Description
B7-H3は、B7免疫制御因子ファミリーにおける1種類の膜タンパク質である。ヒトでは2Ig-B7-H3および4Ig-B7-H3という2種のサブタイプがあるが、マウスでは2Ig-B7-H3という1種のサブタイプしか存在しない。B7-H3は、T細胞からI型インターフェロンの放出を低減することができ、ナチュラルキラー細胞の殺傷機能を阻害することもできると文献に示されているため、B7-H3は免疫抑制因子であると見なされている。また、B7-H3が移植片対宿主反応、心臓移植拒絶反応、気道炎症、自己免疫性脳脊髄炎などの疾患を抑制できることは、他の文献にも確認されている。しかしながら、B7-H3は、自己免疫のインビボ及びインビトロのモデルにおいてT細胞共刺激因子であると説明する文献もある。
マウス神経膠腫モデルによって確認されたように、本発明によって調製されたB7-H3キメラ抗原受容体のT細胞は、ほとんどのマウスの腫瘍を除去し、一定期間内に再発することなく腫瘍を制御することができ、生存期間は、顕著に延長され、本発明に係るB7-H3キメラ抗原受容体のT細胞がインビボで腫瘍細胞を殺傷することができ、潜在的な腫瘍治療法として使用できることを証明している。
本発明では、別段の指定がない限り、本明細書で使用される科学的および技術的用語は、当業者によって一般的に理解される意味を有する。さらに、本明細書で使用されるタンパク質および核酸化学、分子生物学、細胞および組織培養、微生物学、免疫学に関連する用語および試験室の操作手順は、対応する分野で広く使用される用語および日常手順である。一方、本発明をよりよく理解するために、関連用語の定義と説明を以下に示す。
「コードする」とは、遺伝子、cDNAまたはmRNAのようなポリヌクレオチドにおけるヌクレオチドの特異性配列がテンプレートとして機能し、生物学的プロセスにおける他のポリマーおよび大分子を合成する固有特性を指し、前記ポリマーおよび大分子は、ヌクレオチド(即ち、rRNA、tRNAおよびmRNA)の規定された配列またはアミノ酸の規定された配列のいずれか、およびそれ由来の生物学的特性を有する。したがって、遺伝子に対応するmRNAの転写および翻訳が細胞または他の生物学的システムにおいてタンパク質を産生する場合、その遺伝子はタンパク質をコードする。ヌクレオチド配列は、mRNA配列と同等であり、通常、配列リストにおけるコード鎖に提供され、それと遺伝子またはcDNAを転写するためのテンプレートとして機能する非コード鎖の両方も、その遺伝子またはcDNAをコードするタンパク質または他の産物と呼ばれる。
免疫原性組成物の「非経口」投与は、例えば、皮下(s.c)、静脈内(i.v.)、筋肉内(i.m.)または胸骨内の注射または注入技術を含む。
一態様において、本発明は、B7-H3を標的とするキメラ抗原受容体、及びB7-H3を標的とするキメラ抗原受容体を発現する免疫エフェクター細胞を提供する。具体的に、B7-H3を標的とするキメラ抗原受容体を発現する免疫エフェクター細胞は、B7-H3を標的とするキメラ抗原受容体を発現するT細胞である。
腫瘍におけるB7-H3抗原の発現状況を特定するために、まず、異なる腫瘍細胞株を検出のために選択した。原発腫瘍におけるB7-H3の発現をさらに検証するために、複数の患者の神経膠腫組織と正常組織を含む切片におけるB7-H3の発現状況を検出した。
本発明に用いられる細胞株は、10%FBS、1%glutaMAX、および1%ダイアボディを含むIMDM培地で培養されている293T細胞を含む。
WM-266-4、SK-OV-3、LN-229、LN-18、U-87MG細胞培地は、10%FBSのRPMI640又はDMEMであった。
