(A~C)NPのPEG化後、精製前に、PEG-Cy5-C'ドットの反応混合物に、チオール-シランを導入することによって合成した、精製チオール官能化SH-PEG-Cy5-C'ドットのGPCエルグラム(A)、FCS相関曲線及びフィット(B)、及びUV-可視吸光度スペクトル(C)。追加のチオール官能化を行わなかったNPと比べて、250nm未満にわずかに増加した吸収が観察された(C)。
(D~F)チオール-シランの添加後、NP精製前に、チオール-官能化SH-PEG-Cy5-C'ドットの反応混合物に、FITC-NCS色素をさらに導入することによって合成した、精製FITC-PEG-Cy5-C'ドットのGPCエルグラム(D)、FCS相関曲線及びフィット(E)、及びUV-可視吸光度スペクトル(F)。チオール-官能化無しのPEG-Cy5-C'ドットを使用して同じように合成したものと比較して、FITC-PEG-Cy5-C'ドットの吸収シグナルは、450nm付近の波長で実質的に増加する。
(F)このシグナルは、DI水におけるFITC色素の吸光度シグナルと対応し、FITCの付着の成功と、SH-PEG-Cy5-C'ドット上のチオール基のアクセス性を確認する。低いFITCシグナルが、チオール-官能化無しのサンプルでも観察されたが、これは、C'ドットへのFITC色素の非特異的吸収か、又はCy5色素シラン-コンジュゲーション工程で使用した過剰のチオール-シランがもたらした、標準的C'ドット合成で残ったチオール基に起因していると思われる。
(A)PEG-Cy5-C'ドット、c(RGDyC)、DFO、及びFITCからの吸収シグナルの線形結合を使用して十分にフィットされるFITC-DFO-c(RGDyC)-PEG-Cy5-C'ドットのUV-可視吸光度スペクトル。デコンボリューションは、図12のDFO-c(RGDyC)-PEG-Cy5-C'ドットについて記載したのと同様であるが、さらに、解析においてFITCのUV-可視シグナルを含む。そのフィットに基づいて、スペクトル(A)を、PEG-Cy5-C'ドット(B)、c(RGDyC)(C)、DFO(D)、及びFITC(E)からの寄与にデコンボリュートする。フィットにおけるこれらの寄与から、NPあたりのCy5、c(RGDyC)、DFO、及びFITC分子の数が、それぞれ約1.7、23、19及び4と推定される。

この実施例は、本開示の方法及び組成物、並びにその使用の例を示す。
c(RGDyC)-PEG-Cy5.5-C'ドットの合成及び精製。以前に報告されたC'ドットの合成は、異なるタイプの色素及び癌標的化ペプチドを用いてC'ドットの機能化を可能にする多用途アプローチである(図8)。例えば、c(RGDyC)-PEG-Cy5.5-C'ドットと呼ばれる、NIR色素Cy5.5及びαvβ3インテグリン-ターゲティング環状(アルギニン-グリシン-アスパラギン酸-D-チロシン-システイン)ペプチド(c(RGDyC))で機能化したC'ドットを生成するために、オルトケイ酸テトラメチル(TMOS)及びCy5.5 シランコンジュゲート(Cy5.5-シラン)を、まず室温でpH8付近の水酸化アンモニウム水溶液に添加した。加水分解されたTMOS及びCy5.5-シラン分子は、一緒に縮合して、シリカマトリクス中に共有結合的に封入されたCy5.5色素を有する極小シリカNPを形成する。次の工程では、シラン官能化PEG(PEG-シラン)及びc(RGDyC)ペプチド修飾PEG-シラン(c(RGDyC)-PEG-シラン)を一緒に反応混合物に添加した。添加すると、c(RGDyC)-PEG-シラン及びPEG-シランは、表面シラノール基とPEGとの間の水素結合を介してNP表面上に迅速に吸着され、それによってナノ粒子成長を終結させる。反応混合物を室温で一晩放置した後、反応温度を80℃に上昇させ、そこで一晩維持して、NP表面へのPEG-シラン及びc(RGDyC)-PEG-シランの共有結合を増強した。次いで、反応混合物を室温まで冷却し、c(RGDyC)-Cy5.5-Cドットを、最終的にゲル浸透クロマトグラフィー(GPC)を用いて精製し、続いて最終用途のために滅菌フィルターを用いて濾過した。
挿入によるPEG化後表面修飾(PPSMI)を介したC'ドット表面への二次機能基の導入。PEG化の間に異なるリガンド-シラン・コンジュゲートを共縮合させることによってC'ドット表面に二次機能基を導入する試みは、異なる機能基と裸のシリカNP表面との間の劇的に変化する親和性に基づく。生物医学的用途にとって興味深い機能性リガンドの多くは、シリカに対して高い親和性を有するかなりの量のアミン基及び/又はヒドロキシル基を保有する。前節で説明したように、このようなリガンドとシリカとの間の強い会合は潜在的に、それらのアクセス不能性及び不均質な分布につながる可能性がある。
