上述のように、本明細書に記載の方法及び装置はいずれも、個別マイクロ粒子(例えば、単一細胞又は細胞のグループ)の選別、又は選別と定量供給に使用されてよい。必要とされているのは、マイクロ粒子(例えば、細胞)を高速で(理想的には、毎秒100,000個を超える速度で)選別することが可能であって、更に、非常に希少なマイクロ粒子(例えば、がん患者の血液中の循環腫瘍細胞、或いは妊婦の血液中の循環胎児細胞)を隔離することも可能な、シンプルな装置である。このようなケースでは、希少細胞の頻度は、100万個に1個から10億個に1個である場合がある。これらの極めて希少な細胞を直接隔離することは、典型的には選別速度が低すぎる現行の細胞選別装置では達成できない。現在利用可能な選別方法では、細胞1103が選別装置の選別領域内を、照明及び検出1105の為にマイクロ流体チャネルの流動方向1107に、一列縦隊で1つずつ動くことが必要である(例えば、図11を参照)。これらの装置及び方法では、典型的には、各個別細胞が1つずつ、レーザでインタロゲートされて選別されるように、一列縦隊配置の各細胞の間隔が十分あることが必要である。このように「1つずつ」選別することで、細胞を選別する速度が大幅に制限される。本明細書に記載の細胞選別方法の幾つかの変形形態では、マイクロ流体チャネル内で細胞を1つずつではなくバルクで動かしてよい。細胞は、1つずつではなく、数百個又は数千個からなるグループとして連続的に集束レーザビームを通過することが可能であり、これによって、選別速度が劇的に高まる。例えば、図12を参照すると、レーザ照射源1205を含む検知領域を大規模な細胞グループ1202が通過する様子が概略的に示されており、この検知領域では、グループ内の細胞のうちの1つ以上が所定の特性を有するかどうかを判定する為に、1つ以上の検出器で細胞グループの同時インタロゲーションを行うことが可能でありうる。細胞をバルクで選別することにより、選別可能な細胞の数が劇的に増える。この方法は特に、極めて希少な細胞(例えば、循環腫瘍細胞、或いは循環胎児細胞)を隔離することに有用である。図12では、ほとんどの細胞(灰色)は、照射された際に蛍光信号を有しない、不要な細胞である。標的細胞の一例1209を、黒で着色して示す。標的細胞がインタロゲーション領域(レーザ1205)を通過すると、この細胞は強い蛍光信号を発生させうる。本明細書に記載の流動選別の方法及び装置を使用すると、標的細胞1209を単一液滴(例えば、約1μlの液滴)中に捕捉することが可能である。図13A及び13Bは、標的細胞の検出及び捕捉の様子を示す。簡単に言うと、図13Aでは、細胞のバルクが、マイクロ流体チャネルを流動方向1307に動いている。標的細胞が検出領域(レーザビーム1305)を通過すると、標的細胞は蛍光信号を発生させる。蛍光信号を発生させない(又は十分高い強度の信号を発生させない)不要な細胞(灰色の細胞)は、流動方向1307に通過し続ける。これに対し、検出領域1305内の細胞グループから蛍光信号が検出されると、選別装置は、分離(及び/又は定量供給)の為に試料出口流路1308を開いて、その細胞グループの進路を、試料出口流路1308に入るように変更する。後述されるように、任意の適切な弁(例えば、音響弁、電気/磁気弁等)が使用されてよいが、図13A及び13Bの例では流動弁が示されている。図13Bに示されるように、この例では、流動弁は注入流1311を(変形形態によっては短い遅延の後に)短時間開く。標的細胞が注入流エリア1313に入るときだけ弁が開くように、遅延が含まれ、計算されてよい。注入流1311は、標的細胞を、グループ内の隣接細胞とともに、ノズルから外に定量供給してよい。細胞選別が行われるが、完全な選別はまだ達成されていない。そして、更なる隔離ステップが実施されてよく、これは、ただちに(例えば、カートリッジの別の部分で、又は同じ検出領域に再度通して)、又は順次に、例えば別のカートリッジを使用して、実施されてよい。