本発明は、操作されたTregを産生するための方法であって、細胞中に、本明細書で規定されるTCRをコードするポリヌクレオチドをin vitroまたはex vivoで導入するステップを含む方法もまた提供する。適切には、この方法は、本発明のTCR分子の発現を可能にする条件下で細胞をインキュベートするステップをさらに含む。必要に応じて、この方法は、操作されたTreg細胞を精製するステップをさらに含み得る。
TCRをコードするレトロウイルスベクターおよびTCR+FOXP3をコードするレトロウイルスベクターの産生
MS2-3C8 TCR
MS2-3C8 TCRは、本明細書に参照により組み込まれる、Muraro et al., JCI 1997; 100, 2, 339-349によって記載されるように、HLA-DRB1*0401によって提示されたMBP111-129(配列番号3)を認識する。コドン最適化されたMS2-TCRvα、マウス化された定常アルファドメイン、コドン最適化されたMS2-TCRvβおよびマウス化された定常ベータドメインを、レトロウイルス発現カセットpMP71中にクローニングすることによって、MS2-3C8 TCRをコードするコドン最適化されたMS-2 TCR発現カセットを構築した(図1および図2)。本明細書に参照により組み込まれる、Blood. 2007 Mar 15; 109(6): 2331-2338.およびCancer Res. 2007 Apr 15; 67(8): 3898-3903に記載されるように、定常アルファドメインと定常ベータドメインとの間に追加のジスルフィド結合を加えた。定常アルファドメインとMS2-TCRvβドメインとを、P2A配列によって分離した。
FOXP3およびMS-2 TCRをコードする発現カセットを、上記MS-2 TCRを改変することによって構築した(図1および図3)。停止コドンが除去されたFOXP3遺伝子およびT2A配列を、MS-2 TCRの上流のRsrII部位およびEcoR1部位を使用して、pMP71ベクター中に挿入した。
TCRおよびTCR+FOXP3によるT細胞の形質導入
Phoenix-Ampho細胞を、標準的な組織培養条件で、8mLのイスコフ改変ダルベッコ培地(IMDM)中2×106で24時間播種した。1日目に、細胞を、FuGENE(登録商標)トランスフェクション試薬、ならびに2.5μgの適切なプラスミドDNAおよび同種指向性レトロウイルス共受容体をコードする1.5μgのpCL-Ampと混合した最適な培地を使用して、室温で20分間トランスフェクトした。トランスフェクション混合物を、接着性Phoenix Amphotropic(Phoenix-AMPHO)細胞に添加し、さらに24時間インキュベートした。2日目に、培地を5mLのTexMACS(登録商標)(Miltenyi)組織培養培地に交換し、細胞を、さらに24時間インキュベートした。
in vitro拡大増殖の間のFOXP3発現
T細胞を、実施例2に上記したように単離した。0日目に、FOXP3ならびにTreg細胞表面マーカーCTLA-4(CD152としても公知)およびCD25の発現を、フローサイトメトリーによって測定した。
(実施例4)
エフェクターT細胞およびTregのペプチド再刺激
チャイニーズハムスター卵巣(CHO)細胞を、ヒトHLA-DR4およびCD80またはCD86で形質導入した。CD80またはCD86を発現する細胞を、引き続く実験のために等量で一緒に混合した。
TCR形質導入されたT細胞およびTCR-FOXP3形質導入されたT細胞は、コグネートペプチドに対して応答する
TCR形質導入されたTconv、TCR形質導入されたTreg、TCR-FOXP3変換されたTconvおよびTCR-FOXP3変換されたTconv(上記方法)を、ペプチドパルスした照射APCありまたはなしで4日間培養した。上清を培養物から収集し、ELISAによってIL-2およびIFNγについてアッセイした(n=2~4)。図9は、TCR形質導入されたTregおよびTCR-FOXP3変換されたTconvが、コグネートペプチドに対して応答することを示している。
TCR形質導入およびTCR-FOXP3形質導入
CHO細胞を、上記のように調製した。Tconv細胞を、TCRまたはTCR+FOXP3で形質導入した。形質導入されたT細胞を、磁気ビーズ選別によって単離した。形質導入された細胞を、APCにコンジュゲートした抗マウス定常ベータ抗体で染色した。細胞を、十分に洗浄し、第2の抗APC抗体で染色した。細胞を洗浄し、磁気カラムを通過させ、形質導入された細胞を捕捉し、溶出した。決まって、精製された細胞のうち>95%が、APC+であった。
(実施例7)
TCR形質導入されたT細胞の増殖
CD80+CD86+DR4+CHO細胞にペプチドをロードし、上記のように照射し、その後、0.1×106細胞/mlで再懸濁した。形質導入された応答性T細胞を、温PBS中で37度で3分間、CFSE cell trace色素で染色し、その後、等体積の温PBSを添加し、さらに3分間インキュベートした。
実験的自己免疫性脳脊髄炎の養子移入モデルにおける操作されたTreg

請求項1に記載の操作されたTreg。 - 前記TCRの前記α鎖およびβ鎖の定常領域ドメインが各々、前記α鎖と前記β鎖との間での追加のジスルフィド結合の形成を可能にする、さらなるシステイン残基を含む、請求項1または2のいずれか一項に記載の操作されたTreg。
- (a)前記TCRの前記α鎖が、配列番号13に対して少なくとも90%の配列同一性を有するアミノ酸配列を含み、前記配列同一性が、請求項1に記載のCDR配列を含まず;
(b)前記TCRの前記β鎖が、配列番号14に対して少なくとも90%の配列同一性を有するアミノ酸配列を含み、前記配列同一性が、請求項1に記載のCDR配列を含まない、
請求項1から3のいずれか一項に記載の操作されたTreg。 - 被験体から単離されたT細胞に由来する、請求項1から4のいずれか一項のいずれかに記載の操作されたTreg。
- 請求項1から5のいずれか一項のいずれかに記載の操作されたTregを含む医薬組成物。
- 疾患を処置する際の使用のための、請求項6に記載の操作された医薬組成物。
- 医薬の製造における、請求項1から6のいずれかに記載の操作されたTregまたは医薬組成物の使用。
- 疾患を処置または予防することを必要とする被験体において疾患を処置または予防するための、請求項6に記載の医薬組成物。
- 前記疾患が多発性硬化症である、請求項7または9に記載の使用のための医薬組成物。
- 前記被験体が、HLADRB1*0401陽性の被験体である、請求項9または10に記載の使用のための医薬組成物。
- 請求項1から5のいずれか一項に記載のTCRをコードする核酸配列およびFOXP3をコードする核酸配列を含むベクター。
- 請求項1から5のいずれか一項に記載のTCRをコードする核酸配列を含む第1のポリヌクレオチドまたはベクターと、FOXP3をコードする核酸配列を含む第2のポリヌクレオチドまたはベクターとを含む、ポリヌクレオチドまたはベクターのキット。
- 請求項1から5のいずれかに記載の操作されたTregを産生するための方法であって、細胞中に、請求項1から5のいずれかに記載のTCRをコードするポリヌクレオチドをin vitroまたはex vivoで導入するステップを含む、方法。
- 前記細胞中に、FOXP3タンパク質をコードするポリヌクレオチドをin vitroまたはex vivoで導入するステップをさらに含む、請求項14に記載の方法。
- 前記細胞がT細胞である、請求項14に記載の方法。
