Description
本発明は、非常に安定な合成単一ドメイン抗体骨格の同定、及び、合成単一ドメイン抗体ライブラリーの作成におけるその使用に関する。本発明はまた、前記の安定な単一ドメイン抗体骨格を含む抗原結合タンパク質、及び、特に治療薬としてのその使用に関する。
過去十年間にわたって抗体は、特に腫瘍学の分野において最も有望な治療アプローチの1つとしてだけでなく、研究又は診断のツールの重要な入手源としての役目が課されてきた。
本発明の目的は、合成単一ドメイン抗体ライブラリーを作成する方法によって成し遂げられ、該方法は、
i)CDR1、CDR2及びCDR3をコードする多様な核酸を、合成単一ドメイン抗体(これは本明細書において以後、「hs2dAb」と称され得る)のそれぞれのフレームワークコード領域間に導入して、合成単一ドメイン骨格のアミノ酸配列を有する同合成単一ドメイン抗体をコードする多様な核酸を作製する工程を含み、
前記の合成単一ドメイン骨格のアミノ酸配列は、少なくとも以下の元来のラクダ科の動物のVHHアミノ酸残基:F37、E44、R45、F47;及び少なくとも以下のヒト化アミノ酸残基:P14、S49、S74、R83、A84を含有し、場合によりさらに元来のラクダ科の動物のVHH残基であるQ5、Q108及びT89を含む。

d)それは、例えば融合タンパク質(例えば蛍光タンパク質との融合物)として哺乳動物細胞株において発現させた場合に凝集しない。

本明細書では、合成単一ドメイン抗体又はその断片におけるアミノ酸残基の位置は、本明細書において以後に示されているようにKabatの番号付け命名法に従って示される。
i.CDR1、CDR2及びCDR3をコードする多様な合成核酸を、合成単一ドメイン抗体のそれぞれのフレームワークコード領域間に導入して、合成単一ドメイン抗体骨格のアミノ酸配列を有する多様な同合成単一ドメイン抗体をコードする核酸を作製する工程を含み、
前記の合成単一ドメイン骨格のアミノ酸配列は、少なくとも以下の元来のラクダ科の動物のアミノ酸残基:F37、E44、R45、F47;及び、少なくとも以下のヒト化アミノ酸残基:P14、S49、S74、R83、A84を含有し、場合によりさらに、元来のラクダ科の動物のVHH残基のQ5、Q108、及びT89を含む。
本発明は、非常に安定な単一ドメイン抗体骨格を得るための、単一ドメイン抗体のフレームワーク領域における独特な特色の同定、及び、合成単一ドメイン抗体ファージディスプレイライブラリーなどの合成単一ドメイン抗体ライブラリーを作成する際のその使用に関する。前記の独特な骨格を有する、結果として得られたhs2dAbは非常に安定で、かつ免疫原性のリスクが低い。
(i)生殖系列上のラマFR2アミノ酸配列と同一なFR2アミノ酸配列;
(ii)生殖系列上のヒトFR3(VH3)アミノ酸配列と同一なFR3アミノ酸配列;及び
(iii)生殖系列上のラマFR4アミノ酸配列と同一なFR4アミノ酸配列
を含む。
i.それをE.coliペリプラズムにおいて可溶性の単一ドメイン抗体として発現させることができる、
ii.それをE.coli、酵母又は他の真核生物の細胞質基質において可溶性の細胞内発現抗体として発現させることができる、
iii.それは、クロラムフェニコールアセチルトランスフェラーゼ融合アッセイの還元性環境において安定である、

特にCDRコード配列の無作為な合成又は指向的な合成によって、抗体ライブラリーにおいてCDRの多様性を生じさせ、これを対応するフレームワーク配列にクローニングするための方法は、当技術分野において広く記載されている。

結果として、別の態様によると、本発明は、以前の方法によって得ることのできるか又は得られた合成単一ドメイン抗体ライブラリーに関する。
本発明の合成単一ドメイン抗体ライブラリーの高度な多様性を考慮して、当業者は、ファージディスプレイなどの従来のスクリーニング法によって、対象の標的に対して高い親和性及び高い特異性を有する合成単一ドメイン抗体を得ることができる。
(i)生殖系列上のラマFR2アミノ酸配列と同一なFR2アミノ酸配列;
(ii)生殖系列上のFR3(VH3)アミノ酸配列と同一なFR3アミノ酸配列;及び
(iii)生殖系列上のラマFR4アミノ酸配列と同一なFR4アミノ酸配列
を含む。
(i)配列番号1のフレームワーク領域FR1、配列番号2のFR2、配列番号3のFR3、及び配列番号4のFR4、
(ii)1、2又は3個以下のアミノ酸保存的置換を有し、有利な合成単一ドメイン特性を保持した、機能的な変異フレームワーク領域、