多少の不要な細胞(灰色の細胞)が標的細胞(黒色の細胞)に付いてくるとしても、多数の細胞を短時間で選別することが可能である。図13A及び13Bには、1つの層の細胞がマイクロ流体チャネルを通過する様子だけが示されている。実際には、複数の層の細胞が同時にマイクロ流体チャネルを通過することが可能であり、これによって、選別速度を更に高めることが可能である。例えば、マイクロ流体チャネルの寸法は、標的マイクロ粒子の直径の4倍超(幅)×標的マイクロ粒子の直径の2倍超(奥行)(例えば、約100μm×20μm、120μm×30μm、150μm×50μm、200μm×60μm等)であってよい。注入流路の直径は、例えば、150μmであってよい。未希釈血液は、典型的には、マイクロリットル当たり3×106個の赤血球を含む。未希釈血液試料がマイクロ流体チャネルを0.5μL毎秒の速度で通過すると、全体の選別速度は、3×106×0.5=1.5×106個毎秒になりうる。これは、現行の選別方法より10倍以上速い。マイクロ流体チャネルの断面が約150μm×50μmで長さが20mmの場合、流量が0.5μl毎秒に達するのに必要な圧力は約1psiでよい。注入流の直径がチャネルの幅(例えば、150μm)と同じであれば、全部で約3000個の不要な細胞が、必要な細胞と一緒に捕捉されることになる。このバルク選別方法では、純粋な細胞集団は得られないが、それでも、非常に希少な細胞の濃縮を非常に高速に行うことが可能である。得られたバルク選別試料を1つずつの選別により再度選別することによって、純粋な細胞の隔離を達成することが可能である。
本明細書に記載の方法は、バルク流体試料に含まれるマイクロ粒子を選別する方法であり、選別は、本明細書に記載のいずれかのカートリッジと選別装置(例えば、本明細書に記載のフローサイトメトリ装置)との組み合わせにより、マイクロ粒子の所定の特性に基づいて行われてよい。所定の特性は、サイズ、形状、固有モルフォロジ、蛍光強度、元々の光学品質又は実験室で後から導入された光学品質、その他であってよい。本明細書に記載のカートリッジ100を使用するマイクロ粒子選別方法のエレガントさ及びシンプルさは、使い捨て選別カートリッジ100の長さ方向のチャネル及び入口の固有の配置に基づいている。

マイクロ粒子をバルク選別する方法であって、
流体で囲まれたマイクロ粒子のグループを第1のスイッチの検出領域内まで通すステップであって、前記マイクロ粒子グループ内の複数のマイクロ粒子が、前記検出領域内で、前記検出領域を通る前記マイクロ粒子グループの流れの方向に垂直に隣接して配置されている、前記通すステップと、
前記検出領域内で前記複数のマイクロ粒子を同時に検査して、前記マイクロ粒子グループ中の少なくとも1つのマイクロ粒子が所定の特性を有する場合にこれを検出するステップと、
前記マイクロ粒子グループ中の少なくとも1つのマイクロ粒子が前記所定の特性を有する場合には、前記マイクロ粒子グループを試料出口流路内まで通し、そうでない場合には、前記マイクロ粒子グループを前記第1のスイッチを介して廃棄物出口流路内まで通すステップと、
前記所定の特性を有する前記少なくとも1つのマイクロ粒子を前記マイクロ粒子グループから隔離するステップであって、前記試料出口から定量供給される前記マイクロ粒子グループを、一列縦隊配列で第2のスイッチに通し、前記第2のスイッチの検出領域に通すステップと、前記所定の特性を有する前記少なくとも1つのマイクロ粒子が前記第2のスイッチの検出領域内で検出された場合には、前記少なくとも1つのマイクロ粒子を前記第2のスイッチの第2の試料出口流路内まで通すステップと、前記マイクロ粒子グループ中の、前記所定の特性を有しないマイクロ粒子を前記第2のスイッチの廃棄物出口流路内まで通すステップと、によって実施される前記隔離するステップと、
を含む方法。

Claims (12)
- マイクロ粒子をバルク選別する方法であって、
