Description
デバイスの設計および組み立て
一実施例では、消化管のパッシブサンプリングのためのデバイスが、3D印刷を用いて製造された。このデバイスは、生分解性腸溶コーティング、第1のハウジング部分およびキャップを含む3D印刷されたハウジング、サンプリングヒドロゲル、ならびにガス透過性PDMS膜の4つの構成要素からなった。3D印刷されたハウジングは、SolidWorks(Dassault Systemes)で設計され、PreFormソフトウェアパッケージ(FormLabs)を介し、Form 2 3Dプリンターを用い、ステレオリソグラフィーを介して生体適合性のメタクリレート光硬化性ポリマーで印刷された。カプセルの最終的な外径と長さは、それぞれ9mmおよび15mmであった。内径と内径はそれぞれ7mmおよび14mmであった。各カプセルを、サンプリングヒドロゲルと、ヒドロゲルとキャップに形成されたサンプリング開口部との間に配置された厚さ1mmのPDMS膜とを含むように設計した。特に、図3は、組み立て前の種々の成分の写真を示す。カプセルのキャップのサンプリング開口部(直径5mm)は、カプセルへの液体の容易な流れを許容するように設計された。
一実施例では、AA対AMの4つの比率を用いて、どの比率が最も速い吸収および最も高い圧縮力をもたらしたかを決定した:10%AA/90%AM、30%AA/70%AM、50%AA/50%AM、70%AA/30%AM、および90%AA/10%AM。モノマー混合物の総濃度は、溶液の総重量の35wt%であった。脱イオン水は総重量の60wt%を占め、MBAおよびAPはそれぞれ総重量の1wt%および4wt%を占めた。AA/AMの異なる混合物を、20mLのシンチレーションバイアルで脱イオン水と混合した。VWR Digital Vortex Systemを用いて溶液を3分間ボルテックスすることにより、MBAを完全に溶解した。MBAが完全に溶解すると、重合を阻害し得る酸素分子を除去するために、溶液を窒素ガスで30分間バブリングし、続いて開始剤を添加した。溶液を、室温で3時間重合させた。完全に重合した後、直径5mmおよび長さ12mmのシリンダーを切断し、等温オーブンで80℃で一晩乾燥させた。ヒドロゲルが完全に乾燥すると、それらは試験の準備ができているとみなされた。
一実施例では、腸からカプセルの狭いサンプリング開口部への流体の連続的な引き込みを確実にするために、カプセルのポリマー表面は、上記で論じたように、長持ちする親水性コーティングで修飾された。表面改質を評価するために、Rame-Hart Model290 F1 Advanced Goniometerを用いて、未処理サンプル、プラズマ処理サンプル、およびプラズマ+PEG処理サンプルで表面接触角の測定を行いた。初期ならびに後退接触角の測定は、周囲温度で、約10μLの水滴を、異なる表面改質の前後の3D印刷された表面上に置くことにより実施した。
一実施例では、カプセルの密封メカニズムは、2つの独立した漏れ試験で試験された。図11に概略的に示されている最初の試験では、電気伝導率(EC)の経時変化を調べて、膨潤したヒドロゲルで活性化および密封した後のカプセルからの起こり得る漏れを検出した。この試験のために、完全に組み立てたカプセルおよびむき出しのヒドロゲルを、塩化ナトリウムおよび脱イオン水の1M溶液に18時間沈めて、両方の設定でヒドロゲルが完全に膨潤するようにした。次に、それらを新鮮な脱イオン水の別々の20mL溶液に導入し、Gw Instek LCR-821メーターを用いて電気抵抗率の変化を8時間測定した。脱イオン水の導電率が低い(0.05μS/cm)ことを考えると、導電率の大幅な増加は、脱イオン水および塩を注入したヒドロゲルの間の流体の交換に起因していた。図12は、乱されていない脱イオン水(1202)の溶液、むき出しのヒドロゲルに曝された後の脱イオン水(1204)、および密封されたカプセルに曝された後の脱イオン水(1206)の伝導率の変化のプロットを示す。