c(RGDyC)-PEG-C'ドットにDFOを付着させるのと同様のアプローチを使用して、他のタイプの機能性リガンドを導入することもできる。例えば、1,4,7,10-テトラアザシクロドデカン-1,4,7,10-四酢酸(DOTA)は、177Luのような放射性同位体による放射性標識のための最も効率的なキレート剤の1つである。DFO-NCSをDOTA-NCSに置き換えることにより、狭く分散したDOTA機能化c(RGDyC)-PEG-C'ドット、すなわちDOTA-c(RGDyC)-PEG-C'ドットが生成され、それにより、例えば、177Lu放射性標識が可能になった(図4E~H)。このアプローチは、他のタイプのコンジュゲーション化学にも適用することができる。例えば、PEG化後表面修飾工程においてアミノシランをチオールシランで置換することによって、チオール官能化C'ドットを生成することができ、追加のチオール基は、チオール-エンクリック反応を介して、例えばマレイミド-官能化リガンドとさらに反応することができる。原理証明として、このアプローチによって、チオール-シランをPEG-Cy5-C'ドットのシリカ表面上に導入し、狭く分散したSH-PEG-Cy5-C'ドットを生成した(図13A~C、表1)。その後、マレイミド官能化FITC色素(mal-FITC)を、精製工程の前にSH-PEG-Cy5-C'ドットの反応混合物に添加して、チオール-マレイミド・コンジュゲーションを介してpH検知FITC色素でNPを機能化した(図1)。得られたFITC-PEG-Cy5-C'ドットは精製後にクリーンなGPCエルグラムを示し、高品質のNP精製物を示唆した(図13D)。FCS特性評価は、6.7nmの平均粒径を示した(図13E、表1)。精製されたFITC-PEG-Cy5-C'ドットは、PEG-Cy5-C'ドットと比較して、450nm付近の波長でさらなる吸収特性を示し、これはFITC色素の特徴的な吸収に対応していた(図13F)。結果は、チオール-エンクリック反応によるFITC色素の付着の成功と、チオール-PEG-Cy5-C'ドット上のチオール基のアクセス性を実証した。
上述のPPSMIアプローチの最も重要な利点の1つは、それが、十分に確立された、モジュール式の、ならびに直交コンジュゲーション化学の豊富なツールボックスによってサポートされるという事実である。例えば、上記のアミン-シラン/NCS-リガンド及びチオール-シラン/mal-リガンド化学に加えて、類似のアプローチ、例えば、アジド-シラン/アルキン-リガンドを用いて、他のタイプのコンジュゲーション化学を適用することもできる。さらに、異なる化学的性質を用いるこれらのコンジュゲーション反応は、通常、互いに干渉しないので、異なる機能性リガンドをC'ドットに同時に付着させることができる。
我々は、異なるタイプの機能基が、PEG化工程の間又はPEG化後の表面修飾を介して、C'ドット表面に導入できることを記載した。これらの2つのアプローチの組合せは、水性媒体中でのワンポット合成アプローチにおいて、最も多数の表面機能基を有する多機能性C'ドットへの、モジュール経路及び直交経路を提供する。その目的のために、原理証明として、NP表面上に全部で4つの異なる機能性リガンドを有する5機能性C'ドットを合成し(図1)、同時光学トレーシング、特異的細胞標的化、レシオメトリック検知、放射性金属キレート化、及び潜在的薬物送達を可能にした。
全ての材料は、受け取ったままで使用した。ジメチルスルホキシド(DMSO)、イソプロパノール、(3-メルカプトプロピル)トリメトキシシラン(MPTMS)、(3-アミノプロピル)トリエトキシシラン(APTES)、(3-アミノプロピル)トリメトキシシラン(APTMS)、オルトケイ酸テトラメチル(TMOS)、エファビレンツ(EFV)、及びエタノール中2.0Mアンモニアを、Sigma Aldrichから購入した。メトキシ末端ポリ(エチレングリコール)鎖(PEG-シラン、モル質量約500)をGelestから購入した。マレイミド基及びNHSエステル基を有するヘテロ二官能性PEG(mal-PEG-NHS、モル質量約870)をQuanta BioDesignから購入した。