キメラ抗原受容体は、1本鎖抗体領域、CD8αヒンジ領域、CD8α膜貫通領域、CD28又は4-1BB共刺激因子領域及びCD3ζシグナル伝達領域で構成された(図3に示した)。キメラ抗原受容体の1本鎖抗体領域(scFV)は、各モノクローナルハイブリドーマ細胞によって産生されたB7-H3に対する抗体に由来し、前記scFVアミノ酸配列は、SEQ ID NO:2、14、26、38、50、62、74、86、98又は110に示した。キメラ抗原受容体は、遺伝子合成によって生成され、レトロウイルスベクターにクローニングされた。クローニングが完了した後、配列決定によりその配列の正しさを確認した。

1、T細胞のトランスフェクション及び増幅:
健常者から提供された末梢血を使用して、末梢血単核細胞を分離した。リンパ球密度勾配遠心法により単独の単核細胞を単離した。分離された単独の単核細胞をCD3及びCD28の抗体により活性化された後、実施例4で回収されたレトロウイルス(CD19.28 CAR、B7-H3.BB CAR及びB7-H3.28 CAR)でT細胞を感染させた。次に、B7-H3キメラ抗原受容体の発現及び腫瘍の殺傷を検出するために、キメラ抗原受容体をトランスフェクションしたT細胞を増幅させた。
トランスフェクションから3日後のB7-H3キメラ抗原受容体-T細胞の一部を回収し、B7-H3と抗体Fcフラグメントとの融合タンパク質を利用して、B7-H3キメラ抗原受容体の発現を検出し、二次抗体はAPC蛍光染料で標識されたヤギ抗ヒトのFc抗体であった。フローサイトメトリーを利用してB7-H3キメラ抗原受容体のT細胞での発現を検出した。

本発明に係るキメラ抗原受容体がT細胞を介してB7-H3高発現の腫瘍細胞を殺傷したか否かを確認するために、T細胞と腫瘍細胞との共培養試験を設計して該T細胞による様々な腫瘍細胞の殺傷状況をテストした。腫瘍細胞が壁に接着してから24時間後に、コントロール(CD19.28CAR-T)及びB7-H3キメラ抗原受容体を発現するT細胞(B7-H3.28CAR-T)を添加し、その後フローサイトメトリーでこれらによる様々な腫瘍細胞の殺傷状況を検出した。
既存の特許に同じ抗原を標的とする他の2つのキメラ抗原受容体が開示されていることを考慮して、本発明に係るCARとこれらの2種類のCAR(CAR1は、WO2017/044699に由来し、CAR2は、US10,233,226B2に由来した)のB7-H3抗原に対する親和性及び3種類のCAR-T細胞による腫瘍細胞の殺傷過程におけるサイトカイン放出状況を比較した。
293T細胞にそれぞれB7-H3-FLAG又はFLAG空ベクターコントロールをトランスフェクションした。また、別の293T細胞に対して、それぞれ3種類の異なるB7-H3 CARをその発現量を一致するようにトランスフェクションした。48時間後、RIPA溶解液でB7-H3-FLAG又はFLAG空ベクターコントロールをトランスフェクションした293T細胞を溶解し、遠心分離して上清を回収し、抗-FLAG-M2アフィニティーゲルを添加し、4℃で2時間インキュベートし、RIPA溶解液で洗浄して置いた。RIPA溶解液で異なるB7-H3 CARをトランスフェクションした293T細胞を分解し、遠心分離して上清を回収した後、それぞれB7-H3-FLAG又はFLAG空ベクターコントロールをインキュベーションした後の抗-FLAG-M2アフィニティーゲルと4℃で1時間インキュベートした。RIPA溶解液で4回洗浄した後、溶出液(0.1M glycine-HCl、pH 3.5)で室温で10分間振動溶出し、上清を取り、1M Tris-HCl(pH 7.5)で中和反応をした後、SDSローディング緩衝液を加え、95℃で5分間変性し、ウエスタンブロットで親和性を分析した。