図示するように、線1202および1206は互いに重なり合っている。溶液中にイオンが不足しているため、脱イオン水のベースライン伝導率は0.5μSに近くなる。むき出しのヒドロゲルが導入されると、媒体の伝導率は、ヒドロゲルと環境との間の流体の迅速な交換のために急速に増加する。このシナリオは、遊離のナトリウムイオンおよび塩化物イオンが新鮮な媒体に放出されてシステムの伝導率が増加する、密封メカニズムの完全な故障をシミュレートする。カプセル内のヒドロゲルの結果は、8時間にわたって伝導率の増加がないことを示し、カプセルから新鮮な媒体への漏れがないことを示唆している。
密封メカニズムの有効性を試験することに加えて、ヒドロゲルが細菌を収集する能力を評価し、回収された細菌の培養が達成可能かどうかを判断することが重要であった。このデバイスの最も関連性の高い応用には、消化管の現在アクセス不可能な領域にアクセスして、その後の培養のために生存細菌サンプルを収集する機能が挙げられる。摂取するとさらに液体交換が起こるかどうかを判断するだけでなく、収集した後に細菌が生存し得るかどうかを判断することも重要である。
デバイスの設計および組み立て
一実施例では、カルプロテクチンのための消化管のパッシブサンプリングのためのデバイスが、3D印刷を用いて製造された。このデバイスは、生分解性腸溶コーティング、最初のハウジング部分およびキャップを含む3D印刷されたハウジング、サンプリングヒドロゲル、ならびにガス透過性ポリジメチルシロキサン(PDMS)膜の4つの成分からなった(図2-3を参照)。3D印刷されたハウジングを、SolidWorks(Dassault Systemes)で設計し、Form 2プリンター(FormLabs)を用いて、光造形法により、50μmの厚みで、Formlabsから購入した生体適合性樹脂(EN-ISO 10993-1:2009/AC:2010、USP Class VI)を用いて印刷した。3D印刷後、モデルを、99%純粋なイソプロピルアルコール(IPA)中で15分間すすいで残留物をすべて除去し、その後、UV光硬化デバイス(Formlabs Inc.)で60分間後硬化して、重合を完了させた。カプセルの最終的な外径および長さは、それぞれ9mmおよび15mmであった。内径および内径は、それぞれ5.6mmおよび13mmであった。各カプセルを、サンプリングヒドロゲルと、ヒドロゲルとキャップ中に形成されたサンプリング開口部との間に配置されたPDMS膜と、を含むように設計した。サンプリング開口部は、直径4mmであった。
消化管のpHは、一般的に、1から7.5まで変化する。本発明者らは、カルボキシル基を含むアニオン性ポリマーは、胃の低pHでは不溶性であるが、腸の中性pHでは可溶性であるので、消化管を介した選択的サンプリングを作動させるためのサンプリングデバイスの腸溶コーティングとして使用し得ることを認識した。
一実施例では、架橋剤としてのアクリルアミド(4.5g;Sigma-Aldrich)およびN,N’-メチレンビスアクリルアミド(MBA)(0.15g;Sigma-Aldrich)を、Signature Digital Vortex Mixer 945303(VWR、Radnor,PA,USA)を用いて、溶液を室温で3分間ボルテックスすることにより、10mLの水に溶解した。窒素ガスを10分間バブリングして、重合阻害剤としての溶存酸素を置換することにより、溶液を完全に脱気した。次に、過硫酸アンモニウム(0.1g;Sigma-Aldrich)を、開始剤として溶液に添加した。プレゲル溶液を150μLの容量でPDMSモールドシリンダーに移し、モールドを等温オーブン内に70℃で一晩置いて、アクリルアミドネットワークを形成し、完全に硬化させた。膨潤率については、3つの乾燥した同一のヒドロゲルを秤量し、100rpmの振とう台を備えたインキュベーター内で37℃のpH6.8および7.4の25ml緩衝液に36時間浸漬した。ヒドロゲルを10分間隔で緩衝液から除去し、外面に存在する過剰な緩衝液を、ワイパーで穏やかに吸い取った。各ヒドロゲルの質量を測定した。膨潤比は、膨潤比=(Wt-Wi)/Wiとして定義したが、ここで、WtおよびWiは、それぞれ膨潤したヒドロゲルおよび乾燥したヒドロゲルの重量である。
