Sperms collected from 2-3 zebrafish were suspended in ice-cooled medium of Comparative Example 1 and Comparative Example 2 shown in Table 1 to prepare a sperm suspension. 10 μL of this sperm suspension was sampled and the number of sperms was counted. Then, the sperm suspension adjusted to 74000/μL using the medium was stored in a refrigerator at 4°C for 4 days. On the day of fertilization, eggs were squeezed from a zebrafish female into a glass dish, 40 μL of sperm suspension was applied to about 100 of these eggs, and 800 μL of sterilized fish breeding water was further applied and left for 1 and a half minutes. 7日間室温または8日間4℃で保存した後、受精実験にはメダカ雌から準備した卵を用いて1チューブ分100μl(45,000精子)を用いて行った。この卵を嚢胚期まで培養した後、受精率をカウントした。結果を図7に示す。図7中、縦軸は受精率(%)を示す。実施例4の培地を用いて室温で保存したものは、一部、一週間目頃から精子保存液の色が黄色〜オレンジ色っぽくなった。精巣に由来する体細胞が増殖したものと推測される。それ以外は、精子の受精能が維持されていることを確認した。

After culturing the eggs up to the cyst embryo stage, the fertilization rate was counted. The results are shown in Fig. 7. In FIG. 7, the vertical axis represents the fertilization rate (%). A part of the culture medium stored at room temperature using the medium of Example 4 had a part of the sperm storage solution turned yellow to orange from about the first week. It is presumed that somatic cells derived from the testis proliferated. Other than that, it was confirmed that the fertility of sperm was maintained.

表7に示すように、塩化マグネシウム六水和物、無水硫酸マグネシウム、無水リン酸二水素カリウム、塩化ナトリウム、無水リン酸水素二ナトリウム、ペニシリン/ストレプトマイシン、ウシ胎児血清(FBS)、ウシ血清アルブミン(BSA)、グルコース、乳酸、アミノ酸類、ビタミン類、D−ガラクトース、フェノールレッドナトリウム塩、ピルビン酸ナトリウム塩については、実施例4のゼブラフィッシュに用いた培地の分量と同じで、塩化カルシウム二水和物、塩化カリウムについては、実施例4のゼブラフィッシュに用いた培地の分量の1/3量で、20mM Hepes(pH 8.8)の培地を用意した(実施例7)。マウス精巣上体尾部を剪刀ばさみで切り、絞り出した精子を上記培養液0.5 mlに懸濁して精子懸濁液を作成した。
室温(22〜23℃)で4日間保存した後、下層部約0.02 mlを、精子前培養用ディッシュ中のCARD FERTIUPマウス精子前培養培地 0. 2mlドロップに加えて、60分37℃で前培養し、その精子を、未受精卵を含む受精用mHTFドロップに加えて媒精した。媒精から3時間後、形態的に正常な卵子のみをmHTFドロップで洗浄した。翌日、2cellに発生した胚を胚培養用培地KSOMに移し、この卵を胚盤胞まで培養した後、受精率をカウントした。結果を表8に示す。

After storing at room temperature (22 to 23° C.) for 4 days, about 0.02 ml of the lower layer was added to CARD FERTIUP mouse sperm preculture medium in a sperm preculture dish. In addition to the 2 ml drop, preculture was carried out at 37° C. for 60 minutes, and the sperm was inseminated by adding to the fertilization mHTF drop containing unfertilized eggs. Three hours after the insemination, only morphologically normal eggs were washed with mHTF drops. The next day, the embryos developed in 2 cells were transferred to the embryo culture medium KSOM, and the eggs were cultured up to the blastocyst, and the fertilization rate was counted.