(iii)それぞれ配列番号1~4に対して少なくとも60、70、80、90、95、96、97、98、又は99%の配列同一率を有し、有利な合成単一ドメイン特性を保持した、機能的な変異フレームワーク領域FR1、FR2、FR3及びFR4。
i.それをE.coliペリプラズムにおいて可溶性の単一ドメイン抗体として発現させることができる
典型的には、pelBリーダーペプチドを用いて5mg/Lを超える収率が好ましくはE.coli株において得られる。
ii.それをE.coliの細胞質基質において可溶性の細胞内発現抗体として発現させることができる
例えば、抗体は、T7プロモーターを用いて50mg/Lを超える収率でE.coliのBL21株(DE3)において発現させることができる。
iii.それは、クロラムフェニコールアセチルトランスフェラーゼ融合アッセイにおいて示されるように還元性環境において安定である
上記の特性についての機能的アッセイが実施例に記載されている。C末端タグとしてクロラムフェニコールアセチルトランスフェラーゼと融合した本発明の合成単一ドメイン抗体を含有する、該抗原結合タンパク質を発現している細菌細胞は、クロラムフェニコールに対して、特に培養培地中で300μg/mlより高いクロラムフェニコール濃度で耐性であろう。
iv.それは、蛍光タンパク質との融合物として哺乳動物細胞株において発現させた場合に凝集しない
好ましくは、合成単一ドメイン抗体を含有する抗原結合タンパク質が蛍光タンパク質との融合物として発現される場合に、凝集は全く検出されないだろう。

機能的アッセイ
E.coliペリプラズムにおける可溶性発現
単一ドメイン抗体断片を、pHEN6に由来する細菌ペリプラズム発現ベクターにサブクローニングし、pelB分泌配列の下流において発現させた。新たに形質転換されたコロニーを、1%グルコース及び30μg/mlの抗生物質カナマイシンの補充されたテリフィックブロス培地中で、A600=0.6~0.8に達するまで増殖させた。その後、6つのHisでタグ化された抗体断片の発現を、500μMのイソプロピルβ-D-チオガラクトピラノシドを用いて28℃で16時間かけて誘導し、その後、遠心沈殿した。遠心分離後、細胞ペレットをトリス-EDTA-スクロース浸透圧ショック緩衝液中でインキュベートし、再度遠心分離にかけた。細胞溶解液を清澄にし、ポリヒスチジンタグのためにIMAC樹脂親和性カラムにローディングした。溶出した画分を透析し、タンパク質の純度をSDS-PAGEによって分析した。
単一ドメイン抗体断片を、T7プロモーターの制御下で細菌発現ベクターにサブクローニングした。プラスミド構築物を、E.coli BL21(DE3)細胞に形質転換した。1つのコロニーを、1%グルコース及び30μg/mlの抗生物質カナマイシンの補充されたLB培地中で、A600=0.6~0.8に達するまで増殖させた。その後、抗体断片の発現を、500μMのイソプロピルβ-D-チオガラクトピラノシドを用いて28℃で6時間から8時間かけて誘導し、その後、遠心沈殿した。遠心分離後、細胞ペレットを溶解し、再度遠心分離にかけた。細胞溶解液を清澄にし、ポリヒスチジンタグのためにIMAC樹脂親和性カラムにローディングした。溶出した画分を透析し、タンパク質の純度をSDS-PAGEによって分析した。
単一ドメイン抗体断片を、pAOCAT細菌ペリプラズム発現ベクターにサブクローニングした。クロラムフェニコール耐性アッセイを、pAOCAT-VHH融合構築物を用いて形質転換されたBL21(DE3)細胞を使用して行なった。細菌を、カナマイシン(35μg/mL)及びグルコース(0.2%)を含有するLB 500μLへの接種のために使用し、37℃で、OD600が0.8となるまで増殖させた。VHH-CAT-融合タンパク質の細胞質内発現を、0.2mMのIPTGの添加によって2時間かけて誘導した。誘導期間の終了時に、4μLの細菌アリコートを、IPTG(0.1mM)及び0~500μg/mlの範囲のクロラムフェニコール漸増濃度を含有するLB寒天プレート上に蒔いた。細菌を30℃で20時間インキュベートし、その後、コロニーの形成を定量した。耐性レベルを、様々なクロラムフェニコール濃度におけるコロニー増殖速度に従って評価した。500μg/ml以下のコロニーを与えるいくつかのVHHを、GFP(nb GFP4)又はラミン(Lam)に対して生じた以前に特徴付けられた細胞内発現抗体と、並びに、熱安定性VHH Re3及び細胞内発現抗体ではないC8と比較した。上記のように2時間の間に誘導された液体培養液を連続希釈によって希釈し、250μg/mlのクロラムフェニコール(Cam)を含有する寒天プレート上に10μlをスポットし、30℃で20時間インキュベートした。各希釈液についてコロニーを定量し、D10クローンで常に見られたより多くの量に対して標準化した。