ヘテロ官能性アジド-PEG-チオール(約600のモル質量)は、Nanocs Inc.から購入した。Cy5及びCy5.5蛍光色素をGEから購入し、CW800蛍光色素をLi-corから購入した。S-2-(4-イソチオシアナトベンジル)-1,4,7,10-テトラアザシクロドデカンテトラ酢酸(DOTA-NCS)、及び1-(4-イソチオシアナトフェニル)-3-[6,17-ジヒドロキシ-7,10,18,21-テトラオキソ-27-(N-アセチルヒドロキシルアミノ)-6,11,17,22-テトラアザヘプタエイコシン]チオ尿素(DFO-NCS)を、Macrocyclicsから購入した。環状(アルギニン-グリシン-アスパラギン酸-Dチロシン-システイン)ペプチド(c(RGDyC))は、Peptide Internationalから購入した。Millipore Milli-Qシステムを用いて脱イオン水(DI水)を生成した。
本明細書で導入され、異なるC'ドット化学を区別するために以下で使用される命名法は、図7に示される(表1も参照)。より詳細には、左側からの第1の語がNP生成物プラットフォーム、例えば、水ベースのC'ドット又は単孔メソポーラスシリカナノ粒子(mCドット)。シリカマトリクス中に共有結合的に封入された特定の色素は、NPプラットフォームに続く括弧内で特定される(例えば、図7のCy5)。括弧の右側の内容は、NP表面に付着した成分を、それらの付着の順序で記述する(ドットは最初に合成されるので、それらは既存の短縮された命名法におけるように、右側ではなく左側に現れる)。粒子表面上の機能基の結合性を強調するために、我々は以下の区別を導入する:ダッシュの右側に現れる成分は、それがそのダッシュの左側の成分に直接付着していることを示す。対照的に、成分がアンダースコアの右側に現れる場合、この成分は、そのアンダースコアの左側の先の成分に付着されるのではなく、シリカNP表面に直接付着される。コンジュゲーション化学に使用される基(例えば、マレイミドとチオール基との間のコンジュゲーションを表すmal-チオール)とは対照的に、機能を規定する成分(例えば、標的リガンド又は他の蛍光色素)は太字であり、イタリック体である(図7及び表1を参照のこと)。機能基がPEG鎖を介してシリカNP表面に連結される場合、エチレングリコールモノマー単位の数が示される(例えばPEG6又はPEG12)。序論で概説したように、このようにして、ナノ粒子名は、(i)NPプラットフォーム、(ii)封入された蛍光色素、(iii)特定の表面機能性及び粒子へのその結合性、(iv)特定のコンジュゲーション化学、(v)合成順序、又は(vi)特定のPEG鎖長に関する正確な情報を有する。
c(RGDyC)機能化C'ドットの詳細な合成は、我々の以前の刊行物に記載されている。様々な種類のNIR色素を封入するために、シラン官能化色素、すなわち、Cy5-シラン、Cy5.5-シラン及びCW800-シランを、C'ドット合成の第一工程の間にTMOSと一緒に、反応混合物に添加した。NP合成の残りは、c(RGDyC)機能化C'ドットについて以前に報告したプロトコルと同じままとした。
c(RGDyC)-PEG-シランの濃度を、最初の0.69mMから(これは、C'ドット当たりのc(RGDyC)ペプチドの数を、典型的には16~25とする)、1.73mM(この濃度は、NP当たりのc(RGDyC)の数をさらに増加させる)に増加させた。NP凝集を防ぐために、水酸化アンモニウムの反応濃度も2mMから6mMに増加させた。これは、6mMの水酸化アンモニウム濃度でC'ドット合成を開始するか、又はPEG化工程の直前に追加の水酸化アンモニウムを添加して水酸化アンモニウムの濃度を6mMに増加させるかのいずれかによって、達成することができる。これらの2つのわずかに異なるアプローチによって生成された最終的なC'ドット生成物には、わずかな差異のみが観察された。したがって、製造工程を簡略化するためには、アンモニウム濃度6mMで合成を開始することが好ましい。
