実施形態
一実施形態では、イヌ科動物における早期変性性僧帽弁疾患を診断する方法は、エリシペラトクロストリジウム、ルミノコッカスUCG014、ブチリシコッカス、フィーカリバクテリウム、及びこれらの組み合わせからなる群から選択されるバイオマーカーの、正規化した相対存在量を測定する工程と、エリシペラトクロストリジウムの正規化した相対存在量が0~0.5である場合、ルミノコッカスUCG014の正規化した相対存在量が0~0.1である場合、ブチリシコッカスの正規化した相対存在量が0~0.1である場合、又はフィーカリバクテリウムの正規化した相対存在量が0~0.1である場合に、該イヌ科動物が早期変性性僧帽弁疾患を有すると判定する工程と、を含み得る。
実施例1-イヌのMMVD研究
4つのグループのイヌから糞便試料を採取した:グループ1、心臓疾患のエビデンスがないイヌ;グループ2、ステージB1の僧帽弁粘液腫様変性(MMVD)を有するイヌ;グループ3、ステージB2のMMVDを有するイヌ;及びグループ4、ステージCのMMVDを有するイヌ。アメリカ獣医内科学学会(ACVIM)による合意声明に従って、委員会認定の心臓専門獣医による身体検査及び心エコー図に基づき、イヌを検査及び分類した。かかる4つのグループにおいて、イヌの年齢、性別、ボディコンディションスコア(BCS)を揃えた。以下の条件を有するイヌは除外した:1)60日以内に抗生物質を投与されている、2)活発な下痢又は嘔吐がある、3)癌、真性糖尿病、腎不全、又は全身性高血圧症などの重大な全身性疾患がある、4)先天性心疾患などの心臓異常がある、5)BCSが8以上又は3以下である、6)7歳未満である。投薬の履歴を記録した。
腸内細菌の多様性及び存在量
系統学的多様性(PD)とは、系統学的な分類群の豊かさと同義である。ステージAのイヌは、ステージB1、B2、又はCのイヌと比較して区別可能なPD指数を有する。さらに、ステージAのイヌの有する属の数(116)は、ステージB1、B2、及びCのイヌ(それぞれ99、81、及び90)よりも一貫して多い。
差次的に豊富な分類群
エリシペラトクロストリジウム、ルミノコッカスUCG014、ブチリシコッカス、及びフィーカリバクテリウムの4つの属は、様々なステージのDMVDのイヌよりも、健康なステージAのイヌにおいて、より多量であることが示された(表1)。興味深いことに、これらの存在量は左心臓のサイズと逆相関し、心臓が肥大するにつれて細菌量は減少を示した。
メタゲノム的な機能予測
2つのメタゲノム経路であるカルジオリピン生合成及びバリン分解は、ステージB2のDMVDのイヌでは、健康なステージAのイヌと比較して過剰であったのに対し、グルタミン/グルタメート生合成及びトリプトファン生合成は、健康なイヌでは、B2のイヌと比較して少なかった。
Claims (8)
- イヌ科動物における早期変性性僧帽弁疾患を診断する方法であって、
前記イヌ科動物から糞便試料を採収する工程と、
前記糞便試料におけるエリシペラトクロストリジウム、ルミノコッカスUCG014、ブチリシコッカス、フィーカリバクテリウム、及びこれらの組み合わせからなる群から選択されるバイオマーカーの、正規化した相対存在量を測定する工程であって、前記糞便試料から糞便DNAを抽出し、前記抽出された糞便DNAの配列決定を行い、分類学的割り当て及び配列アライメントを実施することを含む工程と、
前記エリシペラトクロストリジウムの正規化した相対存在量が0~0.5である場合、前記ルミノコッカスUCG014の正規化した相対存在量が0~0.1である場合、前記ブチリシコッカスの正規化した相対存在量が0~0.1である場合、又は前記フィーカリバクテリウムの正規化した相対存在量が0~0.1である場合に、前記イヌ科動物が早期変性性僧帽弁疾患を有すると判定する工程と、
