Description
(1)第1の試料として、RO水を使用した。
次に示す除外項目に当てはまらない健常成人(男性15名(33.3±3.5歳) 女性15名(35.9±9.7 歳))を対象とした。
2)鼻炎等で日常的に鼻づまりの方
3)他のボランティア試験に参加されている方
4)妊娠中および授乳中の方、あるいは試験期間中に妊娠の意思のある方
試料をガラス製バイアルに入れ、被験者には、メトロノームに合わせて4秒間に1回呼吸のリズムで1分間にわたり、そのにおいを嗅いでもらった。
ストレスの負荷には、内田クレペリンテストを用いた。このテストは、一桁の足し算の答えの下一桁を連続して用紙に記入していく作業を、休憩を挟んで前半と後半で各15分間ずつ合計30分間行うテストである。回答用紙から、試料毎に前半と後半の回答数と正答率を算出した。正答率は、以下の式に従って算出した。
主観的気分状態は、ビジュアルアナログスケール(Visual Analog Scale, VAS)をタッチパネル画面上(X軸に[リラックスした気分-緊張した気分]、Y軸に[疲れた気分-元気な気分])に表示して、被験者には、そのときの気分を表わす位置に自由にタッチしてもらい、その座標値から評価するシステム(KOKOROスケール)を用いて測定した。結果は、4回のサンプリングタイムをそれぞれT1:内田クレペリンテスト前、T2:内田クレペリンテスト中、T3:内田クレペリンテスト後およびT4:休憩後とし、入力のタイミング毎に、その平均値及び標準誤差で示した。また、ノンパラメトリックTukey型検定により群内比較を行った。
Profile of mood status 2短縮版(以下「POMS」という。)は、気分を評価する質問紙法の1つである。短縮版は、被験者に35項目の質問に答えてもらい、[怒り-敵意]、[混乱-当惑]、[抑うつ-落込み]、[疲労-無気力]、[緊張-不安]、[活気-活力]、[友好]の7尺度と、ネガティブな気分状態を総合的に表す[総合的気分状態 (Total mood disturbance, TMD)得点]から、所定の時間枠における気分状態を評価するものである。それぞれの素得点を、T得点と呼ばれる標準化得点に換算して解析する。T得点は、尺度、個人、検査間で妥当な比較が可能になるように、平均値50、標準偏差10となるように正規化した値である。以下の試験例1では[混乱-当惑]について結果を示す。結果は、内田クレペリンテスト前後フェーズ(T1~T3)、休憩前後フェーズ(T3~T4)および試験前後フェーズ(T1~T4)の3つのフェーズに分け、それぞれのフェーズ毎に解析を行い、図には各サンプリングタイム間の差分の平均値と標準誤差を示した。また、時間による変化の有無の検定には1標本のt検定を用い、試料による変化の違いの検定には対応のある2群間のt検定を用いて解析した。
発汗には主に温熱性発汗と精神性発汗がある。温熱性発汗は、体温を調節するためのもので、気温が高い時や運動をした時に体を冷やすために起こる現象であり、手掌や足底部を除いた全身に認められる。一方、精神性発汗は、精神的緊張や情動の変化による現象で、手掌や足底部に認められる。ストレス負荷の指標としては、上記クレペリンテストで鉛筆を持たないほうの手掌の拇指球の電気伝導度を、皮膚角質層水分量測定装置(「SKICON-200EX」、株式会社ヤヨイ製)を用いて測定した。被験者個人間のバラつきが大きかったため、各サンプリングタイム間の変化率を計算した。時間による変化の有無の検定には1標本のt検定を用い、試料による変化の違いの検定には対応のある2群間のt検定を用いて解析した。
一般に、精神的なストレスを評価する試験では、緊張などの理由で初回受験時の値と2回目以降の値が大きく異なることが知られている。そのため、初めて試験を受ける被験者の場合には、必要に応じて本番試験の前に、本番試験を短縮した事前試験を被験者に経験してもらった。
試験例1では、RO水と酵母発酵液を比較した。
試験例2では、乳酸菌発酵液と酵母発酵液を比較した。
試験例1、2では、酵母発酵液のにおいを嗅ぐことにより、RO水や乳酸菌発酵液では得られない、ストレスの緩和や集中力の維持についての効果が示された。よって、酵母発酵液に含まれる香気成分が嗅覚器に作用してそのような効果が得られたものと考えられた。
Claims (5)
- ウィッカーハモマイセス属に属する微生物を乳成分含有培地で培養して得られる培養物又はその培養上清を、香気成分として含有するストレス緩和及び/又は集中力維持用組成物。
- 前記乳成分含有培地は、ホエイ含有培地である、請求項1記載の組成物。
- 前記香気成分が、前記微生物を培養した該培養上清である、請求項1又は2記載の組成物。
- 前記微生物が、ウィッカーハモマイセス・ピジュペリYIT8095株(NBRC 1290)及び/又はウィッカーハモマイセス・ピジュペリYIT12779株(NBRC 1887)である、請求項1~3のいずれか1項に記載の組成物。
- 前記香気成分は、少なくとも安息香酸エチル及び/又はイソアミルアルコールを含む、請求項1~4のいずれか1項に記載の組成物。