C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6に追加の機能性リガンドを導入するために、PEG化工程中に、リガンド-シラン・コンジュゲートを、c(RGDyC)-PEG-シラン及びPEG-シランと共に、反応混合物に添加した。これらのリガンド-シラン・コンジュゲートには、マレイミド官能化ヘテロ二官能性PEG-シラン(mal-PEG-シラン)、DFO機能化シラン(DFO-NCS-アミン-シラン)及びDBCO機能化ヘテロ二官能性(二機能性)PEG-シラン(DBCO-PEG-シラン)が含まれた。これらのリガンド-シラン・コンジュゲートの濃度は、表面リガンド密度の要求に応じて、0.01mM~0.34mMの範囲であった。このアプローチによって産生されたC'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG12-mal_PEG6は、チオール-官能化リガンドとの所望の反応性を示した。しかし、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_アミン-NCS-DFO_PEG6及びC'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG4-DBCO_PEG6は、望ましくない特性プロファイル又は不均一なリガンド分布を示した。
C'ドットにアミン基を導入するために、アミノシラン、すなわちAPTMSを、C'ドット合成の異なる工程の間に、反応混合物中に導入した。1つの方法では、APTMSを、c(RGDyC)-PEG-シラン及びPEG-シランを1.7mMの濃度で添加する直前に、激しい撹拌下で反応混合物に添加したが、合成の残りは同じままとした。得られたC'ドット(色素)-アミン_PEG12-mal-チオール-c(RGDyC)_PEG6は、広がったNPサイズ分布を示した。同時に、Cy5色素の分解が観察された。PEG化後アプローチでは、APTMSをPEG化工程の後に添加した。これは、PEG化工程の最後で、まず反応温度を80℃から室温に下げ、続いて2.3mMの濃度で激しく撹拌しながらAPTMSを添加することによって行った。次いで、反応混合物を、激しく撹拌しながら室温で一晩放置した後、精製した。得られたC'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミンは、GPC及びUV-vis特性評価によって示されるように、所望のNPサイズ分布及び好ましい吸光度特性を示した。このアプローチでは、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミンを、0.3mM程度の低いAPTMS濃度で首尾よく製造することができた(アミン反応性リガンドとの反応性を失ったNP生成物を生じることなしに)。
C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFO、及び、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DOTAは、アミノ-シラン添加の1日後、激しく撹拌しながら室温にて、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミンの反応混合物中に、DFO-NCS又はDOTA-NCSをそれぞれ添加することによって合成した。反応物を、激しく撹拌しながら室温にて一晩放置し、その後精製した。DFO-NCS、及びDOTA-NCSの濃度を、大まかに0.02mM~0.4mMの間で変化させ、NP当たりのDFO又はDBCOの数を変化させた。APTMS及びDFOの両方の濃度は、NP当たりのDFOの数を制御するために変えることができた。
PEG-C'ドットにチオール基を導入して、チオール-官能化C'ドット(色素)-PEG6_チオールを生成するために、アミン-シランをチオール-シランで置き換えること、及び、ベースNPとして、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6の代わりに、C'ドット(色素)-PEG6を使用することによって、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミンの合成を修正した。NPの合成の残りは、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミンの合成と同じとした。さらに、FITC色素を、C'ドット(色素)-PEG6_チオールに付着させて、C'ドット(色素)-PEG6_チオール-mal-FITCを生成するために、DFO-NCSをFITC-malで置き換えることによって、及び、ベースNPとして、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミンの代わりに、C'ドット(色素)-PEG6_チオールを使用することによって、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFOの合成を修正した。NPの合成の残りは、C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFOの合成と同じとした。
