2.加水分解された基を1個だけ有することは、縮合工程中に形成される線形ポリマーゲルの形成に好都合である。線形ポリマー構造は遅い放出プロフィールに好都合である。縮合工程は、線形又は高分岐ポリマーゲル構造の形成を可能にする。単一の加水分解から出発すると、縮合反応中に二つの選択肢が提供される(以下参照)
ii.戦略
1.1対1の比率の水対Siと過剰のメタノールを使用
2.低pH
3.適切な確立されたブリンカー方法論(pH、温度)を用いて複数のシランを別々に加水分解した後、それらが単一メトキシ加水分解を受けていた場合、一緒にする(公表されたプロトコル)
a.得られるヒドロゲルの構造は、アルコール縮合対水縮合の相対量に依存する。
b.アルコール縮合に有利な条件(加水分解シラン上の遊離OHが別のシランのメトキシ基と反応して、メタノールが分離する)は、線形ポリマーと、ヒドロゲル中のポリマーの緊密充填をもたらし、より遅い放出プロフィールを有する粒子が得られる。
a.得られたヒドロゲルモノリスを溶解送達物を含有する溶液中に浸す。送達物の性質によってどの溶媒が最も適切か決まる。溶媒は、凍結乾燥工程中に除去されるように十分揮発性であることが必要である。ヒドロゲルに送達物を保持させたら、材料は乾燥工程の準備が整う。

併用療法
[00126]一定の態様において、本願の修飾ナノ粒子は、少なくとも一つの他の治療薬との併用療法に使用することができる。修飾ナノ粒子と治療薬は、相加的に、又はさらに好ましくは相乗的に作用できる。好適な態様において、修飾ナノ粒子を含む組成物は別の治療薬の投与と同時に投与される。別の治療薬は、修飾ナノ粒子と同じ組成物の一部であっても、又は異なる組成物であってもよい。別の態様において、修飾ナノ粒子を含む組成物は、別の治療薬の投与の前又は後に投与される。修飾ナノ粒子が治療に有用な障害の多くは慢性障害なので、一態様において、併用療法は、例えば特定の薬物に随伴する毒性を最小限に抑えるために、修飾ナノ粒子を含む組成物と別の治療薬を含む組成物の投与を交互に行うことを含む。各薬物又は治療薬の投与期間は、例えば、1ヶ月、3ヶ月、6ヶ月、又は1年でありうる。一定の態様において、修飾ナノ粒子が、毒性だけに限定されないが有害副作用を生じる可能性のある別の治療薬と同時に投与される場合、その治療薬は、有害副作用が引き起こされる閾値未満の用量で投与されるのが有益でありうる。

[00135]本発明は以前の技術に優る利点を提供する。他の技術及びこの究極薬物送達プラットフォーム(UDDP)がいかに異なるかを以下に記載する。
ナノ粒子は、反応物に対して感受性の高い溶液/溶媒相法を通じて製造される。これに対し、UDDPの場合、単純で堅牢なブリンカータイプの化学が使用され、ストーバー法で必要な複雑な製造及び化学物質(例えばアンモニア)の要件もない。物理的性質は、UDDPの場合、ブリンカー法から得られた空のモノリスゾルゲルをpH及び温度変化などのよく確立された単純な工程を通じて操作することにより、容易に調整される。その柔軟性はストーバー法を用いてどのように達成できるか(あるとすれば)明らかでない。
ストーバー法は、親水性及び親油性分子の両方を含むあらゆる中~小サイズの分子の保持が容易なUDDPとは対照的に、送達物の保持能力が非常に限られている。ストーバー法は、粒子形成工程中に発生する物理化学的な力を通じて送達物を保持することを必要とする。ストーバー法の複雑さを考えると、UDDP由来粒子に容易に保持される送達物の大部分を保持するための条件を作り出せるのかさえ不明である。各送達物は、ストーバー法由来の粒子の場合、独自の合成条件のセットも必要とするようである。UDDP法では、ゲル形成のための加水分解と縮合段階の完了後に送達物のゾル-ゲル中への保持を利用するので、ゾル-ゲル化学による送達物の妨害はない。すべてのその後の工程は非化学的である。従って、UDDP法は、ゲル製造段階を保持段階から分離することを可能にするので、送達物とは無関係に、得られる粒子の内部及び外部の特徴の物理的性質の操作に関して、莫大な柔軟性を提供する。