真核細胞における細胞内抗体としての機能的発現
単一ドメイン抗体断片を、蛍光タンパク質との融合物として、及びCMVプロモーターの制御下で発現させるために、哺乳動物発現ベクターにサブクローニングした。哺乳動物細胞株にトランスフェクトし、細胞内の蛍光を、トランスフェクトから24時間後又は48時間後に観察した。トランスフェクトされた細胞におけるこれらの蛍光タンパク質の1つと融合したVHHの蛍光分布は、GFP又はmCherryと融合していないものと比較して、均一に広がり、48時間の構成的発現後に明白な凝集物は示されなかった。
プラスミドは、インターロイキン-2(IL2)シグナル配列を含有するInvivogen社(サンディエゴ、米国)のpFUSE-Fc2(IL2ss)(商標)シリーズをベースとし、哺乳動物細胞によるFc-融合タンパク質の分泌を可能とする。hs2dAbをIgGのヒンジドメインに融合させたので、Fcドメインはジスルフィド結合を形成し、hs2dAb-Fcはダイマーとして発現される。それらは、原核細胞及び真核細胞の両方においてゼオシン(商標)(Zeo)を使用して選択可能である。これらのプラスミドは、部位特異的突然変異誘発及びアダプター挿入によって改変され(Moutel S, et al. BMC Biotechnol. 2009 Feb 26;9:14. doi: 10.1186/1472-6750-9-14)、一般的な組換え抗体の大規模な選別コレクション及び発現プラスミド(例えばpHEN、pSEX、pHAL、pCANTAB、pHOG、pOPE、pSTE)から抽出された組換え抗体の簡単な1工程のカセットクローニングが可能となった。s2dAbが、そのC末端でヒトIgG2(及びIgG1)(h)、マウスIgG2a(m)又はウサギIgG(r)Fcドメイン(Fc領域は、IgG重鎖のCH2ドメイン及びCH3ドメインとヒンジ領域とを含む)のいずれかと融合することができる4つのプラスミドを構築した。
抗原コード配列を、酵母ツーハイブリッドのbaitプラスミドのlexA(Vojtek and Hollenberg (1995). Methods Enzymol. 255:331-42)又はgal4(Fromont-Racine, M., Rain, J.C.、及びLegrain, P. (1997). Nat. Genet. 16: 277-282)にクローニングした。試験しようとするDNA結合ドメインSDAB集団を、酵母ツーハイブリッドのpreyプラスミドのpGADGH(Bartel, P.L., et al (1993) in Cellular interactions in development: A practical approach. ed. Hartley, D.A.. (Oxford University Press, Oxford) pp. 153-179)にPCR及びギャップ修復(Orr-Weaver, T. L. and Szostak, J. W. (1983). Proc. Natl. Acad. Sci. USA 80, 4417-4421)によって導入した:pHEN2-3mycプラスミドプールのDNA調製物(1つのクローンから3×109個まで)を調製した。ミニプレップDNAを、マトリックスとしてDNA 10ngを使用してpFuポリメラーゼ(NEB)を使用して、オリゴヌクレオチド5p 8328 CCCACCAAACCCAAAAAAAGAGATCCTAGAACTAGCTATGGCCGGACGGGCCATGGCGGAAGTGCAGCTGCAGGCTTC(配列番号11)及びオリゴヌクレオチド3p 8329 ACCGGGCCTCTAGACACTAGCTACTCGAGGGGCCCCAGTGGCCCTATCTATGCGGCCGCGCTACTCACAGTTAC(配列番号12)を用いてPCRによって増幅した。チューブの数はDNA品質の必要性に依存し、必要とされる形質転換体の数に関連する。典型的には100万個の酵母形質転換体を得るために、本発明者らは50μlの8回のPCRを行なった。