4種類の異なる腫瘍細胞(LN-229、LN-18、WM-266-4及びSK-OV-3)とコントロールとしてのCD19.28CAR-T、CAR1、CAR2及び本発明のB7-H3.28CAR-Tとの共培養試験において、トランスフェクションしてから10日間のT細胞を添加してから24時間後、上清の一部を回収した。ELISAのキットを用いて上清中のサイトカインを検出した。
本発明におけるB7-H3キメラ抗原受容体を発現するT細胞によるインビボでの腫瘍の殺傷作用を確認するために、ヌードマウスの神経膠腫モデルを選択した。6~8週齢のヌードマウス脳内にluciferase標識されたヒト腫瘍細胞(U-87MG又はLN-229細胞)を注射し、腫瘍を形成した後にさらに溶媒PBS、コントロールとしてのCD19.28 CAR-T、B7-H3 BB CAR-T及びB7-H3.28 CAR-T細胞を同所性注射し、その後、T細胞による腫瘍クリアランスと治療されたマウスの生存状況を、画像化によって検出した。
1、インビトロでのB7-H3キメラ抗原受容体-T細胞による患者由来神経膠腫ニューロスフェアの殺傷
本発明において、B7-H3キメラ抗原受容体を発現するT細胞による患者由来腫瘍の殺傷作用を検証するために、神経膠腫患者の神経膠腫幹細胞を分離し、かつ神経膠腫ニューロスフェアに培養した。さらに神経膠腫ニューロスフェアとコントロール(CD 19-CAR)及びB7-H3キメラ抗原受容体(B7-H3.BBCAR及びB7-H 3.28CAR)を発現するT細胞と共培養し、その後、顕微鏡観察及びフローサイトメトリーによってT細胞による患者神経膠腫ニューロスフェアの殺傷状況を検出した。
インビボでのB7-H3キメラ抗原受容体のT細胞による患者由来神経膠腫ニューロスフェアの殺傷作用をさらに確認するために、患者神経膠腫ニューロスフェアを利用して異種同所性移植腫瘍マウスモデルを構築した。ヌードマウスの脳内に患者の膠腫ニューロスフェア細胞を同所性注射し、2週間後に同様に脳内に溶媒PBS、コントロールとしてのCD19.28CAR-T、B7-H3.BB CAR-T及びB7-H3.28 CAR-T細胞を注射し、その後、各群の治療されたマウスの生存状況を記録した。
1.9種類のマウス由来B7-H3キメラ抗原受容体-T細胞による腫瘍細胞の殺傷作用
本発明において、15C11からクローンされたヒト化1本鎖抗体以外、他の9つのB7-H3抗原を識別するマウス由来1本鎖抗体(番号:53E9D7、15C11C3、6E4D9、31F3E5、21H2E11、24A11A11、42B5C12、14A7A8、41C10E7)をさらに含む。これらのマウス由来キメラ受容体が同様にT細胞を介してB7-H3を高く発現する腫瘍細胞を殺傷することができることを確認するために、共培養試験を設計してこれらのマウス由来CAR-T細胞による腫瘍細胞(LN-229)の殺傷状況をテストし、かつ比較した。腫瘍細胞を壁に接着させた後、コントロールとしてのCD19.28 CAR-T及び9種類のマウス由来B7-H3.28 CARを発現するT細胞を添加し、その後フローサイトメトリーでそれらによる腫瘍細胞の殺傷状況を検出した。
T細胞と腫瘍細胞との共培養試験において、9種類のマウス由来B7-H3 CAR-T細胞による腫瘍細胞の殺傷過程におけるサイトカインドの放出状況を検出した。
腫瘍細胞(LN-229とWM-266-4)とキメラ抗原受容体-T細胞(CD19.28 CAR-T及び9種類のマウス由来B7-H3.28 CAR-T)との共培養試験にCAR-T細胞を添加し、24時間後に、培養上清液の一部を回収した。ELISAキットを用いて上清中のサイトカインを検出した。