シリカベースのナノ粒子は、粒子の受動的な保持によってある種の送達物を保持できるが、保持量、保持できるもの及び放出速度の制御に関して著しい制限がある。
UDDP粒子とハイブリッドヒドロゲル粒子の組成はいくらか重なるところもあるが、二つの方法の間には明白な相違がある。主要な相違は、以前のハイブリッドヒドロゲルプラットフォームの場合、送達物はヒドロゲル製造の初期段階、すなわち加水分解後だが縮合前に保持された。その結果、ゲル形成のための条件は添加された送達物に適合するように操作されねばならず、このことがゾル-ゲル製造段階に大きな制限を課していた。多くの送達物は、添加されると、ゲル化の化学を損ね、保持されたゾル-ゲルモノリスの形成を阻止した。ゲル化の条件は、送達物の損傷を回避し、又は可能性ある望まざる反応を回避するために十分穏やかで迅速でなければならなかった。ゲル形成と適合する分子のための送達物の量は、ゲル化を損なうことなく添加できる量に制限された。従って、UDDP粒子の場合、ハイブリッドヒドロゲルプラットフォームの場合のゲル化の成功に必要な溶媒に比べて、クルクミンに対してより高い溶解度を有する溶媒を使用することにより、得られた粒子1mgあたり少なくとも6倍多いクルクミン保持が達成できた。この制限は、油、脂質などをハイブリッドヒドロゲル粒子に保持させようとした時も非常に明らかであった。これらの送達物は、ゲル化の条件を作り上げるために非常に低濃度である必要があった。これに対し、UDDPの場合、完全形成されたゾル-ゲルモノリスは、はるかに多量のこれらの送達物を保持することができた。
UDDP由来粒子からの放出プロフィールを、
-小PEG鎖の包含
-側鎖(例えば、アミン、チオール、様々なサイズのアルキル基)の共有結合的導入
-ヒドロゲル内のポリマーネットワークの修飾(細孔サイズは、方法の2つの工程で水分含量とpHを厳密に制御することによって制御される)
を通じて調節できることも明らかでなかった。
凍結乾燥:
クルクミンを含むゲルは水/メタノール含量が低いので迅速に凍結乾燥されるが(数時間のオーダー)、SNOゲルは凍結乾燥に1~2日かかる。
遊星ボールミル“Fritsch Pulverisette 6”;最大速度=650rpm
--12mLの窒化ケイ素ボウル又は80mLの酸化ジルコニウムボウル
--粉砕ボールの範囲 直径=0.5mm~10mm
--乾燥粉砕により平均直径=8umの粉末が得られる
--高速湿式粉砕(水、プロピレングリコール、又はアルコールなどの中で)により、粉砕ボール、粉砕速度、及び粉砕時間に応じて、直径=150nmほどの小ささのナノ粒子懸濁液が得られる
少量サンプルの場合、めのう乳鉢及び乳棒を用いて、相当程度の粒径縮小(微粉化)が迅速に達成できる(湿式又は乾式)。

--5mg粉末を5mL緩衝液(pH7.4)中に分散
--容器には高純度窒素が通気されており(200mL/分)、放出されたNOはこれによってオゾン-化学発光ベースの検出器に運ばれる。
[00141]図1に様々な送達物の放出速度を示す。この結果は、形成されたゲル(Br1プロトコルを用いて)に様々な親油性物質を保持させることができることを示している。蛍光標識されたコレステロールとパルミチン酸を用いて保持と放出を示した。プロシアニジンは、骨関節炎の治療における使用について評価中の有力な抗酸化剤である。
[00142]ナノ粒子からの放出は、下記により行われた。