サンプリングカプセルのサンプリング性能を評価するために、緑色蛍光タンパク質(GFP)、ウシ血清アルブミン(BSA)、およびカルプロテクチン(炎症のバイオマーカー)を含む、類似したサイズおよび分子量を有する3つの異なるタンパク質を試験した。GFPを、ヒドロゲル組成物内のタンパク質捕捉を視覚的に表示するために選択した(図20A~D)。
GFP(Sigma-Aldrich、St.Louis,MO)をPBS(Sigma-Aldrich、St.Louis,MO)で希釈して、100μg/mLのGFP濃度を達成した。溶液をマイクロ遠心チューブに移し、14,000×gで1分間遠心分離して(Sorvall Legend Micro 21 Microcentrifuge、Thermo Fisher Scientific、MA,USA)、GFP凝集体を沈殿させた。ヒドロゲルをGFP上清に2時間導入して、ヒドロゲル構造全体のタンパク質サンプリング効率を定性的に表示した。画像を、High Performance 2UV Transilluminator UVP(Upland、CA,USA)およびデジタル倒立顕微鏡(AMG EVOS fl、Bothell,WA)でキャプチャした。ヒドロゲルのGFP抽出挙動をモニターするために、それらを4mLのPBSに2時間曝露し、BMG Clariostarプレートリーダーを介して30分間隔で蛍光分析(最大488nmでの励起および510nmでの最大発光)を実行した。各時点で、3つの100μLをCorning(NY,USA)から購入したUV透過性の96ウェルマイクロプレートに移し、蛍光を測定した。
次に、BSAタンパク質のサンプリングおよび抽出について調査した。BSAは、約7nmの平均直径を有する。ウシ血清アルブミン(Sigma-Aldrich、St.Louis,MO)を、合計5つの濃度(10、5、2.5、1.25、および0.625mμg/mL)について、PBS(Sigma-Aldrich、St.Louis,MO)で10~0.625mg/mLの2倍連続希釈した。腸溶コーティングを有しないカプセルを、100rpmで2時間、BSA溶液に浸漬した。続いて、カプセルを回収し、分解し、2時間の抽出のためにPBS培地に導入した。抽出PBS溶液中のBSAの存在を確認するために、ビシンコニン酸(BCA)アッセイを用いて、タンパク質含有量を検出した。25μLの各標準サンプルおよび未知のサンプルをマイクロプレートウェルに加え、続いて200μLの作業試薬を加えた。37℃で30分間インキュベートした後、プレートを室温まで冷却し、BMG CLARIOstar Plusマイクロプレートリーダーを用いて562nmでの吸光度を測定した。
カルプロテクチン(MyBioSource、San Diego,CA)を、Tris緩衝液(20mM Tris、100mM NaCl、pH7.4)で1mg/mLの濃度に再構成した。タンパク質溶液を、4つの異なる濃度にさらに希釈した。バイアルを100rpmで攪拌しながら、完全に組み立てられたカプセルを各濃度の4mLに2時間浸した。その後、カプセルを開け、内容物を4mLの緩衝液に移し、100rpmで再度2時間抽出した。カルプロテクチンの測定は、標準的な製造者のプロトコル(4~240ng/mL)に従い、BUHLMANN fCAL ELISAを用いて分析された酵素結合免疫吸着測定法(ELISA)であった。抽出緩衝液を、捕捉抗体でコーティングされた96ウェルプレートに装填した。30分間のインキュベーションおよび洗浄の後、西洋ワサビペルオキシダーゼ(HRP)に結合した検出抗体を添加すると、検出抗体がカルプロテクチンに付着した。インキュベーションおよび洗浄の後、テトラメチルベンジジン(TMB)を添加し(青色形成)、その後、停止溶液(黄色に変化)を添加した。吸光度を、BMG CLARIOstar Plusマイクロプレートリーダーを用いて、450nmで測定した。
