオン、ポリビニルピロリドンおよび脂
肪族アミドを配合し、PH5.0〜7.5にすることによ
り、オキシテトラサイクリンの安定化を図つた医
薬組成物を開示している。さらに、米国特許第
3335055号はテトラサイクリンをマグネシウムイ
オンおよびイソニコチン酸アミドなどのピリジン
誘導体で安定化する方法を開示している。
しかしながら、テトラサイクリン系抗生物質の
1種であるミノサイクリンについては、従来その
安定化を検討した例は見当らない。一方、近年、
ミノサイクリンの使用に際して、その安定化が要
望されるようになつている。
このような事情にかんがみ、本発明者らはミノ
サイクリンの医薬組成物への安定化配合について
鋭意検討した。その結果、マグネシウム化合物を
含有する多価アルコールを基剤とすることによ
り、ミノサイクリンが安定化され、これが、テト
ラサイクリン系抗生物質中でもミノサイクリンに
のみ見られる特異的な効果であることを知り、本
発明を完成するにいたつた。
発明の概要
本発明は、マグネシウム化合物を含有する多価
アルコールからなる基剤にミノサイクリンまたは
その医薬上許容される塩を配合したことを特徴と
するミノサイクリンを安定に配合した非水系の局
所投与用組成物を提供するものである。本発明に
おいては、所望により、基剤にさらに水溶性高分
子化合物を添加してもよく、該基剤が水溶性非水
系ペースト状であることに特徴を有する。
発明の詳説
用いるマグネシウム化合物としては医薬上許容
されるものであればいずれでもよく、例えば、塩
化マグネシウム、酢酸マグネシウム、硫酸マグネ
シウム、炭酸マグネシウム、グルコン酸マグネシ
ウムなどが挙げられ、ことに塩化マグネシウムが
好ましい。一般に、マグネシウム化合物は組成物
全体に対して0.5〜10%(重量%、以下同じ)、好
ましくは、1〜5%の割合で配合することによ
り、ミノサイクリンの良好な安定が図れる。
多価アルコールとしては、グリセリン、エチレ
ングリコール、ジエチレングリコール、プロピレ
ングリコール、ジプロピレングリコール、ヘキシ
レングリコール、1,5−ペンタンジオール、
1,3−ブチレングリコールなどが単独で、また
は混合して用いられ、ミノサイクリン安定化上、
ことにグリセリンが好ましい。
また配合するミノサイクリンは遊離のもので
も、例えば、塩酸塩、硫酸塩、トリクロル酢酸塩
などの医薬上許容される酸付加塩いずれでもよ
く、薬効上の観点から、一般に、遊離のミノサイ
クリンとして、組成物全体に対して15%程度ま
で、通常、0.1〜5%程度配合される。
本発明の組成物は、基本的に、マグネシウム化
合物と多価アルコールの混合物にミノサイクリン
が配合された粘稠な液状ないしはペースト状の組
成物である。
本発明の組成物は通常の製剤化技術に従つて製
造することができ、例えば、多価アルコールにマ
グネシウム化合物を溶解し、ついで、ミノサイク
リンまたはその塩を添加して所望の組成物を得る
ことができ、要すれば、加熱を行なつてもよい。
また、所望により、さらに水溶性高分子化合物、
可塑剤、界面活性剤等、ミノサイクリンの安定化
を損うことのない成分を添加してもよい。こと
に、ナトリウムカルボキシメチルセルロース、ア
ルギン酸ナトリウム、ポリビニルアルコール、カ
ラギーナン、ローカストビーンガム、グアーガ
ム、ヒドロキシメチルセルロース、ヒドロキシエ
チルセルロース、キサンタンガム、トラガントガ
ム、澱粉、スクシノグルカンなどの水溶性高分子
化合物を多価アルコールに対して0.2〜3%程度
の割合で添加して水溶性非水系のペースト状組成
物にすることにより、ミノサイクリンの安定性が
さらに向上し、投与もより容易になる。
本発明の組成物においては、用いる基剤各成分
はその安定性が充分に確認されているものであ
り、その適用範囲は特に限定されるものではな
く、例えば、皮膚外用により〓瘡などの治療に、
また、口腔内適用により感染性の歯周疾患の治療
等局所的に用いることができる。
実施例
つぎに実施例を挙げて本発明をさらに詳しく説
明する。

ミノサイクリンの安定化試験
以下の第1表に示す処方の種々のミノサイクリ
ン塩酸塩およびテトラサイクリン塩酸塩配合組成
物を調整し、40℃に保存し、1ケ月後および3ケ
月後のそれら抗生物質の力価を日本抗生物質基準
解説(1982年版)に準じて測定し、配合当初の力
価に対する力価残存率(%)を算出した。結果を
第1表に示す。

In view of these circumstances, the present inventors have conducted extensive studies on stabilizing the incorporation of minocycline into pharmaceutical compositions. As a result, we learned that minocycline is stabilized by using a polyhydric alcohol containing a magnesium compound as a base, and that this is a specific effect seen only in minocycline among tetracycline antibiotics. Summary of the Invention The present invention provides a non-aqueous topical composition stably containing minocycline, which is characterized in that minocycline or a pharmaceutically acceptable salt thereof is blended into a base consisting of a polyhydric alcohol containing a magnesium compound.