本発明者らは、細胞内発現及び高い熱安定性について最適化された、高度に機能的な骨格ファミリーを選択した。この選択は、抗生物質耐性遺伝子とVHHのコレクションとの間の融合タンパク質を使用して行なわれた。
プラスミド及びクローニング
6His-タグ及びトリプルc-mycタグから構成される合成遺伝子(Mister Gene)をpHEN2ファージミドベクター(Griffin 1.ライブラリー)のNotI部位とBamHI部位の間に挿入した。pENTR(商標)4ベクター(Invitrogen)からのccdB遺伝子をpHEN2ベクターのNcoI部位とNotI部位との間に挿入した。哺乳動物発現ベクターについては、VHH又はhs2dAbをNcoIとNotIによって消化し、pAOINT又はpmCheryyベクター(Clontech)にライゲートした。
pAO-CATは、カルボキシ末端のHAでタグ化されたクロラムフェニコールアセチルトランスフェラーゼ(CAT)をVHH配列に融合させることのできる細胞質内発現ベクターである。それは、VHH-CAT配列を、XbaI及びKpnIで消化されたpAOD-Tub1-mGFPベクター(Olichon A, et al. 2007. J Biol Chem. Dec 14;282(50):36314-20)にクローニングして、DsbC-Tub1-mGFPを除去することによって構築された。VHH-CAT配列は、多段階PCR戦略によって得られた。VHHを、5’CCTTGATTCTAGAAATAATTTTGTTTAACTTTAAGAAGGAGATATACCATGCTGATGTCCAGCTGCAGGCGT3’(フォワード、配列番号13)及び5’CCACCGCTACCGCCGCTGCGG CCGCGTGAGGAGACGGTGACCTGG G3’(リバース、配列番号14)を使用して増幅した。CATの2つの配列を独立して、pRillプラスミドを鋳型として使用して増幅し、内部NcoI部位を除去した。N末端については以下のプライマーを使用した:5’ GCGGCCGCAGCGGCGGTAGCGGTGGCGAGAAAAAAATCACTGGATATACC 3’(フォワード、配列番号15)及び5’ GCCCATCGTGAAAACGGGGGCG 3’(リバース、配列番号16)。C末端を5’ CGCCCCCGTTTTCACGATGGGC 3’(フォーワード、配列番号17)及び5’AGAATAGGTACCAGCGTAATCTGGGACATCATAAGGGTAGCCACCCGCCCCGCCCTGCACTCATCG 3’(リバース、配列番号18)を使用して増幅した。3つの配列を最後のPCRで構築し、産物をXbaI及びKpnIで消化し、その後、ベクターにライゲートした。
FR及びCDRの設計に対応する遺伝子コレクションをインビトロで合成した(Sloning, GeneArt)。多様な合成物 1μL(10ng)(1×1010個の分子に相当、したがって、標的ライブラリー多様性の10倍)をPCRによって総容量50μLでPhusion DNAポリメラーゼ(New England Biolabs)1μLを以下のプライマーの等モル混合物と共に使用して増幅した:

ライブラリーからの個々のクローンの異質性を、ion Torrentチップ(Invitrogen)で6×105個の挿入断片をシークエンスすることによって確認した。
ヒトβアクチンはSigmaから購入した。アミノ末端のキチン結合ドメイン又はストレプトアクチン結合ペプチドに融合したRhoA GTPアーゼを、HEK293細胞において産生した。ストレプトアビジン結合ペプチド(SBP)と融合したGFPをインビトロでの翻訳システム(Roche)を通して産生し、精製を必要することなくスクリーニングに直接使用された(Moutel S, et al. 2009. Biotechnol J. Jan;4(1):38-43)。ビオチニル化チューブリンはCytoskeletonから購入した。p53については、NP_000537.3アイソフォームの最初の72アミノ酸を、SNAPタグと共に細菌内で産生させ、インビトロでビオチニル化した。Her2については、天然受容体をSKBR3細胞上の膜タンパク質標的として使用した。