実際の消化管移行時間でカプセルのサンプリング性能を評価するために、完全に組み立てたカプセルを、図22A(左から右)に示すpH7、3、6.8、および図22Aに示す7.4で、異なる水性色および緩衝液中で組み立てた。これらのpH環境は、消化管に沿ったpH変化のモデル化を助ける。消化管滞留時間に基づいて、3つのカプセルの群を、前述の各pH緩衝液中に、1分、2時間、2時間、および18時間ずつ順番に導入した。各タイムスロットの後、カプセルを開け、液体の浸透またはヒドロゲルの色の変化を観察した。胃(pH3)中で2時間保持した後に開けたカプセルは、無傷の腸溶コーティングを示し、ヒドロゲルの色の変化およびハウジング内の液体内容物を生じなかった(図22B)。このことは、酸性環境での溶解に抵抗するポリマーの効率を証明する。逆に、小腸(pH6.8緩衝液)に2時間閉じ込めた後に開いたカプセルは、完全に溶解した腸溶コーティングを示し、サンプリングリザーバー内の緩衝液内容物を有する膨潤した青いヒドロゲルを生じた(図22C)。サンプリング前後のカプセルの密封の性能を評価するために、緩衝液内にカプセルを導入する前後に緩衝液の吸光度を検出した。結果は、ピークの追加がないこと(すなわち、流体交換がないこと)を明らかにし、このことは、カプセルの適切な密封を示唆する(図22D~F)。
BSAおよびカルプロテクチンのサンプリングにおけるカプセル効率を研究するために、2つの実験をインビトロで実施した。4つの異なるpH緩衝液を提供した:pH7、3、6.8、および7.4。タンパク質を、pH6.8の緩衝液(小腸をシミュレートする緩衝液)内にのみ注入した。
次に、エクスビボ実験を実施することにより、カプセルのサンプリング性能に対する腸液の複雑さの影響を調査した。カルプロテクチンを、新たに解剖したブタの小腸に加え、続いてサンプリング抽出手順を行った(図24A~C)。図24Aは、内部に2つのサンプリングカプセルを備えた、カルプロテクチンが装填された腸液で満たされた小腸のセグメントを示す。図24Bは、カプセルの回収を示す。図24Cは、2時間の抽出のためにTris緩衝液に入れられたサンプリングされた腸液を有する、分解されたカプセルを示す。図24Dに示すように、2つの試験環境濃度を、通常(113μg/mL)および上昇(745.5μg/mL)の両方のカルプロテクチンレベルが試験されるように選択した。エクスビボでの3.85μg/mLおよび27.5μg/mLの抽出値は、緩衝液およびインビトロ実験で達成された2.6%のE値に対して有意差を示さない。全体として、サンプリングカプセルは、小腸からカルプロテクチンを部位特異的に捕捉してそれをカプセルが体から排泄されるまで効果的に保護し得る、信頼し得るデバイスであると思われる。本実施例では、デバイスは、小腸に入ってから1時間の腸溶コーティング溶解時間、ならびに1.4時間の膨潤およびカプセル密封時間のヒドロゲルを含んでいた。小腸の滞留時間は通常4~6時間であるので、平均2.4時間のサンプリング時間を選択して1.6時間の残り時間を可能にすることにより小腸内部でのカプセルのサンプリングプロセス中に発生し得るすべての不要な長い段階を許容した。

カプセルハウジングの、サンプリング開口部を有する第1のハウジング部分、および、第2のハウジング部分を提供することであって、これらのハウジング部分は組み合わせられるように形成されている、提供すること、
