Description
1.ヒラメを非破壊試料採取装置を用いて3回、5回繰り返して試料を採取。
2.採取した試料を3次滅菌蒸留水(10mLおよび20mL)に5回繰り返して懸濁。
3.試料が抽出された蒸留水を1mLずつ、遠心分離し、細胞試料を濃縮(遠心分離条件:10,000g、3min、4℃)。
4.濃縮した試料をDNA抽出キット(DNA extraction kit)を用いてDNA抽出(DNA extraction)。
5.抽出したDNAをPCR templateで用いてPCRを行う。
6.電気泳動によって特異なbase pair(bp)のバンドを確認。
N:陰性コントロール(クドア・セプテンプンクタータに感染していない試料)
Claims (8)
- 複数の針が備えられたシリンジと、
前記シリンジの移動を案内するためのガイドと、
前記ガイドの下端に連結され、前記シリンジに弾性復元力を伝達する針復元手段と、
を含み、
前記シリンジは、前記ガイドの中空に挿入され、先端運動が行われ、前記針が備えられたバレルおよび前記バレルの中空に挿入され、先端運動によって試料を吸引するプランジャーからなることを特徴とする、非破壊試料採取装置。 - 前記針復元手段は、前記針が出入りする開口部が形成され、前記ガイドのフランジに結合されるカバーと、前記カバーとフランジとの間に備えられたガイド部材と、前記ガイド部材に沿って移動し、針が横切る貫通孔が形成された弾性部材および前記弾性部材とカバーの間に介在された圧縮ばねからなることを特徴とする、請求項1に記載の非破壊試料採取装置。
- 前記貫通孔の内径は、前記バレルの外径よりも小さく形成されていることを特徴とする、請求項2に記載の非破壊試料採取装置。
- 前記バレルにはガイド突起が形成され、前記ガイドには前記ガイド突起が移動するガイド溝が形成されていることを特徴とする、請求項1に記載の非破壊試料採取装置。
- 前記ガイド溝は、前記ガイド突起が進入する第1ガイド溝と、前記バレルの離脱を防止する第2ガイド溝とからなることを特徴とする、請求項4に記載の非破壊試料採取装置。
- 前記ガイド突起が第1ガイド溝の下端に進入した後に前記バレルを回転させると、前記ガイド突起が第2ガイド溝に進入するように構成されたことを特徴とする、請求項5に記載の非破壊試料採取装置。
- 前記第1ガイド溝と第2ガイド溝との間には離脱防止突起が形成されていることを特徴とする、請求項5または6に記載の非破壊試料採取装置。
- 前記第2ガイド溝の下端に前記ガイド突起が回転によって進入する係止溝が形成されていることを特徴とする、請求項5または6に記載の非破壊試料採取装置。