Claims (21)
- 抗B7-H3結合ドメイン、ヒンジ領域、膜貫通ドメイン及びシグナル伝達ドメインを含み、
前記抗B7-H3結合ドメインは、抗B7-H3抗体又は抗原結合部を含み、前記抗体又は抗原結合部は、下記のCDR組み合わせ(1)及び(2):
(1)アミノ酸配列がSEQ ID NO:5である軽鎖CDR1、アミノ酸配列がSEQ ID NO:6である軽鎖CDR2、アミノ酸配列がSEQ ID NO:7である軽鎖CDR3、アミノ酸配列がSEQ ID NO:10である重鎖CDR1、アミノ酸配列がSEQ ID NO:11である重鎖CDR2、アミノ酸配列がSEQ ID NO:12である重鎖CDR3、及び
(2)アミノ酸配列がSEQ ID NO:29である軽鎖CDR1、アミノ酸配列がSEQ ID NO:30である軽鎖CDR2、アミノ酸配列がSEQ ID NO:31である軽鎖CDR3、アミノ酸配列がSEQ ID NO:34である重鎖CDR1、アミノ酸配列がSEQ ID NO:35である重鎖CDR2、アミノ酸配列がSEQ ID NO:36である重鎖CDR3、から選ばれる一種を含むことを特徴とする、B7-H3を標的とするキメラ抗原受容体(CAR)。 - 前記抗B7-H3結合ドメインは、抗B7-H3抗体又は抗原結合部を含み、前記抗体又は抗原結合部は、アミノ酸配列SEQ ID NO:4、28から選ばれる軽鎖配列、及び、アミノ酸配列SEQ ID NO:9、33から選ばれる重鎖配列を含む、請求項1に記載のCAR。
- 前記抗B7-H3結合ドメインは、抗B7-H3抗体領域(scFv)を含み、前記抗B7-H3の1本鎖抗体のアミノ酸配列が、SEQ ID NO:2、26から選ばれるアミノ酸配列である、請求項1又は2に記載のCAR。
- 前記膜貫通ドメインは、TCRのα、β、ζ鎖、CD3γ、CD3δ、CD3ε、CD3ζ、CD4、CD5、CD8α、CD9、CD16、CD22、CD27、CD28、CD33、CD37、CD45、CD64、CD80、CD86、CD134、4-1BB、CD152、CD154、PD-1膜貫通ドメインから選ばれる1種又は複数種であり、
前記ヒンジ領域は、CD8の細胞外ヒンジ領域、IgG1 Fc CH2CH3ヒンジ領域、IgDヒンジ領域、CD28細胞外ヒンジ領域、IgG4 Fc CH2CH3ヒンジ領域及びCD4の細胞外ヒンジ領域から選ばれる1種又は複数種である、請求項1~3のいずれかに記載のCAR。 - 前記膜貫通ドメインのアミノ酸配列が、SEQ ID NO:124のアミノ酸配列であり、
前記ヒンジ領域のアミノ酸配列が、SEQ ID NO:122のアミノ酸配列である、請求項1~4のいずれかに記載のCAR。 - 前記シグナル伝達ドメインは、TCRζ、FcRγ、FcRβ、CD3γ、CD3δ、CD3ε、CD5、CD22、CD79a、CD79b、CD278(ICOS)、CD66d、CD3ζの細胞内シグナル伝達ドメインから選ばれる1種又は複数種である、請求項1~5のいずれかに記載のCAR。
- 前記シグナル伝達ドメインのアミノ酸配列が、SEQ ID NO:130のアミノ酸配列である、請求項6に記載のCAR。
- 前記シグナル伝達ドメインが、さらに1つ又は複数の共刺激ドメインを含み、前記共刺激ドメインが、CARD11、CD2、CD7、CD27、CD28、CD30、CD40、CD54、CD83、OX40、4-1BB、CD134、CD150、CD152、CD223、CD270、PD-L2、PD-L1、CD278、DAP10、LAT、NKD2C、SLP76、TRIM、FcεRIγ、MyD88、4-1BBの細胞内シグナル伝達ドメインから選ばれる1種又は複数種である、請求項1~7のいずれかに記載のCAR。