・Br(ブリンカー)法、クルクミンは空のBrゾル-ゲルブロック(低速ゲル化プロトコル)に保持されている、
・オリジナルのヒドロゲルプラットフォーム、クルクミンは初期の縮合前混合物に混合されている、これはBrゾル-ゲルよりはるかに速くゲル化する、
・クルクミンはオリジナルの高速ゲル化プラットフォームに保持されているが、得られたクルクミン含有ゾル-ゲルは9日間風乾されている(密度の増強-細孔径の減少を促進するため)。
[00144]オリジナルの高速ゲル化プラットフォーム中へのNACSNOの保持(NACSNOは縮合前に添加される)は、水性緩衝液に添加するとNACSNOのほぼ即時放出を示す粒子をもたらす。同様にして製造されたGSNOは不安定で、これも迅速放出を示した。Br低速ゲル化ゾル-ゲルブロックの製造後に送達物を保持すると、オリジナルプラットフォームの場合よりもはるかに遅い放出を示す粒子が得られる。濃度の増加(4倍)は放出プロフィールの増大をもたらした。NAC及びGSHをまずBrゾル-ゲルに保持させた後、低pH緩衝液中で亜硝酸ナトリウムに暴露することにより、NACSNO及びGSNOを残留ナイトライトなしに形成させる。
[00148]初期ゾル-ゲルブロックを、加水分解されたTMOSと加水分解されたX-トリメトキシシラン(XはSiに結合されたチオール含有アルキル基)を混合することによって導入されるチオールでドープする。混合物は縮合反応を受け、共有結合されたチオールを有するポリマーで製造されたゾル-ゲルを生成する。

・反応性フルオロフォア/造影剤、放射性標識(アミン又はチオールに結合する)の、粒子内(ドープされたゲルに凍結乾燥前に試薬を添加)又は粒子上(粒子の製造後に添加)での共有結合;
・マルチカラーを有する粒子が製造可能:ある色は粒子自体用、そしてある色は粒子の表面に結合したPEG鎖用
が可能になる。
[00155]空の/あらかじめ保持させたゾル-ゲルモノリスを小PEG鎖でドープすることによる放出プロフィールへの影響(NACSNO、クルクミン、NO)。
・1つのゲルはPEGの添加なしであるが、他の3個は様々な量のPEG400が添加されている。図面に示されている添加PEGのパーセンテージは、我々のオリジナルのプロトコルで添加されたものと比較した添加PEGのパーセンテージである。
SNO-np周囲にPEGハローを示す蛍光イメージング
[00158]SNOナノ粒子は、内部マトリックス内に共有結合された蛍光プローブを用いて製造された。蛍光標識されたPEGは、ナノ粒子に結合しているチオールに結合させるためにマレイミド誘導体化蛍光PEGを用いて表面に結合された。二つの蛍光プローブは異なる波長を発する。結果は、顕微鏡でナノ粒子自体からの発光だけをモニターする場合(個別又はclumbed輝点として見られる)、その画像は、同じ画像にPEG(PEG化ナノ粒子の周囲にハローを形成するPEGと一致する)からの信号が含まれる場合よりもはるかに小さいことを示している。
[00159]ゲルモノリス/ブロックは、Br1(ゲル形成のためのブリンカー法)と呼ばれるTMOS(テトラメトキシシラン)のみを用いた純処方か、又はアルキル側鎖(オクチル)のチオール含有基(MPTS)である第4部位を有するトリメトキシシランでドープされたTMOSを用いて形成される。ゲルを完全に形成させてから試験分子を保持させる。誘導体化PEG鎖(チオールに迅速に結合するマレイミドで誘導体化されている)を、MPTS(チオールを含有する)でドープされた完全形成ナノ粒子に結合させる。
[00160]結果は、SNO-np、クルクミン保持及びNACSNO保持ナノ粒子に関する以前の研究と同様で、表面PEG化が放出を遅くすることを示している。
[00161]オクチル基の存在は、水溶性NACSNOの放出も脂溶性クルクミンの放出も遅くする。