流体で囲まれたマイクロ粒子のグループを第1のスイッチの検出領域内まで通すステップであって、前記マイクロ粒子グループ内の複数のマイクロ粒子が、前記検出領域内において、前記検出領域を通る前記マイクロ粒子グループの流れの方向に垂直であり、且つ前記流れ方向を横切る平面内で、横方向に隣接して配置されており、前記検出領域は、前記複数のマイクロ粒子が当該検出領域内を流れる間、前記複数のマイクロ粒子の配置が維持されるように構成されている、前記通すステップと、
前記検出領域内で前記複数のマイクロ粒子を同時に検査して、前記マイクロ粒子グループ中の少なくとも1つのマイクロ粒子が所定の特性を有する場合にこれを検出するステップと、
前記マイクロ粒子グループ中の少なくとも1つのマイクロ粒子が前記所定の特性を有する場合には、前記マイクロ粒子グループを試料出口流路内まで通し、そうでない場合には、前記マイクロ粒子グループを前記第1のスイッチを介して廃棄物出口流路内まで通すステップと、
前記所定の特性を有する前記少なくとも1つのマイクロ粒子を前記マイクロ粒子グループから隔離するステップであって、前記試料出口から定量供給される前記マイクロ粒子グループを、一列縦隊配列で第2のスイッチに通し、前記第2のスイッチの検出領域に通すステップと、前記所定の特性を有する前記少なくとも1つのマイクロ粒子が前記第2のスイッチの検出領域内で検出された場合には、前記少なくとも1つのマイクロ粒子を前記第2のスイッチの第2の試料出口流路内まで通すステップと、前記マイクロ粒子グループ中の、前記所定の特性を有しないマイクロ粒子を前記第2のスイッチの廃棄物出口流路内まで通すステップと、によって実施される前記隔離するステップと、
を含む方法。 - 前記第1のスイッチ及び前記第2のスイッチは、いずれも、前記マイクロ粒子の流量及び/又は前記マイクロ粒子を囲む流体の流量に基づいて、前記マイクロ粒子を選別する流動スイッチである、請求項1に記載の方法。
- 同時に検査する前記ステップは、前記マイクロ粒子グループ中の少なくとも1つのマイクロ粒子が前記所定の特性を有していて、前記スイッチ内に存在する場合に、前記マイクロ粒子グループを囲む前記流体の流量を第1の流量から第2の流量に変化させるステップを含む、請求項1に記載の方法。
- 前記マイクロ粒子グループを囲む前記流体の前記流量を変化させる前記ステップは、前記マイクロ粒子グループを囲む流体を追加するか減らすステップを含む、請求項3に記載の方法。
- 同時に検査する前記ステップは、前記マイクロ粒子グループ中の少なくとも1つのマイクロ粒子が前記所定の特性を有していて、前記スイッチ内に存在する場合に、前記マイクロ粒子グループを囲む前記流体の流量を第1の流量から第2の流量に変化させるステップを含み、前記マイクロ粒子グループを前記試料出口流路内まで通す前記ステップは、前記マイクロ粒子グループを囲む前記流体が前記スイッチ内をほぼ前記第2の流量で移動している場合には、前記マイクロ粒子グループを前記試料出口流路内まで通して、前記マイクロ粒子グループを前記試料出口流路から外に定量供給し、そうでなく、前記マイクロ粒子グループを囲む前記流体が前記スイッチ内をほぼ前記第1の流量で移動している場合には、前記マイクロ粒子グループを前記スイッチを介して前記廃棄物出口流路内まで通すステップを含む、請求項1に記載の方法。
- 前記少なくとも1つのマイクロ粒子を隔離する前記ステップは、前記試料出口から定量供給される前記マイクロ粒子グループを、前記第2のスイッチに通すステップを含む、請求項1に記載の方法。
- 前記マイクロ粒子グループを前記検出領域内まで通す前記ステップは、前記マイクロ粒子グループを、前記スイッチを収容するカートリッジに通すステップを含む、請求項1に記載の方法。
- 前記廃棄物出口流路に沿う流体流に対する抵抗は、前記試料出口流路に沿う流体流に対する抵抗より大きい、請求項1に記載の方法。
- 前記マイクロ粒子グループは細胞のグループである、請求項1に記載の方法。
- 前記検出される所定の特性は、形状、サイズ、及び蛍光強度のうちの1つ以上から選択される、請求項1に記載の方法。