- 前記共刺激ドメインのアミノ酸配列が、SEQ ID NO:126、128から選ばれるアミノ酸配列である、請求項8に記載のCAR。
- 請求項1~9のいずれかに記載のCARをコードするポリヌクレオチド配列を含む、分離された核酸分子。
- 前記核酸分子のヌクレオチド配列が、SEQ ID NO:1、25から選ばれるヌクレオチド配列である、請求項10に記載の分離された核酸分子。
- 請求項10又は11に記載の核酸分子を含む、核酸構築体。
- 前記核酸構築体が、ウイルスベクターであり、前記ウイルスベクターがレトロウイルスベクター、レンチウイルスベクター、アデノウイルスベクター、アデノ随伴ウイルスベクターから選ばれる1種又は複数種である、請求項12に記載の核酸構築体。
- 請求項10又は11に記載の核酸分子あるいは請求項12又は13に記載の核酸構築体を含む、ウイルスであって、前記ウイルスがレトロウイルス、レンチウイルス、アデノウイルス、アデノ随伴ウイルスから選ばれる1種又は複数種である、ウイルス。
- 請求項1~9のいずれかに記載のCAR、請求項10又は11に記載の核酸分子、請求項12又は13に記載の核酸構築体、請求項14に記載のウイルスの、B7-H3を発現する腫瘍細胞を標的とする遺伝子改変免疫細胞を調製するための使用。
- 請求項1~9のいずれかに記載のCAR、又は請求項10又は11に記載の核酸分子、又は請求項12又は13に記載の核酸構築体、又は請求項14に記載のウイルスを含む、分離された宿主細胞。
- 前記宿主細胞がT細胞、NK細胞又は細胞株、γδT細胞、NKT細胞、マクロファージまたは細胞株、PG13細胞株から選ばれる1種又は複数種である、請求項16に記載の分離された宿主細胞。
- 前記宿主細胞は、他のエフェクターをさらに発現し、前記他のエフェクターは、サイトカイン、もう一種のCAR、ケモカイン受容体、PD-1発現を低下するsiRNAあるいはPD-L1を阻害するタンパク、TCR、又は安全スイッチを含む、請求項16又は17に記載の宿主細胞。
- 請求項1~9のいずれかに記載のCAR、請求項10又は11に記載の核酸分子、請求項12又は13に記載の核酸構築体、請求項14に記載のウイルス、請求項16~18のいずれかに記載の宿主細胞から選ばれる1種又は複数種と、薬学的に許容される担体とを含む、医薬組成物。
- 疾患の診断、治療又は予防に使用される、請求項1~9のいずれかに記載のCAR、請求項10又は11に記載の核酸分子、請求項12又は13に記載の核酸構築体、請求項14に記載のウイルス、請求項16~18のいずれかに記載の宿主細胞、又は、請求項19に記載の医薬組成物であって、前記疾患ががん、炎症性疾患、自己免疫性疾患から選ばれる1種又は複数種である、請求項1~9のいずれかに記載のCAR、請求項10又は11に記載の核酸分子、請求項12又は13に記載の核酸構築体、請求項14に記載のウイルス、請求項16~18のいずれかに記載の宿主細胞、又は、請求項19に記載の医薬組成物。
- 前記疾患は、B7-H3を発現する腫瘍であり、前記腫瘍は、脳がん、膵臓がん、卵巣がん、腎臓がん、膀胱がん、皮膚がん、胃がん、腸がん、頭頸部がん、甲状腺がん、前立腺がん、肺がん、カポジ肉腫から選ばれる1種又は複数種であり、前記脳がんが膠芽腫、星状細胞腫、髄膜腫、乏突起膠腫及び神経膠腫から選ばれる1種又は複数種である、請求項20に記載の疾患の診断、治療又は予防に使用される、CAR、核酸分子、核酸構築体、ウイルス、宿主細胞、又は、医薬組成物。