[00162]3%オクチルでドープされたゲルはコレステロール放出を変化させたが(80%の放出から55%の放出へ減少)、パルミテートの放出には何の効果もなかった。
[00163]空のゾル-ゲルモノリス/ブロックは、Br1(ゲル形成のためのブリンカー法)と呼ばれるTMOS(テトラメトキシシラン)のみを用いた純処方か、又はトリメトキシシラン(第4部位はアルキル側鎖のチオール含有基(例えばMPTS);アルキル側鎖のアミン含有基(例えばAPTS);様々な長さのアルキル側鎖のいずれか)でドープされたTMOSを用いて形成される。
[00165]ゲルを、様々なサイズ/形状のアルキル側鎖を有するトリメトキシシランで、及びチオール含有トリメトキシシラン(MPTS):チオール-MPTS、イソブチル-IBTS、ビニル-VTS、オクチル-OTS オクタデシル-ODTS、ドープされていない基本処方のブリンカー1でドープすることによるクルクミン及びS-ニトロソN-アセチルシステイン(NACSNO)のナノ粒子放出プロフィールの調整。
[00166]pHは、ゲル化の速度及び細孔構造に影響を及ぼすことが知られている。高pHはゲル化時間を加速するが、大きい細孔を有するゾル-ゲルを形成する。低pHはゲル化を遅くし、小さな細孔を有する緻密なポリマー構造に好都合である。初期混合物に塩基を添加すると、速いゲル化と、ゲル化後に保持された送達物の速い放出プロフィールをもたらす。添加された塩基又は酸はゾル-ゲルモノリスが形成されたら洗い流されるので、pHが特定の送達物を劣化させるかもしれないという懸念は排除される。
[00168]アミンは、加水分解されたAPTSを加水分解されたTMOSと混合することによって組み込まれる。得られるゾル-ゲルモノリスに、負に帯電した4個の基を有する水溶性色素エバンスブルーを保持させる。アミンドーピングがなければ、エバンスブルーの放出は粒子を水に添加するとほぼ即座である。アミンの存在は実質的に放出を遅くする。

薬物を保持しているナノ粒子及び/又はマイクロ粒子の製造法であって、
(a)シラン、テトラメトキシシラン(TMOS)、又は加水分解可能シランを、メタ
ノールとpH≦3を有する水を用いて加水分解して混合物を形成させる工程;
(b)5~8のpHを有する水、任意にポリエチレングリコール(PEG)、及び任意
に薬物を、工程(a)の混合物と合わせ、PEG及び/又は薬物を含んでもよい固体のヒ
ドロゲルモノリスを形成させる工程;
(c)得られた固体ヒドロゲルモノリスを取り出す工程;
(d)任意に、前記モノリスを薬物とインキュベートしてヒドロゲル薬物組成物を形成
させる工程、ここで工程(d)の薬物は工程(b)の薬物と同じでも又は異なっていても
よく;
(e)工程(d)の組成物を凍結乾燥して乾燥材料を形成させる工程;
(f)工程(e)の乾燥材料をボールミル粉砕又はジェットミル粉砕して粉末を形成さ
せる;又は湿式粉砕して粒子のスラリーを形成させる工程;及び
(g)任意に、湿式粉砕後に粒子の表面にポリエチレングリコール(PEG)、アニオ
ン、カチオン、又はアルカンの内の一つ又は複数を適用する工程
を含み、それによって一つ又は複数の薬物を保持しているナノ粒子及び/又はマイクロ粒
子を製造する方法。

工程(a)で添加される水が約1.4のpHを有する、請求項1に記載の方法。

加水分解可能シランが置換トリメトキシシランを含み、前記トリメトキシシランは、ア
ルキル鎖か、チオール、アミン、カルボキシル、カルボニル、PEG、ペプチド、糖、又
は多糖を有するアルキル鎖か、又はそれらの組合せの一つ又は複数で置換されている、請
求項1~6のいずれか1項に記載の方法。
[請求項8]
ペプチド、抗体、造影剤又はそれらの組合せを含むターゲティング分子がPEGに結合