C'ドット(色素)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFOのために記載した合成アプローチにより、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG6にまずDFO基を導入することによって、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFO_チオール-mal-FITCを合成した。その後、前記合成アプローチにより、チオール基を、C'ドット(色素)-PEG6_チオールに添加して、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFO_チオールを生成した。最後の工程で、前記合成アプローチにより、FITC色素を、C'ドット(色素)-PEG6_チオール-mal-FITCに付着させた。得られたC'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFO_チオール-mal-FITCを、最終的に、GPCによって精製し、FCS及び光学特性評価に供した。同じC'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFO_チオール-mal-FITCはまた、別のアプローチでも合成され、そのアプローチでは、アミン-シランとチオール-シランを、別々に添加する代わりに、反応混合物中に一緒に添加した。反応物をその後、激しく撹拌しながら室温で4時間放置し、続いて、DFO-NCS及びFITC-malを同時に添加した。反応の残りは同じとした。
DMSO中でのアジド-アルキン・クリックケミストリーによって、まずEFVにアジド-PEG-チオールをコンジュゲートすることによって、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG12-mal-チオール-PEG10-EVF_PEG6_アミン-NCS-DFO_チオール-mal-FITCを合成した。この際、モル比は4EFV:1アジド-PEG-チオールとし、EFVの濃度は約0.021Mとした。反応混合物を、窒素下で6日間放置し、所望のコンジュゲーション率を達成した。得られたチオール-官能化EFVを、次いで、PEG工程の直後、すなわち、80℃での熱処理工程後に、1.15mMの濃度で、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG12-mal_PEG6の反応混合物中に添加した。反応混合物をその後激しく撹拌しながら、室温でさらに6日間放置し、チオール-官能化EFVを、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG12-mal_PEG6上のマレイミド基に付着させて、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG12-mal-チオール-PEG10-EVF_PEG6を生成した。その後、C'ドット(Cy5)-PEG12-mal-チオール-c(RGDyC)_PEG6_アミン-NCS-DFO_チオール-mal-FITCのために上述したのと同じ手順によって、DFO及びFITCを導入した。
C'ドットの詳細なGPC特性評価及び精製工程は、我々の以前の刊行物に記載されている。
試料の吸光度スペクトルを、Varian Cary 5000分光光度計で測定した。FCS測定は、前述のように家庭用FCS/FCCSセットアップを使用して行った。635nmの固体レーザーを用いてCy5及びCy5.5ドットを励起し、785nmの固体レーザーを用いてcw800ドットを励起した。
