A.ドーパミン-アクリルアミド(DA)の合成
ドーパミン-アクリルアミド(DA)を、Patil et al.(N.Patil,C.Falentin-Daudre,C.Jerome,C.Detrembleur,Polym.Chem.,2015,6,2919-2933.)により記載される手順に従って、いくつかの修飾を含めて合成および特徴化した。簡単に説明すると、31.6mmolのNa2B4O7・10H2Oおよび47.2mmolのNa2CO3を、475mLの「Milli-Q」グレードの水に溶解し、溶液を1時間窒素バブリングにより脱気した。15.8mmolの塩酸ドーパミンを窒素雰囲気下で添加し、混合物を30分間撹拌し続けた。次いで、溶液を0℃に冷却し、63.2mmolの塩化アクリロイルを滴加した。溶液をpH9を超えて維持するために、84.9mmolのNa2CO3を添加し、溶液を室温で12時間撹拌した。その後、HCIを酸性化溶液のpHが1~2になるように添加し、開放容器内で1時間撹拌し続けた。混合物を酢酸エチルで5回抽出し、抽出から得られた茶色がかった有機溶液を収集し、Na2SO4で乾燥させた。その後、溶液の体積を回転蒸発によって低減させ、DAをヘキサンで沈殿させた。形成された懸濁液を、結晶化を助けるために冷蔵庫で一晩保存した。最後に、懸濁液の濾過によって得られた固体を、真空オーブン内で室温で一晩乾燥させた。合成を図1Aに示す。
ミクロゲルを、250mLの3つ口丸底フラスコでの沈殿重合によって合成した。簡単に説明すると、83.90mmolのMeO2MA(ジ(エチレングリコール)メチルエーテルメタクリレート)、0.573mmolのOEGMA7-8(オリゴ(エチレングリコール)メチルエーテルメタクリレート)、0.117mmolのOEGDA4-5(ポリ(エチレングリコール)ジアクリレート)、および57.5gの「Milli-Q」グレードの水を250mLの3つ口丸底フラスコに入れた。構造については図1Bを、合成概略図については図1Cを参照されたい。反応器の内容物を150rpmで撹拌し、窒素で45分間パージして、室温で酸素を除去した。次いで、2mLの「Milli-Q」グレードの水に溶解した0.305mmolのMAA(メタクリル酸)および2mLのエタノールに溶解した3モル%のDAをジャケット付きガラス反応器に添加し、混合物を70℃に加熱した。開始剤(2.5mLの脱気水に溶解した14.3mgのペルオキシ二硫酸カリウム(KPS))を添加した後、6時間撹拌しながら窒素雰囲気下で重合反応を続けた。
上述のように生成された精製粘膜付着性ミクロゲルを、100μg/mlのヒトHB-EGFと組み合わせた。得られた混合物を数時間維持し、カプセル化効率は93%であることが観察された。得られたミクロゲルの概略図を図2Aに提供し、得られたミクロゲルのAFM画像を図2Bに示す。得られたミクロゲルを、質量分析によって決定されるように、30分で5μg/mlのHB-EGFを放出することができた(図2Cを参照)。
上記IIで生成されたHB-EGFが装填された粘膜付着性ミクロゲル製剤がインビボで送達されること、および製剤が放射線により誘発される口腔粘膜炎の予防に有効である
ことを示す研究が行われた(図3および4を参照)。
PET CTスキャンを実行し、HB-EGFをCu64で標識化して、その生体内分布を評価した。結果を図5に示す。群Aは、Cu64-HB-EGF溶液であり、Cu64で標識化された唯一のHB-EGFであり、溶液(ゾル)の形態で局部的に(マウスの口)に投与された。群Bでは、Cu64-HB-EGFが粘膜付着性ミクロゲルにカプセル化され、24時間後にマウスに経口投与された。両方の群は、30分、1時間、2時間、3時間、および4時間でPET CTスキャンを受けた。Cu64信号を、マウス画像の体を上部(U)と下部(L)の関心領域(ROI)に分割することによって測定した。信号が上部ROIからの場合、その信号は、化合物が上部消化管に保持されたことを示す。信号が下部ROIからの場合、その信号は、化合物が腎臓および膀胱にあることを示し、排除を示す。群Aの吸収および排除は、Cu64-HB-EGFを口腔内に保持する群Bと比較してはるかに高速であり、全身吸収を回避することがミクロゲルの粘膜付着特性を証明する。4時間でも、ミクロゲルでカプセル化されたcu64-HBEGFは、下部ROIで非常に少量を示し、上部ROIからの強い信号は、標識化された生成物の高い保持および低い排除を示す。