βアクチン、H1ヒストン、又はFITCについてのスクリーニングが、Marks JD et al, 1991 J Mol Biol. Dec 5;222(3):581-97に記載のようにイムノチューブでのパニングによって行なわれた。GFP、チューブリン及びp53についてのスクリーニングが、Nizak et al. 2003 Science. May 9;300(5621):984-7に記載のようなナイーブな条件で行なわれた。Rhoに対するスクリーニングが、HEK293細胞において発現されたタグが恒常的に活性であるRhoAの突然変異体に対してナイーブな条件で行なわれた。Her2についてのスクリーニングが、Even-Desrumeaux K, Chames P. 2012 Methods Mol Biol.; 907:225-35に記載のような表面細胞上で行なわれた。
個々のクローンを、モノクローナルファージELISAによって何処にでも記載されているようにスクリーニングした(Lee. et al; 2007, Nat Protoc. 2(11): 3001-8)。
SDS-PAGEローディング緩衝液中で煮沸した後、試料を12%SDS-PAGEで分離し、ニトロセルロース膜(Whatman GmbH)上に転写した。膜を、室温で1時間又は4℃で一晩かけて、0.2%Tween20を含む3%無脂肪ミルク-PBS中で遮断した。SDABを1/100で使用し、1/3000の抗hisタグ抗体(Sigma)と共に膜に90分間かけて加えた。その後、ブロットを洗浄し、二次抗マウスHRPで標識された抗体(PBS0.1%Tween20中で1/10000に希釈)(Jakson ImmunoResearch Laboratories)と共に1時間インキュベートした。PBS 0.1%Tween20を用いて5回洗浄した後、次いで、二次抗体を、SuperSignal化学発光試薬(Pierce)及びHyperfilm ECL(GE HealthCare)を使用して顕現させた。
免疫蛍光スクリーニングを、以前に記載されているように(Nizak et al. 2003. Science. May 9;300(5621):984-7)HeLa細胞に対して行なった。
カバーガラス上で培養したHela細胞を、1ウェルあたり1μgのDNA(24ウェルプレート)又は10μgのDNA(直径10cm2の皿)を用いてCaPO4手順に従ってトランスフェクトした。細胞をトランスフェクションから12時間後に観察することができる。
細胞表面染色を、1%SFVの補充されたリン酸緩衝食塩水(PBS)中で行なった。100μlの上清(ファージ 80μl+PBS/ミルク1% 20μl)を、1×105個の細胞上で氷上で1時間インキュベートした。ファージの結合を、氷上で1時間かけて1:300の希釈率の抗M13抗体(GE healthcare)によって検出し、次いで、45分間かけてPEにコンジュゲートした1:1000の希釈率の抗マウス抗体(BD Pharmingen)によって検出した。試料を、CellQuest Proソフトウェア(BD Biosciences、サンノゼ、CA)を使用してFACSCaliburでのフローサイトメトリーによって分析した。
ライブラリーから選択されかつGFP及びErBB2に特異的である、hs2dAb抗体の結合親和性を、それぞれProteOn XPR36(BioRad)及びBiacore T200(GE Healthcare)を使用して25℃で行ない、1:1のラングミュア相互作用モデルに当てはめた。リガンドGFP(24kDa)を酢酸ナトリウム緩衝液(pH5.0)で1.6μMに希釈し、730RUでGLCチップ(BioRad)上にアミンカップリングによって固定した。一価単一ドメイン抗体(14kDa)100μLを分析物として使用し、1000~3μMの濃度で100μL/分で注入した(60秒間かけて注入、600秒間かけて解離)。完全な動態の設定が一回の実行(ワンショット)で収集され、それ故、表面を再生する必要がなかった。ErbB2細胞外ドメイン-Fc(96kDa)を酢酸ナトリウム緩衝液(pH5.0)で400μg/mLに希釈し、991RUでCM5チップ(GE Healthcare)上にアミンカップリングによって固定した。一価単一ドメイン抗体(14kDa)をHBS-EP+緩衝液で希釈し、300~3μMの濃度の分析物として30μL/分で1回サイクルの様式を使用して注入した(120秒間かけて注射、120秒間かけて中期の解離、600秒間かけて最後の解離)。独特な注射順序における動態を収集し、表面再生(10mM グリシンHCl、pH2.5、30μL/分で30秒間)は2回の連続したシリーズの間においてのみ行なわれた。
