[メダカ精子の保存]
表6に示すように、実施例4の培地に加え、乳酸のみを0.2mMに増やした培地を用意した(実施例5)。また、無機塩類(塩化カルシウム二水和物、塩化マグネシウム六水和物、無水硫酸マグネシウム、塩化カリウム、無水リン酸二水素カリウム、塩化ナトリウム、無水リン酸水素二ナトリウム)、ペニシリン/ストレプトマイシン、ウシ胎児血清(FBS)、ウシ血清アルブミン(BSA)、10mM Hepes (pH 8.8)、グルコース、乳酸については、実施例4のゼブラフィッシュに用いた培地の分量と同じで、アミノ酸類、ビタミン類、D-ガラクトース、フェノールレッドナトリウム塩、ピルビン酸ナトリウム塩についてはゼブラフィッシュに用いた培地の分量の1/2量の培地を用意した(実施例6)。精巣(1つ)を取り出して、0.5%のbleachで30秒滅菌し、メダカのリンガー液(BSS)で2回wash後に、用意したそれぞれの培地(66μl)の中で、ピンセットを使って精子を絞り出した。その精子懸濁液10μlを1.5mlチューブに移し、そこに90μlの培地を加えて、精子の濃度は450,000/mlで保存した。
7日間室温または8日間4℃で保存した後、受精実験にはメダカ雌から準備した卵を用いて1チューブ分100μl(45,000精子)を用いて行った。この卵を嚢胚期まで培養した後、受精率をカウントした。結果を図7に示す。図7中、縦軸は受精率(%)を示す。実施例4の培地を用いて室温で保存したものは、一部、一週間目頃から精子保存液の色が黄色~オレンジ色っぽくなった。精巣に由来する体細胞が増殖したものと推測される。それ以外は、精子の受精能が維持されていることを確認した。
[Preservation of medaka sperm]
As shown in Table 6, in addition to the medium of Example 4, a medium was prepared in which only lactic acid was increased to 0.2 mM (Example 5). In addition, the amounts of inorganic salts (calcium chloride dihydrate, magnesium chloride hexahydrate, anhydrous magnesium sulfate, potassium chloride, anhydrous potassium dihydrogen phosphate, sodium chloride, anhydrous disodium hydrogen phosphate), penicillin/streptomycin, fetal bovine serum (FBS), bovine serum albumin (BSA), 10 mM Hepes (pH 8.8), glucose, and lactic acid were the same as those in the medium used for zebrafish in Example 4, and the amounts of amino acids, vitamins, D-galactose, sodium phenol red, and sodium pyruvate were half those in the medium used for zebrafish (Example 6). A testis (one) was removed, sterilized with 0.5% bleach for 30 seconds, washed twice with medaka Ringer's solution (BSS), and sperm were squeezed out using tweezers in each of the prepared media (66 μl). 10 μl of the sperm suspension was transferred to a 1.5 ml tube, and 90 μl of medium was added thereto, and the sperm concentration was maintained at 450,000/ml.
After storage at room temperature for 7 days or at 4°C for 8 days, fertilization experiments were performed using eggs prepared from female medaka, with 100 μl (45,000 sperm) per tube. The eggs were cultured until the sac-embryo stage, and the fertilization rate was counted. The results are shown in Figure 7. In Figure 7, the vertical axis indicates the fertilization rate (%). In some of the sperm stored at room temperature using the medium of Example 4, the color of the sperm storage solution became yellow to orange after about one week. This is presumed to be due to proliferation of somatic cells derived from the testes. It was confirmed that the fertilization ability of the sperm was otherwise maintained.
[マウス精子の保存]
表7に示すように、塩化マグネシウム六水和物、無水硫酸マグネシウム、無水リン酸二水素カリウム、塩化ナトリウム、無水リン酸水素二ナトリウム、ペニシリン/ストレプトマイシン、ウシ胎児血清(FBS)、ウシ血清アルブミン(BSA)、グルコース、乳酸、アミノ酸類、ビタミン類、D-ガラクトース、フェノールレッドナトリウム塩、ピルビン酸ナトリウム塩については、実施例4のゼブラフィッシュに用いた培地の分量と同じで、塩化カルシウム二水和物、塩化カリウムについては、実施例4のゼブラフィッシュに用いた培地の分量の1/3量で、20mM Hepes(pH 8.8)の培地を用意した(実施例7)。マウス精巣上体尾部を剪刀ばさみで切り、絞り出した精子を上記培養液0.5 mlに懸濁して精子懸濁液を作成した。
室温(22~23℃)で4日間保存した後、下層部約0.02 mlを、精子前培養用ディッシュ中のCARD FERTIUPマウス精子前培養培地 0. 2mlドロップに加えて、60分37℃で前培養し、その精子を、未受精卵を含む受精用mHTFドロップに加えて媒精した。媒精から3時間後、形態的に正常な卵子のみをmHTFドロップで洗浄した。翌日、2cellに発生した胚を胚培養用培地KSOMに移し、この卵を胚盤胞まで培養した後、受精率をカウントした。結果を表8に示す。
[Preservation of mouse sperm]
As shown in Table 7, magnesium chloride hexahydrate, anhydrous magnesium sulfate, anhydrous potassium dihydrogen phosphate, sodium chloride, anhydrous disodium hydrogen phosphate, penicillin/streptomycin, fetal bovine serum (FBS), bovine serum albumin (BSA), glucose, lactic acid, amino acids, vitamins, D-galactose, sodium phenol red, and sodium pyruvate were used in the same amounts as those used for the zebrafish in Example 4, and calcium chloride dihydrate and potassium chloride were used in 1/3 the amounts of those used for the zebrafish in Example 4. A 20 mM Hepes (pH 8.8) medium was prepared (Example 7). The mouse cauda epididymis was cut with scissors, and the sperm squeezed out were suspended in 0.5 ml of the above culture solution to prepare a sperm suspension.
After storing at room temperature (22-23°C) for 4 days, about 0.02 ml of the lower layer was added to a 0.2 ml drop of CARD FERTIUP mouse sperm preculture medium in a sperm preculture dish, precultured at 37°C for 60 minutes, and the sperm was added to an mHTF drop for fertilization containing unfertilized eggs for insemination. Three hours after insemination, only morphologically normal eggs were washed with the mHTF drop. The next day, embryos that had developed into 2 cells were transferred to embryo culture medium KSOM, and the eggs were cultured to blastocysts, after which the fertilization rate was counted. The results are shown in Table 8.
表8中、実施例4-1~4-5は、実施例4の培地を用いて、5回の繰り返し実験を行った結果を示し、実施例7-1~7-5は、実施例7の培地を用いて、5回の繰り返し実験を行った結果を示し、
表8に示すように、5回の繰り返し実験で実施例7の培地は、保存能力に優れていることが確認された。