されている、請求項1~7のいずれか1項に記載の方法。
[請求項9]
請求項1~8のいずれかの方法によって製造された、一つ又は複数の薬物を保持してい
るナノ粒子及び/又はマイクロ粒子。
[請求項10]
疾患又は障害を有する対象の治療法であって、前記対象に治療上有効量の請求項9のナ
ノ粒子及び/又はマイクロ粒子を投与することを含む方法。
Claims (9)
- 薬物を保持しているナノ粒子及び/又はマイクロ粒子の製造法であって、
(a)テトラメトキシシラン(TMOS)、又は加水分解可能シランを、メタ
ノールとpH≦3を有する水を用いて加水分解して混合物を形成させる工程;
(b)5~8のpHを有する水、任意にポリエチレングリコール(PEG)、及び任意に薬物を、工程(a)の混合物と合わせ、PEG及び/又は薬物を含んでもよい固体のヒドロゲルモノリスを形成させる工程;
(c)得られた固体ヒドロゲルモノリスを取り出す工程;
(d)前記モノリスを薬物とインキュベートしてヒドロゲル薬物組成物を形成させる工程、ここで工程(d)の薬物は工程(b)の薬物と同じでも又は異なっていてもよく;
(e)工程(d)の組成物を凍結乾燥して乾燥材料を形成させる工程;
(f)工程(e)の乾燥材料をボールミル粉砕又はジェットミル粉砕して粉末を形成させる;又は湿式粉砕して粒子のスラリーを形成させる工程;及び
(g)任意に、湿式粉砕後に粒子の表面にポリエチレングリコール(PEG)、アニオン、カチオン、又はアルカンの内の一つ又は複数を適用する工程
を含み、それによって一つ又は複数の薬物を保持しているナノ粒子及び/又はマイクロ粒子を製造する方法。 - 工程(a)で添加される水が約1.4のpHを有する、請求項1に記載の方法。
- 工程(a)において、メタノールが25~75%の濃度で存在する、請求項1又は2に記載の方法。
- 工程(a)において、メタノールが約45%の濃度で存在する、請求項1~3のいずれか1項に記載の方法。
- 工程(a)が、約60℃の温度で約1.5時間実施される、請求項1~4のいずれか1
項に記載の方法。 - PEGがPEG200ダルトン~PEG10Kダルトンである、請求項1~5のいずれか1項に記載の方法。
- 加水分解可能シランが置換トリメトキシシランを含み、前記トリメトキシシランは、アルキル鎖か、チオール、アミン、カルボキシル、カルボニル、PEG、ペプチド、糖、又は多糖を有するアルキル鎖か、又はそれらの組合せの一つ又は複数で置換されている、請求項1~6のいずれか1項に記載の方法。
- ペプチド、抗体、造影剤又はそれらの組合せを含むターゲティング分子がPEGに結合されている、請求項1~7のいずれか1項に記載の方法。
- 薬物を保持しているナノ粒子及び/又はマイクロ粒子の製造法であって、
(a)テトラメトキシシラン(TMOS)、又は加水分解可能シランを、メタ
ノールとpH≦3を有する水を用いて加水分解して混合物を形成させる工程;
(b)5~8のpHを有する水、薬物、及び任意にポリエチレングリコール(PEG)を、工程(a)の混合物と合わせ、PEGを含んでもよく、そして薬物を含む固体のヒドロゲルモノリスを形成させる工程;
(c)得られた固体ヒドロゲルモノリスを取り出す工程;
(d)前記固体ヒドロゲルモノリスを凍結乾燥して乾燥材料を形成させる工程;
(e)工程(d)の乾燥材料をボールミル粉砕又はジェットミル粉砕して粉末を形成させる;又は湿式粉砕して粒子のスラリーを形成させる工程;及び
(f)任意に、湿式粉砕後に粒子の表面にポリエチレングリコール(PEG)、アニオン、カチオン、又はアルカンの内の一つ又は複数を適用する工程
を含み、それによって一つ又は複数の薬物を保持しているナノ粒子及び/又はマイクロ粒子を製造する方法。