ライブラリーの設計
非常に安定しかつ機能的な抗体断片の豊富な大きな単一ドメイン抗体ライブラリーを作成する観点から、本発明者らは、単一のVHH骨格を同定することを目標とした。本発明者らは以前に、免疫性の又はナイーブなラマVHHライブラリーから数百個のクローンを選択した(Monegal A, et al. 2012 Dev Comp Immunol. Jan;36(1):150-6)。本発明者らは、細菌細胞質において凝集又はアンフォールディングしがちなクローンから非常に安定なクローンを区別するクロラムフェニコールフィルターアッセイ(Olichon)を使用して、一連の高度に発現されたクローンをスクリーニングした。本発明者らは、カルボキシ末端のHAでタグ化されたクロラムフェニコールアセチルトランスフェラーゼ(CAT)をVHH配列に融合させることを可能とする、pAO-CAT細胞質内発現ベクターを使用した。公表されている熱安定性VHH(Olichon A, et al BMC Biotechnol. 2007 Jan 26;7:7)又は細胞内発現抗体(Rothbauer U, et al Nat Methods. 2006 Nov;3(11):887-9)と比較することにより、ある骨格であるクローンD10は、クロラムフェニコールに対するより高い耐性を示した(図1a)。D10のVHHはさらに本発明者らの溶解度、熱安定性、及び、どのような公知の抗原も全く認識しない状況で哺乳動物細胞内で細胞内発現抗体として発現されても凝集しないという全ての基準に当てはまった。その後、本発明者らは、骨格の部分的なヒト化(Vincke C, et al. J Biol Chem. 2009 Jan 30;284(5):3273-84)が、ヒトVH3に見られる7個の残基に標的化することによって、その固有の特性に影響を及ぼすかどうかを評価した。固有の可溶性特性にとって重要であると思われたフレームワーク-2領域における4個のVHH特異的アミノ酸(42位、49位、50位及び52位)の特徴は(図1c)はそのままとした。
本発明者らは、CDR1及びCDR2の各々の位置について(公表されているVHH結合物質に見られるCDRの統計学的分析に基づいて)、天然の多様性を依然として模倣している一連のアミノ酸を合理的に設計することによって3つのCDRに合成的な多様性を導入した。合成CDR1及びCDR2のアミノ酸残基は以下の原則:

によって決定された。CDR3のアミノ酸配列は、以下のアミノ酸:S、T、F、G、A、Y、D、E、N、I、H、R、Q、L、P、V、W、K、Mの1つ以上の中から選択された9、12、15又は及び18個のアミノ酸を含む。
本発明者らはまず、315個のランダムなクローンをサンガーシークエンス法を用いてシークエンスすることによって機能的な多様性を評価した。9個の配列が終止コドンを有するか又は1塩基が欠失しているかのいずれかで見い出され、1つの配列が全てのCDR1を欠失し、もう1つはCDR1、FR1及びCDR2を欠失し、最後の2つの配列は空であることが認められた。したがって、ほんの非常に少数(4.1%)の欠陥クローンしか得られなかった。このことは、3×109個のクローンの大半が組換えhs2dAbを発現しているであろうことを示唆する。
hs2dAb-L1ライブラリーを、表1に報告された一連の異なる抗原に対して標準的な方法(Hoogenboom HR, et al. 1998 Jun;4(1):1-20)を使用してファージディスプレイによってスクリーニングした。様々なスクリーニングアプローチにおけるhs2dAb-L1の使用を検証するために、本発明者らはビーズ上での選択(これは大容量の抗原提示をもたらし、天然タンパク質に近い抗原のコンフォメーションを保つ)、イムノチューブ上でのパニング(標準的な方法として参照される)又は哺乳動物細胞の表面上に天然に提示される天然抗原上でのパニング(治療上関心のあるものに対してインビトロで選択をかける場合にしばしば行なわれるように)のいずれかを行なった。
GFP、Her2及びp53に対する単一ドメイン抗体の親和性を、ProteOn XPR36(BioRad)又はBiacore T200(GE Healthcare)を使用して測定した。親和性はナノモル範囲であると推定され(抗GFP抗体については3.06×10-8Mで、抗Her2抗体については1.94×10-8Mで、抗p53抗体については3.25×10-8M)、このことは、高親和性結合物質が、本発明者らの合成で非免疫原性の単一ドメイン抗体のhs2dAb-L1ライブラリーから得ることができたことを実証する(図5)。