本明細書で導入され、異なるC'ドット化学を区別するために以下で使用される命名法は、図7に示される(表1も参照)。より詳細には、左側からの第1の語がNP生成物プラットフォーム、例えば、水ベースのC'ドット又は単孔メソポーラスシリカナノ粒子(mCドット)。シリカマトリクス中に共有結合的に封入された特定の色素は、NPプラットフォームに続く括弧内で特定される(例えば、図7のCy5)。括弧の右側の内容はNP表面に付着した成分を、それらの付着の順序で記述する(ドットは最初に合成されるので、それらは我々の既存の命名法におけるように、右側ではなく左側に現れる)。粒子表面上の機能基の結合性を強調するために、我々は以下の区別を導入する:ダッシュの右側に現れる成分は、それがそのダッシュの左側の成分に直接付着していることを示す。対照的に、成分がアンダースコアの右側に現れる場合、この成分は、そのアンダースコアの左側の先の成分に付着されるのではなく、シリカNP表面に直接付着される。コンジュゲーション化学に使用される基(例えば、マレイミド基とチオール基との間のコンジュゲーションを表すmal-チオール)とは対照的に、機能を規定する成分(例えば、標的リガンド又は他の蛍光色素)は太字であり、イタリック体である(図7及び表1を参照のこと)。機能基がPEG鎖を介してシリカNP表面に連結される場合、エチレングリコールモノマー単位の数が示される(例えばPEG6又はPEG12)。序論で概説したように、このようにして、ナノ粒子名は、(i)NPプラットフォーム、(ii)封入された蛍光色素、(iii)特定の表面機能性及び粒子へのその結合性、(iv)特定のコンジュゲーション化学、(v)合成順序、又は(vi)特定のPEG鎖長に関する正確な情報を有する。

低濃度のDFO-SCNを用いて、各C'ドットに、5個未満と少ないDFOキレート剤を導入した。従って、C'ドット表面上の残りのアミン基は、上記のようにDBCO付着のために使用され得る。DFO-SCNの添加の1日後、次いで、DBCO-PEG4-NHSエステルを反応混合物に添加して、アミン-NHSエステル・コンジュゲーション反応を介して、C'ドットのPEG層下の残りのアミン基とさらに反応させた。その結果、DFOとDBCOの両方が、PPSMI法によりPEG鎖間のC'ドット表面に共有結合した。

Claims (18)
- PEG化シリカナノ粒子を機能化して、機能性PEG化シリカナノ粒子を形成する方法であって、
挿入によるPEG化後表面修飾(PPSMI)工程を使用して、シリカナノ粒子を機能化することを含み、
ここで、前記シリカナノ粒子は、10nm未満の直径を有し、ナノ粒子の表面に共有結合した複数のポリエチレングリコール(PEG)基を有しており、及び
前記PPSMI工程は、機能化前駆体を、シリカナノ粒子上のPEG基の間に挿入して、シリカナノ粒子の表面に前記前駆体を共有結合させることを含み、ここで、前記機能化前駆体は、シラン基及び反応基を有するものであり、及び、前記方法は水性媒体中で実施されるものであり、
これにより、ナノ粒子の表面に共有結合された反応基を有する機能性PEG化シリカナノ粒子を形成する、方法。 - さらに、1つ以上の追加の機能化前駆体を、前記シリカナノ粒子上のPEG基の間に挿入して、シリカナノ粒子の表面に前記1つ以上の追加の前駆体を共有結合させることを含み、ここで、前記1つ以上の追加の前駆体はそれぞれ、シラン基及び反応基を有するものである、請求項1に記載の方法。
- 各追加の機能化前駆体の挿入が、同じ反応混合物中で行われる、請求項2に記載の方法。

- 前記シリカナノ粒子が、コア-シェルシリカナノ粒子である、請求項1~7のいずれか1項に記載の方法。
- 前記シリカナノ粒子の表面に共有結合された反応基を、機能基前駆体と反応させる工程をさらに含み、それにより機能基を有するナノ粒子を形成する、請求項1~8のいずれか1項に記載の方法。
- 前記機能基前駆体が、色素基、キレート剤基、標的化基、又は薬物基を含む、請求項9に記載の方法。
- 前記標的化基が、腫瘍細胞に特異的結合親和性を有する、請求項10に記載の方法。

- 前記薬物基が、化学療法剤を含む、請求項10に記載の方法。
- 前記機能基前駆体が、さらに、マレイミド基、N-ヒドロキシコハク酸イミド(NHS)エステル基、アジド基、アミン基、チオール基、又はアルキン基を含む、請求項10~13のいずれか1項に記載の方法。
- 前記水性媒体が、極性非プロトン性溶媒以外の有機溶媒を10%以上含まない、請求項1~14のいずれか1項に記載の方法。
- 前記シリカナノ粒子が、その中に被包された1つ以上の蛍光色素分子含む、請求項1~15のいずれか1項に記載の方法。
- 前記蛍光色素分子が、Cy5又はCy5.5である、請求項16に記載の方法。