遮断性抗体の同定は困難な課題である。しかし、その標的機能を阻害するために、非遮断性の細胞内発現抗体を官能基化することが可能である。1つのアプローチは、Caussinus et al. 2011(Caussinus et al. 2011, Nat. Struct. Mol. Biol. 19, 117-121)に記載のような認識された標的のユビキチン化及び分解に依拠する。このアプローチは、タンパク質をプロテアソーム依存性細胞分解へと標的化させる、複合体E1/E2/E3ユビキチン化機序のE3ユビキチンリガーゼである、SCF複合体の一員である、Skip1との相互作用を可能とする、F-ボックスドメインへの細胞内発現抗体の融合に基づく。このアプローチは当初、免疫ライブラリーから初めて単離された頑強な高親和性のGFPラマ細胞内発現抗体である、GFP4と命名された、単一抗GFP細胞内発現抗体を使用してショウジョウバエにおけるいくつかのGFP融合タンパク質を標的化するために開発された(Rothbauer, U. et al.Nat. Methods 3, 887-889)。このようなタンパク質ノックダウンアプローチのためのhs2dAbの相対的機能に関する洞察を得るために、本発明者らは、本発明者らの抗GFP hs2dAbのいくつかをそのアミノ末端においてFボックスドメインに融合させ、その効力をFボックス-GFP4抗体の効力と比較した。Fボックス-細胞内発現抗体の融合タンパク質(F-Ib)を発現している細胞を検出するために、本発明者らは、ミトコンドリアに標的化されたmCherryと一緒に(Mito-mCherry)F-Ibの共発現を駆動するビシストロン性ベクターを構築した。本発明者らは、ヒストンH2Bに融合したGFPを安定に発現しているHeLaクローンにおいてF-Ib抗体を発現し(Sillje, H. H. W., Nagel, S., Korner, R. & Nigg, E. A., 2006, Curr. Biol. CB 16, 731-742)、GFP-H2Bの欠失を探した。予想された通り、degradFPとしても知られる、F-GFP4は、ウェスタンブロットによって分析されるようなH2B-GFP発現の強力な減少を誘導した(データは示されていない)。したがって、核における蛍光強度の強力な減少が、F-GFP4を発現している細胞において観察された。GFP4単独、又は、切断短縮された非機能的なFボックスドメインに融合したGFP4のいずれかを発現している場合には全く効果は観察されなかった。本発明者らが本発明者らの新規ライブラリーから選択された抗GFPクローンを試験した時に、本発明者らは、蛍光細胞内発現抗体として使用された場合に効果的であることが判明したいくつかのhs2dAbは、F-Ibとして発現された場合にH2B-GFPを分解することができなかったことを観察した。このことは、全ての細胞内発現抗体が、F-ボックスを用いて効果的に官能基化されることができるわけではないという事実を強調する。しかし、ある抗GFP hs2dAbは、F-Ibとして発現された場合に核におけるH2B-GFPシグナルの完全な消失を誘導した。FACS分析は、70%も減少した蛍光強度を示した。予想された通り、この効果は、プロテアソーム阻害剤による処理の存在下で逆転した。
Claims (6)
- 合成単一ドメイン抗体ライブラリーを作成する方法であって、該方法は、
i.CDR1、CDR2及びCDR3をコードする多様な核酸を、合成単一ドメイン抗体のそれぞれのフレームワークコード領域間に導入して、その同じ合成単一ドメイン抗体骨格のアミノ酸配列を有する多様な合成単一ドメイン抗体をコードする核酸を作製する工程を含み、
前記の合成単一ドメイン抗体骨格は、フレームワーク領域の全体において1、2又は3つ以下の保存的アミノ酸置換を有する配列番号1のFR1、配列番号2のFR2、配列番号3のFR3、及び配列番号4のFR4からなるフレームワーク領域の機能的な変異を含み、機能的な変異フレームワーク領域は、以下の工程:
(i) アッセイするために、変異フレームワーク領域に移植した配列番号9のクローンD10のCDR1(FTFSSYA)、CDR2(DSSGNHA)及びCDR3(RSDAAGNPSG)を有する基準の単一ドメイン抗体を提供すること、
